Bio-based crosslinkers: Oxidized polysaccharides for improving the colorfastness of natural polyphenol dyes-colored silk fabrics

Shengnan Guo,Meng An,Yidan Liu,Mingbo Ma,Wenlong Zhou

Natural polyphenol dyes are traditionally and widely used for the coloration of natural fibers, e.g. silk. However, the poor affinity between polyphenol dyes and natural fibers often leads to low colorfastness. To address this issue, we developed a green fixation strategy using periodate-oxidized polysaccharides (OPS) as bio-based crosslinkers. Four structurally diverse polysaccharides—pullulan, pectin, starch, and hyaluronic acid—were selectively oxidized to generate reactive aldehyde groups capable of covalently binding with silk fibers and tannins. The successful oxidation and subsequent network formation between tannins, polysaccharides, and silk proteins were verified by FTIR NMR, and XPS analyses. Compared with untreated silk, OPS pretreatment significantly improved dye uptake and colorfastness of dyed silk. Among them, oxidized pullulan (OPullu) exhibited the best performance: achieving a shade depth of K/S = 10.2 and colorfastness grades of 3–4/4 under optimized conditions (pretreatment: pH 6, 80 °C, 6 % OPullu; dyeing: pH 2, 80 °C). Most significantly, OPullu exhibited no cytotoxicity to skin cells, supporting its safe use in textiles. Moreover, the approach was effective for other polyphenolic dyes, including anthocyanins, quercetin, and tea pigment. OPS pretreatment offers a simple, eco-friendly, and versatile approach for improving the colorfastness of silk fabrics dyed with natural polyphenolic colorants.

Preparation, characterization, in vitro digestion, and anti-ulcerative colitis activity evaluation of Pithecellobium clypearia polyphenol nanoliposomes

Chuanju Wang,Qiang Peng,Qianhua Zhang,Shihua Han,Lingyong Zeng,Jiarong Su,Guixin Liang,Zihao Chen,Juan Guo,Yongguang Bi,YuShi Gong,Shaofan Tan,Huaicheng Qin

Pithecellobium clypearia Benth polyphenols (PCBPL) possess potent antioxidant and anti-inflammatory properties, making them promising agents for ulcerative colitis (UC) therapy. However, their clinical translation is hindered by poor water solubility, low stability, and limited bioavailability. To overcome these limitations, Pithecellobium clypearia Benth polyphenols liposomes (PCBPL-Lips) were prepared via thin-film hydration. The resulting PCBPL-Lips exhibited a high encapsulation efficiency of 89.31 ± 1.09%, an average particle size of 124.51 ± 7.61 nm, and a zeta potential of −31.62 ± 2.37 mV. Transmission electron microscopy confirmed their uniform particle distribution, spherical morphology, and smooth surfaces. Stability evaluations showed that PCBPL-Lips remained stable under various pH, temperature, and ionic strength conditions. In vitro digestion studies revealed a sustained-release profile, suggesting potential for prolonged therapeutic action. Biocompatibility assessments showed a hemolysis rate below 5% and cell viability above 80%, indicating excellent safety. In vitro activity assays demonstrated that liposomal encapsulation preserved the antioxidant activity of PCBPL and significantly enhanced its anti-inflammatory efficacy. In a murine model of UC, PCBPL-Lips alleviated clinical symptoms, reduced body weight loss, increased colon length, decreased spleen index and tumor necrosis factor-alpha (TNF-α) levels, and improved colonic tissue architecture. This study demonstrates that liposomal encapsulation markedly enhances the stability, bioavailability, and therapeutic efficacy of PCBPL, providing a promising strategy for its use in the treatment of inflammatory bowel diseases.

Study on the flavor formation of fermented sweet tea and its relation with the polyphenol oxidase

Yong-Kang Wang,Yue-Hua Cen,Jia-Li Li,Cun-Yu Li,Shan-Shan Wu,Xiao-Xiang Li,Xin-Qiang Zheng,Jian-Hui Ye,Yue-Rong Liang,Jian-Liang Lu

