The selective detection of weakly adsorbing, low-abundance molecules in complex biological environments remains a fundamental limitation of conventional surface-enhanced Raman scattering (SERS) technology. Here, we engineered a macrocyclic supramolecular pillararene-bridged gold nanoparticle assembly to create sub-nanometer plasmonic gaps with intense electromagnetic hotspots, enabling ultrasensitive molecule detection in complex matrix. The pillararene-functionalized interface not only enhanced SERS signals (with enhancement factor of 1.3 × 10 7 for curcumin, a model phenolic drug), but also provided selective recognition of phenolic biomarkers through host–guest chemistry, effectively suppressing background noise in water, cell lysate and serum environments. Combined with single-molecule counting, the detection limit reached to 1.03 × 10 −12 mol/L for curcumin. Cellular imaging experiments demonstrated that the pillararene linker facilitated intracellular delivery of curcumin while simultaneously amplifying its Raman signal, enabling real-time tracking of drug distribution at subcellular resolution. This work establishes a versatile detecting platform that integrates supramolecular targeting with plasmonic enhancement, advancing spatially resolved detection of low-abundance molecules in complex biological environments for precision diagnostics and therapeutic evaluation.

The surface-enhanced Raman signals of 4-Aminobenzenethiol (4-ABT) adsorbed on the surface of triangular Au-AuAg hierarchical-multishell nanostructure have been investigated. Here, the approach to produce core-cavity-shell sandwich nanostructures presented as Au-AuAg is the same as preparing metal nanoparticles with hollow morphology, in which the galvanic replacement reaction takes place between silver and chloroauric acid. In this paper, we directly mix 4-ABT with gold nanoparticles and drop it on glass slides to study the effect of nanoparticles on signal enhancement of Raman spectrum , avoiding the cumbersome process of preparing metal-molecular-metal three-layer structure as reported. A significant increase in the SERS intensity of b 2 mode around 1140 cm −1 was observed, which could quantify the concentration of 4-ABT indirectly. In a certain range, the Raman intensity gradually increases with the increasing intermediate gap, which has a strong relationship with dipole plasmon hybridization of core-dielectric-shell sandwich nanostructure. Moreover, Raman spectrum results show that the Au-AuAg substrate can produce signal intensity about 3.8 × 10 2 times stronger than that of 4-ABT alone and the detection limit was as low as 0.1 μM in solution.

Aptamer sensors based on surface-enhanced Raman scattering (SERS) technology have demonstrated great potential in the ultrasensitive and rapid detection of Escherichia coli ( E. coli ). Herein, this paper presents a digital SERS aptamer sensor. This sensor integrates ordered nanoscale array synthesis technology and digital analysis technology, enabling highly sensitive and rapid bacterial quantification. The ordered monolayer gold nanosphere arrays (Au NS) can form uniform and dense “hot spots” on the silicon wafer due to their uniform spherical structures and narrow gaps. Moreover, digital SERS is adopted to further optimize the signal uniformity so as to achieve precise quantification. The sensor modules are combined together through base pairing. The aptamers labeled with Raman tags are detached from the complementary DNA due to the competition of the target substance, thus realizing the detection of E. coli . The digital SERS aptamer sensor has been verified to possess excellent selectivity and reproducibility. It has a wide dynamic linear detection range from 1.0 * 10 1 to 1.0 * 10 9 CFU/ml and a detection limit of 0.657 CFU/ml, maintaining excellent specificity even in the presence of mixed bacterial interference. The spiked recoveries in actual samples range from 98.80 % to 99.81 %. Leveraging different aptamers and digital analysis, the sensor holds promise for food safety and environmental monitoring applications.

