Hyperuricemia (HUA) is widespread, and since conventional long-term treatments can be toxic, safer multi-target alternatives are required. Here, we tested a naturally derived synbiotic pairing of the uricolytic probiotic Lactobacillus gasseri SYP-B4432 with a polyphenol-rich Rumex hanus by. extract in a mouse model of hyperuricemia. In just five weeks of daily treatment, the combination reduced serum uric acid by 57.86%, suppressed serum xanthine oxidase activity by 14.31%, and restored the Bacteroidetes/Firmicutes ratio while lowering pro-inflammatory cytokines and attenuating renal histopathology. Importantly, no hepatic/renal impairment or body weight abnormalities were detected, confirming the safety of this intervention. Collectively, the probiotic–herbal duo alleviates HUA through the synergy of three modes of action: curtailing uric acid synthesis (via XOD inhibition), reshaping gut microbiota (to enhance UA excretion), and resolving renal inflammation (via cytokine modulation). This natural, safe, and multi-target synbiotic provides a clinically translatable strategy that may address the side effect limitations of conventional hyperuricemia drugs.

Prostate cancer (PCa) is one of the most common cancers in males, and its treatment remains challenging due to the tumor microenvironment (TME) with immunosuppressive properties and limited response to anti-PD-1 therapy. Gut microbiota-derived metabolites have recently emerged as modulators of cancer immunometabolism, however, their role in PCa progression and immunotherapy is poorly understood. Here we found that indole-3-lactic acid (ILA), a metabolite produced by Lactobacillus plantarum , exerted dual anti-tumor effects on PCa cells and the TME. Mechanistically, ILA activates the aryl hydrocarbon receptor (AHR), and the resulting AHR/ARNT heterodimer translocates into the nucleus and binds to the promoter of ASF1B. This heterodimer then recruits the HDAC1/2-NuRD complex to reduce H3K27ac levels and suppress ASF1B expression. ASF1B binds to specific residues of ENO1 via its N-terminal core domain and enhances ENO1 enzymatic activity. ILA-induced downregulation of ASF1B impairs this interaction, reduces ENO1 activity, and suppresses the PI3K/Akt pathway, thereby inhibiting the malignant phenotypes of PCa cells. Concurrently, ILA decreased CXCL8 secretion by inhibiting the PI3K/Akt/NF-κB pathway, enhancing CD8 + T cell infiltration and M1 macrophage polarization, thereby remodeling the TME. Additionally, ILA synergized with anti-PD-1 therapy to more effectively suppress tumor growth. These findings reveal a novel mechanism by which gut microbiota-derived metabolites regulate PCa progression and immunometabolism, positioning ILA as a potential therapeutic agent to improve PCa treatment.

A huge number of titanium dioxide nanoparticles (TiO2 NPs) exist in confectionery foods, which is a high-risk factor for development of diet-induced metabolism syndrome (MetS). In this study, we built a high fructose drinking-induced MetS mouse model, and oral intake of 20 mg/kg TiO2 NPs was administered for 8 weeks. Significant pathological changes and inflammatory factors of overproduction were detected in the liver and colon. The 16S rDNA sequencing analysis results indicated that TiO2 NPs evidently and further perturbed the gut microbiota diversity, compositions, and KEGG pathways in MetS mice. Fecal microbiota transplant experiment proved that TiO2 NPs-altered gut microbiota drives liver and colon inflammation damage. More importantly, oral supplementation of Lactobacillus rhamnosus GG (LGG) ameliorated not only the TiO2 NPs-induced inflammation but also the fructose-caused metabolic abnormality. LGG recovered the gut dysbiosis and decreased the abundance of inflammation-related bacteria (Desulfovibrionaceae, Clostridia, and Proteobacteria), thereby protecting against TiO2 NPs-induced severe inflammation damage. Our study suggests the necessity of assessing the toxic effects of foodborne nanoparticles on the chronic disease population and potential usefulness of probiotics as prophylactic and therapeutic.

