Multifunctional zinc phthalocyanine-phenolic resin (ZnPc-PFR)@MSN nanocomposite based fluorescent imaging, photothermal therapy, and pH-sensitive drug release

Xiangfei Sun,Ping Yang,Shaohua Wang,Honglong Xing

Because of the good fluorescence of zinc phthalocyanine-phenolic resin (ZnPc-PFR) photosensitizer and large specific surface area of mesoporous silica nanoparticles (MSNs), a highly efficient nano-drug carrier system, denoted as ZnPc-PFR@MSN, was constructed for photothermal therapy (PTT) and pH-sensitive drug delivery. The facile hydrothermal reaction was used to synthesize ZnPc-PFR nanoparticles in one-step. After loading the as-synthesized ZnPc-PFR nanoparticles into MSNs, and a good high drug-loading rate (143.7 mg g −1 ) to the anticancer drug of Adriamycin (DOX) could be obtained. Thus, a novel nanosphere with the merits of good fluorescence, high drug-loading rate (143.7 mg g −1 ), better sustained-release properties, and photothermal properties (reached 43.23°C within 260 s) was prepared. The as-synthesized multifunctional composites make it a good candidate in fluorescence imaging, PTT, and drug delivery.

Le Wang,Pengxue Sun,Yiyu Yang,Hanzhen Qiao,Hailong Tian,Dapeng Wu,Shuoye Yang,Qipeng Yuan,Jinshui Wang

The field of enzyme cascades in limited microscale or nanoscale environments has undergone a quick growth and attracted increasing interests in the field of rapid development of systems chemistry. In this study, alcohol dehydrogenase (ADH), lactate dehydrogenase (LDH), and mesoporous silica nanoparticles (MSN) immobilized nicotinamide adenine dinucleotide (NAD+) were successfully immobilized on the zeolitic imidazolate frameworks (ZIFs). This immobilized product was named ZIF@ADH/NAD-MSN/LDH, and the effect of the multi-enzyme cascade was studied by measuring the catalytic synthesis of lactic acid. The loading efficiency of the enzyme in the in-situ co-immobilization method reached 92.65%. The synthesis rate of lactic acid was increased to 70.10%, which was about 2.82 times that of the free enzyme under the optimal conditions (40 °C, pH = 8). Additionally, ZIF@ADH/NAD-MSN/LDH had experimental stability (71.67% relative activity after four experiments) and storage stability (93.45% relative activity after three weeks of storage at 4 °C; 76.89% relative activity after incubation in acetonitrile-aqueous solution for 1 h; 27.42% relative activity after incubation in 15% N, N-Dimethylformamide (DMF) solution for 1 h). In summary, in this paper, the cyclic regeneration of coenzymes was achieved, and the reaction efficiency of the multi-enzyme biocatalytic cascade was improved due to the reduction of substrate diffusion. Keywords:confinement effect;multi-enzyme catalytic cascade;in-situ synthesis;coenzyme recycling;co-immobilization enzyme

Improving osteogenic and antibacterial properties of porous titanium scaffolds by facile flow-casting of bioactive glass-Ag@MSN coatings

Yuhan Zhang,Jingjie Yu,Jiabao Guo,Guandi He,Yonghe Zhao,Qinqin Lu,Junzhi Wu,Xinrui Shu,Xiao Lin,Qiang Chen

Modification of porous titanium (PT) implants with biological coatings is being challenged due to their inert surface and complex internal structure. A facile flow-casting technique has been utilized to prepare a bioceramic coating consisting of 45S5 bioactive glass (BG) and core-shelled [email protected] particles homogeneously on the entire surface of PT. The addition of [email protected] nanoparticles facilitated the sintering of BG and subsequently enhanced the adhesion of the coating. Slightly delayed hydroxyapatite formation was also detected probably due to minimized release and consumption of calcium ions . In-vitro assessments of the coating cultured with MC3T3-E1 osteoblasts and Staphylococcus aureus were carried out to assess their potential as multifunctional bone implants with excellent bioactivity and antibacterial properties. A template for producing robust and multi-component ceramic coatings on PT and other porous implants with tailorable biological properties for bone tissue engineering has been established.

