BACKGROUND In this study, a safe and relatively stable γ -cyclodextrin-lysozyme ( γ -CD-Lys) was synthesized using epichlorohydrin as the cross-linking agent, and curcumin was successfully encapsulated in γ -CD-Lys. RESULTS The successful Lys grafting onto γ -CD can be demonstrated by a high grafting ratio (79.02%) and was further confirmed by Fourier transform infrared (FTIR) band shifts and the new signal obtained at δ 2.75 in proton nuclear magnetic resonance. The encapsulation efficiency value of γ -CD-Lys was 76.74%, and the successful encapsulation of curcumin into γ -CD-Lys was confirmed by crystal structure change, increased melting point, and FTIR band shifts. The intermolecular bonds results suggested that associative forces between curcumin and γ -CD-Lys were electrostatic interaction, hydrogen bonds interaction, and hydrophobic interaction. The designed nanoparticles had excellent stability at low pH and low salt concentration. The release rate of these nanoparticles was inhibited in simulated gastric conditions, whereas it increased significantly in intestinal media. Simulated gastrointestinal digestion experiments further confirmed that nanoparticles showed higher bioaccessibility (86.05%) compared with curcumin (58.82%). CONCLUSION Overall, our study showed that the nanoparticles were highly promising for delivering curcumin because of their enhanced functional attributes and stabilization in acid or low salt environments. Also, it was an excellent wall material for targeting hydrophobic bioactive compounds in the intestinal tract via oral administration. © 2022 Society of Chemical Industry.

To change the strong hydrophilicity of alginate and make it have good amphiphilic property, thereby broadening its application as a new pesticide-controlled release system, the hexyl alginate ester derivative (HAED) was synthesized by the bimolecular nucleophilic substitution (S N 2) reaction, using 1-bromohexane as the hydrophobic modifier. The structure and self-aggregate performance of the synthesized HAED were contrastively characterized. Moreover, the loading and release performance of λ-Cyhalothrin (LCH) for the HAED microcapsules prepared by the high-speed shearing method was also studied. The synthesized HAED exhibited enhanced molecular flexibility and good self-aggregate performance, which could form the stable self-aggregated spherical micelles with the average hydrodynamic diameter and zeta potential of 416 nm (PDI = 0.29) and − 39.9 mV, respectively, due to the intra- or intermolecular hydrophobic associations. Furthermore, the obtained HAED microcapsules displayed the high encapsulation efficiency and sustained-release property in comparison with the SA microcapsules for the good affinity of HAED to hydrophobic LCH that retarded the drug diffusion. On the basis of the excellent advantages of HAED, it was expected to become the ideal candidate for hydrophobic drug delivery in controlled-release pesticide formulations.

In recent years, the development of functional packaging from natural active compounds has been a promising strategy for microbial control in foods . In this study, the basil essential oil (BEO) was applied as the principal antibacterial agent , and a controlled-release nanofibrous system was engineered with responsiveness to bacterial phospholipase . It indicated that encapsulation by cationic liposome significantly improved the dispersity and stability of BEO during the electrospinning process, and the prepared BEO-loaded cationic liposomes (BCLs) could maintain structural integrity in polymer nanofibers. The resulting BCLs-loaded nanofibers showed an effective antibacterial activity against L. monocytogenes . Additionally, it is worth noting that incorporation of soybean lecithin in nanofiber substrate could accelerate the release of loaded antibacterial agents when exposed to L. monocytogenes , thus obtaining an enhanced antibacterial activity. This could be attributed to the increased sensitivity of nanofibers to the degradation by bacterial phospholipases after incorporation with soybean lecithin. Finally, the application test showed that the prepared antibacterial nanofibrous mats could help maintain the quality of chilled pork during 4-days storage, which indicated their promising potential as active packaging. Given the serious threat of L. monocytogenes to food safety, the present antibacterial nanofibers have wide application prospect for improving microbial safety of chilled meat.

The wettability of the coating used in controlled release fertilizers (CRFs) is a crucial parameter for the release behavior of nutrients. However, how to control the wettability of coating to tune nutrient release is a big challenge. Herein, the alkylamine-poly (tannic acid) (APTA) building blocks coatings with controlled wettability were prepared as coating materials for CRFs. Alkylamine (octadecylamine (ODA), hexadecylamine (HDA), dodecylamine (DDA), and hexylamine (HEA)) were applied for the modification of poly (tannic acid) (PTA) coating on the silicon wafer and urea surface. The wettability of coatings on silicon wafers were studied and nutrient release of APTA coated fertilizers were characterized in water and soil. The nitrogen nutrient and water vapor permeability coefficient of coatings were characterisized and the mathematical model for predicting the release of nutrients in soil. The alkyl chain length of alkylamine is demonstrated to play a crucial part in controlling the wettability. The rate of nutrient release increases in the order of ODA-PTA < HDA-PTA < DDA-PTA < HEA-PTA coated fertilizer. We envision that APTA building blocks with diverse wettability on urea surface could control the release rate of the nutritional content of fertilizer, which has potential application in the development of controlled-release fertilizers.

