DNAzyme concatemer-catalyzed precipitation on an interdigitated micro-comb electrode for capacitance immunosensing of interleukin-6 with rolling circle amplification

Jianming Zhang,Jia Lin,Tingjin Zheng,Yancheng Jiang,Shimu Luo,Yao Lin,Zhishan Zhang

A novel capacitance immunosensor based on DNAzyme concatemer-amplified signal-generation tags was developed for the sensitive detection of interleukin-6 (IL-6) on an interdigitated micro-comb electrode. To construct such an immunosensor, monoclonal mouse anti-human IL-6 antibodies were covalently conjugated onto a bovine serum albumin (BSA)-coated electrode using the glutaraldehyde cross-linker. Gold nanoparticles heavily functionalized with primer DNA (for the subsequent rolling cycle amplification; RCA) and polyclonal rabbit anti-human IL-6 antibodies were used for the development of a sandwich-type immunoassay on the modified interdigitated electrode in the presence of the target IL-6. Thereafter, the attached primer DNA with the immunocomplex triggered the RCA reaction for the formation of numerous DNAzyme concatemers with the assistance of hemins and G-rich DNA probes. DNAzyme concatemers catalyzed 4-chloro-1-naphthol into insoluble benzo-4-chlorohexadienone and were deposited onto the electrode, thus resulting in the local capacitance variation of the interdigitated micro-comb electrode. The performance and external factors affecting the analytical properties were studied in detail. Under optimum conditions, the capacitance variations of the immunosensor were proportional to the IL-6 concentration in the dynamic range of 1.0–1000 pg mL−1 at a detection limit of 0.68 pg mL−1. Good reproducibility, high specificity and acceptable storage stability were acquired for the analysis of the target IL-6. Moreover, the capacitance immunosensor was utilized to determine IL-6 in human serum specimens and gave well-matched analytical results in reference to the human IL-6 ELISA kit.

A sensitive DNAzyme-based electrochemical sensor for Pb2+ detection with platinum nanoparticles decorated TiO2/α-Fe2O3 nanocomposite as signal labels

Xianjiu Liao,Jianjun Luo,Jing Wu,Taotao Fan,Yao Yao,Fenglei Gao,Yong Qian

In this work, a sensitive DNAzyme-based electrochemical sensor had been developed for Pb 2+ detection with platinum nanoparticles decorated TiO 2 /α-Fe 2 O 3 composite as signal labels. The morphological and compositional properties of the PtNPs/TiO 2 /α-Fe 2 O 3 were examined by using transmission electron microscopy , X-ray diffraction, and X-ray photoelectron spectroscopy. The result showed that the shape of α-Fe 2 O 3 nanocrystals were the cubic, the average length ranged from 150 to 300 nm, and the PtNPs with about 3–4 nm in diameter were well dispersed on the surface of TiO 2 /α-Fe 2 O 3 . TiO 2 /α-Fe 2 O 3 were used as a “carrier” to load the PtNPs tracers as well as for Apt3 immobilization. In turn, the DNAzyme structure of Apt1 and Apt2 was immobilized on the gold electrode surface as the capture probes. Subsequently, DNAzyme modified electrodes reacted with target Pb 2+ and the Apt3/PtNPs/TiO 2 /α-Fe 2 O 3 composite were deposited on the electrode surface. The performance of the DNAzyme-based electrochemical sensor was tested by recording the electrocatalytic reduction current toward H 2 O 2 . Enhanced sensitivity was achieved by TiO 2 /α-Fe 2 O 3 nanocomposites as a nanocarrier in order to PtNPs at a high ratio. The DNAzyme-based electrochemical sensor described here exhibited excellent sensitivity and specificity for target ion. This work represented a promising potential for on-site testing Pb 2+ in real drinking water and serum sample analysis.

Detection of ultralow concentration of specific protein plays a key role in biotechnological applications and medical diagnostics. In this study, we demonstrate an amplified chemiluminescence biosensing platform for sensitive detection of protein. The biosensing platform ingeniously combines target-catalyzed hairpin assembly and Exo III-assisted signal amplification . A catalytic G-quadruplex-hemin DNAzyme is further employed to stimulate the generation of chemiluminescence in the presence of luminol and H 2 O 2 . These designs together allow a high sensitivity for the biotarget, human α-thrombin, resulting in a detection limit of 0.92   pM, and it is much lower than previous reported studies. In addition, the proposed biosensing platform is versatile. By conjugating with various recognition units, it could be employed to sensitive detect various DNA-binding proteins and might find wide applications in biomedical fields .

