Polylactic acid-glycolic acid (PLGA) porous microspheres have attracted significant scholarly interest as cell carriers, attributed to their exceptional biocompatibility and three-dimensional porous architecture. Nonetheless, the modulation of surface morphology and degradability remains a pivotal factor affecting cell adhesion and proliferation. In this study, PLGA (75:25) was synthesized through an anhydrous, oxygen-free ring-opening polymerization process, employing a lactic acid/glycolide molar ratio of 3:1. Subsequently, PLGA porous microspheres, with diameters ranging from 200 to 250 µm, were fabricated via the double emulsion technique. The microspheres were produced using 4% sodium bicarbonate as the pore-forming agent, with a stirring speed of 300 r/min and a 2% polyvinyl alcohol (PVA) concentration in the outer aqueous phase. The resultant microspheres demonstrated a narrow particle size distribution, uniform surface porosity, and a high pore-forming efficiency. In comparison to solid microspheres, the degradation rate of PLGA porous microspheres was markedly elevated, as evidenced by a decrease in pH from 7.28 to 7.17 over a 28 days period. Additionally, the surface morphology of the microspheres exhibited significant collapse. Furthermore, the viability of L929 cells cultured with PLGA microspheres surpassed 95%, indicating superior biocompatibility and significantly promoting cell adhesion and proliferation. This investigation effectively optimized the preparation process, resulting in PLGA microspheres characterized by uniform particle size and high porosity, thus offering an optimal scaffold material for biomedical applications.

Poly (lactic -co -glycolic acid) (PLGA) is a promising material for creating microrobots for drug delivery due to their biodegradability, biocompatibility, and FDA approval for clinical use. Nevertheless, traditional spherical PLGA drug carriers continue to face challenges, including limited motion efficiency and slow degradation. This study introduces rod-like Fe 3 O 4 @PLGA microrobots fabricated via a low-cost, scalable stretching method, to address these limitations. Their rod-like structures enhance their motion efficiency under rotating magnetic fields, outperforming their spherical counterparts by ≈58%. Thermogravimetric, Fourier transform infrared, and magnetization analyses confirm their excellent structural integrity and ample magnetic content of 39.8%. In vitro biocompatibility tests reveal no significant cytotoxicity toward L929 and HeLa cells. Their rod-like structures exhibit accelerated degradation, with swelling size 1.59 times higher than spheres by day 40, enabling rapid drug release with a 40.2% improvement. Fluorescence imaging validates uniform doxorubicin encapsulation and retention post-deformation. These results demonstrate that rod-like PLGA microrobots synergize precise magnetic navigation, high drug-loading capacity, and tunable degradation, offering a transformative platform for targeted therapies, particularly in acute conditions like localized cancer intervention.

Introduction: Guided bone regeneration (GBR) is widely used for maxillofacial defects, but fabricating membranes that enhance osteoinduction and antimicrobial resistance remains challenging. This study addresses critical bone defect therapy by developing collagen (Col) and zeolitic imidazolate framework-8 (ZIF-8) reinforced poly(lactide-co-glycolide) (PLGA) nanofibrous membranes. Methods: Material characterization analyzed the composite nanofibrous membranes’ morphology, structure, wettability, tensile strength, in vitro degradation, and ion release. Biocompatibility and osteogenesis were evaluated using live/dead staining, cytoskeleton staining, CCK-8 assay, alkaline phosphatase (ALP) activity quantification, ALP staining, alizarin red S (ARS) staining and ARS quantification. Antibacterial efficacy was assessed via agar plate counting and bacterial growth kinetics. In vivo bone regeneration was examined in a rat cranial critical defect model treated with the membrane; bone formation was evaluated by Micro-CT and hematoxylin-eosin staining after 4 weeks. Results: Optimization of the PLGA/Col weight ratio (100:3) yielded composite membranes demonstrating superior tensile strength. The PLGA/Col/ZIF-8 nanofibrous composite incorporating 1 wt% ZIF-8 nanoparticles exhibited optimal biocompatibility with sustained Zn2+ release kinetics. In vitro experiments demonstrated that sustained release of Zn2+ has the dual effects of stimulating osteogenic differentiation and effectively preventing early bacterial infection. In vivo a rat calvarial defect model further confirmed the positive bone regeneration effect of the PLGA/Col/ZIF-8 composite nanofibrous membrane.Discussion: PLGA/Col/ZIF-8 composite nanofibrous membranes have great potential for application in guiding bone tissue regeneration.