A mesoporous magnetothermal nanopomegranate with acid-tolerant hierarchical structure for remote controlled drug delivery

Yanxia Yang,Yingzi Li,Xin Xia,Shengkai Li,Shen Wang,Liang Zhang,Yuqi Cheng,Long Chen,Qian Dong,Zhuo Chen

Developing acid-tolerant magnetothermal nanoplatform with robust drug loading capacity for remote controlled drug release is challenging in the harsh acid environment of stomach. Herein, a nanopomegranate-type magnetothermal controllable drug release platform (FeCo@G@MSN), consisted of magnetothermal convertor (graphene-isolated FeCo nanocrystal core, FeCo@G) and drug reservoir (mesoporous silica, MSN), with acid-tolerant hierarchical structure was developed. Benefiting from the acid tolerance of graphene and MSN, FeCo@G@MSN was stable in 1 M hydrochloric acid for more than 12 h. The nanopomegranate-type hierarchical structure endowed FeCo@G@MSN with high saturation magnetization (114.9 emu/g) to generate a magnetothermal effect and large specific surface area (293.7 m 2 /g) to provide ample space for drug loading. As a proof of concept, a heat-triggered nitric oxide (NO) precursor was loaded, and NO were controlled released by breaking of S-NO bonds through localized heat derived from magnetothermal effect of FeCo@G, resulting in an inhibition effect towards Helicobacter pylori . A chemotherapeutic drug (doxorubicin, DOX) was also loaded and controlled released by breaking of hydrogen bonds between silica and DOX under remote an alternating magnetic field, thus exhibiting an inhibition effect towards gastric cancer cells. This work provides an efficient strategy to exploit magnetothermal controlled drug release platform and offers inspiration for the treatment of gastric diseases.

Electrochemical immunosensor for detection of prostate specific antigen based on an acid cleavable linker into MSN-based controlled release system

Dawei Fan,Na Li,Hongmin Ma,Yan Li,Lihua Hu,Bin Du,Qin Wei

A mesoporous silica nanoparticle (MSN)-based controlled release system with acid cleavable linkage was developed to fabricate an electrochemical immunosensor for the quantitative detection of the prostate-specific antigen (PSA). 3,9-Bis(3-aminopropyl)-2,4,8,10-tetraoxaspiro[5.5]undecane functionalized mesoporous silica nanoparticles (MSN-Acetal) were used to immobilize the electron mediator thionine (Th). The encapsulation of Th molecules was achieved by capping the pores of MSN-Acetal with carboxylic acid modified Au nanoparticles (defined as MSN-Th-Au). Under the acidic conditions, the capped Au nanoparticles were removed from MSN-Th-Au through the hydrolysis of the acid-labile acetal linker, resulting in the release of encapsulated Th. In this work, the pH-responsive cargo release system was firstly used as the label of secondary anti-PSA for developing an electrochemical immunosensor, and amination Fe 3 O 4 was used as the sensing matrix for immobilizing primary anti-PSA on magnetic carbon electrode surfaces . The specific recognition of PSA resulted in the attachment of MSN-Th-Au-secondary anti-PSA (MSN-Th-Au-Ab 2 ) onto the electrode surfaces. Subsequently, the released Th was detected by differential pulse voltammetry under the acidic conditions. The developed cargo release system provided an innovative and reliable method for the detection of PSA because the response signal was correlated with the concentration of PSA. Under the optimal conditions, the electrochemical immunosensor exhibited a wide linear range of 0.001–5.0   ng/mL with a low detection limit of 0.31   pg/mL. Moreover, the developed immunosensor showed superior reproducibility and long-term stability, which has promising applications in bioassay and biosensing.

