Chemiluminescence of Cl-NCDs and KMnO4 systems based on waste pomelo peel for erythromycin detection

Hao Xue,Hong Zheng,Jing Xiang,Ke Cheng,Biyue Zhu

In this work, using discarded white pomelo peel as the green precursor and hydrochloric acid (HCl) as the dopant, water-soluble Cl-N-doped carbon dots (Cl-NCDs) with excellent performance were synthesized by one-step hydrothermal method. The Cl-NCDs function as both luminescent agents and catalysts, significantly enhancing the chemiluminescence intensity of the HCl-HCHO-KMnO 4 system, forming a novel HCl-HCHO-(Cl-NCDs)-KMnO 4 chemiluminescence system. The addition of erythromycin (EM) solution inhibits the chemiluminescence intensity of new system. By optimizing the conditions, we established a simple, cost-effective, and reliable chemiluminescence method for EM detection, offering a linear range of 5-100 μM and a detection limit of 0.0149 μM. In addition, this method was successfully applied to the analysis of EM in various water simples, achieving recovery rates ranging from 97.6% to 106%.

Thermosensitive Polyhedral Oligomeric Silsesquioxane Hybrid Hydrogel Enhances the Antibacterial Efficiency of Erythromycin in Bacterial Keratitis

Lan Zheng,Ying Chen,Yi Han,Jingwei Lin,Kai Fan,Mengyuan Wang,Ting Teng,Xiuqin Yang,Lingjie Ke,Muyuan Li,Shujia Guo,Zibiao Li,Yunlong Wu,Cheng Li

Bacterial keratitis is a serious ocular infection that can impair vision or even cause blindness. The clinical use of antibiotics is limited due to their low bioavailability and drug resistance. Hence, there is a need to develop a novel drug delivery system for this infectious disease. In this study, erythromycin (EM) was encapsulated into a bifunctional polyhedral oligomeric silsesquioxane (BPOSS) with the backbone of the poly-PEG/PPG urethane (BPEP) hydrogel with the aim of improving the drug efficiency in treating bacterial keratitis. A comprehensive characterization of the BPEP hydrogel was performed, and its biocompatibility was assessed. Furthermore, we carried out the evaluation of the antimicrobial effect of the BPEP-EM hydrogel in S. aureus keratitis using in vivo mouse model. The BPEP hydrogel exhibited self-assembling and thermogelling properties, which assisted the drug loading of drug EM and improved its water solubility. Furthermore, the BPEP hydrogel could effectively bind with mucin on the ocular surface, thereby markedly prolonging the ocular residence time of EM. In vivo testing confirmed that the BPEP-EM hydrogel exerted a potent therapeutic action in the mouse model of bacterial keratitis. In addition, the hydrogel also exhibited an excellent biocompatibility. Our findings demonstrate that the BPEP-EM hydrogel showed a superior therapeutic effect in bacterial keratitis and demonstrated its potential as an ophthalmic formulation.

Portable smartphone-assisted amperometric immunosensor based on CoCe-layered double hydroxide for rapidly immunosensing erythromycin

Bin Hu,Yifei Wang,Haosen Jia,Xiaohong Shang,Fenghe Duan,Chuanpan Guo,Shuai Zhang,Minghua Wang,Zhihong Zhang

Herein, we have manufactured a newly designed bifunctional impedimetric and amperometric immunosensor for rapidly detecting erythromycin (ERY) in complicated environments and food stuffs. For this, bimetallic cobalt/cerium-layered double hydroxide nanosheets (CoCe-LDH NSs), which was derived from Co-based zeolite imidazole framework via the structure conversion, was simultaneously utilized as the bioplatform for anchoring the ERY-targeted antibody and for modifying the gold and screen printed electrode. Basic characterizations revealed that CoCe-LDH NSs was composed of mixed metal valences, enrich redox, and abundant oxygen vacancies, facilitating the adhesion on the electrode, the antibody adsorption, and the electron transfers. The manufactured impedimetric and amperometric immunosensor based on CoCe-LDH has showed the comparable sensing performance, having a wide linear detection range from 1.0 fg mL −1 to 1.0 ng mL −1 with the ultralow detection limit toward ERY. Also, the portable, visualized, and efficient analysis of ERY was then attained at the smartphone-assisted CoCe-LDH-based SPE.

