Photochemical ROS-Driven Autocatalytic Transformation of Heavy Metal–Xanthate Complex Pollutants in Tailings Pond

Jiacheng Sun,Shuchen Tu,Zemin Li,Ke Li,Yuting Zhang,Qinzi Huang,Tao Chen,Hongxing Chen,Lingtian Xie,Bo Yan

Heavy metal–xanthate complexes (HMXs), formed during processing in nonferrous metal mines, have attracted increasing attention due to their complex transformation pathways and ecological risks. Solar radiation plays a key role in driving the transformation of HMXs in tailings ponds. This study revealed an overlooked transformation pathway and environmental impact of mining pollutants─the photochemical ROS-Driven autocatalytic transformation of typical HMXs such as cadmium–xanthate (CdX). CdX conversion pathways and their kinetic characteristics were investigated by the comprehensive material characterization and substance analysis. Charge transferring and radical formation-attacking were demonstrated through the joint survey of electrochemical measurements, oxygen activation monitoring, and quantum chemical calculations. Results shows that 94.93% of CdX transformed after 28 days of solar exposure. The autocatalytic kinetic matches the accelerated characteristics described by the logistic model, with its half-lives varied significantly among different types of HMX: zinc–xanthate (ZnX) (4.3 days) < CdX (19.2 days) < plumbum–xanthate (PbX) (23.6 days). Light-induced charge carriers in CdX initiated the ROS production and its conversion to cadmium sulfide (CdS). CdS accumulation enhanced the photochemical activity and the ROS generation, resulting in the autocatalytic conversion. This work provides molecular-level insights into the transformation mechanisms of HMXs, and offers theoretical foundations for controlling mine-related pollutants.

An immunomodulatory hydrogel featuring antibacterial and ROS-scavenging functions for enhanced burn wound healing

Zhao Xian-Sheng,Zhang Li-Ping,Wang Geng,Yang Qu-Yang,Cheng Xiu-Jun,Wu Chang-Yue,Wu Dan,Dong Can-Bin,Yang Yu-Tian,Wang Yi-Fan,Cai Xiu-Jun,Zhu Ning-Wen

Infectious wounds induce a cycle of bacterial proliferation, oxidative stress accumulation, and dysregulated macrophage polarization, collectively hindering tissue repair. Conventional wound dressings typically address these pathological factors in isolation, resulting in suboptimal therapeutic outcomes. Here, we report the design of a biocompatible multifunctional hydrogel (GAPC). This system integrates self-assembled Proanthocyanidin/Chlorhexidine nanoparticles into a dual-network GelMA/ADM scaffold. Consequently, the hydrogel exhibits simultaneous antibacterial activity, ROS scavenging, and immunomodulatory capacity. In vitro, GAPC hydrogel has superb antibacterial, antioxidant, and anti-inflammatory effects. On the other hand, GAPC hydrogel promotes M1-to-M2 macrophage transition and preserved cellular viability under oxidative stress. Furthermore, in vivo it accelerated infected burn wound closure, enhanced collagen remodeling, and stimulated neovascularization. Collectively, GAPC hydrogel interrupts the “infection-oxidative stress-inflammation” loop, offering a safe and promising option for managing infected wounds. Graphical

ROS-triggered smart nanoplatform promotes osteoarthritis repair by modulating macrophage polarization and mitochondrial homeostasis

Weijian Chen,Jifeng Zhao,Jiasheng Tao,Xiaodong Lin,Yizhe Song,Wenxiang Weiyang,Chao Lu,Peng Peng,Tao Jiang,Chuanxi Zhao,Wengang Liu

