The H2S-releasing multifunctional composite ionic liquid facilitates topical melittin delivery for psoriasis treatment through RONS scavenging and inflammation inhibition

Meng Zeng,Ping Deng,Qin Li,Qi Tang,Jixiang Li,Liangke Zhang

Psoriasis, an immune-mediated disease, commonly utilizes topical drug administration in clinical treatments, yet the thickening of skin presents a challenge to conventional topical drug delivery methods. In this study, we have successfully developed a multifunctional composite ionic liquid (CIL) that enhances the local delivery of melittin (CIL@Mel) by modifying the fluidity of the stratum corneum and the structure of keratin. In addition, CIL@Mel can transport H 2 S to the psoriatic microenvironment, effectively regulating the redox imbalance within it. The in vivo experimental results demonstrated that CIL@Mel effectively mitigates the progression of psoriasis, provides relief from oxidative stress, reduces the production of inflammatory factors, and promotes the repolarization of macrophages. Computational simulations and experimental validation revealed that CIL@Mel exerts its effects by inhibiting the expression of EGFR, STAT3, IL-6, SRC, and HSP90AA1. In conclusion, this study unveils the transdermal promotion mechanism of CIL@Mel and validates its efficacy in psoriasis.

The effect of in vitro simulated digestion on peptide release of Torreya grandis meal proteins and bioactivity assessment of the hydrolysates

Song Shiqing,Wu Yuying,Wang Huatian,Xie Kun,Liu Hao,Sun Min,Shao Li,Yue Heng,Feng Tao,Yao Lingyun

The Torreya grandis meal (TGM) is a protein-rich by-product during the seed oil manufacturing process. In this study, the T. grandis meal protein (TGMP) was extracted from the defatted TGM by alkali extraction and isoelectric point precipitation, and the effect of in vitro digestion on peptide release of the TGMP was evaluated using static digestion model. The TGMP was composed of 17 amino acids with the molecular weight was major located at 35 kDa by SDS-PAGE. Under simulated digestion, the variation in peptide content, amino acid composition, and molecular weight distribution of the hydrolyzed TGMP were determined. The degree of hydrolysis (DH) of TGMP was revealed to be 16.52% after 120 min of gastric digestion and 18.85% after 270 min of gastrointestinal digestion, indicating that the simulated digestion is insufficient to hydrolyze the TGMP thoroughly. Meanwhile, the bioactivity assessment suggested that the antioxidant and anti-tyrosinase activity of the TGMP was significantly enhanced after simulated digestion. The obtained results would provide valuable information on the digestive characteristics of TGMP in the gastrointestinal tract, and would also help to develop functional peptides from the T. grandis meal.

Synergistic Reversal of Inflammation-Mediated Degeneration in Intervertebral Discs: Phenylboric Acid-Grafted Hyaluronic Acid Hydrogel as an Anti-Oxidative Vehicle for Timp-3 Delivery and Promotion of Extracellular Matrix Synthesis

Yan Li,Yuxiang Zhang,Shuqin Wang,Xiaojing Ma,Chengxin Dai,Yifan Wang,Chenyi Ye,Sunwen Pan,Changyou Gao,Weixu Li

Background : Intervertebral disc degeneration (IDD) is intricately linked to the aging process, wherein reactive oxygen species (ROS) and inflammatory responses markedly contribute to matrix degradation and hyperplasia. Injectable antioxidant hydrogels loaded with pharmacological agents hold immense promise for clinical translation in early intervention of IDD. Our previous study revealed that the tissue inhibitor of metalloproteinase-3 (TIMP3) is a pivotal regulator of matrix remodeling and inflammation. Results : We developed a biodegradable ROS-responsive hydrogel functionalized with phenylboronic acid (R-gel) as a controlled release carrier of TIMP3 (R-gel-TIMP3). R-gel-TIMP3 effectively scavenged ROS and provided sustained TIMP3 delivery, thereby attenuating inflammation-driven disc degeneration. In vitro, R-gel-TIMP3 exhibited negligible cytotoxicity, reduced ROS levels in the nucleus pulposus cells, and alleviated cellular senescence and apoptosis. In vivo, it decreased ROS accumulation, inflammatory M1 macrophages, matrix degradation, and neovascularization, significantly ameliorating IDD pathology. Conclusion : The synergistic action of ROS-responsive TIMP3 delivery markedly amplified the therapeutic efficacy against IDD, underscoring the therapeutic potential of R-gel-TIMP3 in IDD management. Statement of Significance 1 We synthesized an injectable bioactive ROS-responsive hydrogel as an anti-oxidative vehicle for TIMP3 protein delivery. 2 The hydrogel enabled sustained release of TIMP-3 in situ and acted as an efficient ROS scavenger to protect NPs against oxidative stress. 3 This treatment could effectively intervene in the progression of IDD from early stage, promote extracellular matrix synthesis, and ultimately reduce IDD in a rat model.