Background β 2 −agonists, also known as β 2 −adrenoceptor agonists, are secreted by the adrenal medulla of the animal body and have a characteristic catecholamine structure. When inhaled, the drug rapidly relaxes the smooth muscles of the airways in asthma patients, improves ventilation, and temporarily increases alertness during exercise and helps relieve fatigue. However, the use of β 2 −agonists to improve physical performance in a short period of time can lead to extreme dependence and irritation, and there are still many unknown hazards. They are now on the World Anti−Doping Organization (WADA) prohibited list. Results Surface−enhanced Raman spectroscopy (SERS) can directly detect the characteristic peaks associated with β 2 −agonist compounds in a fast and sensitive manner. This study is the first to utilize a direct Raman detection technique, achieving successful acquisition of the fingerprint spectra of three target analytes (vilanterol, fenoterol, bambuterol) as well as the establishment of a corresponding fingerprint spectral analysis method. To further improve its performance, in this study, gold nanoparticles were synthesized via redox methods and integrated with covalent organic framework (COF) to construct a novel highly sensitive detection platform for β 2 −agonists. The Au−COF composite exhibits high adsorption performance and can significantly enhance the SERS signal of the target, thereby achieving low detection limits, including 9.25 × 10 −6 g·mL −1 for bambuterol, 9.76 × 10 −6 g·mL −1 for vilanterol, and 7.54 × 10 −6 g·mL −1 for fenoterol, which makes it promising for improving the overall sensitivity of β 2 −agonist detection. Significance This study develops a novel analytical method utilizing Au−COF composites for highly sensitive SERS detection. It pioneers the application of this strategy in the qualitative and quantitative analysis of trace β2-agonists. The research not only significantly expands the scope of Au−COF composites but also provides a technologically promising approach with substantial practical value for ensuring medical safety through clinical medication monitoring and for detecting banned substances in sports.

Organized two-dimensional polystyrene bead arrays perform ion etching, and protruding nanostructures are created on polystyrene beads due to the shadow effects from the ring beads, leading to nucleus selection and growth in Au nanostructure deposition. Ag nanostructures are prepared via plasmon-mediated chemical reactions (PMCRs), leading to the Ag nanocavity geometry of the honeycomb pattern when the etching time and Ag growth time are tuned. Due to the strong electromagnetic coupling, the Ag honeycomb-shaped nanocavity array works as the SERS substrate with high sensitivity and good repeatability, which is used to detect thiram pesticide residues with a concentration down to 10−9M.

Early identification and timely intervention of precancerous lesions of gastric cancer are the core strategies to block their malignant transformation into gastric cancer (GC). This study developed a dual functional sensor that combined surface-enhanced Raman scattering (SERS) activity and nanoenzyme catalytic properties. The sensor was driven by catalytic hairpin self-assembly (CHA) technology triggered by the target microRNA-21 (miR-21), promoting the conversion of 3,3′,5,5′-tetramethylbenzidine (TMB) and signal amplification of TMB oxidation (ox-TMB). This work used Au–platinum nanoparticles (Au–Pt NPs) as nanozyme probes and silica-encapsulated ferrite (Fe 3 O 4 @SiO 2 ) as capture probes. In the presence of target miR-21, CHA technology induced Au–Pt NPs to assemble on the Fe 3 O 4 @SiO 2 surface, endowing the sensor with SERS enhancement capability and peroxidase-like (POD-like) simulated catalytic performance. This catalyzed the conversion of TMB and hydrogen peroxide (H 2 O 2 ) into the oxidized product ox-TMB with characteristic absorption. CHA technology, acting as an intrinsic driving force for sensor assembly, further facilitated exponential amplification of the ox-TMB detection signal. This SERS sensor demonstrated outstanding detection performance, achieving a detection limit as low as 20.24 aM for miR-21. It also exhibited excellent clinical applicability and accuracy in clinical sample testing. Therefore, the proposed SERS sensor holds significant clinical potential for diagnosing gastric precancerous lesions. Early identification and timely intervention of precancerous lesions of gastric cancer are the core strategies to block their malignant transformation into gastric cancer (GC).

Surface-enhanced Raman scattering (SERS) has emerged as a powerful tool for food safety analysis. We present a three-dimensional particle-in-cavity substrate that utilizes plasmonic cavity resonances for highly sensitive detection. Fabricated by a convenient transfer of AuNPs onto an anodic aluminum oxide (AAO) template, this design facilitates scalable production. The synergistic cavity-particle interplay yields a 4.24-fold SERS enhancement over a conventional flat Si substrate. The substrate demonstrated high sensitivity with a Rh6G enhancement factor of 1.77 × 10(Raman and Krishnan, 1928 [ 9 ] ) , excellent uniformity showcasing relative standard deviations of 9.17% for Rh6G and 8.54% for crystal violet, and strong log-concentration linearity with correlation coefficients reaching 0.988 for Rh6G and 0.989 for Crystal Violet. It maintained 60% of its initial signal intensity after 30 days of storage and achieved an aspartame detection limit as low as 0.0078 g/L, demonstrating its excellent detection capability.