Oral squamous cell carcinoma (OSCC) is the most prevalent malignancy of the head and neck. Current treatment strategies, including surgery, radiotherapy, and chemotherapy, are limited by moderate efficacy and severe side effects, highlighting the need for novel therapeutic approaches. Wogonin (Wo), a natural flavonoid, has shown antiproliferative, pro-apoptotic, and antimetastatic activities in various cancer models; however, its effect on OSCC has not been investigated. In this study, Wo-loaded poly(lactic-co-glycolic acid) nanoparticles (PW NPs) were prepared to enhance water solubility and achieve sustained drug release. In vitro assays, including CCK-8, flow cytometry, live/dead staining, and wound healing assays, revealed that PW NPs significantly inhibited OSCC cell proliferation, induced apoptosis, and reduced cell migration. Network pharmacology analysis and qRT-PCR further suggested that Wo exerts its therapeutic effects through the EGFR signaling pathway. Hemolysis testing and histological examination of major organs confirmed the favorable biocompatibility of the nanoparticles. This study provides the first systematic evidence of the therapeutic potential of Wo in OSCC and lays a foundation for its further preclinical development.

The formation of a Protein Corona (PC) on the surface of nanoparticles (NPs) is a critical event that shapes their biological identity and governs interactions with the immune system. In this study, we investigated the composition of the PC formed on mixtures of PLGA and PEG–PLGA NPs, aiming to elucidate the link between NPsurface chemistry, proteomic fingerprint in cell culture medium, and uptake by bone marrow-derived macrophages (BMDMs). NPs showed different sizes but comparable actual PEG amount exposed on the surface, which is significantly lower than the theoretical values. The PC, isolated using a standardized microfiltration protocol, revealed distinct patterns of protein adsorption as a function of the PEG density. Uptake studies in BMDMs revealed a strong inverse relationship between PEG surface density, PC composition, and macrophage internalization, supporting the hypothesis that the opsonin/dysopsonin balance is more critical than a single protein interaction. In conclusion, this work demonstrates that the PEG surface density is not the only determinant of PC composition. These findings underscore the importance of rigorous surface characterization and PC profiling to predict and tune nanocarrier performance in vivo.

Drug (Fenbufen, FBF)-loaded poly( d , l -lactide-co-glycolide) (PLGA) and PLGA/gelatin nanofibrous scaffolds were fabricated via electrospinning technique. The influences of gelatin content, fiber arrangement, crosslinking time and pH value of the buffer solution on FBF release behavior of the resulting nanofibrous scaffolds were investigated, with the corresponding FBF-loaded PLGA and PLGA/gelatin solvent-cast films as controls. The release rate of FBF was found to be increased with the increment of gelatin content for all the composite samples, and the FBF release rate of aligned nanofibrous scaffold was lower than that of randomly oriented scaffold. Moreover, the crosslinking treatment depressed effectively the burst release of FBF at initial release stage of PLGA/gelatin (9/1) nanofibrous scaffold. In addition, the pH value of the buffer solution could change the physical state of the polymer and affect the FBF release rate.

Graphene oxide (GO)/poly (lactide-co-glycolic acid) (PLGA) scaffolds have promising applications in the biomedical field. However, greater attention is focused on the incorporated system and its applications in normal cells. In this work, a novel GO immobilized PLGA nanofibrous scaffold assisted by polydopamine (PLGA-PDA-GO) is developed for growth inhibition of HT-29 colon cancer cells. The interactions between GO and PDA are attributed to a π–π conjugate interaction and electrostatic attraction. In addition to the enhancement of thermal stability and mechanical strength, the surface roughness, hydrophilicity, and electro-activity of the scaffolds are significantly improved by immobilization of GO. The scaffolds show good inhibition of HT-29, and immobilized GO is observed to be in contact with but not internalized in HT-29 cells. The cytotoxicity mechanism of scaffolds toward HT-29 is attributed to intracellular activated reactive oxygen species that result from the physical interaction of the sharp GO edges and electrical signals of π–π stacking between PDA and GO.

Polymer functionalized nanoparticles (NPs) have a great attention in biomedical applications owing to their unique properties like regenerative antioxidant, anti-inflammatory, auto-catalytic properties, and biocompatibility. In this current work, we demonstrated a facile synthesis of Se-CeO 2 via chemical method followed by precipitation method. The prepared Se NPs were characterized by ultraviolet–visible (UV-vis) spectroscopy, and the size and morphology of the NPs were analysed using transmission electron microscopy (TEM). Meanwhile, Se-CeO 2 NPs loaded on polylactic-co-glycolic acid (PLGA) nanocarrier were characterised by Fourier transform infrared (FT-IR), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), TEM, and energy dispersive X-ray analysis (EDAX) mapping techniques. The morphological and spectroscopic investigations of prepared nanomaterials have exhibited favourable morphological structure and chemical interactions with respective polymeric molecules, which established that nanovesicle suitability for the SCI functional recovery. We first investigated Se nanoformulated CeO 2 material for the potential healing of in vitro spinal cord study. Our results demonstrated that preparation of NPs loaded PLGA nanocarrier has provide effective spinal cord regeneration and imply that it was explored that promising nanocarrier in the SCI treatment. Se-NPs encapsulated CeO 2 nanostructures administrations for SCI therapies have greatly suppressed oxidative stress and induced anti-inflammatory action, which leads to prospective therapeutic benefits of spinal cord regeneration. These investigative results demonstrate that Se-CeO 2 NPs with PLGA carrier could have great attention for effecient functional recovery treatment and care for spinal cord injury.