Oxidative stress and inflammation intertwine to form a self-amplifying “vicious cycle”, posing the core factor driving the progression of osteoarthritis (OA). Disrupting this cycle by regulating macrophage polarization, reducing oxidative stress, and maintaining mitochondrial homeostasis is a key strategy to fundamentally halt OA progression. In this study, a ROS-triggered intelligent nanodelivery system (CS@mPDA@Res) was developed to effectively promote OA repair. The system employs mesoporous polydopamine (mPDA) loaded with resveratrol (Res) as the core. By leveraging the high specific surface area and acid resistance of mPDA, this system enables sustained drug release within the OA joint microenvironment. Subsequently, the mPDA@Res nanoparticles were further coated with thioketal (TK)-modified chitosan (CS) as an outer shell. The CS shell disassembles upon ROS stimulation, enabling on-demand release of Res to efficiently promote OA repair through its anti-inflammatory and antioxidant activities. The results demonstrated that CS@mPDA@Res exhibits ROS-triggered intelligent drug release properties and excellent free radical scavenging capacity. In vitro cellular experiments showed that CS@mPDA@Res effectively modulates macrophage polarization, and alleviate oxidative stress-induced mitochondrial dysfunction and cartilage matrix degradation in chondrocytes. In vivo studies further revealed that CS@mPDA@Res treatment significantly alleviates OA joint damage. On day 28 of treatment, OA rats in the CS@mPDA@Res group exhibited 74.1% and 68.69% reductions in Mankin score and International Cartilage Repair Society (ICRS) score, respectively, compared to the model group. In summary, CS@mPDA@Res offers a novel and highly promising therapeutic approach for OA treatment.

ROS-Responsive Hydrogel Delivering Bio-Nanoselenium for Targeted Therapy of UVB-Induced Skin Photodamage

Kongdi Zhu,Jiamin Zhao,Tong Li,Shidi Zhang,Chuanming Liu,Zijian Xu,Cong Lv,Guijie Zhang,Yunlu Kang,Jiaqiang Huang

Ultraviolet B (UVB) radiation is a major environmental factor that induces skin photodamage through oxidative stress and inflammation. To address this, we developed a reactive oxygen species (ROS)-responsive hydrogel, SeNPs@HPTA, for the intelligent delivery of biogenic selenium nanoparticles (BioSeNPs) synthesized by Bifidobacterium animalis. The hydrogel was fabricated by cross-linking phenylboronic acid-grafted hyaluronic acid (HA-PBA) with tannic acid (TA), forming dynamic boronic ester bonds that cleave under high ROS conditions to enable on-demand BioSeNPs release. Transcriptomic analysis revealed that B. lactis H15 efficiently reduces selenite (Se(IV)) to elemental SeNPs by upregulating ion transport and NADPH-regenerating pathways. In vitro, SeNPs@HPTA enhanced HaCaT cell viability, scavenged ROS, and promoted cell migration post-UVB irradiation. In vivo, topical application alleviated skin lesions, improved hydration, reduced epidermal hyperplasia, and suppressed collagen degradation. Mechanistically, the hydrogel conferred therapeutic effects by enhancing antioxidant defenses, inhibiting inflammation and attenuating ferroptosis via GPx4/Nrf2 activation. Critically, genetic knockdown of GPx4 abolished its therapeutic effects, establishing a strict dependence on this key selenoenzyme. The formulation exhibited an excellent biosafety profile. This study presents a microbiome-inspired, stimulus-responsive strategy for precise skin photodamage treatment. We anticipate this microbiome-inspired, intelligent delivery system will offer a powerful and precise strategy for treating oxidative stress-related dermatoses.

A Hydrogen-Releasing Nanozyme Engineers a Mitochondrial ROS Amplifier for Self-Sustaining Catalytic Immunotherapy

Mingfan Shi,Jingrui Cao,Tong Wu,Guang Yang,YaWen Yang,Shixin Zhang,Wenwen Su,Hongyu Chu,Yangyang Zhao,Shan Jiang,Qiong Wu,Dongxu Jiao,Fangfang Chen

The efficacy of nanocatalytic therapy is constrained by the limited availability of endogenous hydrogen peroxide (H 2 O 2 ) as a reaction substrate, finite catalytic activity of nanozymes and rapid scavenging by intracellular antioxidants, hindering their accumulation at target sites to therapeutic concentrations. To address the core bottleneck, we developed a hydrogen-doped rhodium-palladium alloy (RhPd‑H) nanozyme that integrates enhanced peroxidase (POD)-mimetic catalytic activity with thermally triggered hydrogen gas (H 2 ) release. Under near-infrared (NIR) irradiation, the RhPd-H performs POD activity to efficiently produce exogenous hydroxyl radicals (·OH), inducing initial oxidative stress. Concurrently, the released H 2 flux could reduce the level of reactive oxygen species (ROS) within mitochondria, thereby mitigating oxidative damage and reprogramming mitochondria into endogenous ROS generator that continuously leak superoxide anion (·O 2 − ). This dual-path ROS generation mechanism sustains prolonged intracellular ROS burst to efficiently kill tumor cells. Further, in vivo evaluations demonstrated that RhPd-H nanoenzyme exhibited long-term tumor retention, significant suppression of tumor growth and activation of antitumor immunity. By differentially regulating ROS across space and time, RhPd‑H nanozyme establishes a persistent and overwhelming oxidative stress that effectively disrupts redox homeostasis. Our work advances beyond conventional catalytic therapy, proposing a new concept of metabolically amplified nanozyme therapy.

