Curcumin (CUR) possesses notable health benefits but suffers from poor solubility, instability, and low bioavailability. To address these limitations, this study developed a novel starch-chitosan composite cross-linked with citric acid (CA). The modified starch was employed in the fabrication of emulsion and emulsion gel beads, which were systematically evaluated based on rheological properties, texture, moisture distribution, and in vitro digestibility. The starch-based emulsion exhibited higher storage modulus and loss modulus compared to the emulsion. Texture analysis revealed that the hardness and adhesiveness decreased to 1.89 ± 0.13 N and 0.66 ± 0.05 N, respectively, while cohesion, elasticity, and chewiness were increased. The LF-NMR results indicated that the CA combined CS modified starch-based emulsion beads (CA/CS-SEB) had a diminished proportion of free water and a correspondingly greater proportion of bound water compared to starch-based emulsion beads (SEB). The encapsulation efficiency of the CUR-loaded beads reached 86.50 ± 0.25 %. In simulated gastrointestinal digestion, CA combined CS modified starch-based emulsion beads loaded with curcumin (CA/CS-SEB-CUR) exhibited the slowest release ratio, which decreased from 23.20 ± 0.03 % to 17.03 ± 0.77 %. These findings provide a theoretical basis for the use of modified starch and propose a viable strategy for curcumin delivery.

Addressing limitations of conventional dressings in real-time monitoring and active repair, this study developed a smart, multifunctional nanofiber membrane by electrospinning curcumin (Cur) into a polyvinyl alcohol (PVA) matrix. The PVA/Cur membranes integrated antibacterial, antioxidant, and pH-responsive colorimetric properties. Increasing Cur content (0–1.5 wt%) reduced fiber diameter and enhanced hydrophilicity. The PVA/Cur1.0 membrane exhibited optimal mechanical properties (2.84 MPa tensile strength, 43.31% elongation). Biologically, PVA/Cur1.5 achieved a > 5-log reduction within 10 min and over 70% DPPH radical scavenging. Leveraging Cur's pH sensitivity, the dressing enabled visual wound monitoring via a distinct color transition from yellow (acidic, pH 4–6) to reddish-brown (alkaline, pH 7.5–8.9), synchronized with pH-triggered Cur release. Excellent biocompatibility was confirmed (hemolysis < 3%, fibroblast viability > 85%). This work presents an integrated strategy for a smart theragnostic dressing, merging real-time diagnosis and active therapy to advance beyond passive wound care. Graphical

Porous microneedles (PMNs) can efficiently load drugs via capillary action within their porous structure and enable controlled drug release in deep wound layers, thereby significantly promoting wound healing. However, increasing the porosity of PMNs typically compromises mechanical strength, leading to needle tip deformation during skin penetration. To overcome this limitation, a mechanically robust porous microneedle (PMN) was fabricated by lyophilizing a chemically and physically dual-crosslinked hyaluronic acid/polyvinyl alcohol hydrogel formed in situ within a microneedle mold. The resulting PMN exhibited a single-needle bearing force exceeding the minimum threshold for skin penetration (>0.1 N) while maintaining a relatively high porosity (28.7%). Furthermore, a curcumin-loaded PMN (PMN@Cur) was easily prepared by adsorbing a curcumin/β-cyclodextrin complex solution into the abundant pores of PMN. The PMN@Cur showed sustained drug release, along with superior antioxidant and antibacterial activities. In infected wounds models, the PMN@Cur attained a wound contraction rate of up to 87.2% by day 14, demonstrating its high therapeutic efficacy in wound healing. These findings suggest that PMN@Cur, which combines high porosity with favorable mechanical properties, holds significant promise as a drug delivery system for advanced wound care.