Microwave (MW) hyperthermia has been widely studied in tumor therapy, while the lack of specificity, and the potential toxicity induced by instability or difficulty in degradation of existed MW thermal sensitizers still limits the application. Herein, a new biocompatible Poly(lactic-co-glycolic
acid) (PLGA)-based nanosensitizer of Dtxl-Gd@PLGA-PEG-TPP (DGPPT) with capacities of magnetic resonance (MR) imaging and mitochondrial targeting for MW hyperthermia combined with chemotherapy was constructed via a double emulsion solvent evaporation method. The modified TPP significantly enhanced
the specificity of sensitizer for targeting mitochondria, a heat-sensitive energy supply plant in cells. Thus the MW thermal damage induced by the loaded Gd in PLGA nanospheres was also strengthened. Together, the system could also achieve MR imaging due to the existence of Gd. In addition,
the encapsulated Dtxl performed the chemotherapy of inhibiting mitochondrial function for assisting with MW hyperthermia. In vivo experiments demonstrated that PLGA had high biocompatibility that no obvious damage occurred even the dose was up to 200 mg/kg. Meanwhile, DGPPT+MW representing
the combination of mitochondrial targeting and MW hyperthermia-chemotherapy has also been proved to shrink tumor size effectively. This study provides a new direction for building biosafe and multifunctional MW sensitizer with active targeting ability to impede tumor growth.

Background Transcatheter arterial chemoembolization (TACE) is extensively used in the treatment of advanced hepatocellular carcinoma (HCC). However, the efficacy of TACE is usually limited to secondary tumor hypoxia and hypoxia-related tumor angiogenesis. Methods In this study, poly(lactic-co-glycolic acid) (PLGA) microspheres (SOR-CAT-PLGA MSs) encapsulating sorafenib (SOR) and catalase (CAT) were prepared by double-emulsion solvent diffusion method. Sorafenib inhibits tumor angiogenesis, and catalase decomposes hydrogen peroxide (H 2 O 2 ) to generate oxygen in the tumor. Results In vitro and in vivo , SOR -CAT-PLGA MSs could significantly improve the efficacy of hepatic artery embolization in the treatment of rabbit VX2 liver tumors, regulate tumor hypoxia and immunosuppressive microenvironment, then achieved near-complete and rapid necrosis of liver tumors. Conclusions The application of new SOR -CAT-PLGA MSs in hepatic artery chemoembolization of rabbit VX2 liver tumor is a promising approach to improve the therapeutic effect of liver tumors and has a broad clinical application prospect.

Summary Background/Objective A novel porous scaffold poly (lactide- co -glycolide) and tricalcium phosphate (PLGA/TCP) was developed by three-dimensional printing technology for bone defect repair. As a Class 2 solvent with less severe toxicity, content of residual 1,4-dioxane in this newly developed scaffold should be rigorously controlled when it is translated to clinical use. In this study, a headspace gas chromatography-mass spectrometric (HS-GC-MS) method and related testing protocol were developed for quantitative determination of 1,4-dioxane in the PLGA/TCP composite scaffolds. Methods Matrix effect analysis was used to optimise the pretreatment method of the scaffolds. Then, the procedure for testing 1,4-dioxane using HS-GC-MS was set up. The accuracy, precision, and robustness of this newly developed quantitative method were also validated before quantification of 1,4-dioxane in the scaffolds with different drying procedures. Results Dimethyl formamide (DMF) was the optimal solvent for dissolving scaffolds for GC-MS with proper sensitivity and without matrix effect. Then, the optimised procedure was determined as: the scaffolds were dissolved in DMF and kept at 90°C for 40 minutes, separated on a HP-5MS column, and detected by mass spectroscopy. Recovery experiments gave 97.9–100.7% recovery for 1,4-dioxane. The linear range for 1,4-dioxane was determined as 1–40 ppm with linear correlation coefficient ≥ 0.9999. Intraday and interday precision was determined as being within relative standard deviation of below 0.68%. The passable drying procedure was related to lyophilising (−50°C, 50 Pa) the scaffolds for 2 days and drying in vacuum (50 Pa) for 7 days. Conclusion This is the first quantitative method established to test 1,4-dixoane in a novel scaffold. This method was validated with good accuracy and reproducibility, and met the methodological requirements of the Guideline 9101 documented in the Chinese Pharmacopoeia 2015 Edition. The translational potential of this article This quantitative method for determination of residual 1,4-dioxane in the novel scaffolds is a key technical method during its translation into clinical use because this method is an important and indispensable file in the enterprise standard when the porous scaffold is registered as a Class III implanted medical device for bone defect repair, which is used to guarantee the safety of the scaffolds. It is also applied to optimise the drying process of scaffolds and to monitor the quality of scaffolds in the industrialisation process. Further, this method provides references for other solvents quantitative determination in porous scaffolds or materials.