Combined effects of nanosized polystyrene and erythromycin on bacterial growth and resistance mutations in Escherichia coli

Qing Ning,Dali Wang,Jiahui An,Qi Ding,Zhiyi Huang,Yue Zou,Fan Wu,Jing You

Toxicological effects of nanoplastics have been demonstrated in a variety of organisms, yet their impacts on bacteria, especially on the antibiotic resistance evolution remain under explored. Herein, we report individual and combined effects of nano-polystyrene (nano-PS) and erythromycin (ERY) on growth and resistance mutations of Escherichia coli . The toxicity of nano-PS was dependent on size and functional modifications, with 30 nm and amino-modified PS (PS-NH 2 , 200 nm) showing the greatest toxicity. Adsorption of nano-PS onto bacterial surface and the subsequent increase of intracellular ROS or the probable mechanical damage were considered as the primary toxic mechanisms. Furthermore, nano-PS increased the bacterial resistance mutations, which was due to the oxidative damage to DNA and the SOS response. In addition, PS-NH 2 presented synergistic effects with ERY while non-modified PS had no impact, although both of them showed adsorption capacity to ERY. This was likely because the positively charged PS-NH 2 acted as a carrier of ERY and enhanced the interactions between ERY and the bacteria. Our findings raised the concerns about the risk of nanoplastics in accelerating the bacterial resistance evolution, and highlighted the necessity of including combined effects of nanoplastics and co-contaminants in risk assessment.

SiO2-MIP core-shell nanoparticles containing gold nanoclusters for sensitive fluorescence detection of the antibiotic erythromycin

Zhang Yufeng,Zhou Zhiping,Zheng Jiahong,Li Hongji,Cui Jiuyun,Liu Siwei,Yan Yongsheng,Li Chunxiang

The authors describe a fluorescence based assay for determination of the macrolid antibiotic erythromycin (ERY). It is based in the use of fluorescent gold nanoclusters (AuNCs) coated first with silica (to obtain AuNC@SiO 2 ) and then with a molecularly imprinted polymer (MIP). The MIP was synthesized from 3-aminopropyltriethoxysilane as the functional monomer, ERY as the template, and tetraethoxysilane as the cross-linker via a sol-gel process that leads to surface imprinting. By using this method, the strong fluorescence of AuNCs was maintained in the resultant MIPs. After optimization of the experimental conditions, the signal (measured at excitation/emission wavelengths of 396/585 nm) decreases linearly with increasing concentration of ERY in the 0.1 to 11.9 μM ERY concentration range. The limit of detection is 12 nM, and the imprinting factor is 4.0. The method was successfuly applied to the determination of ERY in spiked human urine and saliva. Schematic of the preparation of a core-shell sensor material with the silica coated by fluorescent gold nanoclusters as core and a molecularly imprinted polymer. It can be used to selectively detect residual erythromycin in biological samples.

Thermal stability enhancement of erythromycin esterase C from Enterobacter hormaechei through a combinatorial greedy accumulated strategy

Yulei Zhang,Zhaolin Huang,Wenli Zhang,Dawei Ni,Wei Xu,Cuie Guang,Wanmeng Mu

The presence of macrolides in food and the environment poses a substantial threat to the sustainable development of natural ecosystems and presents risks to human health. Previous research has demonstrated that microbial esterases can degrade erythromycin (Ery), roxithromycin (Rox), and clarithromycin (Clr), rendering them candidates for bioremediation of macrolide residues. However, the efficacy of esterases is limited by insufficient thermal stability, restricting their widespread application. To address this challenge, this study employed a combinatorial strategy integrating computational molecular dynamics (MD) simulations, server-assisted virtual mutation prediction, and a greedy algorithm for protein engineering to enhance the thermal stability of esterase C from Enterobacter hormaechei ( Ehno -EreC). After four rounds of screening, we successfully obtained a four-point mutant, M4 (D245P/S141A/D154I/T292W), which exhibited significantly improved thermal stability compared to the wild-type enzyme. Notably, the M4 mutant not only extended the enzyme's half-life at 40 °C from 7 to 1095 min, but also increased its degradation activity against Ery, Rox, and Clr. Structural analyses revealed enhanced rigidity in the loop between the major and minor lobes and the strengthened hydrophobic interactions post-mutation, contributing to the enzyme's structural stability. This finding was consistent with the observed reduction in correlated and anti-correlated motions in the MD trajectory.

