Endowed with superior tailorability and functionality, metal–organic frameworks (MOFs) have been exploited for their diverse applications in biomedicine. However, the instability of MOFs under an aqueous environment causes an accelerated degradation preventing their clinical applications and commercialization. To address this issue, studies have reported the incorporation of hydrophobic functional groups into MOF to enhance its stability. In this study, we aimed to fabricate a multifunctional coating on titanium implants by exploiting the synergy between Zn-based MOFs and raloxifene (Ral). We theorized that the integration of MOF and Ral will not only locally deliver the drug, but also take advantage of the hydrophobic functional group of Ral to enhance the stability of MOF in an aqueous environment. The release studies showed a sustained release of both Zn 2+ and Ral for more than 14 d. In-depth in vitro investigations also verified that MOF/Ral coatings significantly enhanced the cell viability and osteogenic differentiation of osteoblasts, as well as decreasing the tartrate-resistant acid phosphatase activity and osteoclastic biomarker expressions. In addition, both micro-computed tomography and immunohistochemical analysis further confirmed the efficacy of MOF/Ral coatings in the formation of new bone within the femurs of osteoporotic rats. The feasibility of MOF/Ral coated titanium implants in inhibiting osteolysis and promoting osteogenesis has undoubtedly proved their clinical prospects in the treatment of osteoporotic bone injury.

Gossypol is a poisonous substance in cottonseed which seriously harms the internal organs, blood vessels, nerves, and reproductive systems of organisms. Therefore, detecting gossypol is of great significance in the field of food safety. This manuscript constructs a novel target-oriented gossypol fluorescence probe based on the energy competition absorption mechanism. The probe is a zinc-based metal organic framework material (Zn-MOF) with blue fluorescence, which can achieve rapid and sensitive detection of gossypol through fluorescence “turn-off” effect. It has the advantages of low detection limit (16.6 nM), ultra-fast response time (8 s), and favorable selectivity. Interestingly, this probe can be reused and the fluorescence can still be recovered after five cycles when detecting gossypol. In addition, the application of the fluorescent probe in cottonseed oil and fetal bovine serum is reliable, with the recovery of 99.35%–101.5% and the relative standard deviation of less than 2.72%. These results indicate that the developed sensor has prominent application potential in the field of food and biological detection.

Diabetic wound healing is a great clinical challenge due to the microenvironment of hyperglycemia and high pH value, bacterial infection and persistent inflammation. Here, we develop a cascade nanoreactor hydrogel (Arg@Zn-MOF-GOx Gel, AZG-Gel) with arginine (Arg) loaded Zinc metal organic framework (Zn-MOF) and glucose oxidase (GOx) based on chondroitin sulfate (CS) and Pluronic (F127) to accelerate diabetic infected wound healing. GOx in AZG-Gel was triggered by hyperglycemic environment to reduce local glucose and pH, and simultaneously produced hydrogen peroxide (H 2 O 2 ) to enable Arg to release nitric oxide (NO) for inflammation regulation, providing a suitable microenvironment for wound healing. Zinc ions (Zn 2+ ) released from acid-responsive Zn-MOF significantly inhibited the proliferation and biofilm formation of S.aureus and E.coli . AZG-Gel significantly accelerated diabetic infected wound healing by down-regulating pro-inflammatory tumor necrosis factor (TNF)-α and interleukin (IL)-6, up-regulating anti-inflammatory factor IL-4, promoting angiogenesis and collagen deposition in vivo . Collectively, our nanoreactor cascade strategy combining “endogenous improvement (reducing glucose and pH)” with “exogenous resistance (anti-bacterial and anti-inflammatory)” provides a new idea for promoting diabetic infected wound healing by addressing both symptoms and root causes. Statement of Significance A cascade nanoreactor (AZG-Gel) is constructed to solve three key problems in diabetic wound healing, namely, hyperglycemia and high pH microenvironment, bacterial infection and persistent inflammation. Local glucose and pH levels are reduced by GOx to provide a suitable microenvironment for wound healing. The release of Zn 2+ significantly inhibits bacterial proliferation and biofilm formation, and NO reduces wound inflammation and promotes angiogenesis. The pH change when AZG-Gel is applied to wounds is expected to enable the visualization of wound healing to guide the treatment of diabetic wound. Our strategy of “endogenous improvement (reducing glucose and pH)” combined with “exogenous resistance (anti-bacterial and anti-inflammatory)” provides a new way for promoting diabetic wound healing.

L-lactate is an essential biomarker in clinical diagnostics and food quality assessment. This study introduces a novel ratiometric fluorescence sensor, RhB@Zn-MOF, which was specifically designed for the sensitive and selective detection of L-lactate. Through the strategic incorporation of Rhodamine B (RhB) into Zn-MOF, RhB@Zn-MOF was synthesized, exhibiting dual-emission properties and could effectively distinguish L-lactate in complex biological and food matrices such as milk and sweat based on the competitive absorption mechanism. Notably, the sensor achieves a low detection limit of 0.091 μM and demonstrates excellent stability and reproducibility in varied conditions. Furthermore, the integration of the sensor with smartphone technology enables rapid, real-time analysis, showcasing potential applications in sports medicine, clinical environments, and the food industry.