Fermentation process can significantly improve the flavor of sweet tea. • Flavor improvement results from key chemical fluctuation related to taste and aroma. • Flavor formation is modulated by fermentation-triggered gene differential expression. • Differentially expressed LlPPO plays a key role for forming flavor in fermentation. Sweet tea, produced from the tender shoots of Lithocarpus litseifolius using the green tea process, usually exhibits an unacceptable flavor. In this study, the effect of fermentation on the sweet tea flavor and its mechanism had been comprehensively investigated. Results showed that fermentation could effectively reduce the sweetness of sweet tea by significantly lowering the level of trilobatin, and collectively weaken the bitterness and astringency by downregulating the contents of trilobatin and (+)-catechin. Fermentation could greatly develop the sweet fragrance through significantly altering the level and proportion of volatiles with different odors, leading to high flavor acceptability of sweet tea. Chemical fluctuations were closely associated with differential expression of the genes and the corresponding enzymatic reactions triggered by fermentation and its leading step rolling. The differential oxidation of trilobatin and phlorizin during fermentation was verified by catalytic characteristics of prokaryotically expressed polyphenol oxidase (LlPPO). The findings elucidated the potential mechanism underlying the quality improvement of the fermented sweet tea based on metabolite profiling, gene expression and catalytic property of key enzyme, which laid a solid foundation for developing high quality sweet tea. Antimicrobial/Anti-Inflammatory Peptidomimetic-Polyphenol Co-Assemblies with pH-Triggered Activation for S. aureus Abscess Therapy

Junfeng Song,Anming Yang,Yang Song,Xirui Huang,Xingjin Li,Tianzhao Xu,Tiancong Zhao

The treatment of Staphylococcus aureus subcutaneous abscesses is hindered by the protective abscess microenvironment and persistent bacterial colonization. Herein, we present a pH-responsive supramolecular nanoplatform fabricated via single-step coassembly of cationic arginine-derived antimicrobial peptidomimetics (AMPM-1) and natural polyphenols. This process, governed by hydrogen bonding, electrostatic interactions, and hydrophobic packing, effectively masks the cationic charges of AMPM-1, substantially reducing intrinsic hemolytic and cytotoxic properties. In vitro studies demonstrate that the assemblies maintain potent, membrane-disruptive activity against methicillin-resistant S. aureus (MRSA), including dormant and persister subpopulations. In a murine MRSA abscess model, local administration of the quercetin-based assemblies (AMPM-1-QA) simultaneously reduces bacterial burden and attenuates excessive inflammatory signaling. Histological examination confirms accelerated tissue regeneration, characterized by enhanced collagen deposition and anti-inflammatory macrophage polarization. By integrating acid-triggered bactericidal action with polyphenol-mediated immunomodulation, this dual-functional supramolecular strategy offers a promising approach for the stimuli-responsive treatment of recalcitrant localized infections.

Aggregation and conformational change of mushroom (Agaricus bisporus) polyphenol oxidase subjected to atmospheric cold plasma treatment

Yifan Zhu,Mubango Elliot,Yanhong Zheng,Jing Chen,Dongzhi Chen,Shanggui Deng

Atmospheric cold plasma (ACP) is a novel nonthermal technology with potential applications in maintaining and improving food quality. The effect of ACP on the activity and structure of mushroom ( Agaricus bisporus ) polyphenol oxidase (PPO) was evaluated. Results demonstrated that the dielectric barrier discharge (DBD) based plasma technology could inactivate PPO (up to 69%) at 50 kV with the increased concentrations of H 2 O 2 and NO x . An obvious enhancement of surface hydrophobicity was observed, whereas a gradual reduction of total sulfhydryl content was recorded with the increasing exposure time. Data from circular dichroism, atomic force microscopy, particle size distribution and fluorescence spectra displayed the rearrangement of secondary structure and disruption of the tertiary structure. Red shifts of fluorescence spectra showed positive correlations with the inactivation rate of PPO. Therefore, ACP treatment could be served as an alternative approach to inactivate undesirable enzymes to minimize the loss of food nutrition and quality.

Enhanced stability and antioxidant activity of pickering emulsions stabilized by chayote tuber starch-tea polyphenol composites

Xiaoxue Wang,Jia Huang,Xingyuan Zhu,Qing Guo,Qiang Cui,Jie Yu,Xiaoyan Hou,Hong Liu,Guanghui Shen,Zhongqun He,Zhiqing Zhang

