Sandalwood ( Santalum album L.) is a hemi-parasitic tree species famous for its santalol and santalene, which are extracted from its heartwood and roots. The ability to understand root functionality within its branched root system would benefit the regulation of sandalwood growth and enhance the commercial value of sandalwood. Phenotypic and anatomical evidence in this study indicated that seed germination stage 4 (SG4) seemed pivotal for lateral root (LR) morphogenesis. Small RNA (sRNA) high-throughput sequencing of root tissues at three sub-stages of SG4 (lateral root primordia initiation (LRPI), lateral root primordia development (LRPD), and lateral root primordia emergence (LRPE)) was performed to identify microRNAs (miRNAs) associated with LR development. A total of 135 miRNAs, including 70 differentially expressed miRNAs (DEMs), were screened. Ten DEMs were selected to investigate transcript abundance in different organs or developmental stages. Among 100 negative DEM-mRNA interaction pairs, four targets (Sa-miR166m_2, 408d, 858a, and novel_Sa-miR8) were selected for studying cleavage sites by 5′ RLM-RACE validation. The expression mode of the four miRNA-mRNA pairs was investigated after indole-3-acetic acid (IAA) treatment. IAA enhanced the abundance of homeobox-leucine-zipper protein 32 ( HOX32 ), laccase 12 ( LAC12 ), myeloblastosis86 ( MYB86 ), and pectin methylesterase inhibitor6 ( PMEI6 ) target transcripts by reducing the expression of Sa-miR166m_2, 408d, 858a, and novel_Sa-miR8 in the first 10 min. A schematic model of miRNA-regulated LR development is proposed for this hemi-parasitic species. This novel genetic information for improving sandalwood root growth and development may allow for the cultivation of fast-growing and high-yielding plantations.

In this work, a novel label-free and signal-off electrochemical biosensor based on efficient target-trigger T-structure recycling with dual strand displacement amplification (T-DSDA) was constructed for sensitive detection of miRNA-155 that related to cardiovascular diseases. Compared with traditional SDA, the proposed strategy not only accomplished simple polymerization and shearing of dsDNA without continuous consumption of temple DNA , but also ingeniously designed two analogous sequences that produced double cleavage sites, thereby obtaining extensive output DNA for further cleaving Y-structure assembled on electrode with significantly improved target conversion efficiency. Since the redox probe methylene blue (MB) attach to Y-structure via electrostatic interaction and tessellation, an obviously decreased current with a typical signal-off mode could be acquired for sensing miRNA-155 in the assistant of DNAzyme cleavage recycling. As a result, the electrochemical biosensor realized the rapid and sensitive detection of miRNA-155 with a low detection limit down to 166.7 fM. In addition, the biosensor displayed a strong anti-interference ability in bovine whole blood, and thus was expected to be applied to detect blood samples of various miRNA-155 related diseases in clinical diagnosis.

The tumor immunosuppressive microenvironment (TIM) greatly hindered the efficacy of cancer immunotherapy. Overexpressed indoleamine 2,3-dioxygenase-1 (IDO1) in tumor tissues plays a vital role in TIM generation, and downregulation of IDO1 expression may reverse TIM. Inspired by the Watson–Crick base-pairing rule, a versatile noncationic miRNA vector (miDAC@PDA) is developed for cancer immunotherapy. Doxorubicin (DOX), adenosine triphosphate (ATP), and copper ions (Cu2+) are coassembled into coordination polymer nanoparticles (DAC) and bind miRNA via the hydrogen bond interaction (miDAC) between adenine residues (ATP) and uracil residues (miRNA). Polydopamine (PDA) is deposited onto the surface of miDAC for photothermal therapy. miDAC@PDA can efficiently accumulate into tumor tissues for cellular uptake. Under laser irradiation and high intracellular GSH levels, the PDA shell of miDAC@PDA can dissociate from miDAC for miRNA release due to local hyperthermia. Cu2+-mediated GSH consumption and intracellular ATP release can amplify the DOX-based immunogenic cell death (ICD) cascade, together with miR-448-mediated IDO1 inhibition, and these versatile nanoplexes will not only restrain primary tumor growth but also display a remarkable abscopal effect on distant tumors. Collectively, our study provides a unique strategy for intracellular gene delivery and an inspirational approach for multimechanism cancer management.

