Combretastatin A4 phosphate (CA4P), the water-soluble phosphate pro-drug of combretastatin A4 (CA4), acts as a novel vascular disrupting agent (VDA). It strongly inhibits tubulin polymerization and selectively exerts action against tumor blood vessels. However, its quite short half-life (∼30 min) results in a short action time and consequently limits its application. Therefore, it is beneficial to develop a sustained-release preparation of CA4P for relatively long action duration. On the other hand, conventional PLGA microspheres display poor encapsulation capability for water-soluble small molecules due to rapid diffusion outside. Hence, we developed CA4P-loaded microspheres (CA4P-MS) composed of methoxy poly(ethylene glycol)-b-polylactide (PELA) reverse micelles and PLGA with a nano-in-micro sea-island structure. The CA4P encapsulation efficiency of CA4P-MS reached 92.88%. More interestingly, sustained drug release was achieved, which contributed to the prolonged inhibition effect on S180 tumor during a 30-day period with one-time subcutaneous administration. In conclusion, our study established a biodegradable polymer based platform to construct microspheres for water-soluble small molecular compounds with sustained release and a long-lasting action.

Large scale of Cd-contaminated soil has become a serious threat to environmental safety and human health, whereas field applicability of existing remediation technologies has encountered numerous obstacles, such as poor stability, secondary pollution, long operation time, and high chemical cost. Herein, a novel activated calcite with significantly enhanced immobilization stability of Cd in soil has been synthesized based on mechanochemical technology. Our results demonstrate that the passivation rate of bioavailable Cd in Cd-contaminated soil (initial concentration 3 mg/kg) could be maintained at 65 % after 50 days of passivation cultured with activated calcite. Mechanistic studies have revealed that the outstanding performance is attributed to the enhancement of the CO 3 2– and Ca 2+ release properties of calcite through mechanical activation, leading to a 550 % increase in Ca 2+ release. The CO 3 2– fraction binds in situ to the free Cd in the soil, forming a stable carbonate-bound state. Simultaneously, the OH – released from the continuous hydrolysis of CO 3 2– effectively regulates and stabilizes the soil pH within the neutral range. Further, SEM-EDS and sequential extraction procedures (SEP) analyses implied that the specific adsorption of Cd by in-situ formed iron oxides in soil was visibly related to the release of OH – from activated calcite and immobilized the free state Cd in the more stable iron oxide-bound state. The current work shows the high sustainability and stability of Cd-contaminated soil immobilization by activated calcite materials.

Pickering emulsions (PEs) have wide applications in delivering nutraceuticals. However, the impact of extracting nanocellulose from corn stover on stabilizing PEs and delivering nutraceuticals remains unclear. In this study, four types of nanocellulose, cellulose nanocrystals (CNC), cellulose nanofibers (CNF), lignin-containing cellulose nanocrystals (LCNC), and lignin-containing cellulose nanofibers (LCNF) were successfully prepared from corn stover, an agricultural waste. Among them, LCNC and LCNF exhibited stable reticulated microstructures, lower crystallinity, and excellent thermal stability. Besides, lignin enhanced the nanoparticles' hydrophobicity, promoting the formation of more ideally amphiphilic particles, resulting in denser emulsions at the oil-water interface. Furthermore, emulsions stabilized by LCNC and LCNF demonstrated remarkable resistance to quercetin degradation under UV light exposure (with residual level exceeding 90 %) and improved quercetin's bioaccessibility during the in vitro digestion tests, achieving the highest bioaccessibility of 48.3 %. This study provided an innovative perspective on utilizing stover-derived materials for stabilizing PEs and delivering lipophilic nutrients.

