A electrochemical biosensor was designed utilizing a CRISPR Cas9n-driven DNA walker combined with gold-nanosphere-like covalent organic frameworks (COFs-AuNPs) to detect breast cancer markers (PIK3CA E545K ctDNA). The DNA walker probe is activated only in the presence of circulating tumor deoxyribonucleic acid (ctDNA), binding to a support probe to form a double strand that is then specifically cleaved by the Cas9n/sgRNA complex. This cleavage produces numerous DNA fragments for signal amplification. The COF-AuNPs as electrode materials facilitate electronic transfer and provide additional active sites for the immobilization of nucleic acid probes. This setup achieves a detection limit of 1.76 aM, demonstrating high sensitivity. Additionally, Cas9n improves the specificity of the sensor, accurately distinguishing a pair of base-mismatched sequences, and reducing the occurrence of false positives. Overall, the sensor exhibits excellent selectivity, reproducibility, and potential for early diagnosis of breast cancer. Graphical Abstract

Given the frequent occurrence of multidrug-resistant bacteria (MDRB), which pose a severe threat towards public hygiene, it is important to construct facile and sensitive strategies for the detection of MDRB. We developed a fluorescent/colorimetric dual-mode biosensing platform for the analysis of MDRB by integrating the remarkable peroxidase-like activity of boric acid-decorated fluorescent framework hybrid (MOF@COF) nanozyme and enhanced bacterial affinity of DNA programmable multivalent aptamer scaffold. The efficiency and sensitivity of the proposed analysis approach were dramatically increased due to richer aptamers and high-performance MOF@COF nanozyme. Compared with commonly used single recognition methods, multivalent aptamers and bacteria-affinity boric acid groups on the nanozyme surface could identify multiple sites of MDRBs to improve site utilization for ultrasensitive biosensing. The platform realized selective detection of Acinetobacter baumannii (AB) and Pseudomonas aeruginosa (PA) in two modes with consistent results. In colorimetry and fluorescence modes, the linear ranges of AB were 10 2 -10 8 and 10–10 8 CFU/mL with corresponding detection limits of 12 and 2 CFU/mL, and the linear ranges of PA were 10 2 -10 8 and 10–10 8 CFU/mL with corresponding detection limits of 10 and 3 CFU/mL, respectively. More importantly, the biosensing platform could be applied for MDRB quantification in biological specimens, including human serum, cerebrospinal fluid, and urine with remarkable recoveries (96.8–105.0%) in both modes, indicating the bright prospect of the biosensing platform for MDRB determination in clinical diagnosis.

The development of highly active photocatalysts for H 2 O 2 production is of significant practical importance for addressing environmental issues and energy shortages. In this study, an S-scheme heterojunction photocatalyst with strong redox performance was synthesized through the in-situ growth of the TpPa-Cl covalent organic framework (COF) material on the surface of Cu-loaded ZnO nanorods. The incorporation of Cu disrupts the charge shielding effect of ZnO, promotes the spatial separation of charge carriers, and enhances piezoelectric catalytic activity. Furthermore, the porous ultra-thin COF layer extends carrier lifetime and increases the contact area with reactants, endowing the composite with strong light absorption capacity and excellent oxygen reduction performance. Under the combined influence of the piezoelectric-photocatalytic dual electric field, the H 2 O 2 yield of the composite in pure water reached 1838.8 μmol g −1 h −1 , and the resulting H 2 O 2 solution can be directly utilized for pollutant removal and water disinfection. In situ X-ray photoelectron spectroscopy, density functional theory calculations, and electron spin resonance studies elucidate the S-scheme electron transfer mechanism, which accelerates the transfer of photogenerated carriers and enhances the utilization of electron-hole pairs. This research offers a novel approach for the design and development of catalysts aimed at efficient H 2 O 2 production.

The increasing presence of pyrethroid pesticides (PYs) in agricultural products necessitates advanced analytical methods for food safety monitoring. This study introduces a novel metal–organic framework–covalent organic framework (MOF–COF) hybrid adsorbent, NH₂–MIL–101(Fe)@TpBD, for the rapid extraction of PYs from eggplants via pH/salt–independent dispersive solid–phase extraction (DSPE) coupled with high-performance liquid chromatography (HPLC). The optimized method achieved efficient extraction of four PYs (permethrin, cyfluthrin, esfenvalerate, and bifenthrin) within 4 min, using 20 mg of adsorbent and 1.5 mL of acetonitrile. The method demonstrated excellent linearity (0.12–8.00 mg·kg −1 ), high sensitivity (LODs: 0.0416–0.0515 mg·kg −1 ), and robust recoveries (85.5–107.6 %), with enrichment factors of 389–410. Adsorption mechanisms were dominated by chemisorption, facilitated by π–π stacking and hydrogen bonding. The rapidity and efficiency of this method highlight its potential for high-throughput food safety analysis. This work underscores the synergistic integration of MOF/COF hybrids as advanced adsorbents for pesticide monitoring in complex food matrices.

