The mortality rate of cardiovascular diseases (CVD) remains alarmingly high. Studies have shown that reactive oxygen species (ROS)-mediated inflammatory responses and oxidative stress are key pathological mechanisms driving CVD. Nanozymes, nanomaterials with enzyme-like activities, offer several advantages over natural enzymes, including enhanced stability and scalability. Previous research has demonstrated that nanozymes containing cerium (Ce) or manganese (Mn) possess excellent ROS-scavenging capabilities. Dopamine, a positive inotropic agent commonly used in clinical practice to improve cardiac function in patients with heart failure, has also been shown to self.

Photodynamic therapy (PDT) is a promising anticancer strategy but remains limited by inefficient reactive oxygen species (ROS) generation and insufficient subcellular targeting. Here, we report a cationic engineering strategy to enhance mitochondrial targeting and ROS-amplified photodynamic tumor therapy. A neutral aggregation-induced emission (AIE) photosensitizer (PS) was stepwise converted into monocationic (PS-Bu) and dicationic (PS–PPh) derivatives, enabling precise regulation of molecular charge density. Cationic modification modulates excited-state energetics, promotes intersystem crossing, and simultaneously enhances Type I and Type II ROS generation under visible-light irradiation. The increased positive charge drives preferential mitochondrial accumulation, leading to pronounced mitochondrial membrane depolarization, intracellular ROS amplification, cell-cycle arrest, and apoptosis. Consequently, PS–PPh exhibits markedly enhanced phototoxicity toward HepG2 cells while maintaining negligible dark toxicity. In vivo, PS–PPh achieves superior tumor growth suppression in HepG2 xenograft models, accompanied by reduced proliferation, enhanced apoptosis, and alleviation of tumor hypoxia, without detectable systemic toxicity. This work establishes cationic modulation as an effective molecular design paradigm for mitochondria-targeted and ROS-enhanced photodynamic cancer therapy.

Diabetic wound healing represents a highly prevalent
and economically
burdensome complication of diabetes mellitus. The healing is hampered
by two pathophysiological barriers: persistent bacterial biofilms
and impaired angiogenesis. While reactive oxygen species (ROS)-mediated
strategies are promising, their efficacy is limited by exogenous H 2 O 2 dependency and rapid oxidant decomposition.
In this study, we engineer a multifunctional nanoplatform (SNP/ZnO 2 @lipo), which synergizes potent antibacterial ability and
angiogenesis capacity. Its architecture features polyvinylpyrrolidone-modified
zinc peroxide (ZnO 2 ) cores with sodium nitroprusside (SNP)
adsorbed on their surface, which are further enveloped within cationic
phospholipid bilayers. In infected, mildly acidic microenvironments,
bacterial phospholipases and membrane instability destabilize the
lipid shell, thereby exposing the SNP/ZnO 2 core. The exposed
ZnO 2 and SNP then generate H 2 O 2 ,
Zn 2+ , Fe 2+ , and NO in situ .
The orchestrated ROS and reactive nitrogen species (RNS) cascade achieves
synergistic bactericidal efficacy through dual mechanisms: Fe 2+ -mediated Fenton reactions and NO-derived ·NO formation.
The continuous in situ production of oxidants overcomes
the need for exogenous H 2 O 2 . In vitro , the platform shows high biocompatibility, a strong antibacterial
capacity, and remarkable angiogenic potential. In diabetic murine
wound models, SNP/ZnO 2 @lipo accelerates wound healing through
concurrent clearing of infection and promoting neovascularization.
These data identify SNP/ZnO 2 @lipo as an intelligent wound
microenvironment-responsive nanoplatform that couples microbial eradication
with vascular regeneration, offering a promising strategy for treating
infected diabetic wounds.