Semaglutide has the advantages of high efficacy and fewer side effects in the treatment of Type 2 diabetes (T2DM). However, the marketed formulations of semaglutide have issues such as poor compliance in individuals who require repeated injections and low oral bioavailability, limiting its use. To overcome the aforementioned issues, semaglutide microspheres with single-phase zero-order release behavior were created using two methods: polymer blending and surfactant addition. Their characteristics, in vitro release, and in vivo pharmacodynamics were investigated. Two types of semaglutide microspheres show a desirable encapsulation effectiveness and smaller particle size. When compared to ordinary semaglutide microspheres (SEM-MS), due to the introduction of hydrophilic substances such as low molecular weight PLGA and Tween 20, the lag phase disappears, it exhibits zero-order release, with better in vitro release behavior. When injected subcutaneously to T2DM rats, the drug was continuously released for about a month, and the blood drug concentration fluctuated little, exhibiting good hypoglycemic and repair β cell effects. The efficacy is superior to SEM-MS and consistent with injection. The semaglutide microspheres created by two ways in this investigation can address the issues with commercial formulations while remaining safe.

Polycyclic Aromatic Hydrocarbons (PAHs) are persistent organic pollutants known for their carcinogenic, teratogenic, and mutagenic properties. A considerable amount of PAHs has been released into groundwater. Persulfate-based advanced oxidation processes (PS-AOP) are commonly employed to remediate such contamination. However, traditional in-situ chemical oxidation (ISCO) faces challenges such as concentration rebound and tailing. To address these issues, sodium persulfate sustained release materials (PDS@SA) and ferrous sulfate sustained release materials (FeSO 4 @SA) were developed using stearic acid (SA) as a binder. Batch and column experiments were conducted to evaluate the efficacy of pyrene degradation through PDS@SA activated by FeSO 4 @SA. The batch experiments showed that the optimal mass ratio of FeSO 4 @SA to PDS@SA was 1:2, achieving degradation efficiencies over 95 % for pyrene with various concentrations (5, 10, 15, 20 mg⸱L −1 ) within 2 h. The column experiments showed that sustained release materials (FeSO 4 @SA and PDS@SA) with a particle size of 5 mm achieved an average removal rate of 89.3 % during days 7–15, while those with particle sizes of 7.5 mm and 10 mm achieved average removal rates of 68.5 % and 74.6 % during days 15–23, respectively. Additionally, electron paramagnetic resonance (EPR) analysis confirmed sulfate radicals SO 4 - ⸱ and hydroxyl radicals ⸱OH as the primary oxidative species during pyrene degradation. These findings suggested that the sustained activation of PDS by sustained release Fe 2+ represents a viable method for degrading PAHs in groundwater.

Autologous fat transplantation is increasingly applied in plastic and reconstructive surgery. Stromal vascular fraction cells (SVFs) combined with angiogenic factors, such as VEGF (vascular endothelial growth factor A) and Ang-1 (angiogenin-1), can improve angiogenesis, which is a critical factor for graft survival. However, direct transplant with such a mixture is insufficient owing to the short half-life of angiogenic factors. In this study, we evaluated whether a double sustained release system of VEGF/ANG-1-PLGA (poly (lactic-co-glycolic acid)) microspheres plus SVFs can improve angiogenesis and graft survival after autologous fat transplantation. VEGF/ANG-1-PLGA-sustained release microspheres were fabricated by a modified double emulsion–solvent evaporation technique. Human aspirated fat was mixed with SVF suspension plus VEGF/ANG-1 sustained release microspheres (Group C), SVF suspension (Group B) alone, or Dulbecco’s modified Eagle’s medium as the control (Group A). Eighteen immunocompromised nude mice were injected with these three mixtures subcutaneously at random positions. After 8 weeks, the mean volume of grafts was greater in the SVFs plus VEGF/ANG-1-PLGA group than in the control and SVFs groups (1.08 ± 0.069 ml vs. 0.62 ± 0.036 ml, and 0.83 ± 0.059 ml, respectively). Histological assessments showed that lower fibrosis, but greater microvascular density in the SVFs plus VEGF/ANG-1-PLGA group than in the other groups, though the SVFs group also had an appropriate capillary density and reduced fibrosis. Our findings indicate that SVFs plus VEGF/ANG-1-PLGA-sustained release microspheres can improve angiogenesis and graft survival after autologous fat transplantation.