Idiopathic pulmonary fibrosis (IPF) is a prototype of chronic, progressive, and fibrotic lung disease with high morbidity and high mortality. Menstrual blood-derived stem cells (MenSCs) have proven to be an attractive tool for the treatment of acute lung injury and fibrosis-related diseases through immunosuppression and antifibrosis. However, whether MenSC-derived exosomes have the similar function on pulmonary fibrosis remains unclear. In the present study, exosomes secreted from MenSCs (MenSCs-Exo) were verified by transmission electron microscope (TEM), nanoparticle tracking analyzer (NTA), and western blotting. And MenSC-Exo addition significantly improved BLM-induced lung fibrosis and alveolar epithelial cell damage in mice, mainly reflected in BLM-mediated enhancement of the fibrosis score, blue collagen deposition, dry/wet gravity ratio, hydroxyproline and malondialdehyde levels, and downregulation of glutathione peroxidase, which were all robustly reversed by MenSC-Exo management. Additionally, BLM- and TGF-β1-evoked cellular reactive oxygen species (ROS), mitochondrial DNA (mtDNA) damage, and cell apoptosis were rescued by MenSCs-Exo in vivo and in vitro. Further study indicated that the MenSCs-Exo could transport miRNA Let-7 into recipient alveolar epithelial cells. Let-7 inhibitor administration significantly blocked the exosome-mediated improvement role on lung fibrosis in mice. Mechanistically, Let-7 was able to regulate the expression of lectin-like oxidized low-density lipoprotein receptor-1 (LOX1) through binding to its 3-UTR region. Forced expression of LOX1 promoted the expression of apoptosis-related protein and mtDNA damage markers via regulating NLRP3 which was also confirmed in BLM model mice under the combination therapy of the exosome and Let-7 inhibitor. Collectively, this study demonstrates that exosomal Let-7 from MenSCs remits pulmonary fibrosis through regulating ROS, mtDNA damage, and NLRP3 inflammasome activation. This provides a new approach of exocytosis on the treatment of fibrotic lung disease.

The detection of low-abundance microribonucleic acid (miRNA) frequently adopted nucleic acid sequence-based amplification detection, which was found to have poor selectivity for the nonspecific amplification of template-dependent ligation in enzyme-mediated cascade reactions. Here, a highly selective detection of miRNAs was developed that combined microsphere-enhanced fluorescence (MSEF) and solid-phase base-paired hybridization. The target miRNA could be accurately and quantitatively identified through the solid-phase hybridization assay on the surface of an optical microsphere, while the detected fluorescence signal could be physically amplified by MSEF. Hereinto, the optical microsphere acted as the fluorescence amplifier and whose surface supplied the space to carry out base-paired hybridization to recognize the target miRNA via the immobilized capture DNA sequence. The detected fluorescence signal of the single-base mismatched miRNA-21 sequence was just around 12% of that of the target miRNA-21 sequence in the measurement of model miRNA-21, while the limit of detection of miRNA-21 could be 1.0 fM. The developed detection of miRNA on an optical microsphere was demonstrated to be an excellent physically amplified method to selectively and sensitively detect the target miRNA and magnificently avoid the nonspecific amplification and false-positive results, which is expected to have wide applications in pathematology, pharmacology, clinic diagnosis, and on-site screening fields as well.

Tumor markers (TMs) are crucial indicators of preneoplastic and neoplastic diseases, which provide the basis for the accurate diagnosis of malignant tumors. We present a diagnostic platform utilizing magnetic photonic microspheres (mPCMs) for simultaneous detection of various sorts of TMs, specifically for reliable breast cancer identification. The multiplex detection capability of mPCMs stems from the color encoding function of the photonic crystal structure formed by self-assembled silica nanoparticles, while their magnetic responsiveness is achieved through ferric oxide nanoparticle doping. The ordered nanostructure offers a large specific surface area for biological detection, and the magnetically controlled movement enhances the capture efficiency of target biomarkers. Specifically, CA 15–3, miRNA-21, and exosomes are used as target biomarkers, reflecting different aspects of breast cancer biology. Our multivariate detection platform effectively identifies proteins, nucleic acids, and exosomes through specific binding and base complementary pairing. Thus, mPCMs provide a novel approach for the combined analysis of diverse biomarkers.

