The rapid emergence of difficult-to-treat multidrug-resistant pathogens, combined with the scarcity of antibiotics possessing novel mechanisms, poses a significant threat to global public health. Here, we integrate the synthetic-bioinformatic natural product approach with peptide optimization to unveil the antibiotic-producing potential of Paenibacillaceae bacteria. Our culture-independent approach led to the discovery of paenimicin, a novel 11-mer depsi -lipopeptide featuring an unprecedented dual-binding mechanism. By sequestering the phosphate and hydroxyl groups of lipid A in Gram-negative bacteria, as well as the phosphate groups of teichoic acids in Gram-positive bacteria, paenimicin exhibits potent and broad-spectrum efficacy against MDR pathogens in vitro and in vivo models. Paenimicin demonstrates no detectable resistance, favorable pharmacokinetics and low nephrotoxicity, positioning it as a promising candidate for treating severe and urgent MDR infections.

Biofilm infections present escalating clinical challenges due to their intrinsic resistance to conventional treatments, underscoring the urgent need for innovative therapeutic strategies. Here, we show a pioneering approach that leverages sacrificial nitric oxide (NO) to enhance hydrogen peroxide (H 2 O 2 ) retention for antibiofilm therapy. H 2 O 2 has the ability to produce hydroxyl radicals (•OH), the most potent oxidants in nature, but it is often neutralized by H 2 O 2 scavengers before •OH formation. Our design utilizes ultrasound irradiation to release NO from the biofilm-anchoring nano-xanthiums (G-B@NXs), which subsequently interact with biofilm-associated H 2 O 2 scavengers, including catalase and glutathione, to suppress the decomposition ability toward H 2 O 2 . Additionally, glucose oxidase on the surface of G-B@NXs supplements H 2 O 2 production, facilitates its conversion into •OH, and exhibits effective biofilm sterilization in different infection models. Our strategy not only shows the promise for advancing antibiofilm applications but also provides novel insights into the development of NO-assisted biomaterials.

Radiotherapy (RT), essential for treating various cancers, faces challenges from tumor hypoxia, which induces radioresistance. A tumor-targeted “prosthetic-Arginine” coassembled nanozyme system, engineered to catalytically generate nitric oxide (NO) and oxygen (O 2 ) in the tumor microenvironment (TME), overcoming hypoxia and enhancing radiosensitivity is presented. This system integrates the prosthetic heme of nitric oxide synthase (NOS) and catalase (CAT) with NO-donating Fmoc-protected Arginine and Ru 3+ ions, creating HRRu nanozymes that merge NOS and CAT functionalities. Surface modification with human heavy chain ferritin (HFn) improves the targeting ability of nanozymes (HRRu-HFn) to tumor tissues. In the TME, strategic arginine incorporation within the nanozyme allows autonomous O 2 and NO release, triggered by endogenous hydrogen peroxide, elevating NO and O 2 levels to normalize vasculature and improve blood perfusion, thus mitigating hypoxia. Employing the intrinsic O 2 -transporting ability of heme, HRRu-HFn nanozymes also deliver O 2 directly to the tumor site. Utilizing esophageal squamous cell carcinoma as a tumor model, the studies reveal that the synergistic functions of NO and O 2 production, alongside targeted delivery, enable the HRRu-HFn nanozymes to combat tumor hypoxia and potentiate radiotherapy. This HRRu-HFn nanozyme based approach holds the potential to reduce the radiation dose required and minimize side effects associated with conventional radiotherapy.

Tea polysaccharides have attracted widespread attention in regulating gut microbiota. A highly efficient enzymolysis and ultrasonic-assisted extraction method was developed and optimized for the extraction of polysaccharides from Zhonghuang No. 1 etiolated-green tea (GTP) with a-single factor and response surface methodology, i.e, with a pH of 5.5, an enzyme concentration of 7%, an ultrasonic time of 2.5 h, a solid-liquid ratio of 1:20 g/mL, and an ultrasonic temperature of 80°C. GTP exhibited a triple helix structure and demonstrated the ability to bind with Congo red. Its surface was observed to be spherical with slender chain fragments. GTP remained intact throughout the simulated in vitro digestive process, which encompassed the oral, gastric, and small intestinal phases. The relative abundance of Bacteroidota was significantly increased, while the Firmicutes/Bacteroidota ratio was reduced following the administration of GTP. The contents of Faecalibacterium and Bacteroides were increased, while the contents of Escherichia_Shigella and Dorea were reduced after GTP treatment. Furthermore, GTP were degraded and utilized by gut microbiota to produce SCFAs and reducing pH value. GTP regulated the composition of the gut microbiota and has the potential to act as a prebiotic, with the possibility of its use as a functional food.