A nerve guidance conduit (NGC) based on MXene-MnO₂ hydrogel is developed for the regeneration of peripheral nerves. • The NGC possesses the dual advantages for promoting Schwann cell proliferation and alleviating chronic inflammation. • The NGC can actively regulate the neural regeneration microenvironment and promote angiogenesis. • The NGC can promote neurofilament formation, remyelination and prevent muscle atrophy. Peripheral nerve regeneration remains a major challenge in clinical due to persistent inflammatory microenvironments and oxidative stress that impede functional recovery. In this study, we designed a bio-responsive nerve guidance conduit (NGC) based on a MXene-MnO₂ nanocomposite hydrogel that actively orchestrates the regenerative microenvironment through material-mediated biological crosstalk. Unlike conventional passive conduits, our system leverages the unique properties of two-dimensional MXene to enhance Schwann cell proliferation, while MnO₂ nanoparticles act as catalytic anti-oxidative modules that continuously neutralize ROS and resolve chronic inflammation. This dynamic intervention promotes a timely immune transition to shift macrophages from M1 to M2 phenotype, which facilitates angiogenesis within the injury site. In vivo tests further revealed that the treatment group not only exhibited increased neurofilament number and reduced muscle atrophy but also showed significantly enhanced remyelination. The modular design and catalytic microenvironment-regulation strategy represent a new direction in neural interface engineering. This work provides a scalable and effective platform for advanced neural repair applications and material-guided immunomodulation in regenerative medicine. Illustration of nerve regeneration promoted by MXene-MnO₂ nanocomposite hydrogel NGCs via scavenging ROS and facilitating macrophage polarization towards the M2 phenotype. The therapeutic efficacy of glucose oxidase (GOx)-mediated starvation therapy and β-Lapachone (LPC)-induced oxidative stress is significantly constrained by tumor hypoxia and elevated antioxidant defenses in the tumor microenvironment. To address these limitations, a multifunctional nanoplatform, LPC/GOx@ZIF-8@MnO 2 , was developed through the integration of LPC and GOx within a zeolitic imidazolate framework-8 (ZIF-8) carrier followed by surface modification with manganese dioxide (MnO 2 ), and its antitumor efficacy and mechanistic basis were systematically evaluated using HeLa cells in vitro and U14 tumor-bearing mouse models in vivo . Comprehensive in vitro and in vivo studies demonstrated significant tumor growth inhibition with excellent biosafety, while mechanistic investigations revealed that this nanocomposite achieves synergistic antitumor effects through four coordinated mechanisms: MnO 2 -catalyzed conversion of endogenous hydrogen peroxide to oxygen alleviates hypoxia while sustaining GOx-mediated glucose depletion; a self-amplifying cascade reaction enhances LPC-induced reactive oxygen species (ROS) generation; photothermally activated upregulation of NAD(P)H:quinone oxidoreductase 1 (NQO1) via the heat shock protein 70 (HSP70)/NQO1 axis enhances LPC efficacy; and depletion of glutathione disrupts cellular antioxidant defenses. Furthermore, it was also revealed that LPC/GOx@ZIF-8@MnO 2 acts on caspase-3, high mobility group box 1 (HMGB1), and matrix metalloproteinase-1 (MMP-1) to coordinate apoptosis induction and metastasis suppression, with molecular docking further clarifying the key binding sites of LPC with these three targets. Collectively, this work presents an innovative strategy to overcome therapeutic limitations imposed by tumor hypoxia, provides molecular-level insights into multimodal synergistic therapy, and offers new ideas for precision tumor therapy.

Puerarin (PU) helps slow down and control the harmful progression of atherosclerosis (AS) by reducing inflammation, inhibiting foam cell formation, and exhibiting anticoagulant effects. However, its clinical application is limited due to its low bioavailability and other drawbacks. In recent years, nanoparticle-based therapeutic strategies targeting reactive oxygen species (ROS) at AS pathological sites and enhancing drug bioavailability and biocompatibility have shown great potential for AS treatment. In this study, we developed ROS-responsive cinnamaldehyde polymer nanoparticles loaded with PU (RGCP@PU NPs) for the treatment of AS. These nanoparticles demonstrate good stability and biocompatibility in physiological environments and can respond to ROS in the AS pathological environment, thereby releasing the encapsulated PU. In vitro experiments showed that these nanoparticles effectively inhibited oxidative stress and inflammation levels. In in vivo studies, RGCP@PU NPs were able to stabilize atherosclerotic plaques, prevent their further deterioration, and delay the progression of AS. Moreover, compared to free PU, these nanoparticles exhibited superior therapeutic effects in inhibiting AS progression. Therefore, RGCP@PU NPs represent a promising nanocarrier that not only effectively suppresses AS progression but also provides a new approach for the design of responsive drug delivery systems for AS treatment.

Hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide. The main treatment methods include surgery and chemotherapy, but these often lead to significant drug resistance. Therefore, there is an urgent need to find new, low-toxicity, and efficient anti-HCC drugs. In this study, we developed a reactive oxygen species (ROS)-responsive nanoparticle drug delivery system to enhance the anti-tumor efficacy of radiotherapy through local ROS-activated chemotherapy. We synthesized a ROS-responsive polymer, PLA-TDE-PLA, for drug delivery, and encapsulated the newly synthesized compound 1 within it, constructing PLA-TDE-PLA@1. The non-polar sulfide residues in the drug delivery system are oxidized to polar sulfoxide units upon ROS activation, causing structural changes in the PLA-TDE-PLA carrying compound 1, including swelling and partial dissociation, leading to significant drug release. Cytological experiments showed that PLA-TDE-PLA@1 significantly inhibited the proliferation of HCC cells and exerted its effects by reducing the expression of Bcl-2. Through the molecular docking simulation, the proposed compound has been demonstrated to have good biological effect, further, based on the proposed compound, a generative study has been performed and some outstanding optimized molecules are analyzed in detail.

Cataracts are one of the primary causes of blindness worldwide; however, their pathogenesis remains unclear. Oxidative stress and apoptosis are two dominant inducers in the progression of cataracts; however, little is known about the specific mechanisms associated with mitophagy. This study aimed to investigate the role of PTEN-induced putative kinase 1(PINK1)-mediated mitophagy in cataract development. Initially, we induced a rat cataract model using sodium selenite and observed the upregulated expression of PINK1 and other autophagy-related proteins within lens epithelial cells, accompanied by apoptosis. Furthermore, the survival rate of human lens epithelial cells was significantly reduced by H 2 O 2 treatment. However, PINK1 overexpression reduced ROS levels, allowing cells to survive. This reduction in reactive oxygen species (ROS) levels led to a decrease in cleaved caspase-3 and Bcl-2-associated X protein (Bax) expression and an increase in B-cell lymphoma 2 (Bcl-2) levels. In summary, PINK1 maintains mitochondrial functional stability and inhibits apoptosis by activating mitophagy, thus potentially playing a crucial protective role in cataract pathogenesis.

Sonodynamic therapy (SDT) has emerged as an encouraging route in tumor treatment, due to its exceptional tissue penetration depth and favorable safety profile. Nevertheless, the clinical translation of conventional organic sonosensitizers is hindered by intrinsic limitations, including pronounced hydrophobicity, insufficient chemical stability, and low reactive oxygen species (ROS) production. In contrast, hollow covalent organic frameworks (HCOFs) exhibit exceptional cargo-loading capabilities, structural robustness, and biocompatibility, positioning them as ideal nanoplatforms for advanced therapeutic applications. Herein, we engineered a bowl-shaped HCOF architecture designed to amplify ultrasonic cavitation effects. This nanostructure was subsequently functionalized with the sonosensitizer (Hemin) and subjected to strategic metallization via metal ion incorporation, culminating in the development of a high-efficiency antitumor nanosystem (FeHHCA). FeHHCA can achieve dual-mode ROS generation, namely, sonodynamic synergistically generating 1O2 and being specifically activated by a tumor microenvironment (TME) to generate ˙OH through a Fenton-like reaction, achieving an 78.7% tumor inhibition rate in vivo. These findings offer innovative approaches and strategies for the design of hollow COFs and offer great potential for the application of SDT in cancer treatment.