Highly sensitive electrochemical sensing platform for lead ion based on synergetic catalysis of DNAzyme and Au–Pd porous bimetallic nanostructures

Qian Zhou,Youxiu Lin,Yuping Lin,Qiaohua Wei,Guonan Chen,Dianping Tang

In this work, a sensitive and specific electrochemical biosensor for lead ion (Pb 2+ ) detection was developed by coupling with synergetic catalysis of porous Au–Pd bimetallic nanoparticles and hemin/G-quadruplex-based DNAzyme. In the presence of target Pb 2+ , the substrate strand was cleaved and the enzyme strand was released. Subsequently, G-rich DNA-labeled Au–Pd bimetallic nanoparticle was linked with the released enzyme strand through the helper DNA . Upon addition of hemin , a large number of hemin/G-quadruplex-based DNAzyme molecules were formed on the electrode to serve as nicotinamide adenine dinucleotide hydrogen (NADH) oxidase and peroxidase mimics. DNAzyme could catalyzed the reduction of H 2 O 2 , generated from NADH in the presence of O 2 , to produce an electrochemical signal when using thionine as the electron mediator. Introduction of porors Au–Pd bimetallic nanoparticles could enhance the detectable signal and cause the increase in the sensitivity. Experimental results showed that the variations (∆ I ) in the cathodic peak currents of the biosensor were linearly dependent on target Pb 2+ concentrations from 1.0   pM to 100   nM with a detection limit (LOD) of 0.34   pM. The excellent performance of the sensing platform indicated its promising prospect as a valuable tool for simple and cost-effective Pb 2+ detection in practical application.

Electrochemical aptasensor for thrombin using co-catalysis of hemin/G-quadruplex DNAzyme and octahedral Cu2O-Au nanocomposites for signal amplification

Shuai Chen,Pin Liu,Kewen Su,Xuan Li,Zhen Qin,Wei Xu,Jun Chen,Chaorui Li,Jingfu Qiu

In this work, novel octahedral Cu 2 O-Au nanocomposites were synthesized and first applied in an electrochemical aptasensor to detect thrombin (TB) with the aid of a DNAzyme for signal amplification . The octahedral Cu 2 O-Au nanocomposites have not only simultaneously served as signal amplifying molecules but have also been utilized as an ideal loading platform to immobilize a large number of electroactive substances and recognition probes. Gold nanoparticles (AuNPs) were grown directly on the surface of the octahedral Cu 2 O nanocrystals , and the Cu 2 O-Au nanocomposites obtained had the advantages of large surface areas and excellent biocompatibilities. The hemin/G-quadruplex, which was formed by intercalating hemin into the amino terminated thrombin binding aptamer (NH 2 -TBA), and the electroactive toluidine blue (Tb) were immobilized onto the Cu 2 O-Au nanocomposite surfaces through a stable Au-N bond. AuNPs, Cu 2 O and hemin/G-quadruplex co-catalyse the H 2 O 2 in the working buffer to promote the electron transfer of Tb as a multiple signal amplification strategy in order to improve the performance of the electrochemical aptasensor. Under optimal conditions, the designed aptasensor exhibited sensitive detection of TB from 100 fM to 20   nM with a lower detection limit of 23fM. This proposed aptasensor exhibited good sensitivity, high specificity and acceptable reproducibility and could be widely applied in bioassay analysis.

Label-free colorimetric detection platform based on catalytic hairpin self-assembly and G-quadruplex/hemin DNAzyme for comprehensive biomarker profiling

Changjiang Li,Yuqiang Hu,Tianzi Shi,Kejun Dong,Tongbo Wu

The expression level of human apurinic/apyrimidinic endonuclease 1 (APE1) is closely associated with the onset of various diseases, establishing it as a crucial clinical biomarker and a target in anti-cancer efforts. This study accomplished colorimetric and visual detection of APE1 by harnessing its endonuclease activity through catalytic hairpin self-assembly (CHA) and G-quadruplex/hemin DNAzyme. Optimization of the freedom degrees of the G-rich sequence significantly improved the detection performance of the strategy by influencing DNAzyme formation. Additionally, we replaced the signal reporting system with a molecular beacon to develop a fluorescence detection strategy, which served as an extension of the signal amplification system for validation and signal readout. The fluorescent probe method achieved a detection limit of 3.37 × 10 −4 U/mL, while the colorimetric method yielded a detection limit of 6.5 × 10 −3 U/mL, with a linear range spanning from 0.01 to 0.25 U/mL. Subsequently, the colorimetric approach effectively assessed APE1 activity in biological samples and facilitated the screening of APE1 activity inhibitors. Furthermore, this CHA/G-quadruplex/hemin DNAzyme strategy was adapted for the colorimetric detection of adenosine, showcasing its broad applicability across various biomarkers. The developed colorimetric analytical strategy represents a pivotal biosensing platform for diagnosing and treating diseases.