Pulmonary hypertension (PH) is a severe pulmonary vascular disease that ultimately leads to right heart failure, which involves multiple core mechanisms, including oxidative stress, inflammation, and pathological cellular proliferation. However, current therapies are limited in simultaneously modulating multiple core mechanisms, leading to poor clinical outcomes and high mortality. Herein, we introduce tetrahedral framework nucleic acids-Curcumin (tFNAs-Cur) as a novel therapeutic for PH, targeting multiple synergistic pathways including TGF β1, NF-κB, and PI3K-Akt. The proposed tFNAs-Cur featured the advantages of high drug loading capacity, good water solubility, strong stability, and excellent biocompatibility. Cellular experiments demonstrate that tFNAs-Cur is efficiently internalized by pulmonary artery smooth muscle cells, alleviates oxidative stress, and inhibits their proliferation and migration. In the monocrotaline-induced PH rat model, tFNAs-Cur significantly improves pulmonary artery acceleration time, reduces vascular wall thickness, and medial pulmonary artery diameter. Cardiac assessments show improved exercise capacity, reduced Fulton index, and alleviated right ventricular fibrosis. Further transcriptomic and biochemical analyses reveal that the therapeutic effects of tFNAs-Cur were mediated via the ROS/TGF β1 antioxidant, NF-κB anti-inflammatory, and PI3K-Akt anti-proliferative pathways. The proposed tFNAs-Cur provides an efficient and safe method with potential for clinical translation that simultaneously utilizes multiple synergistic mechanisms for the treatment of PH.

Objective: Inhibition of tumor angiogenesis has become a new targeted tumor therapy. In this study, we established a micellar carrier with a tumor neovascularization-targeting effect modified by the neovascularization-targeting peptide NGR. Methods: The targeted polymer poly(ethylene glycol)-b-poly(lactide-co-glycolide) (PEG-PLGA) modified with Asn–Gly–Arg (NGR) peptide was prepared and characterized by 1 H nuclear magnetic resonance and Fourier-transform infrared spectrometry. NGR-PEG-PLGA was used to construct curcumin (Cur)-loaded micelles by the solvent evaporation method. The physicochemical properties of the micelles were also investigated. Additionally, we evaluated the antitumor efficacy of the polymer micelles (PM) using in vitro cytology experiments and in vivo animal studies. Results: The particle size of Cur-NGR-PM was 139.70 ± 2.51 nm, and the drug-loading capacity was 14.37 ± 0.06%. In vitro cytological evaluation showed that NGR-modified micelles showed higher cellular uptake through receptor-mediated endocytosis pathways than did unmodified micelles, leading to the apoptosis of tumor cells. Then, in vivo antitumor experiments showed that the modified micelles significantly inhibited tumor growth and were safe. Conclusions: NGR-modified micelles significantly optimized the therapeutic efficacy of Cur. This strategy offers a viable avenue for cancer treatment. Export

Foodborne spoilage bacteria are the key factor causing food deterioration, seriously restricting the shelf life of food and threatening consumer safety. In this study, we investigated the synergistic bactericidal effect of curcumin-mediated photodynamic inactivation technology (Cur-PDI) combined with eugenol (Eug) on two typical f foodborne spoilage bacteria Leuconostoc mesenteroides and Carnobacterium maltaromaticum , and further evaluated its application in bacon preservation. The results indicated that under the combined treatment conditions (20 μΜ Cur, 25 μg/mL Eug, 20 min of light exposure), the viable bacterial counts of two bacteria decreased significantly by 7.11 log CFU/mL and 7.12 log CFU/mL compared with the blank control. As both Cur and Eug are lipophilic, the combination exhibited a stronger bactericidal effect in a lipid system than the water-soluble photosensitizer erythrosine (ERY). Mechanistic studies revealed that, compared with PDI alone, the combined treatment more significantly disrupted bacterial membrane integrity, induced higher levels of reactive oxygen species (ROS), and severely interfered with antioxidant defense and energy metabolism. In the bacon preservation experiment, the combined treatment effectively reduced the initial colony count of the product and continuously inhibited the growth of microorganisms during storage for 25 days. This study systematically clarified the synergistic antibacterial mechanism of Cur-PDI and Eug, and verified its potential in the preservation of bacon, providing a theoretical basis for the development of natural and efficient preservation strategies for foods rich in fat.