Herein, a novel covalent-organic framework (COF) coated Fe doped hollow carbon nanosphere (HCNS), Fe-HCNS@COF, was prepared for selective adsorption and photocatalytic reduction on dye pollutant in water. The Fe-HCNS@COF displays a high affinity to malachite green (MG) with a distribution coefficient ( K d ) of 6.931, which can be attributed to the confinement effect by the π−π stacking between the benzene ring in MG and triazine ring in COF. Pseudo second order (PSO) kinetics ( R 2 = 0.9914) and Langmuir isotherm model ( R 2 = 0.970 9) match better to Fe-HCNS@COF on MG adsorption indicating a chemisorption with mono-layer interface. Additionally, enhanced catalytic efficiency of the nanospheres on MG reduction at 96.3% was observed under sunlight irradiation with improved reusability after five cycles (74.4%). The catalytic mechanism investigation indicates that 1 O 2 , •O 2 − , and •OH play significant role in the catalytic process, and Fe-HCNS@COF exhibits a smaller E g at 1.53 eV compared to COF (1.86 eV), signifying enhancement for separation of hole-electron pairs. A continuous flow device was established to investigated the real application in dye removal, achieving an 88.7% removal rate for MG degradation after 40 h continuous operation. Moreover, an improved bacterial inhibition rate over 99% was found against both Gram-negative and Gram-positive bacteria. In conclusion, the as prepared Fe-HCNS@COF provides us a novel nano-platform with multiple-function for the potential application of dye removal in real water management.

The rational design of efficient photocatalysts to achieve artificial nitrogen fixation is an urgent challenge. Herein, we combined donor–acceptor covalent organic framework with iron-doped nitrogen vacancy graphitized carbon nitride (D-A COF/Fe-g-C 3 N x ) for photocatalytic nitrogen fixation. The photocatalyst exhibited good crystallinity, high porosity, and a large specific surface area. Without a sacrificial agent, the optimal 40 % D-A COF/Fe-g-C 3 N x exhibited an excellent rate of ammonia production (646 μmol h −1 g −1 ) at 420 nm, and durable stability after successive cycling. Exhaustive experimental research and theory calculations verified that the D-A unit and Fe doping redistributed the distribution of the charge, which enhanced the visible light utilization and provided chemisorption sites for further polarization. Besides N -vacancies can serve as electron-trapping active sites to promote the directional migration of carriers. The reaction mechanism demonstrated that superoxide radical and hydrogen peroxide were formed by electron and hole, respectively, which promote the reduction of nitrogen to ammonia. This work provides a new idea for the rationalizing design of efficient catalysts for photocatalytic nitrogen fixation under mild conditions.

Simple and efficient bacterial detection is important for the diagnosis of foodborne diseases caused by bacteria. Herein, we proposed a new electrochemical biosensor based on Zn 2+ -triggered swing-arm robot and multifunctional covalent organic framework-based nanocomposite (COF@Au-Thi/H4) for ultrasensitive detection of Escherichia coli ( E. coli ). COF, as an efficient nanocarrier for DNA and signal molecules, has efficient hybridization and loading capabilities. Its high porosity provides a large number of attachment sites for signal molecule thioredoxin (Thi). The biosensor used poly(diallyldimethylammonium chloride)-functionalized graphene/Ag@Au nanosheets (PDDA Gr/Ag@AuNS) as the electrode modification materials and COF@Au-Thi/H4 as the signal probes. The aptamer-containing arms and S2 probe formed triple helix molecular switch (THMS). When E. coli were present in the system, specific interaction between the aptamer and target caused the S2 probe to be released from the THMS, which triggered strand displacement reaction (T-SDR) to release S1 probe and excess Zn 2+ under acid dissolution. These released probes activated the swing-arm robot. The Zn 2+ -dependent DNAzyme cycle cleaved the substrate DNA strand, causing the exposure of DNA complementary to hybridize with COF@Au-Thi/H4 signal probe, thus generated significant current signal for bacteria detection, with the detection limit (LOD) of 10 CFU/mL. The detection time was less than 3 h. In addition, the method could detect real samples, which has broad prospects in biological analysis and disease diagnosis applications.

In order to study the adsorption interaction and behavior of organophosphate pesticides (OPPs) on the surface of different microplastics, a Zr-covalent organic framework (COF)-based magnetic solid-phase extraction (MSPE) was established. Due to various interactions (π-π interaction, hydrogen bonding and Zr-O-P coordination interactions), Zr-COF exhibited higher adsorption capacities towards five OPPs than COF without Zr through the theoretical calculation and analysis. The established MSPE-HPLC method showed the detection limit of 0.011–0.019 mg L −1 for OPPs. It was used to determine the adsorption performance of four microplastics towards OPPs. Small sized and aging microplastics showed more adsorption capacities than the large sized and original microplastics. Polylactic acid exhibited higher adsorption capacities than most OPPs, especially with the N element, -NO 2 , and P=O groups, due to the presence of hydrogen bonding. Besides, the method matrix effect was between 2.90%-17.84% in real samples, and the adsorption capacity in juice was significantly higher than that in aquatic samples because of the acidic environment.