DNAzyme-adsorbed polydopamine@MnO2 core–shell nanocomposites for enhanced photothermal therapy via the self-activated suppression of heat shock protein 70

Yang Xi,Xin Xie,Ying Peng,Peng Liu,Jinsong Ding,Wenhu Zhou

Photothermal therapy (PTT) is a promising tumor treatment modality, but its efficacy is strictly hindered by abnormally upregulated heat shock proteins (HSPs) in tumor cells under heat stress. Herein, we developed a flower-like MnO2-coated polydopamine (PDA@MnO2) core–shell nanoplatform with the surface adsorption of HSP70-silencing DNAzyme (DZ) for enhanced PPT. The PDA core acted as a robust photothermal agent, and also as a reductant to allow the surface growth of MnO2via an in situ reduction of KMnO4. The MnO2 shell enabled a rapid and efficient adsorption of DZ, and more importantly, acted as a metal reservoir to release Mn2+ in response to intracellular stimuli for the in situ activation of DZ, which addressed the key limitation of DZ for biological applications, i.e., metal-dependent activity. As a result, HSP70 was remarkably suppressed for improved PTT efficacy upon laser irradiation, which was explicitly demonstrated both in vitro and in vivo. Upon intravenous injection, the nanosystem could effectively accumulate in the tumor, and impose potent PTT for complete tumor elimination via inducing tumor cell apoptosis, but without any noticeable toxicity. This work provides a promising nanosystem for enhanced PTT via silencing resistance-related genes, and offers ideas for the design of self-activated gene therapy platforms using DZ.

Ultrasensitive electrochemical/visual colorimetric dual-mode biosensing of Pb2+ based on G-quadruplex DNAzyme and 2D bimetallic porphyrin MOF derived nanocomposites

Chuang Yang,Fuyan Chen,Yajie Zhao,Wushuang Bai

Rapid and accurate detection of lead (II) ions (Pb 2+ ) is of great importance for food and environmental safety. In this work, we developed a dual-mode biosensing strategy for Pb 2+ determination based on a bimetallic two-dimensional porphyrinic MOF nanocomposite and G-quadruplex DNAzyme, combined with enzyme-like synergistic catalysis and DNA hybridization chain reaction (HCR) amplification. The sensing platform provides both electrochemical and visual colorimetric readouts, enabling highly sensitive detection under different application scenarios. Benefiting from the coupling of HCR with the cooperative catalysis of the G-quadruplex DNAzyme and MOF nanocomposite, the assay shows markedly enhanced sensitivity. In the electrochemical mode, the sensor achieves a limit of detection of 0.4 nM with a linear range from 1 to 10,000 nM. In the colorimetric mode, the assay affords a detection limit of 2.6 nM with a linear range from 10 to 10,000 nM. This dual-mode Pb 2+ biosensing strategy shows promising potential for food safety monitoring and environmental pollution analysis, and provides a useful reference for the development of new techniques for trace heavy-metal ion detection.

One-step electrochemical aptasensor for lactate detection in athlete training using AuPt NPs and PEI-rGO composite with Na+-dependent DNAzyme

Xu Han,Haozhen Li,Ling Niu

This study introduces a novel electrochemical aptasensor designed for the sensitive detection of lactate in athlete sweat. The sensor is based on a PEI-rGO-modified SPCE coupled with AuPt NPs, and utilizes Na+-assisted DNAzyme for signal amplification. The PEI-rGO modification enhances the aptasensor's conductivity, while the AuPt NPs efficiently load MB and LDNA, generating stable electrochemical signals. Furthermore, the Na+-dependent DNAzyme leverages the Na+ naturally present in sweat to activate the cleavage process, amplifying the detection signal. The sensor exhibits a strong linear relationship between current response and lactate concentration in the range of 5 × 10−3 to 10 mM, with a limit of detection (LOD) of 2.974 μM. It demonstrates excellent specificity, stability, and repeatability. The optimized aptasensor offers significant potential for real-time, non-invasive lactate monitoring, particularly in athletic performance and health assessment. This work provides a promising approach for the development of reliable, portable biosensors for tracking lactate metabolism, with implications for sports science and health management.