Improving the Therapeutic Efficiency of Hypoxic-Activated Prodrugs by Enhancing Hypoxia in Solid Tumors

Gaoqian Zhao,Yan Jin,Shutao Gao,Tingshan Xiao,Miao Fan,Dandan Liu,Jinchao Zhang,Zhenhua Li,Xiaohan Zhou,Huifang Liu

The low sensitivity of hypoxic regions in solid tumors to radiotherapy and chemotherapy remains a major obstacle to cancer treatment. By taking advantage of hypoxic-activated prodrugs, tirapazamine (TPZ), generating cytotoxic reductive products and the glucose oxidase (GOx)-based glucose oxidation reaction, we designed a nanodrug-loading system that combined TPZ-induced chemotherapy with GOx-mediated cancer-orchestrated starvation therapy and cancer oxidation therapy. In this work, we first prepared mesoporous silica (MSN) loaded with TPZ. Then, in order to prevent the leakage of TPZ in advance, the surface was coated with a layer of carMOF formed by Fe3+ and carbenicillin (car), and GOx was adsorbed on the outermost layer to form the final nanosystem MSN-TPZ@carMOF-GOx (MT@c-G). GOx could effectively consume oxygen and catalyzed glucose into gluconic acid and hydrogen peroxide. First, the generated gluconic acid lowered the pH of tumor tissues, promoted the decomposition of carMOF, and released TPZ. Second, oxygen consumption could improve the degree of hypoxia in tumor tissues, so that enhanced the activity of TPZ. Furthermore, GOx could generate cancer-orchestrated starvation/oxidation therapy. Therefore, our study provided a new strategy that TPZ combined with GOx achieved starvation/oxidation/chemotherapy for enhancing anticancer effects in hypoxic regions.

Targeted polydopamine/MSN delivery system to enhance the anti-breast tumor efficacy by inducing ferroptosis and synergistic chemotherapy and immunology effect

Shuoye Yang,Li Su,Suxi Liu,Siyu Zheng,Beibei Zhang,Xin Wang,Ning Li,Lu Zhang,Peng Li

The combination of ferroptosis with chemotherapy and immunotherapy is a novel cellular mechanism with significant implications for antitumor therapeutic strategies. It can mitigate toxic damage to normal tissues and cells, while potentiating antitumor immune responses. In this study, we propose a multi-mechanism nanosystem that utilizes amino-modified mesoporous silica (MSNs-NH 2 ) as carriers. These nanoparticles are loaded with doxorubicin (DOX) and the immunomodulator all-trans retinoic acid (ATRA), followed by coating with polydopamine (PDA). A hybrid nanosystem featuring a core/shell structure was fabricated by chemically chelating ferrous lactate onto PDA coating. It was subsequently conjugated with RGD-targeting peptides (RFP/MN@D/A). In vitro experiment results revealed that RFP/MN@D/A possessed pH/H 2 O 2 dual-responsive drug release characteristics, and could be efficiently internalized by MCF-7 cells. Exogenous Fe 2+ activated the Fenton reaction in the acidic tumor microenvironment (TME), generating an abundance of reactive oxygen species (ROS) that attack membrane phospholipids. This attack led to cytotoxic free radical production and thereby induced ferroptosis. Additionally, following gradual rupture of the PDA coating, DOX and ATRA were released responsively. This release triggered immunogenic cell death (ICD) and apoptosis through multiple mechanisms, including HMGB1 release, cell cycle arrest, and mitochondrial dysfunction. In tumor-bearing mice, RFP/MN@D/A exhibited superior tumor growth inhibitory activity alongside good biosafety profiles. Collectively, the functionalized delivery system developed in this study has significant potential for modulating immune responses and inducing ferroptosis through targeted delivery coupled with multiple responsive release strategies. This approach synergizes effectively with chemotherapy to achieve enhanced therapeutic efficacy against breast tumors, with broad application prospects for solid cancer treatment. The flame retardant and smoke suppressant effect of transition metal hydroxystannate (MSn(OH)6, M = Fe, Co, Mn) for epoxy resin