Hexagonal boron nitride (hBN) was used as a CuOX/TiO2 catalyst carrier and its effect on NO reduction with NH3 was studied. After hBN was treated with concentrated HNO3, CuOX/TiO2 nanoparticles were dispersed well onto hBN, and the addition of hBN was found to promote NO oxidation, while at the same time suppress NH3 oxidation to NO, and thus promoted the selective catalytic reduction of NO at reaction temperatures between 150 to 350 °C, and a high de-NOX efficiency of 90.6% was achieved at 275 °C. Our study indicates that hBN is a promising catalyst promoter and carrier with excellent stability compared to carbonaceous materials.

Diabetic wounds represent a common and challenging complication of diabetes mellitus, characterized by a complex pathological microenvironment that includes excessive reactive oxygen species (ROS), chronic inflammation, hypoxia, and impaired nitric oxide (NO) synthesis. These factors collectively contribute to delayed wound healing, increased infection risk, and potential progression to chronic non-healing ulcers. Herein, we constructed a multifunctional injectable hydrogel (Prussian blue nanozyme and S-nitrosothiol-loaded hydrogel, PBE&SNO@HG) co-loaded with multi-enzyme-mimicking Prussian blue nanozymes (PBE) and an ultrasound-responsive nitric oxide (NO) donor (SNO). This composite hydrogel exhibited favorable injectability and tissue adhesiveness. The incorporated PBE nanozymes simultaneously mimicked superoxide dismutase (SOD) and catalase (CAT), enabling the scavenging of superoxide anions (⋅O 2 - ) and hydrogen peroxide (H 2 O 2 ), thereby reducing oxidative stress levels at the wound site. Additionally, the ultrasound (US)-responsive release of NO promoted angiogenesis and alleviates inflammatory responses in the wound area. Animal experiments demonstrated that this hydrogel system significantly enhances collagen deposition, accelerates re-epithelialization, promotes hair follicle regeneration, and accelerates wound healing. This study demonstrated the synergistic effect of combining antioxidant strategies with gas therapy in diabetic wound treatment, while also indicating that US-responsive hydrogel materials can achieve on-demand release of therapeutic molecules via external field stimulation, thereby providing a novel strategy for chronic wound management.

Rationale: Chronic diabetic wounds present significant therapeutic challenges due to biofilm resistance and dysregulated metabolism of glucose, reactive oxygen species (ROS), and nitric oxide (NO), which collectively exacerbate immunosuppression and impair tissue repair. Methods: This study developed a glucose-driven cascade nanozyme-loaded dissolvable microneedle system (PPLG@MN) that enables spatiotemporal regulation of the wound microenvironment through a closed-loop mechanism involving nutrient deprivation, gas modulation, and immune reprogramming. The system consists of porous Prussian blue nanozymes (PPB) loaded with glucose oxidase (GOx) and L-arginine (L-arg), which is precisely delivered into the tissue via microneedles. Results: PPLG@MN initiates a self-sustaining therapeutic cycle within the biofilm microenvironment, in which GOx catalyzes the oxidation of glucose to generate hydrogen peroxide (H 2 O 2 ), inducing bacterial starvation. Subsequently, H 2 O 2 reacts with PPB and L-arg to release oxygen (O 2 ) and biofilm-disrupting NO, thereby alleviating local hypoxia and enhancing antibacterial efficacy. Furthermore, the synergistic action of O 2 and NO reprograms macrophages toward an anti-inflammatory M2 phenotype (approximately 30-fold increase compared to the model group), effectively resolving inflammation and promoting angiogenesis. In vivo studies confirmed that the system achieved > 99.9% biofilm eradication efficiency and accelerated wound healing by 25.6% compared to the model group. Conclusion: This nanoplatform offers a clinically translatable therapeutic strategy for biofilm-associated diabetic wounds by synergistically combining gas therapy and immune reprogramming.

Considering that nitrogen oxides (NO x ) and carbon monoxide (CO) usually meantime exist in industrial flue gas, it is meaningful to develop a collaborative control technology for the simultaneous purification of NO x and CO. In this work, the catalysts xCu-1V-5W/Ti were evaluated in the simultaneously selective catalytic reduction by NH 3 (NH 3 -SCR) and CO oxidation . The introduction of CuO provided the active sites for the oxidation of CO, while inhibited the NH 3 -SCR process and facilitated the generation of N 2 O by-product at high temperature. Accordingly, a catalyst coupling system was designed by employing 1V-5W/Ti and 2Cu-1V-5W/Ti catalysts in one reactor, and high NO conversion (>90 %) and CO removal efficiency (>80 %) could be simultaneously realized within 350–500 °C. This work would provide valuable insights for achieving simultaneous removal of NO x and CO using the catalyst coupling method.