Starch has emerged as a promising Pickering emulsion stabilizer due to its low cost, renewability, and biocompatibility; however, its poor interfacial activity and lack of functional properties have limited its broader applications. In the present work, composites of chayote tuber starch (CTS) and tea polyphenols (TP) were prepared via a co-gelatinization method. The effects of varying TP concentrations (0–20 %) on the physicochemical, structural, and emulsifying properties of the CTS/TP composites as Pickering emulsion stabilizers were systematically investigated. FT-IR and XRD analyses revealed that hydrogen-bonding interactions between TP and CTS disrupted both short-range and long-range molecular order, resulting in a looser and more porous structure. The addition of TP increased particle size while decreasing the ζ-potential, gelatinization enthalpy, and viscosity of the composites. CTS/TP10 composites exhibited optimal interfacial properties, as the interfacial tension decreased from 64.48 to 33.53 mN/m and the contact angle increased from 60.27° to 93.55°. Compared with emulsions stabilized by pure CTS particles, emulsions stabilized by CTS/TP10 composites exhibited significantly superior physical stability. Moreover, TP, as a potent antioxidant, inhibited free radical-induced damage, thereby suppressing lipid oxidation within the emulsion system. Specifically, when the TP concentration in the CTS/TP composites was 10 %, the peroxide value (POV) and thiobarbituric acid reactive substances (TBARS) decreased by 29.95 % and 84.94 %, respectively, after 15 days of accelerated oxidation testing. In conclusion, CTS/TP composites exhibited excellent interfacial properties and antioxidant activity, highlighting their potential as multifunctional emulsifiers capable of inhibiting oil oxidation.

Shape-adaptive multifunctional hydrogel based on in situ crosslinking of natural polyphenol-gelatin for wound treatment

Qin Yang,Yuchun Liu,Ya Tian,Peng Guo,Pengkun Lei,Siwei Xiong,Jun Zhao,Bing Chen,Chen Zhang,Yan Qu

Traditional wound dressings have problems such as a lack of bioactive substances, poor shape adaptability and secondary damage, which make it difficult to meet clinical needs. Therefore, this study developed a new type of photocurable hydrogel preparation (Ferulic acid-modified gelatin, GelFA) to promote wound healing. GelFA can be solidified in situ under ultraviolet light to form a hydrogel. Inspired by the compatibility of classical drug pairs, it is loaded with Bletilla striata polysaccharide (BSP) and notoginsenoside R1 (R1), which improves the moisturizing performance of GelFA but also synergistically promotes wound repair. GelFA/BSP-R1 hydrogel has excellent environmental adaptability, can prevent wound infection. At the same time, the hydrogel has anti-inflammatory, antioxidant, hemostatic and other functional properties. The results of cell experiments showed that GelFA/BSP-R1 hydrogel had good biocompatibility and significantly promoted angiogenesis, which laid a solid foundation for tissue repair. In wound models of rats and Bama pigs, it accelerates tissue repair by reducing inflammation, promoting collagen deposition, and accelerating angiogenesis. Inspired by the above research results, based on the new photocuring material GelFA, this study further developed various dosage forms to expand its clinical application. In summary, this study provides a promising new treatment strategy for clinical tissue regeneration.

Hollow Conductive Polymer Nanospheres with Metal–Polyphenol Interfaces for Tunable Hydrogen Peroxide Activation and Energy Conversion

Ruolan Du,Shuyan Liu,Yuanzhe Li

Hydrogen peroxide (H2O2) is a key oxidant for green chemical processes, yet its catalytic utilization and activation efficiency remain limited by material instability and uncontrolled radical release. Here, we report a dual-functional, hollow conductive polymer nanostructure that enables selective modulation of H2O2reactivity through interfacial physicochemical design. Hollow polypyrrole nanospheres functionalized with carboxyl groups (PPy@PyCOOH) were synthesized via a one-step Fe2+/H2O2oxidative copolymerization route, in which H2O2simultaneously served as oxidant, template, and reactant. The resulting structure exhibits enhanced hydrophilicity, rapid redox degradability (>80% optical loss in 60 min (82.5 ± 4.1%, 95% CI: 82.5 ± 10.2%), 10 mM H2O2, pH 6.5), and strong electronic coupling to reactive oxygen intermediates. Subsequent tannic acid–copper (TA–Cu) coordination produced a conformal metal–polyphenol network that introduces a controllable Fenton-like catalytic interface, achieving a 50% increase in ROS yield (1.52 ± 0.08-fold vs. control, 95% CI: 1.52 ± 0.20-fold) while maintaining stable photothermal conversion under repeated NIR cycles. Mechanistic analysis reveals that interfacial TA–Cu complexes regulate charge delocalization and proton–electron transfer at the polymer–solution boundary, balancing redox catalysis with energy dissipation. This work establishes a sustainable platform for H2O2-driven redox and photo-thermal coupling, integrating conductive polymer chemistry with eco-friendly catalytic pathways.