Inulin and metal-phenolic network single-cell encapsulation: Boosting the tolerance and intestinal colonization capability of Bifidobacterium lactis

Mengjun Wang,Shuo Zhang,Siyan Liu,Linyu Nian,Suilou Wang,Chongjiang Cao

Probiotics play a vital role in human health, but maintaining their high viability during processing and oral administration remains challenging. In this study, we utilized inulin and tannic acid (TA)/Ca 2+ metal-phenolic networks (MPNs) as coating materials to encapsulate anaerobic Bifidobacterium lactis (BL) using a layer-by-layer self-assembly strategy to improve their tolerance to the adverse gastrointestinal microenvironment and colonization in the intestinal tract. The results demonstrated that the three-dimensional gel coating formed by inulin could protect the MPNs coating from degradation by gastric acid and improve the ability of BL to resist gastric acid and bile salts, significantly increasing the survival rate of probiotics. Additionally, MPNs could effectively scavenge free radicals and resist cell damage caused by oxidative stress. Importantly, TA in MPNs could form strong interactions (hydrogen bonds) with mucin in the intestine, greatly prolonging the residence time of BL (more than 96 h). Moreover, the raw materials used to prepare BL@TA@Inulin are food-grade, thus ensuring high biosafety. In summary, BL@TA@Inulin represents a novel probiotic single-cell encapsulation approach to achieve highly active and adhesive delivery of probiotics, with potential applications in developing healthy foods and treating gastrointestinal tract-related diseases.

Purification, identification, and mechanism of action of novel antioxidant peptides from quinoa (Chenopodium quinoa wild.) protein hydrolysate

Huan-Huan Xiao,Xue-Ying Li,Ying-Ying Chen,Jin-Xiang Zhou,Ting Zhang,Qiang-Ming Li,Li-Hua Pan,Jian-Ping Luo,Xue-Qiang Zha

The aim of this study was to identify novel antioxidant peptides from quinoa ( Chenopodium quinoa wild.) protein hydrolysates, which were prepared using enzyme combination methods. Eleven novel antioxidant peptides were screened through in silico evaluation, and their structure-activity relationships were further investigated. Molecular docking studies indicated that GPGGGGKGEMF (GF-11) exhibited a higher binding affinity and interaction capacity. GF-11 significantly reduced reactive oxygen species (ROS) levels to 43.20 and malondialdehyde (MDA) levels to 1.92 nmol/mg protein in oxidatively damaged RAW264.7 cells, while also significantly enhancing SOD, GSH-Px, and CAT activities to 25.33, 133.99, and 43.20 U/mg protein, respectively. Furthermore, GF-11 competed with Nrf2 for the binding site on Keap1, promoting the expression of Nrf2, HO-1, and NQO1 proteins, which ultimately activated the Keap1-Nrf2 pathway and mitigated oxidative damage. These findings provide a scientific basis for the application of quinoa antioxidant peptides in the development of functional foods.

Alfalfa Flavonoids Mitigate Salmonella-Induced Colitis via the Keap1-Nrf2 and TLR4/NF-κB/COX-2 Pathways

Xiaoli Qin,Yan Lu,Yawen Luo,Yafang Cui,Kai Zhao,Yang He,Muhammad Aziz ur Rahman,Shengnan Min,Wenfang Wang,Fuyu Yang,Binghai Cao,Huawei Su

Alfalfa is rich in flavonoid compounds, which are known for their antioxidative and anti-inflammatory properties, suggesting therapeutic potential for alfalfa flavonoids (AF) in inflammation-related diseases. This study investigated the effects of AF on Salmonella -induced colitis, a severe inflammatory bowel disorder characterized by oxidative damage and inflammatory response. In vitro, antioxidant assays revealed AF's concentration-dependent radical scavenging, significantly reducing electron paramagnetic resonance (EPR) signals for HO • and O 2 •− by 42% and 54%, respectively. In vivo, AF treatment significantly mitigated body weight (BW) loss by 6%, increased colon length by 11%, and reduced liver and spleen weights by 19% and 81%, respectively, compared to the colitis group. Mechanistically, AF suppressed inflammation by downregulating the Toll-like receptor 4 (TLR4)/IκB/nuclear factor-κB (NF-κB)/cyclooxygenase-2 (COX-2) pathway and inhibiting nucleotide-binding domain-like receptor protein 3 (NLRP3) inflammasome activation, thereby lowering levels of pro-inflammatory cytokines (tumor necrosis factor alpha [TNF-α], interleukin 6 [IL-6], interleukin 1 beta [IL-1β]). Concurrently, AF enhanced antioxidant defense via the Kelch-like ECH-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels while increasing catalase (CAT) (69%), glutathione peroxidase (GPx) (83%), and superoxide dismutase (SOD) (21%) activities. Moreover, AF preserved epithelial and mucosal barriers by reducing apoptosis and upregulating tight junction proteins (Claudin1, ZO-1, E-cadherin) and goblet cell marker Ulex europaeus (Gorse) Agglutinin I (UEA-1). Microbiota analysis revealed that AF significantly enriched beneficial bacteria, including Akkermansia , Oscillibacter , and butyrate-producing taxa, thereby counteracting Salmonella -induced dysbiosis. Furthermore, AF restored the disrupted profile of short-chain fatty acids (SCFAs), strengthening the relationship between symbiotic microbiota and mucosal defense. Overall, AF exerted multifaced protection against Salmonella -induced colitis by alleviating oxidative stress, stabilizing intestinal homeostasis, and thus attenuating inflammation. These findings make AF a promising phytopharmaceutical for the prevention and treatment of inflammatory diseases.