Thioether-functionalized polycarbonate nanosponges mitigate cisplatin-induced ototoxicity via ROS scavenging and cisplatin deactivation

Hang Yan,Bo Liu,Zilong Chen,Yunni Tu,Yaqin Tu,Xiaoting Zeng,Hongjun Xiao,Lesan Yan

Cisplatin remains a first-line chemotherapeutic for a wide spectrum of solid and metastatic tumors; however, its dose-limiting ototoxicity severely compromises patients' quality of life and causes irreversible deficits in language development and socio-cognitive function in children. To address this challenge, we developed a multifunctional amphiphilic polycarbonate-based nanosponge platform (NPs@AST) loaded with astaxanthin, designed to mitigate cisplatin-induced ototoxicity. This system is synthesized through one-step ring-opening polymerization of a trithioether-containing bicyclic carbonate monomer, resulting in a core-crosslinked nanosponge with densely distributed thioether groups. These groups confer intrinsic ROS scavenging, ROS-triggered drug release, and cisplatin-chelating detoxification in a single, chemically defined platform. Upon transtympanic administration, NPs@AST exhibits prolonged cochlear retention and robust otoprotection via two synergistic mechanisms: (i) thioether groups chelate cisplatin, deactivating it and reducing intracellular accumulation; and (ii) intracellular ROS scavenging, coupled with ROS-triggered astaxanthin release, enhancing antioxidant defense against oxidative stress. In C57BL/6 mice, NPs@AST preserved auditory thresholds at ~35 dB on day 21, compared to ~70 dB in untreated controls, as assessed by auditory brainstem response. Therefore, this ROS-responsive polycarbonate nanoplatform offers a promising strategy to mitigate cisplatin-induced ototoxicity and lays the foundation for developing effective, low-toxicity inner-ear drug delivery systems.

Smart glucose-ROS responsive CPOD/MGF@GMs hydrogel for mangiferin-mediated diabetic wound healing

Minna Wen,Xinshuo Liu,Yijia Tian,Zexiang Zheng,Shuyun Zhang,Tengfeng Zhuang,Lihua Li,Changren Zhou

Mangiferin (MGF), a natural glucosylxanthone with pro-angiogenic, anti-inflammatory, antioxidant, and anti-diabetic properties, is a promising candidate for diabetic wound healing. However, its topical application remains underexplored due to delivery limitations. Herein, we developed a smart dual-responsive chitosan/alginate hydrogel (CPOD) encapsulating MGF-loaded gelatin microspheres (MGF@GMs) for glucose- and reactive oxygen species (ROS)-triggered drug release. The CPOD/MGF@GMs hydrogel exhibited excellent self-healing capability (95% strain recovery), tissue adhesiveness (23.12 ± 6.58 kPa), and on-demand MGF release under pathological microenvironmental conditions (high glucose/ROS). In vitro assays confirmed its biocompatibility and potent pro-angiogenic effects (4–6-fold upregulation of CD31, FGF, and VEGF mRNA in HUVECs). In a diabetic full-thickness skin injury model, the hydrogel significantly accelerated wound closure (92.57% ± 2.60% healing rate within 14 days), enhanced re-epithelialization, promoted collagen deposition, and suppressed pro-inflammatory markers (84.21% reduction in TNF-α, 71.43% reduction in IL-6). Mechanistically, dual-responsive MGF release synergized with the bioactive matrix to modulate the wound microenvironment and drive tissue regeneration. This study integrates natural pharmacotherapy with smart material design, offering a clinically translatable solution for diabetic wound management.