Molecular mechanism of crisaborole combined with erythromycin against methicillin-resistant Staphylococcus aureus in vivo and in vitro

Yinuo Zou,Lei Yu,Jinjing Xue,Tianqi Fang,Liping Sun,Jianfeng Wang,Minhe Cui,Peng Zhang,Yonglin Zhou

The widespread presence of methicillin-resistant Staphylococcus aureus (MRSA) severely threatens convenient therapeutic options in the postantibiotic era. The use of combinations of existing drugs at this stage may be a viable strategy for dealing with complex drug-resistant MRSA infections. An checkerboard MIC (Minimum Inhibitory Concentration) assay, growth curve assay, bactericidal test and scanning electron microscope (SEM) assays were performed to determine whether crisaborole (AN2728), a PDE4 inhibitor for treating atopic dermatitis (AD), produced bactericidal effect with different antibiotics. Here, we identified (AN2728) produced a significant synergistic bactericidal effect with erythromycin, cefuroxime and rifampicin against different bacterial strains of Staphylococcus aureus (S. aureus), especially MRSA (FIC < 0.5) (p < 0.05). Transcriptome analysis, bacterial biofilm assay and several kit assays revealed that AN2728 could also simultaneously affect the membrane and transporter capacity of MRSA. Moreover, in a mouse skin infection model of MRSA, the combination of AN2728 and erythromycin showed remarkable treatment benefits, as shown by significantly reduced bacterial loading (p < 0.05), pathological lesions of the skin and an obvious anti-inflammatory effect (p < 0.05). To our knowledge, this study is the first to establish that AN2728 can cooperate with antibiotics such as erythromycin to completely kill MRSA and that AN2728 can be used to extend the usage life of different antibiotics to address the inevitability of severe MRSA infection.

Preparation of erythromycin imprinted polymer by metal-free visible-light–induced ATRP and its application in sensor

Sun Yue,Zhao Mengyuan,Liu Yutong,Fu Liye,Li Siyu,Yang Yifei

Erythromycin (ERY) molecular imprinted polymer was prepared on the surface of Au electrode modified with Ni and Au nanoclusters (MIP/AuNCs/Ni/Au) via metal-free visible-light–induced atom transfer radical polymerization, which was very easy, controllable, and environmentally benign. MIP/AuNCs/Ni/Au was examined by energy dispersive spectrometer (EDS), scanning electron microscope (SEM), cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). Further, MIP/AuNCs/Ni/Au was successfully used for the determination of ERY by differential pulse voltammetry (DPV). Under the optimal conditions, MIP/AuNCs/Ni/Au gave a linear correlation over the ERY concentration from 1.0 × 10 −8 to 1.0 × 10 −1  mg/L with the detection limit of 3.2 × 10 −9  mg/L ( S / N  = 3). Comparing with the similar sensors, the larger linear range and lower detection limit indicated the promising prospect of this electrochemical sensor. Hence, this study was of great importance for the polymer-modified electrodes and the sensitive detection of ERY.

Short-term performance of enhanced biological phosphorus removal (EBPR) system exposed to erythromycin (ERY) and oxytetracycline (OTC)

Zhetai Hu,Peide Sun,Zhirong Hu,Jingyi Han,Ruyi Wang,Liang Jiao,Pengfei Yang

The effects of Erythromycin (ERY) and oxytetracycline (OTC), including individual and combinative effect, on enhanced biological phosphorus removal (EBPR) system within a short-term (24   h) were evaluated in this study. Results showed that the P-removal efficiency decreased to 34.6% and 0.0% under the effect of ERY (10   mg/L) and OTC (10   mg/L) for 24   h. OTC concentration higher than 5   mg/L was sufficient to cause serious adverse impact on the EBPR performance. While the performance of EBPR system will be impacted by ERY above 10   mg/L. OTC, due to its special antibacterial action to the gram-negative bacteria which most PAOs belong to, has more serious negative effect on the EBPR performance than ERY does. Moreover, in the combined antibiotics test, neither synergistic nor antagonistic effect was detected between ERY and OTC. Finally, ERY (10   mg/L) and OTC (10   mg/L) could inhibit the microorganisms’ activity, while couldn’t induce serious microorganisms death within 24   h.