Peritoneal fibrosis is a complication of long-term peritoneal dialysis (PD) that restricts its clinical application for the treatment of end-stage renal disease. Lactobacillus casei Zhang (LCZ), a probiotic strain isolated from traditional fermented koumiss, exhibits health benefits such as anti-inflammatory and antioxidative effects, improvement of insulin resistance and mitigation of renal injury. However, whether LCZ can prevent peritoneal fibrosis remains unknown. Here, we assessed the effects of LCZ in a mouse model of PD-induced peritoneal fibrosis. Our results showed that the administration of LCZ significantly ameliorated peritoneal fibrosis in experimental mice. Macrophage infiltration, inflammatory M1 polarization and inflammatory cytokines in peritoneal dialysis effluents were effectively reduced by LCZ. Meanwhile, LCZ corrected gut dysbiosis and enriched beneficial bacteria that produce short-chain fatty acids, specifically Dubosiella, Lachnospiraceae, Parvibacter, and Butyricicoccus. Correspondingly, the local butyrate level in peritoneal dialysis effluents was significantly elevated by LCZ. Mechanistically, we found activation of PPARγ and inhibition of the NF-κB pathway in LCZ-treated mice, an observation that was replicated in a butyrate-treated macrophage cell line. In conclusion, our study suggests that LCZ is beneficial for preventing PD-induced peritoneal fibrosis through modulating the gut microbiota, enhancing butyrate production, activating PPARγ, and suppressing NF-κB-mediated inflammation.

The effects of a Tween 80 and CaCl 2 mixture on the hypocholesterolemic activity of bile salt hydrolase (BSH)-active Lactobacillus casei F0422 were investigated in rats fed a cholesterol-rich diet for 3 weeks. This mixture significantly enhanced the hypocholesterolemic activity of strain F0422 in rats. The group administered drinking water supplemented with both strain F0422 and the Tween 80 and CaCl 2 mixture had significantly lower levels of total serum cholesterol, low-density lipoprotein cholesterol and total liver cholesterol than did the group administered drinking water supplemented only with strain F0422 or the Tween 80 and CaCl 2 mixture. The hypocholesterolemic activity of the combined strain F0422 and Tween 80 and CaCl 2 mixture was attributed to the increase in fecal bile acid excretion through enhancement of bile salt tolerance and BSH catalytic activity of strain F0422 by Tween 80 and up-regulation of the adhesion of strain F0422 to intestinal epithelial cells by CaCl 2 .

BACKGROUND Phenolic compounds are increasingly recognized for their diverse health benefits, particularly their potent antioxidant capacities. To enhance its lipophilicity, stability and bioavailability, hydroxytyrosol has been successfully esterified with various alkyl chains lengths and different isomers (branched-chain and straight-chain), forming hydroxytyrosol-fatty acid acyl esters (Hty-Es). These synthesized Hty-Es, have garnered significant interest and are widely explored in fields such as food preservation because of their superior antioxidant properties. However, little is known about the digestion, fermentation characteristics and the effects on the intestinal microbiota in vitro . RESULTS This study evaluated the influence of hydroxytyrosol (HTy) esterified with fatty acids of varying chain lengths (C2:0, C3:0, C4:0 n , C4:0 i , C6:0, C8:0, C12:0, C14:0 and C16:0) on polyphenol sustained-release properties and gut microbiota modulation during simulated in vitro digestion, fecal fermentation and Lactobacillus fermentation. The findings demonstrated partial hydrolysis of HTy-Es in both simulated gastric fluid and intestinal fluid to slowly release a small amount of polyphenols, suggesting their potential to reach the colon and cecum. Analysis of fermented fecal samples revealed that HTy-Es supplementation modulated microbiota composition, elevating the diversity of Actinobacteriota, Firmicutes and Bacteroidetes , whereas it decreased the diversity of Desulfobacterota, Actinobacteriota, Chloroflexi and Verrucomicrobiota . At the genus level, the growth of detrimental genera ( Escherichia shigella ) was successfully suppressed. Notably, HTy-Es with distinct acyl chain lengths and isomer configurations exhibited divergent effects on polyphenol bioaccessibility and gut microbiome diversity, underscoring their structure-dependent functional potential for health applications. CONCLUSION Conclusively, the slow release of fatty acids and HTy clearly demonstrated that HTy-Es would be an effective approach for enhancing the beneficial health effects of fatty acids and HTy. © 2025 Society of Chemical Industry.