Monitoring seafood freshness is critical for ensuring nutritional value and food quality, but targeted analysis in complex matrices remains challenging. Herein, a thiol-functionalized calix[6]arene covalent organic framework grafted with AuNPs, denoted as CX6-SCOF-AuNPs, was strategically designed for rapid, sensitive and selective detection of tryptamine in seafood by surface-enhanced Raman spectroscopy (SERS). CX6-SCOF-AuNPs featured extensive pocket-like hydrophobic cavities integrated within an ordered porous structure, enabling specific capture of tryptamine with exceptional enrichment capacity (203.4 mg/g) and rapid adsorption kinetics (within 2 min). The selectivity mechanism stemmed from the formation of a ground-state charge-transfer complex driven by specific “pocket effect” between CX6-SCOF and size-matched tryptamine. CX6-SCOF-AuNPs was successfully applied to enrichment and detection of tryptamine in seafood, achieving high sensitivity (LOD of 3.4 μg/L) and satisfactory recoveries (90.6 % – 107.4 %). This work provides a promising strategy for supramolecular recognition and targeted SERS analysis of trace hazards in complex food matrices.

It is a major challenge to combine the advantages of two kinds of two-dimensional materials to construct a heterojunction and achieve efficient photocatalytic antifouling. In this work, we covalently connected two materials MXenes and covalent organic frameworks (COFs) through the Schiff base reaction and anchored Ag nanoparticles (NPs) to prepare a Ti 3 C 2 /TpPa-1/Ag composite material with high efficiency bactericidal properties. The covalent bonding between MXene and COF greatly improved the stability of the material. Ti 3 C 2 /TpPa-1/Ag composite showed an excellent antibacterial property against S. aureus and P. aeruginosa . The fluorescence spectra of Ti 3 C 2 /TpPa-1/Ag proved that the electron transfer channels formed between the ternary materials could greatly improve the efficiency of carrier separation and prolong the life of photogenerated carriers. Density functional theory calculations showed that the synergistic catalytic effect of Ag and Ti 3 C 2 could greatly reduce the work function along the interface, and the built-in electric field between the layers drive carrier fast migration, which effectively improve the catalytic performance.

Most of the reported covalent organic frameworks (COFs) are hydrophobic, limiting their adsorption application in sample pretreatment field. In this work, sulphonate functionalized magnetic covalent organic frameworks (COFs) composites were first synthesized by loading gold nanoparticles on Fe 3 O 4 @COF(TpBD) surface and then functionalized by sodium 3-mercaptopropanesulphonate immobilization via Au-S bonding formation (denoted as Fe 3 O 4 @COF(TpBD)@Au-MPS nanocomposites), which were further utilized as adsorbents for magnetic solid-phase extraction (MSPE) of fluoroquinolones. Compared with Fe 3 O 4 @COF(TpBD), the composites exhibited higher affinity to fluoroquinolones. Under optimized conditions, the developed MSPE method coupled with HPLC–MS/MS showed good linearity (R 2 ≥0.9989) and yielded low limits of detection (0.1–1.0 μg kg− 1 ) for fluoroquinolones. Moreover, the proposed method was successfully applied to extract fluoroquinolones from spiked meats (pork, chicken and bovine). The satisfactory recoveries were in the range of 82–110.2% with the relative standard deviations (RSDs) lower than 7.7%. These results indicated that the Fe 3 O 4 @COF(TpBD)@Au-MPS is a promising magnetic adsorbent for trace fluoroquinolones determination in meat samples. This work not only provided a facile strategy for COF functionalization, but also developed an efficient method for detecting fluoroquinolones in foodstuffs.

High-performance proton exchange membranes (PEMs) that simultaneously possess high proton conductivity and low methanol permeability is crucial for direct methanol fuel cell (DMFCs) applications. Herein, a sulfonated covalent organic framework (COF–SO 3 H) was synthesized and incorporated into a sulfonated polybenzimidazole (SPBI) matrix to fabricate composite proton exchange membranes. Acid-base interactions between sulfonic acid and protonated imidazole groups under acidic conditions to form ionic self-crosslinked structure, effectively reducing free volume size to suppress methanol permeability. The hydrophilic COF–SO 3 H improved water uptake and facilitated continuous proton-conducting pathways through its two-dimensional structure. The optimized membrane exhibited a high proton conductivity of 106.13 mS cm −1 at 80 o C. Furthermore, the SPBI/COF–SO 3 H-1.5 % membrane with thickness of 10 μm achieved a low methanol crossover current density of 187.10 mA cm −2 and outstanding power density of 152.67 mW cm −2 under 2 M methanol concentration, significantly outperforming those of commercialized Nafion 115. This study presents a promising strategy for high-performance PEMs in DMFCs system.