Bio-inspired natural polyphenol cross-linking poly(vinyl alcohol) films with strong integrated strength and toughness

Yu Guan,Leishan Shao,Dongyu Dong,Fei Wang,Yuliang Zhang,Yinghan Wang

Polymer composites exhibit very high strength but this is usually found to be at the expense of toughness, which greatly limits their application in many fields. Inspired by the hierarchical architectures in spider silk, we attempted to combine simultaneous high strength and toughness via a hydrogen bonding network. PVA composites with tannic acid (TA) molecules were fabricated by a green and easy-to-scale-up water casting method. The results showed that the hydrogen bonding crosslinking between PVA and TA has strong effect on the Tg, crystallinity, thermal stability and mechanical properties of PVA. In addition, the PVA composite films with 3 wt% TA exhibited, simultaneously, the best tensile strength and toughness, improving by 46% and 27% compared to the pure PVA, respectively.

Characterisation of polyphenol constituents of Linderae aggregate leaves using HPLC fingerprint analysis and their antioxidant activities

Chanjuan Xu,Bingxian Yang,Wei Zhu,Ximin Li,Jingkui Tian,Lin Zhang

The leaves of Linderae aggregate (LAL) has been used as a type of tea in China and other Southeast Asian countries. In this study, 11 polyphenols in LAL were clarified for the first time using multiple high-performance liquid chromatographic techniques. An optimal extracting method was developed through the comparison of the amount of quercetin-3- O -α- l -rhamnoside using a uniform design method. From the fingerprint liquid chromatography data, 11 common peaks in the 8 samples collected from April to November were semi-determined. The antioxidant capacities were examined using the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging assay and the ferric reducing/antioxidant power assay. All 8 samples contained the same 11 polyphenols in similar ratios. Three samples, S2, S5 and S6 contained higher amount of quercetin-3- O -α- l -rhamnoside and were demonstrated to have stronger antioxidant capacities in both antioxidant assays. These results are critical in optimising harvest time and quality control of LAL.

Injectable sodium alginate hydrogel loaded with plant polyphenol-functionalized silver nanoparticles for bacteria-infected wound healing

Qinsheng Hu,Yong Nie,Jun Xiang,Jinwei Xie,Haibo Si,Donghai Li,Shaoyun Zhang,Mei Li,Shishu Huang

A novel injectable hydrogel dressing (GA@AgNPs-SA) with long-term antimicrobial effect is developed that can accelerate the closure of bacteria-infected wounds. The hydrogel dressing was prepared by cross-linking sodium alginate molecular chains and gallic acid functionalized silver nanoparticles (GA@AgNPs) via calcium ions to form a three-dimensional network. The hydrogel dressing demonstrates excellent biocompatibility and can achieve a sustainable release of silver ions, ensuring a long-term antibacterial activity and inhibiting biofilm formation. Moreover, an in vivo study demonstrates that the GA@AgNPs-SA hydrogel can effectively decrease the expression of IL-6 and TNF-α to alleviate the inflammatory response, and promote angiogenesis by upregulating CD31, α-SMA and VEGF expression, thus significantly accelerating the repair of infected wounds. Given these interesting properties, this antibacterial hydrogel has great potential for application in the clinical care of bacteria-infected wounds.

Epigallocatechin-3-gallate (EGCG) based metal-polyphenol nanoformulations alleviates chondrocytes inflammation by modulating synovial macrophages polarization

Hong Wei,Jun Qin,Quanxin Huang,Zhiqiang Jin,Li Zheng,Jinmin Zhao,Zainen Qin

The activation of M1-type macrophages are dominant cells secreting proinflammatory present within the inflamed synovium in the progression of osteoarthritis (OA). Increased oxidative stress, such as redundant ROS and hydrogen peroxide (H 2 O 2 ), are important factors in driving macrophages to polarize into M1 type. In this study, metal-polyphenol nanoformulations (Cu-Epigallocatechin-3-gallate (Cu-EGCG) nanosheets) were synthesized through the coordination interaction between EGCG and copper ions, which possessed the antioxidant effect of EGCG and anti-inflammatory of Cu 2+ . Results showed that Cu-EGCG nanosheets were biocompatible and the Cu 2+ could be sustained released from the nanoparticles. Cu-EGCG nanosheets with multienzyme-like antioxidative activity could effectively scavenge the excessive intracellular ROS, leading to significantly decreased expression of the pro-inflammatory cytokines, which could reduce the expression of M1-type macrophages and exhibit excellent promotion on shifting macrophages to M2 phenotypes. Moreover, the secreted factor from the cell supernatant of Cu-EGCG treated macrophages exhibited anti-inflammatory potential in chondrocytes of inflamed synovial joints. This study suggests a novel strategy for OA therapy by using metal-polyphenol nanoformulations targeting macrophages.