Degradation of antibiotics and antibiotic resistance genes in erythromycin fermentation residues using radiation coupled with peroxymonosulfate oxidation

Libing Chu,Dan Chen,Jianlong Wang,Zhiling Yang,Yunpeng Shen

Ionizing radiation coupled with peroxymonosulfate (PMS) oxidation was developed to degrade antibiotics and antibiotic resistance genes (ARGs) from the erythromycin fermentation (EryF) residual wastes. The experimental results showed that the ERY content and ARGs abundance decreased with increase of the absorbed dose and PMS dosage and gamma irradiation was more effective to abate ARGs from the EryF wastes. The removal efficiency of ERY reached 49–55% and more than 96–99% of ARGs (1.32–2.55 log) was eliminated with the absorbed dose of 25–50 kGy and PMS dosage of 50–100 mM. Illumina pyrosequencing revealed that 3 bacterial phyla, Proteobacteria , Firmicutes and Fusobacteria were highly enriched and the ARGs-linked hosts were affiliated to the genera Aeromonas , Enterobacteriaceae and Enterobacter in the phylum Proteobacteria. The abundance of the ARGs-linked bacteria decreased by gamma/PMS treatment. Ionizing radiation/PMS treatment with the doses of 25 kGy and 50 mM PMS is proposed for potential practical application.

Fabrication of magnetic imprinted sorbents prepared by Pickering emulsion polymerization for adsorption of erythromycin from aqueous solution

Yanzhuo Zhu,Dayu Jiang,Dongshu Sun,Yongsheng Yan,Chunxiang Li

In this work, erythromycin-based (EM-based) magnetic molecularly imprinted polymers (MMIPs) were prepared by magnetic eggshells-stabilized Pickering emulsion imprinting polymerization. The feature of easy separation caused by good magnetism made the product separation and adsorption process more time-saving and convenient. More importantly and interestingly, the magnetic eggshell particles were not prepared beforehand, but were found to form instantly and spontaneously in water phase. The magnetic eggshell particles created in this way were used as stabilizers of emulsion polymerization for the first time, which may be benefit for the preservation of magnetism of MMIPs. The imprinting process was conducted by radical polymerization and the MMIPs was characterize by various characterization methods such as Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and vibrating sample magnetometer (VSM). The results sufficiently demonstrated the formation of the approximately spherical and wrinkled magnetic imprinted polymers. A series of adsorption experiments showed that the as-prepared MMIPs exhibited not only outstanding adsorption capacities, but also excellent selective adsorption effect toward EM molecules, and the adsorption mechanism were also discussed in detail with different adsorption models.

The key molecular mechanisms of antagonism induced by combined exposure to erythromycin and roxithromycin in Chlorella pyrenoidosa

Feifan Wu,Xiangjie Pan,Yuhao Zhou,Yan Zhu,Kai Liu,Wei Li,Jiangang Han

Emerging pollutants such as antibiotics have raised great concern in recent years, but the complex coexistence of multiple antibiotics in the environment poses a new challenge in the accurate assessment of the toxicity of antibiotics to aquatic organisms such as microalgae. In this study, the mechanism of action of a combination of erythromycin (ERY) and roxithromycin (ROX) on Chlorella pyrenoidosa was illustrated based on the physiological–biochemical response and transcriptomic analysis. The results revealed an inhibitory effect on the biomass of C. pyrenoidosa at 14 d in all treatment groups, whereas an antagonistic effect was observed in the coexposure groups. The photosystem was the main target despite the existence of multiple compensatory mechanisms, such as expanding the antenna size and initiating alternative electron carriers. The intercept of electrons on the donor side of PSI limited the production of energy, whereas the adjustment of the content and ratio of pigments strengthened microalgal adaptation. Enzymes and genes related to the degradation of exogenous compounds, including cytochrome P450 (P450), glutathione S-transferase (GST) and ABC transporters, mediated the detoxification of antibiotics. The upregulated expression of related genes induced by coexposure increased resistance and explained the antagonistic effects. The shift in energy allocation by increasing the proportion of lipids met the urgent requirements of microalgal physiological activities. This study reemphasizes the modes of interactions between multiple antibiotics and provides new insights into the mechanisms of antagonism induced by combinations of antibiotics.