Targeting the ROS-senescence-fibrosis axis: A dual-pronged strategy against intervertebral disc degeneration

Yifan Ding,Sheng Zhang,Ziheng Yang,Junfei Wu,Yucheng Wang,Zhiping Zhang,Songwei Tan,Yixin Cai

Intervertebral disc degeneration (IDD) is a prevalent spinal disorder with unclear etiology and limited treatment options. Through integrative analysis of single-cell transcriptomics and validation in clinical specimens, we identified a self-sustaining “ROS–senescence–fibrosis” vicious axis that critically promotes IDD progression, accompanied by significant upregulation of PDE4B in degenerative nucleus pulposus cells (NPCs). To disrupt this pathogenic circuit, we repurposed the clinical PDE4B inhibitor Nerandomilast (Nd) and constructed a biomimetic dual-pronged system for its precise delivery. This system comprises NPC membrane-derived nanovesicles for targeted intracellular Nd delivery to break the ROS–senescence–fibrosis axis, combined with a type II collagen (Col2)-loaded hyaluronic acid hydrogel for synergistic remodeling of the degenerative microenvironment. Both in vitro and in vivo experiments demonstrated excellent biocompatibility and significant efficacy in delaying IDD progression. This research establishes a mechanism-target-material strategy by integrating pathological insights with advanced nanocarrier design, which not only offers a promising therapeutic paradigm for IDD but also provides a conceptual framework for treating other fibrosis-driven degenerative diseases.

Yuying Xie,Yangchun Ge,Zhuochao Shen,Yupeng Qiu,Haohua Zhang,Changmeng Sun,Jiangkuan Tang,Honglei Wang,Bingke Li,Junran Xu,Ping Liang,Xuqiang Peng,Lanlan Zhai,Zhongyu Li,Zhihui Li

ROS-triggered release of avermectin from cinnamaldehyde-based delivery system; • Enhanced insecticidal efficacy against Plutella xylostella larvae; • Improved leaf adhesion and favorable plant safety profile; • Stable carriers with high drug loading capacity and encapsulation efficiency. To achieve precise delivery and controlled release of pesticides, this study designed and developed a reactive oxygen species (ROS)-responsive nano-pesticide triggered by the excessively high ROS levels in pests, enabling precise and efficient control of target pests. Using the natural compound cinnamaldehyde as the core, an ROS-responsive amphiphilic small molecule was synthesized, and hydrophobic pesticide avermectin-loaded micelle suspension were successfully prepared through self-assembly. Characterization results showed that the avermectin-loaded micelle suspension had an average particle size of 136.0 nm, a drug loading capacity of 17.78 ± 0.13%, and an encapsulation efficiency of 77.05 ± 0.55%, significantly enhancing the adhesion of pesticides to leaf surfaces. In vitro release experiments confirmed that under micelle suspension, the cumulative release rate was minimal over 50 h, while under simulated high ROS conditions in pests, the cumulative release rate reached 86.02% ± 1.69% over 50 h. Indoor bioassays demonstrated that the micelle suspension exhibited superior efficacy against Plutella xylostella larvae compared to conventional pesticides. Plant safety experiments indicated that the micelle suspension possesses favorable safety profiles. ROS-activated prodrug nano-assemblies based on cinnamaldehyde-modified paclitaxel for treating breast cancer

Chen Yali,Qian Lei,Xu Chunyun,Huo Hongjie,Sun Zhenya,Zhang Hongyang,Yuan Shuang,Qi Yan,Chu Peng,Chen Lixue,Ding Yanfang,Li Lei

Purpose One of the hallmarks of cancer cells is their elevated levels of reactive oxygen species (ROS) compared with normal cells. Capitalizing on this, a novel paclitaxel (PTX, a first-line chemotherapeutic agent) prodrug was designed and synthesized by attaching a cinnamaldehyde (CA) molecule to PTX via a ROS-responsive thioacetal group. Methods The paclitaxel prodrug (PTX-S-CA) prodrug was synthesized and characterized, followed by self-assembly into nanoparticles (NPs) using DSPE-PEG 2K as a stabilizer, forming PTX-S-CA/DSPE-PEG 2K NPs. The physicochemical properties of the NPs were evaluated, which included drug-loading capacity, encapsulation efficiency, and plasma stability. In vitro cytotoxicity assays were conducted on cancer cells, and in vivo anti-tumor efficacy was assessed in a murine tumor model. The pharmacokinetics of the NPs were also studied in rats. Results In the tumor microenvironment, the ROS-sensitive thioacetal group in PTX-S-CA was degraded due to high ROS levels, leading to the release of both PTX and CA. The released CA synergistically enhanced the anti-tumor effects of PTX by inducing additional ROS production within the mitochondria. The NPs demonstrated a high loading capacity and encapsulation efficiency for PTX-S-CA, high cytotoxicity against cancer cells, excellent anti-tumor effects on mice, and high plasma stability in rats. Conclusion The use of the NP drug delivery platform served as a potentially effective therapeutic strategy for treating tumors, especially breast cancer.