With the rise of electronic cigarettes ( E -cigarettes), certain illegal additives have gradually emerged in these products, posing significant health risks to humans. Consequently, there is an urgent need for rapid and highly sensitive detection of illegal additives in E -cigarettes. This study combines deep learning algorithms with Surface-Enhanced Raman Spectroscopy (SERS) to achieve the sequential detection following phase separation of the illegal additive nicotine and the new psychoactive substance etomidate. First, by utilizing prepared gold nanoparticles as an enhancement substrate, we accomplished highly sensitive detection of nicotine and etomidate standards. Additionally, through density functional theory calculations, we established the attribution of the characteristic peaks of both substances, providing a basis for their qualitative differentiation. Additionally, by developing a liquid-liquid extraction method, we successfully isolated nicotine and etomidate from e-cigarettes, achieving highly sensitive SERS detection with lowest detectable concentrations of 0.18 ng/mL and 1 μg/mL, respectively. Finally, by integrating one-dimensional convolutional neural network (1D-CNN), we attained a classification accuracy of 94.0% for real samples. Therefore, this strategy may provide a reference for the detection of illegal substances in the future.

Long-term exposure to aromatic volatile organic compounds (VOCs) will create a lot of damage in the body, making their efficient collection and highly sensitive detection a crucial task. Herein, we developed a flexible surface-enhanced Raman scattering (SERS) substrate by loading gold nanoparticle multimers onto oil blotting paper (Au NPs-OBP) for the highly sensitive detection of aromatic VOCs. Leveraging a hydrophobic surface and similarity compatibility principle, the substrate facilitates efficient adsorption of trace aromatic VOCs from human skin. The coagulation of the OBP further causes tight aggregation of Au NPs, anchoring them firmly to the fibers and generating dense hot spots for SERS enhancement. The Au NPs-OBP substrate confirms high sensitivity, with limits of detection (LOD) of 4.628 × 10 −11 M and 9.422 × 10 −9 M for Rhodamine 6G (R6G) and p-nitrophenol (4-NP), respectively. Finite difference time domain (FDTD) simulations demonstrated that the substrate possesses a homogeneous high local electromagnetic field. Excellent reproducibility was achieved across randomly selected spots on the same substrate and among substrates from different batches. The LOD for catechol was found to be 6.657 × 10 −12 M, and the recovery rate improved further, varying between 89.26% and 97.09%. These results indicate that the Au NPs-OBP substrate is a promising platform for the highly sensitive detection of the aromatic VOCs.

Enhancement of the light-matter coupling is a promising approach to improve the sensitivity and universality of semiconductor-based surface-enhanced Raman scattering (SERS). Herein, it is demonstrated that hollow multi-shelled V 2 O 5 microstructure can be utilized as an active substrate to integrate multiple synergistic contributions. The experiments and numerical simulations clearly reveal that the excellent SERS activity originates from the unique double-shelled hollow structures, which allow multiple reflections of electromagnetic waves. This light-trapping effect provides an efficient resonance absorption for charge-transfer (CT) and exciton enhancements. More importantly, the coupling of outer and inner shells remarkably generates high electric field contribution for SERS enhancement. Besides, the symmetry analysis of vibrational modes based on density functional theory well explains the origin of the CT coupling of Herzberg–Teller selection rules. The synergy of all factors contributes to a significant increase in the sensitivity and universality of semiconductor SERS. It is anticipated that the strategy demonstrated in this study will widen the scope of currently available methodologies for designing substrates with high semiconductor SERS activity.

The nanostructures with uniform long-range ordered structure are of crucial importance for performance standardization of high-quality surface-enhanced Raman scattering (SERS) spectra. In this paper, we described the fabrication and SERS properties of Au decorated Cu (Au@Cu) nanoarrays. The Cu nanoarrays with uniform long-range ordered structure were first synthesized by in-situ electrochemistry assembly on insulated substrate. The Cu nanoarrays can reach a size of centimeters with strictly periodic nano-microstructure, which is beneficial for the production and performance standardization of SERS substrates. Then Au nanoparticals were decorated on the Cu nanoarrays by galvanic reaction without any capping agent. The obtained Au@Cu nanoarrays exhibit excellent SERS activity for 4-Mercaptopyridine, and the sensitivity limit is as low as 10−8M. Therefore, this facile route provides a useful platform for the fabrication of SERS substrates based on nano ordered arrays.