In this study, carboxymethyl chitosan (CMCS) with excellent biocompatibility was used as the “gatekeeper” to design and fabricate a pH-responsive drug delivery system (CMCS-DFNS) as paclitaxel carriers. Characterization results showed that CMCS-DFNS was successfully prepared and the nanocarriers displayed excellent drug loading efficiency of 19.8 %, and the results of the adsorption mechanism revealed that the adsorption of PTX was consistent with the Freundlich isotherm and pseudo-second-order kinetic model. Furthermore, the pH-responsive controlled release behavior at different pH (pH = 7.4, 6.5, and 5.0) was evaluated, and the results demonstrated that the cumulative release at pH 5.0 was 58.8 %, which was 2.7 times higher than that at pH 7.4, suggesting that the carrier exhibited a good pH sensitivity. The results of in vitro cellular experiments further indicated that CMCS-DFNS significantly improved the drug uptake efficiency in breast cancer MCF-7 cells. Importantly, the results of in vivo and cellular pharmacokinetic revealed that CMCS-DFNS can improve the circulation time and enhance the relative bioavailability of paclitaxel. Therefore, the fabricated pH-responsive drug delivery system has potential applications in the delivery of anti-tumor drugs, and provides a new delivery pathway for other compounds with low bioavailability.

The purpose of this study was to construct a delivery system using a microfluidic chip to protect procyanidins (PCs) and to achieve their pH-controlled release in simulated gastrointestinal fluid. The microfluidic chip was designed and fabricated to generate water-in-water-in-oil (W/W/O) templates for the preparation of sodium alginate/chitosan microparticles with a uniform size and core–shell structure, using an internal–external gelation method. Compared with free PCs, the stability of PCs embedded in microparticles was improved and a pH stimulus-responsive release of PCs from microparticles was observed under neutral pH conditions. The delivery system of microparticles was nontoxic and showed an inhibitory effect on the decrease of mitochondrial membrane potential in Caco-2 cells caused by H2O2 and acrylamide. This work provided a method for fabricating compact microfluidic chips to prepare a pH stimulus-responsive PCs delivery system with improved stability, which may have potential applications in the delivery of other nutrients.

Avermectin (AVM) is a highly effective and safe biopesticide but is very sensitive to ultraviolet (UV) light and exhibits poor water solubility. Developing green and multifunctional adjuvants is important for the protection and controlled release of AVM. In this work, a number of water-soluble enzymatic hydrolysis lignins (W-EHLs) were prepared via grafting basic amino acids and used as emulsifiers with co-surfactants to prepare high-internal phase emulsions (HIPEs). The results showed that W-EHLs with co-surfactants could be prepared with HIPEs that contained 90 vol % green oil phases such as turpentine, and the stability of the HIPEs first increased and then decreased when the rate of grafting of basic amino acids on lignin increased from 0.26 to 1.46 mmol/g. The more polar oil droplets were less deformable due to their higher viscosity, thereby affording a stability advantage to HIPEs. Subsequently, the relations between the stability and interfacial viscoelasticity of the emulsion were effectively correlated by interfacial rheology, droplet size, and physical stability tests. The results showed that HIPEs with smaller droplets had poor fluidity and strong interfacial viscoelasticity due to their higher droplet packing density, which resulted in good macroscopic stability. Like the AVM carrier, the retention rate of AVM in HIPEs was 80.1% after UV radiation for 72 h, which represented the highest UV protection efficiency in AVM delivery systems. The release curves showed that the rate of release of AVM from HIPEs was adjusted by controlling the pH value of the medium. In addition, the release of HIPEs is completely in accord with both diffusion and the matrix erosion mechanism. The strategy could be extended to other sensitive pesticides and used to promote the development of sustainable agriculture.