Hui Xianghe,Fang Dong,Wang Guifang,Ran Feng,Zhang Xin,Fu Zewei,Ruzimuradov Olim

Perovskite hydroxystannate (MSn(OH) 6 ) has garnered considerable interest in recent years as a novel flame retardant characterized by its low toxicity and environmentally benign properties. In order to improve the flame retardancy and smoke suppression properties of epoxy resin (EP) matrices, transition metal hydroxystannate (MSn(OH) 6 , M = Fe, Co, Mn) was prepared by a ultrasonic-coprecipitation method and used as flame retardants. The synthesized composites were assessed for their flame retardant characteristics and mechanical properties through the measurement of the limiting oxygen index (LOI), the cone calorimetry test (CCT), and universal tensile testing. Upon the incorporation of 10 wt% of the MSn(OH) 6 flame retardants, the flame retardant of EP composites exhibited marked enhancement, especially manganese hydroxystannate (MHS). Compared with pure EP, the EP/MHS-10 composite demonstrated the best performance, reducing the peak of heat release rate (pHRR), total heat release (THR), total smoke production (TSP) by 60.0%, 14.4%, and 32.6%, respectively. During the process of combustion, the decomposition of MSn(OH) 6 results in the generation of non-flammable gases and water vapor. In the condensed phase, tin (Sn) and transition metals contribute to the formation of a more protective char residue. The residue serves as a physical barrier, effectively isolating the underlying epoxy (EP) matrix material from heat and oxygen.

Investigation of nirmatrelvir epimerization occurred in analytical sample solution by using high resolution LC-MSn, NMR, and hydrogen/deuterium exchange study

Lewei Zheng,Lingqiao Wang,Dan Li,Chongjun Xie,Zhiyuan Gao,Jinsheng Lin,Jian Ye,Wenbin Chen,Min Li

During the related substances testing of nirmatrelvir , an unknown peak was observed and the level of the peak increased over time during the storage of the sample solution. By using a strategy including LC-PDA/UV-MS n analysis, the degradant was rapidly identified as an epimer of nirmatrelvir, a solution degradation product that is caused by the trace amount of alkaline impurities leaching from the glass HPLC vials. In addition, by using hydrogen/deuterium exchange NMR spectroscopy analysis, the epimerization position was determined to be the carbon α to the adjacent cyano group . Further investigation indicated that the occurrence of the solution degradation can be suppressed when the glass HPLC vials were replaced by plastic HPLC or mass spectrometric grade vials.

Amino-functionalized mesoporous silica nanospheres (MSN-NH2) as sorbent for extraction and concentration of synthetic dyes from foodstuffs prior to HPLC analysis

Peige Qin,Yixin Yang,Wenqi Li,Jing Zhang,Qian Zhou,Minghua Lu

In this work, amino-functionalized mesoporous silica nanospheres (MSN-NH2) were synthesized and demonstrated as useful sorbents of dispersive solid-phase extraction (dSPE) for extraction and preconcentration of synthetic dyes (tartrazine, amaranth, sunset yellow, allura red, and orange II) from foodstuffs prior to high-performance liquid chromatography (HPLC) analysis. The MSN-NH2 was characterized by transmission electron microscopy, Fourier-transform infrared spectroscopy, nitrogen adsorption and desorption isotherms, and elemental analysis. To achieve optimized extraction efficiency, several critical factors including amount of MSN-NH2, sample pH, adsorption time, adsorption temperature, desorption time, and desorption solvents, were systematically investigated. Under the optimized conditions, the enrichment factors were higher than 120. The limits of detection were found to be 0.10–0.30 ng mL−1. The method showed excellent linearity in the range of 0.45–1000.0 ng mL−1 with correlation coefficients (R2) ≥ 0.9951. The intra-day and inter-day precision (n = 5) were 1.72–3.64% and 1.45–4.52%. The recoveries at three concentration levels ranged from 82.7 to 116.3%. The proposed dSPE by using MSN-NH2 as sorbent coupled with HPLC was successfully applied to the analysis of synthetic dyes in real samples.