Background Previous studies have demonstrated that numerous medicine and food homology (MFH) possess the potential to regulate purine metabolism disorders, promote uric acid excretion, and alleviate hyperuricemia symptoms. Examples include CS (Chaenomeles speciosa (Sweet) Nakai), SR (Smilax glabra Roxb.) and PL (Pueraria montana var. lobata). Methods Metabolomics was employed to analyze the compositional changes in medicinal and edible extracts before and after fermentation. Network pharmacology and molecular docking studies were further utilized to elucidate the interactions between these differential metabolites and the core targets of hyperuricemia. In vitro enzyme activity assays were conducted to confirm the therapeutic effects. Results A total of 283, 248, and 18 differential metabolites were identified in CS,SR and PL samples, respectively. Among these, 54 significantly upregulated differential metabolites were selected for screening. Based on these metabolites, 53 HUA-related targets were identified for CS, SR and PL. Functional enrichment analysis revealed their roles in inflammatory stress and uric acid production pathways, particularly the MAPK signaling pathway and purine metabolism regulated by XDH. Additionally, other targets in the purine metabolism pathway, such as ADA, PNP, AMPD3, and IMPDH2, were co-regulated. Enzyme activity assays indicate that fermented MFH more effectively inhibits XOD, thereby regulating the conversion of xanthine and hypoxanthine into uric acid. Molecular docking revealed two significantly upregulated compounds in CS; and five in PL; and four in SR. exhibit strong binding to XOD.Conclusion These findings provide theoretical support for FMFH as a potential effective component in preventing and treating hyperuricemia. Our research demonstrates that FMFH targets multiple pathways associated with hyperuricemia, offering a promising approach for preventing this condition.

Soluble dietary fiber (SDF) is a key functional component in rice bran, but its low natural abundance limits its application potential. In this study, two Lactobacillus johnsonii strains with distinct feruloyl esterase (FAE) activities were selected to investigate their effects on the modification of rice bran SDF through fermentation. The results showed that fermentation with the high-FAE strain L. johnsonii 46 (FSDF-46) increased the SDF yield from 6.39% to 11.45%. Structural characterization indicated that fermentation was associated with the formation of a porous structure, reduced particle size, and decreased crystallinity. Furthermore, FSDF-46 exhibited higher water holding capacity (0.87 g/g) and oil holding capacity (3.789 g/g). The glucose adsorption capacity of FSDF-46 increased by 44.8% and 37.52%. In vitro assays showed that FSDF-46 exhibited enhanced inhibitory effects on α-amylase (52.33%) and α-glucosidase (91.63%). These findings suggest a potential role of FAE-related fermentation in dietary fiber modification and provide a basis for the development of value-added products from rice bran.

In this study, we demonstrated the role of Pickering high internal phase emulsions (HIPEs) stabilized with the whey protein isolate (WPI)/(-)-epigallocatechin-3-gallate (EGCG) covalent conjugates on the storage and GIT passage viability of probiotics powder ( Lactobacillus Plantarum ). At pH 9 value, the WPI-EGCG covalent conjugates by free-radical induction reaction formed hydroxyquinone or hydroxyquinone dimers with better wettability, and digestive enzyme inhibition effect and smaller particle size. Then, the WPI-EGCG covalent conjugates were used to stabilize the O/W type Pickering emulsion. The oil fraction ( ϕ ) addition led to an increase in the droplet size and modulus of Pickering emulsions toward to generate gel-like structure. On the other hand, at a constant ϕ value, the droplet size of these Pickering HIPEs reduced with increasing particle concentration ( c ) from 0.5% to 5%, indicating that high c can stabilize more oil droplets. Encapsulation of Lactobacillus Plantarum powder with the Pickering HIPEs was successful to enhance the viable cell count after 14 days storage and GIT digestion. The protective effect of Pickering emulsions was improved with the addition of the c and/or ϕ value. Therefore, Pickering HIPEs can be applied as a promising carrier for enhancing the viability of probiotic powder during storage and GIT digestion by reason of the high oil phase fraction and the adsorption of the WPI-EGCG covalent conjugates on the droplet interface.