Multifunctional magnetic MOF under cover of erythrocytes for ROS scavenging and magnetic hyperthermia thrombolysis

Pengpeng Guan,Caijie Yuan,Jiali Li,Zhou Wen,Enling Hu,Guangqian Lan,Bowen Cheng,Yinggui Huang,Fei Lu

The inadequacy of current antithrombotic therapies lies in the limited efficacy of pharmacotherapy and the poor targetability of drugs to thrombotic sites. To address these challenges, we developed a multifunctional nanoplatform, Fe 3 O 4 @MC-DEA-PBAP (FMDP), which integrates magnetic responsiveness and erythrocyte targeting for magnetic hyperthermia thrombolysis (MHT). FMDP is constructed using metal–organic frameworks (MOFs) crosslinking CaCO 3 /cyclodextrin as the base carrier, modified with 2-(Diethylamino)ethyl methacrylate (DEA) and 4-hydroxyphenylboronic acid pinacol ester (PBAP), then conjugated with nano-ferric oxide for MHT. DEA and PBAP make FMDP to adhere onto erythrocytes and scavenge reactive oxygen species (ROS), respectively. In vitro experiments proved that FMDP exhibits high photothermal conversion efficiency and robust magnetic responsiveness, enabling the thrombolytic efficiency of FMDP was three times that of uPA group. In addition, FMDP adhere to erythrocytes (83.5 %) and reach the thrombi under the cover of erythrocytes for MHT. In addition, FMDP effectively scavenges ROS, mitigating endothelial damages and promoting vascular regeneration. Overall, FMDP represents a promising therapeutic strategy for the comprehensive management of thrombosis, offering improved efficacy and reduced bleeding risks compared to conventional thrombolytic therapies.

Microenvironment-adaptive nanomedicine MXene promotes flap survival by inhibiting ROS cascade and endothelial pyroptosis

Yang Ningning,Hua Rongrong,Lai Yingying,Zhu Peijun,Ding Jian,Ma Xianhui,Yu Gaoxiang,Xia Yiheng,Liang Chao,Gao Weiyang,Wang Zhouguang,Zhang Hongyu,Yang Liangliang,Zhou Kailiang,Ge Lu

In the field of large-area trauma flap transplantation, preventing avascular necrosis remains a critical challenge. Key mechanisms for improving flap viability include angiogenesis promotion, oxidative stress inhibition, and cell death prevention. Recently, two-dimensional ultrathin Ti 3 C 2 T X (MXene) nanosheets have gained attention for their potential contributions to these processes, though MXene’s physiological impact on flap survival had not been previously investigated. This study is the first to confirm MXene's biological effects on the ischaemic microenvironment post-skin flap transplantation. Findings indicated that MXene significantly decreased the necrotic area in ischaemic flaps (37.96% ± 2.00%), with reductions of 30.40% ± 1.86% at 1 mg/mL and 20.19% ± 2.11% at 2 mg/mL in a concentration-dependent manner. Mechanistically, MXene facilitated in situ angiogenesis, mitigated oxidative stress, suppressed pro-inflammatory pyroptosis, and activated the PI3K-Akt pathway, particularly influencing vascular endothelial cells. Comparative transcriptome analysis of skin tissues with and without MXene treatment provided additional evidence, highlighting mechanisms such as pro-inflammatory pyroptosis, ROS metabolic processes, endothelial cell proliferation regulation, and PI3K-Akt signaling pathway activation. Overall, MXene demonstrated biological activity, effectively promoting ischaemic flaps survival and presenting a novel strategy for addressing ischaemic necrosis in skin flaps. Graphical abstract