Surface fouling and bacterial contamination present significant challenges to daily life and industrial production, motivating the development of multifunctional surfaces with antifouling and antimicrobial properties. In this context, the development of multifunctional surfaces based on environmentally friendly green materials, which possess both superhydrophobic anti-fouling and highly efficient antibacterial properties, has become one of the approaches to address the aforementioned issues. In this study, superhydrophobic antimicrobial multifunctional coatings were successfully prepared by using environmentally friendly materials chitosan (CS) and curcumin (Cur) as antimicrobial agents, shellac and rosin instead of the traditional fluoride as low surface energy substances and adhesives, and nano‑calcium carbonate (Nano-CaCO 3 ) as hydrophobic particles. The multifunctional coating exhibits a contact angle exceeding 150° for both water and other liquids, demonstrating exceptional liquid-repellent properties and superior antifouling performance. Under an 80 g load, the coating withstands 45 cycles of sandpaper abrasion testing and 50 cycles of adhesive tape peeling testing. Furthermore, the composite coating samples show a 28% increase in tear strength and a 24% enhancement in puncture resistance compared to the pristine counterpart. The coating exhibits significant antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), achieving antibacterial efficiency rates of 99.1% and 99.3%, respectively. In strawberry preservation tests, the shelf life of the samples was extended to the sixth day. Composed of non-toxic and sustainable chemicals, the coating integrates superhydrophobicity, antifouling properties, and antimicrobial functionalities, thereby demonstrating broad applicability across diverse sectors, including paper protection, packaging, biomedical applications, and the food industry.

The repair of irregular bone defects poses a significant challenge for orthopedic surgeons. This study presents a customizable, novel bone tissue engineering scaffold capable of promoting bone regeneration, which is composed of curcumin-quercetin-piperine-loaded poly(lactic-co-glycolic acid) nanoparticles (Cur-Que-Pip-PLGA NPs) loaded onto 3D-printed porous tantalum (3DPTa). The Cur-Que-Pip-PLGA NPs were prepared via the emulsification-solvent evaporation method, and subsequently loaded onto the 3DPTa scaffold via the physical impregnation method. The morphology and structure of the composite scaffold were characterized by scanning electron microscopy (SEM), and its drug release profile was evaluated via high-performance liquid chromatography (HPLC). The biocompatibility and osteogenic potential of the scaffold were evaluated. Cell proliferation assays and live/dead cell staining demonstrated that the composite scaffold exhibited excellent biocompatibility. Both in vitro and in vivo osteogenic experiments demonstrated that the composite scaffold significantly promoted the osteogenic differentiation of MC3T3-E1 cells and new bone formation. These results indicate that this novel Cur-Que-Pip-PLGA NPs 3DPTa scaffold has excellent biocompatibility and osteogenic potential, thus representing a promising candidate material for bone defect repair.

Background Ulcerative colitis (UC) is a common type of inflammatory bowel disease, where the vicious cycle of inflammation and oxidative stress poses a major challenge in its treatment, and existing therapies have limitations. Berberine (BBR) and curcumin (CUR) have the potential for synergistic treatment of UC, but this potential has not been verified in UC. Additionally, both BBR and CUR suffer from poor water solubility and low bioavailability. This study aims to construct prebiotic microcapsules (BBR/CUR@MC) for the co-delivery of BBR and CUR and explore their therapeutic mechanism in UC. Methods Network pharmacology was used to predict the targets and pathways of BBR and CUR in UC. BBR/CUR@MC was prepared using microfluidic electrospray technology, and its colon targeting and biocompatibility were evaluated through in vivo experiments. In a Dextran sulfate sodium (DSS)-induced UC mouse model, the therapeutic effect was assessed using multiple indicators, and the mechanism of action was explored by transcriptome analysis. Results Network pharmacology showed that BBR and CUR can exert therapeutic effects on UC through synergistic regulation of TNF and AGE-RAGE signaling pathways. The successfully constructed BBR/CUR@MC had good colon targeting and biocompatibility. In the mouse colitis model, oral administration of BBR/CUR@MC inhibited ADAM17 in the TNF signaling pathway and MAPK11/13, COL4A1, and COL1A1 in the AGE-RAGE signaling pathway, thereby downregulating pro-inflammatory cytokines such as TNF-α and IL-1α, upregulating IL-10, scavenging ROS, significantly alleviating colonic inflammation and repairing the intestinal barrier in mice, with a therapeutic effect superior to that of single-drug microcapsules and 5-ASA. Conclusion BBR/CUR@MC can exert synergistic anti-inflammatory and antioxidant effects in UC treatment by regulating TNF and AGE-RAGE signaling pathways, providing a new multi-mechanism therapeutic strategy for UC.