An Intelligent Cupreous Nanoplatform with Self-Supplied H2O2 and Cu2+/Cu+ Conversion to Boost Cuproptosis and Chemodynamic Combined Therapy

Xiangjie Tian,Hui Xu,FangFang Zhou,Xiyu Gong,Songwen Tan,Yongju He

Cuproptosis is a newly identified copper-dependent cell death and holds great promise for cancer therapy. However, transporting enough copper into cancer cells is a challenge. Herein, an intelligent cupreous nanoplatform (denoted as CuO2-MSN@TA-Cu2+), consisting of in situ formation of CuO2 within mesoporous silica nanoparticles (MSN) and then deposition with a tannic acid (TA)-Cu2+ complex, is designed and developed to realize on-demand copper delivery for cuproptosis-based combination therapy. CuO2-MSN@TA-Cu2+ exhibits tumor microenvironment-triggered therapeutic activity, wherein the outer TA-Cu2+ complex is readily disassembled to release Cu2+ and liberate the internal CuO2 to produce Cu2+ and H2O2. The overloaded Cu2+ can not only directly convert endogenous H2O2 and self-supplied H2O2 into highly toxic hydroxyl radicals for chemodynamic therapy (CDT) via Cu-based Fenton-like reaction but also undergo glutathione-mediated reduction into Cu+ species to induce potent cellular cuproptosis and enhance CDT. The experimental results indicate that CuO2-MSN@TA-Cu2+ produces remarkable cytotoxicity against cancer cells and significantly suppresses tumor growth by 93.42% in mice-bearing 4T1 breast tumors. This work provides a new paradigm to boost cuproptosis-related therapy and may also inspire the design of advanced therapeutic nanoplatforms.

Hybrid Mesoporous–Microporous Nanocarriers for Overcoming Multidrug Resistance by Sequential Drug Delivery

Liucan Wang,Haidi Guan,Zhenqiang Wang,Yuxin Xing,Jixi Zhang,Kaiyong Cai

Combination chemotherapy with a modulator and a chemotherapeutic drug has become one of the most promising strategies for the treatment of multidrug resistance (MDR) in cancer therapy. However, the development of nanocarriers with a high payload and sequential release of therapeutic agents poses a significant challenge. In this work, we report a type of hybrid nanocarriers prepared by polydopamine (PDA) mediated integration of the mesoporous MSN core and the microporous zeolite imidazolate frameworks-8 (ZIF-8) shell. The nanocarriers exploit storage capacities for drugs based on the high porosity and molecular sieving capabilities of ZIF-8 for sequential drug release. Particularly, large amounts of an anticancer drug (DOX, 607 μg mg–1) and a MDR inhibitor curcumin (CUR, 778 μg mg–1) were sequentially loaded in the mesoporous core via π–π stacking interactions mediated by PDA and in the microporous shell via the encapsulation during ZIF-8 growth. The sustained release of DOX was observed to follow earlier and faster release of CUR by acid-sensitive dissolution of the ZIF-8 shell. Furthermore, the nanoparticles showed good biocompatibility and effective cellular uptake in in vitro evaluations using drug-resistant MCF-7/ADR cancer cells. More importantly, the preferentially released CUR inhibited the drug efflux function of the membrane P-glycoprotein (P-gp), which subsequently facilitated the nuclear transportation of DOX released from the PDA-MSN core, and, in turn, the synergistic effects on killing MDR cancer cells. The hybrid mesoporous–microporous nanocarrier holds great promise for combination chemotherapy applications on the basis of sequential drug release.