Currently, undeveloped diagnosis and delayed treatment of bacteria-infected sites in vivo not only expand the risk of tissue infection but are also a major clinical cause of multiple drug-resistant bacterial infections. Here, an efficient nanoplatform for near-infrared (NIR)-light-controlled release and bacteria-targeted delivery of nitric oxide (NO) combined with photothermal therapy (PTT) is presented. Using maltotriose-decorated mesoporous polydopamine (MPDA-Mal) and BNN6, a smart antibacterial (B@MPDA-Mal) is developed to combine bacterial targeting, gas-controlled release, and PTT. Utilizing bacteria's unique maltodextrin transport system, B@MPDA-Mal can accurately distinguish bacterial infection from sterile inflammation and target the bacteria-infected sites for efficient drug enrichment. Moreover, NIR-light causes MPDA to generate heat, which not only effectively induces BNN6 to produce NO, but also raises the temperature to harm the bacteria further. NO/photothermal combination therapy effectively eliminates biofilm and drug-resistant bacteria. The myositis model of methicillin-resistant Staphylococcus aureus infection is established and indicates that B@MPDA-Mal can successfully eradicate inflammation and abscesses in mice. Meanwhile, magnetic resonance imaging technology is used to monitor the treatment procedure and healing outcomes. Given the aforementioned advantages, the smart antibacterial nanoplatform B@MPDA-Mal can be used as a potential therapeutic tool in the biomedical field against drug-resistant bacterial infections.

Nanocatalysis plays an important role in chemical industrial production, environmental protection and etc. However, traditional catalyst preparation methods are generally labor-intensive, time-consuming and need high-temperature calcination, which are not conducive to the preparation of nanostructured catalyst. Therefore, developing a green and facile method for nano-sized catalyst preparation is very significative. In this work, a series of nano-sized Co 3 O 4 /CeO 2 catalysts with high specific surface area, high oxygen storage and homogeneous distribution of active species were synthesized via a glucose-urea (G-U method), and applied in NO oxidation The NO oxidation performance indicates that the 12.5% CoCe G−U catalyst prepared by the G-U method shows higher activity than those of catalysts prepared by the traditional methods (precipitation, hydrothermal and impregnation methods). The characterization results revealed that the CoCe G−U catalysts have the highest specific surface area, pore volume, content of surface Ce 3+ and Co 3+ among the catalysts. Moreover, the nano Co 3 O 4 is better dispersed on CeO 2 , which resulted in more active oxygen species on the surface of the catalysts prepared by the G-U method. Herein, the higher specific surface area, the smaller Co 3 O 4 nanoparticles, the more active oxygen species and the higher Ce 3+ and Co 3+ contents of the CoCe G−U catalyst lead to the more excellent NO oxidation performance. This work provides a green and efficient approach for the industrial production of nano-sized oxide catalysts.

Photocatalysis is an effective technology for NO removal even at low concentrations in the ambient atmosphere. However, the low efficiency of this advanced process and the tendency of producing toxic byproducts hinder the practical application of photocatalysis. To overcome these problems, the Bi@Ba2TiO4/BaBi4Ti4O15photocatalytic composites were successfully prepared by a one-step hydrothermal method. The as-synthesized photocatalysts exhibited an efficient photocatalytic performance and generated low amounts of toxic byproducts. X-ray diffraction studies show that Bi3+is successfully reduced on the surface of Ba2TiO4/BaBi4Ti4O15(BT/BBT). After L-Ascorbic acid (AA) modification, the photocatalytic NO removal efficiency of Bi@Ba2TiO4/BaBi4Ti4O15is increased from 25.55% to 67.88%, while the production of the toxic byproductNO2is reduced by 92.02%, where the initial concentration of NO is diluted to ca. 800 ppb by the gas stream and the flow rate is controlled at 301.98 mL·min−1in a 150 mL cylindrical reactor. Furthermore, ambient humidity has little effect on the photocatalytic performance of theBi@Ba2TiO4/BaBi4Ti4O15, and the photocatalyst exhibits excellent reusability after repeated cleaning with deionized water. The improved photocatalytic effect is attributed to the addition of AA in BT/BBT being able to reduce Bi3+ions to form Bi nanoparticles giving surface plasmon effect (SPR) and generate oxygen vacancies (OVs) at the same time, thereby improving the separation efficiency of photogenerated carriers, enhancing the light absorption, and increasing the specific surface areas. The present work could provide new insights into the design of high-performance photocatalysts and their potential applications in air purification, especially for NO removal.Keywords:photocatalysis;BaBi4Ti4O15;surface plasmon effect;NO removal