Biofouling poses a great challenge to marine facilities and causes serious economic losses, making the development of green and non-toxic anti-fouling coatings a high priority. In this study, leveraging the broad-spectrum bacteriostatic and economic properties of the natural product curcumin (Cur), it was chemically grafted onto the surface of xanthate-functionalized silica nanoparticles with the hydrophilic monomer vinylpyrrolidone (NVP) using a reversible addition-fragmentation chain transfer (RAFT) polymerization strategy. The resulting modified particles were incorporated as active fillers into a terminal hydroxy-polydimethylsiloxane (PDMS) matrix, successfully constructing a functional coating labeled PDMS/SiO 2 -X. This coating exhibits a synergistic antifouling mechanism combining self-polishing, static antifouling agent mitigation and dynamic fouling release. The presence of modified silica significantly improves the mechanical properties of the coating. The low surface energy surface imparts excellent fouling-release characteristics, while the hydrolysis of ester bond enables the sustained release of Cur to achieve efficient bacterial inhibition. Additionally, NVP effectively resists protein adhesion by forming a hydration layer. A 180-day real-sea field test verifies the long-lasting and stable antifouling performance of the coating. This work provides a new idea for developing highly efficient and environmentally friendly marine antifouling coatings.

Hyperglycemia can lead to excessive production of reactive oxygen species (ROS), contributing to diabetes and its complications, such as diabetic cardiomyopathy (DCM). This study aims to elucidate the protective mechanisms of Curcumin (CUR) against hyperglycemia-induced cardiomyocyte injury. We constructed a high-glucose (HG) model using primary cardiomyocytes and determined the optimal concentration of CUR by assessing cell viability with the CCK-8 assay. Enzyme-linked assays were used to measure the activities of HO-1, T-SOD, and GSH-Px, investigating the antioxidant effects of the Nrf2 signaling pathway. Flow cytometry was employed to measure apoptosis and mitochondrial membrane potential (ΔΨm). Mitochondria were isolated from cardiomyocytes to examine cytosolic and mitochondrial cytochrome c (cytc) expression. Western blotting was used to analyze the expression levels of Nrf2, PI3K, AKT, Keap1, HO-1, Bcl-2, and Bax proteins. Our findings suggest that CUR enhanced the viability and antioxidant enzyme activity of primary cardiomyocytes under HG conditions, stabilized ΔΨm, reduced ROS production, and decreased apoptosis. Moreover, CUR alleviated HG-induced cardiomyocyte damage by upregulating Nrf2, PI3K, AKT, HO-1, and Bcl-2 expression, while downregulating Bax. These findings demonstrate that the protective effect of CUR against diabetic cardiac injury may be mediated through activation of the Nrf2 and PI3K/AKT signaling pathways and upregulation of antioxidant enzyme expression, aiming to provide a theoretical foundation for further research into the therapeutic application of CUR in DCM.

This magnetic drug delivery system is based on droplet microfluidic technology, with uniform size of the microspheres and controllable mode. • MNPs enabled esophageal retention under magnetic fields, while GelMA component enables controlled curcumin release under reflux conditions. • These multifunctional microspheres effectively address the challenges associated with targeted drug delivery to the esophagus. Activated molecular pathways with elevated chemokine secretion are critical in driving pathogenesis of the mucosal inflammation in gastroesophageal reflux disease (GERD). The therapeutic efficacy of acid-suppressants is often constrained in GERD patients due to the high prevalence of non-acid reflux. Curcumin, a natural polyphenol with robust anti-inflammatory and antioxidant effects, offers promising therapeutic potential. In this study, leveraging the favorable biocompatibility and bioactive property of gelatin methacryloyl (GelMA), we developed GelMA microspheres as microcarriers encapsulating curcumin nanoparticles (Cur) for local treatment of GERD. The further incorporation of magnetic nanoparticles (MNPs) facilitates esophageal localization of the microspheres under an external magnetic field. Additionally, Cur-MNPs@GelMA microspheres sustain drug release in simulated gastric fluid, ensuring effective local concentrations. In vitro and in vivo investigations corroborated that Cur-MNPs@GelMA microspheres markedly inhibited the generation of inflammatory cytokines and reactive oxygen species (ROS). Mechanistically, the microspheres can restore mitochondrial function and inhibit NF-κB pathway. Furthermore, the multifunctional microspheres can effectively ameliorate histopathological damage and suppress inflammation in a mouse model of GERD. These findings endorse the therapeutic potential of Cur-MNPs@GelMA microspheres as a novel clinical intervention for GERD. Download: Download high-res image (192KB) Download: Download full-size image