The clinical efficacy of platinum-based chemotherapeutics, such as cisplatin, has been compromised by the prevalent emergence of drug resistance. This has propelled the search for alternative metal-based anti-tumor agents. Herein, we introduce PolyRu, a novel amphiphilic polymer containing a ruthenium complex and thioketal bonds in its main chain, for lung cancer treatment. PolyRu can self-assemble into nanoparticles (NP@PolyRu) in aqueous solutions and degrade within the reactive oxygen species-rich tumor microenvironment. The ruthenium complex in PolyRu specifically targets mitochondria and induces cancer cell apoptosis. The efficacy of NP@PolyRu was validated in a patient-derived xenograft model of human lung cancer, where NP@PolyRu significantly suppressed tumor progression with favorable biocompatibility, and showed excellent anti-tumor immune effect. NP@PolyRu emerges as a promising candidate for addressing cancer drug resistance, signifying a substantial leap forward in the realm of metal polymer-based cancer therapeutics.

The central challenge in gene delivery systems lies in achieving efficient intracellular delivery of plasmids and sustained release. In this study, a cyclic reactive oxygen species (ROS)-responsive gene delivery platform based on silicon nanowire arrays (SN) was developed. The high-density positive charged polyethylenimine (PEI) polymers were covalently grafted onto the SN surface, allowing for electrostatic adsorption of both plasmid DNA and trans-cinnamaldehyde precursor (TAC). When cells are co-cultured on this material, the needle-like nanostructures of SN would penetrate the cell membrane through physical effect, efficiently delivering the loaded plasmids into stem cells. The release mechanism of this system was based on an ROS-triggered cascade response, TAC oxidized by ROS to generate cinnamaldehyde, and then induced mitochondrial oxidative stress to promote cascade production of more ROS. This triggered the dissociation of SN-grafted PEI from the nanowires, enabling sustained plasmid release. In vitro release assays showed that the treatment of 1 mM H 2 O 2 (exogenous ROS stimulant) for only 10 min induced 79 % plasmid release from SN surfaces. For cells that are difficult to be transfected, such as mouse embryonic stem cells (mESC) and mesenchymal stem cells (MSC), this platform exhibited excellent transfection efficiencies of 94 % and 74 %, respectively, while maintaining good cytocompatibility. This study developed an SN-based ROS-responsive gene delivery platform, achieving efficient plasmid delivery and sustained release via synergistic physical penetration and biochemical-responsiveness. It provides a solution with important application value for gene transfection in hard-to-transfect stem cells and demonstrates promising translational potential in the fields of gene therapy and regenerative medicine.

When a burst of reactive oxygen species (ROS) is induced after radiation exposure, a sufficient radioprotector drug must be quickly released to neutralize the ROS . An oral polymeric carrier of poly(ε-caprolactone)-b-poly(ethylene glycol)-b-poly(ε-caprolactone) was synthesized via the ROS-response linker thioketal (TK-linked PCEC) and used for delivery of the drug WR-1065. The drug-loaded nanoparticles (PCEC/WR-1065 NPs) had high WR-1065 content (19.7 % w/w) in the core and a particle size of 183 nm. The PCEC/WR-1065 NPs could protect the drug from degradation at gastric pH. More than 70 % of the TK-linked PCEC copolymer could be cleaved in 1 h. Therefore, a sufficient amount of WR-1065 could be quickly released from the PCEC/WR-1065 NPs to neutralize the burst of free radicals generated under irradiation. The in vitro cell uptake results of TK-linked PCEC NPs indicated that the NPs could be efficiently endocytosized by Caco-2 cells. Oral administration of the PCEC/WR-1065 led to survival benefit as well as protective effects on hematopoiesis and main organs with respect to the irradiation and oral free WR-1065 groups after 7.2 and 8.0 Gy total body irradiation, respectively. In vivo biodistribution studies indicated that the PCEC/WR-1065 NPs accumulated mainly in intestinal tissue. These results suggest that the design presented here provides a promising oral ROS-sensitive platform for acute radiation syndrome.