In this study, a novel polymer based on aggregation-induced emission (AIE) fluorogen , biotin and disulfide bonds modified chitosan (TPE-bi(SS-CS-Bio)) was designed and synthesized. The polymer could self-assemble into micelles in aqueous media and encapsulate paclitaxel (PTX) into the core with high drug loading. Fluorescence study indicated that the micelles exhibited excellent AIE feature with intense blue fluorescence emitted. In vitro drug release study indicated that the micelles could disassemble rapidly in the presence of high level of glutathione . The modification by biotin could enhance the cellular uptake of the micelles. The drug-loaded micelles possessed remarkable cytotoxicity against MCF-7 cells, and their distribution in the cells could be traced due to the excellent AIE feature. In vivo antitumor efficacy study demonstrated the superior antitumor activity of the PTX-loaded TPE-bi(SS-CS-Bio) micelles. These results indicated that TPE-bi(SS-CS-Bio) has the ability of biological imaging and can be used as a potential carrier for PTX.

The treatment of bone defects caused by various reasons is still a major problem in orthopedic clinical work. Many studies on osteogenic implant materials have used various biologically active factors such as osteogenic inducers, but these biologically active factors have various side effects. Therefore, in this study, silk fibroin (SF) was used as a scaffold material, mesoporous bioactive glass nanoparticles (MBGNs) as a sustained release carrier, and the traditional Chinese drug icariin (ICA) was loaded to promote bone formation. The experiments in this study have proven that SF/MBGNs-ICA scaffolds can successfully load and release ICA for a long time, and the sustained-release ICA can promote the proliferation and differentiation of BMSCs for a long time. This controlled-release ICA organic/inorganic two-component scaffold material is expected to become a new bone grafting solution.

The periosteum plays an important role in vascularization and ossification during bone repair. However, in most studies, an artificial periosteum cannot restore both functions of the periosteum concurrently. In this study, a novel nanofiber that can sustain the release of vascular endothelial growth factor (VEGF) and bone morphogenetic protein-2 (BMP-2) was fabricated to enhance the durability of angiogenesis and osteogenesis during bone regeneration. A cell-free tissue engineered periosteum based on an electrospinning poly-l-lactic acid (PLLA) nanofiber was fabricated, on which VEGF and BMP-2 were immobilized through a polydopamine (PDA) coating conveniently and safely (BVP@PLLA membrane). The results indicated a significantly improved loading rate as well as a slow and sustained release of VEGF and BMP-2 with the help of the PDA coating. BMP-2 immobilized on nanofibers successfully induced the osteogenic differentiation of human bone marrow mesenchymal stem cells (BMSCs) in vitro with high expression of runt-related transcription factor 2 (Runx2), osteopontin (OPN), and alkaline phosphatase (ALP). Similarly, angiogenic differentiation of BMSCs with the expression of fetal liver kinase-1 (Flk-1) and vascular endothelial cadherin (VE-cadherin) was observed under the environment of VEGF sustained release. Moreover, an in vivo study revealed that the BVP@PLLA membrane could enhance vascular formation and new bone formation, which accelerates bone regeneration in rat femoral defects along with a massive periosteum defect. Therefore, our study suggests that the novel artificial periosteum with dual growth factor controlled release is a promising system to improve bone regeneration in bone defects along with a massive periosteum defect.

The redox-responsive hollow mesoporous silica sphere (HMSS) as smart nanocontainer demonstrates great potential for active agent storage and controlled release. In the present work, HMSS was firstly prepared by surfactant assisted selective etching method. Then, target corrosion inhibitors, 2-mercaptobenzothiazole (MBT) molecules, were loaded and encapsulated with redox-responsive supramolecular nanovalves, ZnO quantum dots. Results reveal that obtained HMSS exhibits a hierarchical mesoporous structure consisting of hollow cavity and mesoporous shell. Corrosion inhibitors were successfully loaded and capped in HMSS nanocontainers, showing on-demand release behavior under specific environment. Moreover, the release efficiency gradually increased with increasing dosage of the reducing agent. Therefore, the novel smart nanocontainers with redox-responsive characteristics will offer a great promise for the development of new generation nano delivery system for corrosion protection accompanying redox environment change.

Pluronic F127 is a thermosensitive polymer that has been extensively studied and utilized in biomedicine. To improve the gelation properties of F127, α-cyclodextrin was introduced by physical interactions, such as forming inclusion complexes, hydrogen bond self-assembly, and hydrophobic interactions, to prepare F127−α-CD hydrogels. This study explored the temperature-dependent sol–gel transition behavior, gelation mechanism, interior morphology, and controlled release of the anticancer drug 5-fluorouracil. Results showed that hydrogel could be obtained at 1.0%–8.0% weight content of F127. The cross-linking points in this system were PEO−α-CD microcrystalline region and micelle with poly(propylene oxide) as the core. The network inside F127−α-CD hydrogels made it stable and conducive for controlled release. Therefore, this hydrogel is a promising drug release system.