A visualized assay for miRNAs detection has been developed in this work. The presented method is based on a combination of enzyme-free amplification cascades of catalyzed hairpin assembly (CHA) and hybridization chain reaction (HCR) and fluorescence quenching of dual-emission ratiometric fluorescent probes (RF probes). MiRNAs can efficiently initiate enzyme-free amplification reactions (CHA and HCR) and produce the long nicked dsDNAs with a lot of glucose oxidases (GOD) on the surface of dynabeads bridged by the GOD-labeled hairpin DNA probes . Hydrogen peroxide (H 2 O 2 ) is generated by oxidation of glucose catalyzed by GOD, which can quench the outer green fluorescence without affecting the internal red fluorescence of RF probes. Therefore, increased miRNA amount can result in change of the two fluorescence intensity ratios of RF probes with continuous color changes from green to red under a UV lamp, which can be easily recognized by naked eye. The proposed assay exhibits high sensitivity toward let-7a with dynamic range from 10 −13 M to 10 −8 M, and which is applied successfully to detecting let-7a in the small RNA samples.

Evidence indicates that microRNAs (miRNAs) play vital roles in regulating osteogenic differentiation and bone formation. Methods : Here, we show that a polyethyleneimine (PEI)-functionalized graphene oxide (GO) complex efficiently loaded with the miR-214 inhibitor is assembled into silk fibroin/hydroxyapatite (SF/HAP) scaffolds that spatially control the release of the miR-214 inhibitor. Results : SF/HAP/GO scaffolds with nanosized GO show high mechanical strength, and their hierarchical microporous structures promote cell adhesion and growth. The SF/HAP/GO-PEI scaffolds loaded with mir-214 inhibitor (SF/HAP/GPM) were tested for their ability to enhance osteogenic differentiation by inhibiting the expression of miR-214 while inversely increasing the expression of activating transcription factor 4 (ATF4) and activating the Akt and ERK1/2 signaling pathways in mouse osteoblastic cells (MC3T3-E1) in vitro . Similarly, the scaffolds activated the osteoblastic activity of endogenous osteoblast cells to repair critical-sized bone defects in rats without the need for loading osteoblast cells. Conclusion : This technology is used to increase osteogenic differentiation and mineralized bone formation in bone defects, which helps to achieve cell-free scaffold-based miRNA-inhibitor therapy for bone tissue engineering.

Children exposed to common aeroallergens may develop asthma that progresses into adulthood. Inflammation regulated by T helper 2 (Th2) cells, a specific subpopulation of CD4+ T lymphocytes, is involved in asthmatic injury. Herein, our microarray data indicated that microRNA-451a-5p (miRNA-451a) expression decreased by 4.6-fold and ETS proto-oncogene 1 (ETS1) increased by 2.2-fold in the peripheral blood lymphocytes isolated from asthmatic children (n = 4) as compared to control individuals (n = 4). The negative correlation between miRNA-451a and ETS1 was further validated in 40 CD4+ T cell samples (10 healthy vs. 30 asthmatic samples). In vitro, naïve CD4+ T cells isolated from control individuals were cultured under Th2 cell polarizing condition. miRNA-451a expression decreased while ETS1 increased in CD4+ T cells in the setting of Th2 cell polarization. Moreover, miRNA-451a knockdown enhanced Th2 cell polarization – cells positive for both GATA3 (GATA binding protein 3, a Th2-transcription factor) and CD4 increased, and the generation of Th2 cell cytokines, interleukin (IL)5 and IL13, increased. In contrast, miRNA-451a overexpression inhibited Th2 cell differentiation. Interestingly, dual-Luciferase assay proved ETS1 as a novel target of miRNA-451a. Moreover, enforced expression of ETS1 partially restored miRNA-451a-induced inhibition of IL5 and IL13, and increased the GATA3+CD4+ cell population. Collectively, our work demonstrates that downregulation of miRNA-451a upregulates ETS1 expression in CD4+ T cells, which may contribute to Th2 cell differentiation in pediatric asthma