A ratiometric electrochemical zearalenone aptasensor based on Fc-Au-MoS2 nanosheet and triple-helix molecular switch-triggered cleavage effect of DNAzyme

Yihao Li,Fengzhen Jin,Bowen Li,Shun Wu,Yuanqing Wang,Yaohui Wu,Yonghong Wang,Ge Ning

Zearalenone (ZEN) is a mycotoxin that exerts multiple toxic effects on humans through the contamination of grains such as corn, posing a significant threat to food safety and human health. This work proposed a dual-signal ratiometric electrochemical aptasensor utilizing ferrocene-carboxylic acid‑gold nanoparticle‑molybdenum disulfide nanosheets (Fc-Au-MoS 2 ), hybridization chain reaction (HCR), and the cleavage effect of DNAzyme triggered by triple-helix molecular switch (THMS) to detect ZEN. The specific interaction with ZEN triggers the release of the DNAzyme from THMS, which then removes the immobilized primer chain on the electrode surface with the assistance of Pb 2+ , thereby preventing HCR occurrence. This inhibitory mechanism reduces methylene blue (MB) insertion while exposing the active sites of Fc-Au-MoS 2 , generating an opposite signal response to achieve high-precision ratio detection. The aptasensor demonstrated a low detection limit of 29.4 pM, with satisfactory recoveries in corn flour (97.3 %–106.4 %), confirming its potential for monitoring mycotoxin residues in food safety.

Signal-on fluorescent sensing strategy for Pb2+ detection based on 8–17 DNAzyme-mediated molecular beacon-type catalytic hairpin assembly circuit

Wang Junyang,Liu Zheng,Li Ying,Yang Chuanyu,Ma Xinyue,Li Hongxia,Sun Chunyan

Based on a Pb 2+ -specific 8–17 DNAzyme-induced catalytic hairpin assembly (CHA), a simple signal-on fluorescence strategy for lead ion detection was established. 8–17 DNAzyme was used as the recognition element of Pb 2+ , which catalyzed the cleavage of the RNA base embedded in the DNA substrate strand, while releasing part of the substrate strand (S’) as CHA initiator. And two hairpin probes (H1 and H2-FQ) were designed according to the sequence of S’ for CHA, in which H2-FQ was labeled with the fluorophore FAM and quencher BHQ-1 as fluorescent “molecular switch” based on fluorescence resonance energy transfer (FRET). In the presence of Pb 2+ , the CHA reaction was triggered to form a large number of H1-H2 complexes, enabling enzyme-free isothermal amplification and a signal-on fluorescence strategy. In the concentration range of 0.5–1000 nM, the fluorescence signal increases with the increase of Pb 2+ concentration. The quantitative detection limit of Pb 2+ by this method is 0.5 nM, which has better detection performance compared with the FQ-labeled 8–17 DNAzyme method. The established biosensor exhibits good specificity and can be effectively used for the detection of Pb 2+ in real samples of river water and grass carp. Through ingenious nucleic acid sequence design, DNAzyme and CHA reactions are integrated to realize the enzyme-free isothermal amplifications and sensitive detection of Pb 2+ , which holds potential versatility in food supervision and environmental monitoring. Dual DNAzyme-catalytic assembly of G-quadruplexes for inducing the aggregation of gold nanoparticles and developing a novel antibiotic assay method

Wang Xiaojun,Yang Jingru,Xie Yiming,Lai Guosong

By utilizing a target biorecognition reaction to induce the self-assembly of G-quadruplexes and the aggregation of gold nanoparticles (Au NPs), this work develops a novel colorimetric biosensing method for kanamycin (Kana) antibiotic detection. The compact G-quadruplex structure was assembled from its two half-split sequences which were designed in two hairpin substrates of the Mg 2+ -dependent DNAzyme (MNAzyme). Besides hybridizing with the aptamer strand, the MNAzyme sequence was also split into two half fragments to be designed in the two substrates. Upon the aptamer-recognition reaction toward Kana, the MNAzyme strand could be quantitatively released to cause the exposure of the split G-quadruplex-sequences on two hairpin substrate-modified Au NPs and simultaneous release of two half fragments of the MNAzyme-sequence. Thus, the K + -assisted self-folding of G-quadruplexes causes the cross-linking of the two Au NPs to realize the Au NP aggregation–based colorimetric signal output (measured at the largest absorption peak near 520 nm). Meanwhile, the self-assembled formation of the second MNAzyme drastically amplified the signal response. Under the optimal conditions, a wide linear range from 0.1 pg mL −1  to 10 ng mL −1 and an ultrahigh sensitivity with the detection limit of 76 fg mL −1 were obtained. The dose-recovery experiments in real samples showed satisfactory results with recoveries from 98.4 to 105.4% and relative errors compared with the ELISA method less than 4.1%. Due to the high selectivity, excellent repeatability and stability, and simple manipulation, this method indicates a promising potential for practical applications. Graphical abstract A novel homogeneous biosensing method was developed for the convenient detection of the kanamycin antibiotic. The target biorecognition-induced and dual DNAzyme-catalytic assembly of G-quadruplexes enabled the amplified aggregation of gold nanoparticles for the simple, cheap, stable, and ultrasensitive colorimetric signal transduction of the method.