Synthetic polymer fluid loss additive (FLA), an important type of admixture, was broadly applied in modern well cementation. However, the filter loss volume and fluidity of cement pastes containing polymer-FLA would deteriorate remarkably when sea water was used in mixing slurries instead of fresh water. In this study, a novel sustained-release-type fluid loss additive(S-FLA) was synthesized by means of anion-exchange intercalation reaction between an anionic type-copolymer and a calcium/aluminum type-Layered Double Hydroxide (Ca/Al-LDH). Based on the fluidity and compressive strength of experiments, it was found that in seawater mixing conditions, this composite material not only utilized its sustained release effect to significantly improve retention fluidity performance, but also the seed crystal effect of the Ca/Al-LDH effectively alleviated the declining in compressive strength of the slurries caused by the carboxyl group in the polymer. More interestingly, the realization of the slow release function increased actual adsorption capacity of the anionic polymer on the surface of cement hydrated particles, which made its controlling water loss effect was also better than that of the conventional FLA. The above advantages of this hybrid materials created the possibility to surmount the negative effect of electrolytes present in seawater, so as to provide some useful references for its practical application in the offshore well cement.

This study aimed to investigate the regulatory mechanism of spinning solution composition on the structure and drug sustained-release behavior of inorganic–organic drug-loaded nanofiber membranes fabricated via emulsion electrospinning. By adjusting water content (0, 1, 1.5 g) and zeolite loading, nanofiber membranes with different porous structures were prepared. Results indicated that fiber morphology and release kinetics were significantly influenced by the spinning solution composition: Systems containing water (W 1 -Zeo@DS NFs, W 1.5 -Zeo@DS NFs) formed shallow surface pores due to the emulsion template effect, resulting in finer fiber diameters. The system without water (Zeo@DS NFs) formed deeper internal channels via solvent-induced phase separation, leading to larger fiber diameters. Based on comprehensive release profiles and cumulative release rates, W 1 -Zeo@DS NFs demonstrated the optimal sustained-release performance and the highest drug utilization efficiency (56.85%). This study demonstrates that synergistic regulation of water content and zeolite loading can effectively optimize the multi-level pore structure of fibers, enabling precise control over drug release behavior, thereby providing a reference for designing efficient composite sustained-release drug delivery systems.

Polymer-based drug-loaded microspheres emerged as prevalent clinical modalities, exemplified by transarterial chemoembolization (TACE) and composite wound dressings, leveraging their sub-millimeter dimensions for versatile therapeutic deployment and targeted delivery. However, inherent limitations persist, including constrained drug loading capacity, rapid drug release kinetics, and non-degradability, thereby precluding the feasibility of repeat interventions. Based on these considerations, this study proposes a novel microsphere construction strategy that utilizes anionic monomer 2-acrylamide-2-methylpropanesulfonic acid (AMPS)-modified polyvinyl alcohol (PVA) as the primary raw material and incorporates cationic modified chitosan quaternary ammonium salt (QCS) to form a double-network microsphere. Owing to the abundant dynamic ionic interactions and the synergistic integration of soft and rigid molecular chains within the gel matrix, the resulting microsphere exhibits excellent mechanical properties, including a compressive deformation resistance exceeding 70%, high drug loading capacity, and prolonged sustained-release performance. When loaded with the cationic anticancer drug doxorubicin hydrochloride (DOX) and the antibacterial agent polyhexamethylene biguanide hydrochloride (PHMB), the microspheres achieved loading concentrations of 100.31 mg/mL and 132.25 mg/mL, respectively, with sustained release lasting over 12 days. In a rabbit hepatic artery embolization experiment, DOX-loaded microspheres effectively occluded blood flow and induced liver necrosis, thereby confirming their superior embolic performance. These features collectively demonstrate the promising application potential of this microsphere in the biomedical field.