MicroRNA-135a (miRNA-135a) is a crucial biomarker for the early diagnosis of breast cancer (BC), the leading malignancy among women. However, its sensitive and efficient detection remains challenging due to the time-consuming nature of conventional techniques and the extremely low abundance of miRNA-135a in biological samples. In this work, we developed an ultrasensitive electrochemical aptasensor based on nickel‑embedded nitrogen‑doped carbon nanotubes (Ni‑NCNTs) wrapped carbon fiber (CF) for the rapid detection of miRNA‑135a within only 15 min. The tendril‑like Ni‑NCNTs were grown in situ and uniformly around the CF surface via a Ni 2 + ‑induced controlled chemical vapor deposition process, serving as an effective nanoscale signal amplifier. This unique hierarchical architecture exhibits excellent electrocatalytic activity, rapid electron‑transfer capability, and abundant active sites for immobilizing capture DNA (cDNA), thereby enhancing the recognition and signal response to reverse‑transcribed DNA (rtDNA) corresponding to miRNA‑135a. Under optimized conditions, the aptasensor achieved an ultralow detection limit of 3.24 aM (3.24 ×10 −9 nM) with high reliability, excellent selectivity, and efficient performance in real‑sample analysis. The CF@Ni‑NCNTs based aptasensor demonstrates considerable potential to facilitate rapid, ultrasensitive early detection screening and warning of various cancers, offering a powerful analytical platform to support clinical diagnostics in diverse scenarios.

A low-background paper platform enables cascaded RCA-based ratiometric sensing. • Blue CQDs provide self-calibrated and reliable miRNA-21 quantification. • Smartphone-assisted portable POCT enables noninvasive urinary miRNA analysis. The non-invasive detection of microRNA-21 (miRNA-21), a promising biomarker for bladder cancer, is challenged by its low abundance in urine and the lack of portable, sensitive analytical methods. Herein, we report an integrated smartphone-based paper microfluidic platform that combines rolling circle amplification (RCA), DMSN-based fluorescence enhancement, carbon quantum dots (CQD) -assisted ratiometric fluorescence detection, and smartphone imaging into a portable analytical system for quantitative miRNA-21 detection. A ratiometric fluorescence strategy is employed using blue-emitting CQDs covalently immobilized on glass fiber filter paper as an internal reference, while the RCA product is detected by a DMSN-coated QD probe. Under optimal conditions, the method achieves a limit of detection (LOD) of 1.06 nM for miRNA-21 with a linear range of 0.25–200 nM (R 2 = 0.9891). The assay exhibits excellent specificity against mismatched sequences and other miRNAs, and spike-recovery tests in artificial urine yield recoveries of 88.5–120.3% with relative standard deviations below 15%. The platform shows good batch-to-batch reproducibility (RSD < 7%). Spike recovery tests using clinical urine samples from healthy individuals yielded recoveries of 86.97%–108.5%, confirming the method's high accuracy and reliability in complex urine matrices. Integrated with a 3D-printed portable device and a custom smartphone application, this paper-based sensing platform provides a practical, low-cost, and portable analytical platform for point-of-care testing (POCT) of miRNA-21. The graphical abstract was prepared using SolidWorks, Cinema 4D (C4D), and Microsoft PowerPoint. The development of highly sensitive and reliable biosensors is essential for the early diagnosis and therapeutic monitoring of cancer. Herein, we report a dual-mode fluorescence–colorimetric biosensing platform for the ultrasensitive detection of miRNA-21, integrating AuNP-supported dual-stage rolling circle amplification (RCA) with an RCA-generated multipedal binding-and-cleavage mechanism. The sensing hierarchy is initiated by target-dependent activation of the primary RCA on the gold nanoparticle (AuNP) surface, producing long DNA strands with multiple repeated recognition domains that function as numerous walker “legs” and form a DNA-rich nanogel matrix. Through these repeated domains, the RCA-generated multipedal walker repeatedly and multivalently binds fluorophore-labeled hairpin substrates on the AuNP surface, thereby facilitating APE1-mediated substrate cleavage and fluorescence recovery. The APE1-generated cleavage fragments subsequently initiate the secondary RCA following the addition of the T2 template. The two successive RCA reactions ultimately produce an Au@RCA DNA nanogel containing abundant G-quadruplex/hemin DNAzymes that catalyze a visually detectable color change. By integrating the multipedal walker-mediated multivalent binding-and-cleavage process with dual-stage RCA, the platform enables dual-mode fluorescence–colorimetric detection with efficient signal amplification and low background, achieving limits of detection of 9.854 and 790 fM for the fluorescence and colorimetric modes, respectively. Furthermore, the two complementary outputs provide independent signal readouts for cross-validation. The biosensor demonstrated excellent specificity and distinguished the endogenous responses of MCF-7, HeLa, and HEK293T cell lysates. These results support the potential of the platform for ultrasensitive tumor-associated biomarker detection, while the cell-lysate experiments represent a preliminary proof-of-concept evaluation in complex biological matrices.