Here a new type of ferrocene confined in a metal–organic framework (Fc-Zn-MOF) as an electrochemical signal tag has been synthesized and applied in an immunosensor for highly sensitive amyloid-β (Aβ, as a model protein) detection. In the present work, an electrochemically active molecule of Fc was covalently bonded in a Zn-MOF via postsynthetic modification to obtain a flower-like Fc-Zn-MOF, in which the high-content Fc signal units were periodically arranged in a well-defined porous structure, showing enhanced electrochemical activity. As noticed, Fc molecules are chemically anchored on the Zn-MOF, which avoids leakage of signal molecules, protects those electrochemical activities from potential damage and exhibits good stability. The Fc-Zn-MOF signal tag was further explored in constructing an electrochemical immunosensor for Aβ detection, with a linear range from 0.0001 to 100 ng mL−1 and a detection limit of 0.03 pg mL−1, showing improved analytical performance. This work presents an example of an Fc-MOF as a signal tag for Aβ detection, which would encourage atypical research lines for MOFs in bioassays where the introduction of ordered structures promises new opportunities.

Reactive oxygen species (ROS) play a dual role in cellular processes, serving as essential signaling molecules at moderate levels but inducing oxidative stress and cell death in excess. Coronavirus infections significantly elevate intracellular ROS, disrupting cellular antioxidant defenses and promoting viral replication. Given the challenges posed by coronaviruses, including their high pathogenicity and cross-species transmission, new antiviral strategies are urgently needed. This study reports a newly designed zinc-based metal–organic framework (Zn-MOF) material, NJAU1, featuring a 2-fold interpenetrating helical porous structure synthesized through aqueous self-assembly technology. NJAU1 was characterized using advanced analytical techniques and demonstrated broad-spectrum antiviral activity against various coronaviruses, including PEDV, TGEV, and PDCoV. The underlying antiviral mechanisms of NJAU1 involve direct viral particle inactivation, inhibition of viral internalization and replication, ROS scavenging, anti-inflammatory and antioxidant activities, as well as Zn2+-mediated modulation of ferroptosis and apoptosis. This work highlights NJAU1 as a promising multifunctional antiviral material that offers a novel approach to the prevention and treatment of coronavirus infections.

ZIF-8-derived materials showed great potential for adsorbing and decomposing ozone, making them suitable for application in catalytic ozonation. This study synthesized four X-Zn-MOF-C (X = 4, 6, 8, 10) materials, among which 10-Zn-MOF-C exhibited the best catalytic performance, possibly due to its superior promotion of ozone decomposition into ∙OH. A probe method was employed to quantify ROS (∙OH, O 2 −· and 1 O 2 ) in the four X-Zn-MOF-C/O 3 systems. The results showed that the yield of ∙OH was lower than that of O 2 −· and 1 O 2 , however, ∙OH played a more critical role in IBP degradation. ROS quenching experiments further confirmed the significant contribution of ∙OH to pollutant removal. Further the investigation into the sources of ∙OH generation was conducted from two dimensions, catalyst sites and solution properties. Compared to the surface hydroxyl protonation of the catalyst, the synergistic effect between the exposed Zn 2+ and oxygen vacancy on the catalyst surface was more favorable for ∙OH generation on the catalyst surface, which mainly resulted from the formations of metal and vacancy hydroxyl groups. Meanwhile, the generation of ∙OH in the solution relied on the contribution of H 2 O 2 produced from pollutant degradation.

Self-supporting and degradable (Zn-MOF/SA/CNF) acts as multifunctional photoactive membrane. • Zn-MOF acts as a porous skeleton to enhance flux, SA as a binder, and CNF as a photo-Fenton catalyst. • Membrane shows 96–98% rejection of dye, 80% rejection of tetracycline and a flux of 50–60 L‧m −2 ‧h −1 ‧bar −1 . • Membrane shows antibacterial property, photo-Fenton induced flux recovery and acid-triggered regeneration. In this study, self-supporting multifunctional photoactive membranes with a sandwich structure were prepared via a green route using eco-friendly materials for water treatment. The Zn-BDC synthesized at 25 °C was employed to construct a strippable skeleton with high flux. Sodium alginate (SA) acted as both a binder and cavity filler to repair defects and enhance tensile properties. Meanwhile, C₃N₄-Fe₂O₃ (CNF) was synthesized and applied as a photo-Fenton catalyst. The results showed that both the membrane with SA on the top (MCS) and the membrane with SA in the middle (MSC) achieved over 98% rejection toward Congo red, Alcian blue, and nickel(II) phthalocyanine-tetrasulfonic acid tetrasodium salt, with a flux ranging from 50 to 70 L·m −2 ·h −1 ·bar −1 . While the MSC membrane displayed better photocleaning performance and mechanical flexibility than the MCS membrane. Its flux could be restored via the photo-Fenton reaction. Moreover, the MSC membrane also attained over 80% rejection of tetracycline with the assistance of the photo-Fenton reaction. Furthermore, the membrane showed over 95% dye rejection in ethanol and exhibited favorable antibacterial performance. In addition, the membrane could be dissolved in a citric acid solution, which offers a convenient strategy for membrane disposal or recycling. Collectively, this work provides a green and sustainable membrane candidate for water treatment applications. Download: Download high-res image (129KB) Download: Download full-size image