Developing biodegradable active packaging films that combine mechanical reinforcement, barrier improvement, antibacterial activity, and controlled release of natural antimicrobials remains challenging. Here, TEMPO-oxidized bacterial cellulose (TOBC) was used to stabilize clove essential oil (CEO) Pickering emulsions, which were incorporated into sodium alginate (SA) matrices. The effects of TOBC concentration on emulsion stability and emulsion loading on film-forming solutions, film structure, mechanical performance, barrier properties, antioxidant and antibacterial activities, and CEO release were evaluated. The emulsion stabilized with 0.7 wt% TOBC showed good visual stability after 30 days and was selected for film preparation. Moderate incorporation of the TOBC-stabilized CEO Pickering emulsion improved SA film performance, with SA-E10 showing the best overall balance. SA-E10 reached a tensile strength of 38.56 ± 1.66 MPa and an elongation at break of 10.27%, while the moisture content decreased from 36.48 ± 2.32% to 27.93 ± 0.46%. The films also showed enhanced UV-shielding capacity and lower water vapor permeability. Antioxidant activity increased with emulsion loading, reaching 71.43 ± 2.55% for DPPH and 83.66 ± 1.49% for ABTS. Film-coated paper disks showed visible inhibition zones againstEscherichia coliandStaphylococcus aureus. Compared with direct CEO incorporation, the Pickering emulsion system delayed CEO release, indicating potential for sustained-release active packaging.

Chlorogenic acid (CGA) is a natural polyphenol with various biological activities, but its poor stability and premature release in the gastrointestinal tract limit oral application. Herein, a pH-responsive bilayer hydrogel based on sodium alginate (SA) and carboxymethyl cellulose (CMC) was developed to enhance the gastrointestinal stability and controlled release of CGA. CGA-loaded SA hydrogels were prepared via Ca 2+ -induced ionotropic gelation, followed by CMC coating to form a bilayer structure. The SA/CMC hydrogels showed a drug loading capacity of 15.2–16.7% and pH-dependent swelling behavior. In vitro release studies revealed that the bilayer hydrogel suppressed CGA release in simulated gastric fluid (pH 1.2), with a cumulative release of approximately 30%, while enabling sustained release in simulated intestinal fluid (pH 6.8), reaching about 70% within 10 h. Release kinetics indicated that CGA release was controlled by Fickian diffusion under acidic conditions and by a diffusion-polymer relaxation mechanism under intestinal conditions. Moreover, encapsulation in the SA/CMC hydrogel improved the thermal, light, and pH stability of CGA while maintaining its antioxidant activity and biocompatibility. These results indicate that SA/CMC bilayer hydrogels provide a promising strategy for stabilized gastrointestinal delivery of chlorogenic acid.

Effective vaccination hinges on overcoming persistent challenges in delivery site optimization, sustained antigen release, maintaining antigen integrity, and providing safe adjuvanticity. Here, we report a photothermal self-adjuvant porous microneedle (PSAPMN) patch as an all-in-one platform that comprehensively addresses these issues. Fabricated from polylactic-co-glycolic acid (PLGA) and polydopamine nanoparticles (PDA-NPs), the microneedles possess a unique interconnected porous architecture, enabling exceptionally mild, diffusion-based antigen loading. Upon brief near-infrared (NIR) irradiation, the photothermal effect of PDA-NPs not only accelerates pore self-healing for complete antigen encapsulation within 20 minutes but also creates a biodegradable depot for sustained intradermal release over 15 days. Crucially, the localized NIR-generated heat simultaneously serves as a potent physical adjuvant, promoting dendritic cell activation. In vivo, this integrated strategy elicits significantly more potent and durable antigen-specific immunity and immunological memory than conventional methods. This work presents a versatile platform for developing next-generation vaccines by synergistically solving multiple critical delivery challenges in a single device.