In this study, soyasaponin (Ssa), corn oil, and soybean protein isolate (SPI) were used to prepare a water-in-oil-in-water (W/O/W) double emulsion system that can protect Lactobacillus plantarum (L.P) in the upper gastrointestinal tract for delivery to the small intestine and colon. Incorporating different amounts of L.P (0.1–0.6% (w/v)) in the W/O/W double emulsion caused the viscoelastic modulus and droplet size of the L.P-embedded emulsion system to change, and the emulsion system transformed from a non-Newtonian fluid to a “solid-like” gel. These results revealed that L.P encapsulation led to significantly improved stability of the W/O/W emulsion. In addition, different concentrations of L.P had different effects on the survival rate of L.P in simulated digestive fluids, and after in vitro digestion, L.P was naturally delivered from the stomach into the intestines. The optimal concentration of L.P for obtaining a stable emulsion was found to be 0.3%, and the survival rate of the encapsulated L.P in the small intestine was as high as 78.96% after 1 h. Thus, choosing an appropriate amount of L.P was crucial. The W/O/W emulsion system effectively maintained the viability of L.P and successfully delivered it to the small intestine and colon.

The work aimed to evaluate the effect of lactic acid bacteria, natamycin and hexanoic acid on the fermentation quality, aerobic stability and in vitro digestibility of total mixed ration (TMR) silages containing water bamboo sheath leaves. The treatments were as follows: (1) no additives (control, C), (2) lactic acid bacteria (L), (3) natamycin (N), (4) hexanoic acid (H), (5) lactic acid bacteria + natamycin (LN), (6) lactic acid bacteria + hexanoic acid (LH). Silos were opened and silages were evaluated to assess fermentation quality, aerobic stability and in vitro digestibility. All TMR silages were well-preserved as indicated by dominant lactic acid content, low ammonia nitrogen/total nitrogen, trace propionic acid and negligible butyric acid contents. All additives improved the fermentation quality and in vitro digestibility, evidenced by higher lactic acid/acetic acid ratio, lactic acid, acetic acid, water-soluble carbohydrate contents and lactic acid bacteria count, and lower pH, aerobic bacteria and yeast counts. The LN had the highest lactic acid, acetic acid, lactic acid/acetic acid ratio, in vitro gas production and in vitro digestibility. During aerobic exposure, the LN silage had the highest lactic acid and acetic acid contents, and the lowest pH among all TMR silages. The LN and LH prolonged aerobic stability, reaching 85.5 and 81.5 h, respectively. The LN is recommended to improve the fermentation quality, aerobic stability and in vitro digestibility of TMR prepared with water bamboo sheath leaves.

Probiotics have considerable interest due to their inseparable link to human health. However, probiotic products are seriously challenged during processing, preservation, and intake. Food-grade probiotic delivery systems need to be further explored as an effective way to enhance cell viability. In this study, water-in-oil (W/O) Pickering emulsions were fabricated by adding candelilla wax (CLW) as a network stabilizer based on acylated EGCG derivatives in the crystalline form as a Pickering stabilizer. The effects of acylated EGCG derivatives’ concentration, CLW concentration, and oil phase volume fraction on the droplet size distribution, microstructure, and physical stability of Pickering emulsions were explored. The presence of CLW reduced the particle size and improved the physical stability of acylated EGCG-based emulsions, and the effect was more positive with increasing concentration. The protective effect of emulsions with different oil phase volume fractions on Lactobacillus plantarum during freeze-thaw cycles, storage, and gastrointestinal digestion was evaluated. The outer-phase physical barrier of W/O emulsions co-stabilized with acylated EGCG derivatives and CLW facilitated the sensitivity of probiotics to ice crystal growth, temperature changes, acidic environments, and digestive enzymes. The emulsions formulated with 40% oil phase volume fractions allowed Lactobacillus plantarum to survive up to 7.75 log CFU g−1 in the harsh gastrointestinal environment. The results offer promising strategies for applying W/O emulsion probiotic delivery systems in food processing, storage, and oral administration.