An electrochemical sensor with a dual recognition strategy was designed using molecularly imprinted polymers (MIPs) and boric acid for the highly sensitive detection of rutin. The synthesized metal-organic framework (MOFs) is combined with phenylboronic acid, which increases the recognition area of phenylboronic acid and its recognition ability towards adjacent diol compounds. Meanwhile, the molecularly imprinted polymer (MIP) provides specific recognition for rutin. The dual recognition strategy is widely adopted in sensors, enhancing the specificity of the imprinted cavity and effectively addressing the influence of interfering substances in complex matrices. After optimizing the experimental parameters, the detection range of this sensor is from 1 nM to 200 nM, and the limit of detection is 0.43 nM, indicating that this sensor has high potential for practical applications.

Cu/Zn-MOF nanozyme exhibits robust triple-enzyme activities. • It promotes healing by combining antibacterial, antioxidant, and pro-proliferative effects. • This work provides a strategy for designing multifunctional nanoenzyme platform for efficient wound therapy. The treatment of infected wounds is severely challenged by bacterial resistance and dysregulated healing processes. Herein, we report a bimetallic Cu/Zn-MOF nanozyme fabricated via a facile self-assembly strategy. The material exhibits robust triple-enzyme mimetic activity (superoxide dismutase (SOD), glutathione peroxidase (GPx), and ascorbate peroxidase (APx)) enabling efficient reactive oxygen species (ROS) scavenging to alleviate oxidative stress and inflammation. In vitro, Cu/Zn-MOF demonstrated potent, broad-spectrum antibacterial activity, primarily associated with disruption of bacterial nucleotide metabolism and energy homeostasis, while also promoting fibroblast proliferation and migration. In an infectious wound model, Cu/Zn-MOF treatment significantly accelerated wound closure, enhanced collagen deposition, and facilitated orderly tissue regeneration, achieving a healing rate of 93.4% by day 12. This work presents a versatile nanozyme platform that intelligently orchestrates the sequential stages of wound healing, offering a promising multi-functional strategy for regenerative medicine.

Bone regenerative biomaterials are essential in treating bone defects as they serve as extracellular matrix (ECM) mimics, creating a favorable environment for cell attachment, proliferation, and differentiation. However, the currently used bone regenerative biomaterials mostly exhibit high stiffness, which may lead to difficulties in degradation and thus increase the risk of foreign body ingestion. In this study, we prepared soft fibrous scaffolds modified with Zn-MOF-74 nanoparticles via electrostatic spinning. The soft fibers (only 1 kPa) permit remodeling under cellular adhesive force, optimizing the mechanical cues in the microenvironment to support cell adhesion and mechanosensing. In addition, the incorporation of Zn-MOF-74 nanoparticles enables the stable and sustained release of zinc ions, promoting stem cell mechanotransduction and osteogenic differentiation. Therefore, the hybrid soft fibers facilitate the regeneration of new bone in the rat femoral defect model, which provides a promising approach for regenerative medicine.

In the zinc ion capacitors (ZICs), Zn2+ storage is limited by the common carbon-based cathodes due to the sluggish kinetics of zinc ion diffusion. Porous carbon derived from metal–organic frameworks (MOFs) is a good candidate with the abundant micropores and mesopores. By using a one-step template-free solvothermal method, double-shell hollow Zn-MOF microspheres were directly synthesized, eliminating the traditional multistep activation process using KOH/HCl. During the carbonization procedure, urea has been used to provide an additional carbon source and activate the derived carbon, which is due to urea pyrolysis products, such as NH3, cyanuric acid, and ammelide. The derived porous carbon (U-MPC) exhibits a high specific surface area of 1695.66 m2 g–1. The ZIC device assembled with U-MPC as the cathode has a high specific capacitance of 199.0 F g–1 and a high energy density of 70.8 Wh kg–1 at 0.1 A g–1. The Zn//U-MPC ZICs demonstrate a good antiself-discharge performance, with a low self-discharge rate of only 3.1 mV h–1 over 300 h, and can maintain 59.7% of the initial capacitance. Moreover, the ZICs exhibit good cyclic stability, showing a slight decrease after 20,000 cycles at 5 A g–1, and the Coulombic efficiency is 90%.