Haisha Lu,Tianle Zhang,Jian Liang,Jinxiang Zeng,Junwei He,Jixiao Zhu,Jinbin Yuan,Xuelan Fu,Huiqing Li,Guoyue Zhong

Gastric ulcer (GU) is one of the most common diseases in humans. Liuwei Muxiang powder (LWMXP), a Tibetan medicine, is a traditional compound prescription used for treating GUs. However, there is currently no in-depth report on its effective substances and action mechanisms. In this study, first, the method of serum pharmacochemistry was adopted to identify 61 prototype components and 18 metabolites of LWMXP. Then, network pharmacology was used to predict the core components, key targets, and important pathways of LWMXP for treating GUs. Subsequently, the effects of LWMXP and its core components on the viability, biochemical indices, cell cycle, reactive oxygen species (ROS), and apoptosis rate of gastric epithelial cell line-1 (GES1) cells in the ethanol-induced GU model were explored. After that, molecular docking was utilized to predict the interactions between the positive drug (omeprazole), six core components, and eight key targets of the PI3K-AKT signaling pathway, and Western blot experiment was carried out for verification. Finally, it was determined that LWMXP for treating GES1 cells in the ethanol-induced GU model mainly acts on the PI3K-AKT signaling pathway, and its effective substances include protopine, piperine, eugenol, methyl gallate, gallic acid, and costunolide. This provides ideas for the follow-up research of LWMXP. Graphical First, the main blood-entering components of LWMXP were identified by serum pharmacochemistry methods. Then, the core components, key targets, and important pathways of LWMXP in treating gastric ulcers were screened through network pharmacology. Subsequently, the effects of LWMXP and its core components on the activity, biochemical indicators, cell cycle, ROS, and apoptosis rate of GES1 cells in the ethanol-induced gastric ulcer model were explored. Then, molecular docking was used to validate the interactions between the positive control drug (omeprazole), six core components, and eight key targets in the PI3K-AKT signaling pathway. Finally, Western blot experiments were carried out to verify the effects of LWMXP and the six core components on the aforementioned targets, ultimately identifying the bioactive substances and mechanisms of LWMXP in the treatment of gastric ulcer.

Sequential extraction, structural characterization, and biological activities of polysaccharides from olive (Olea europaea L.) pomace

Xiao-Liang Zhao,Kai-Li Qi,Hui Han,Hai-Dong Wen,Yan-Mei Liu,Qing-Song Zhou,Yuan-Yuan Ma,Cun-Jin Wang,Jing Zhang,Ji Zhang,Wei-Jie Zhang

Olive pomace, a by-product of olive oil production, remains underexplored despite its potential environmental and economic benefits. This study sequentially extracted three polysaccharides (OERC, OERH, and OERA) from olive pomace using water at room temperature, hot water (80 °C), and 2% Na2CO3 solution (60 °C). Characterization through UV–Vis spectroscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis, high-performance liquid chromatography, and Congo red experiment revealed that these polysaccharides are heteropolysaccharides primarily composed of glucose, mannose, rhamnose, and galactose, each with distinct molar ratios. The polysaccharides exhibited significant antioxidant activity by scavenging hydroxyl, DPPH, and superoxide radicals, with OERC showing the highest potency. They also repaired H2O2-induced oxidative damage in HepG2 cells, with OERC at 400 μg/mL and OERH/OERA at 200 μg/mL displaying optimal efficacy. Additionally, these polysaccharides effectively inhibited α-glucosidase activity, enhanced glucose consumption, and increased glycogen content in insulin-resistant models, thereby exerting hypoglycemic effects. Furthermore, they promoted proliferation, phagocytosis, and nitric oxide release in RAW264.7 macrophages, demonstrating immunomodulatory effects. These results indicate that sequential extraction under varying conditions is an effective method for preparing polysaccharides. The three polysaccharides isolated in this study show great potential for development as functional products with antioxidant, hypoglycemic, and immunomodulatory applications.

Development of metal-organic frameworks and HPβCD/erythritol inclusion complexes loaded onto efficient nanometer fiber membranes for active food packaging

Lingling Bai,Chen Yang,Xiaomin Feng,Xianteng Meng,Hui Yi,Yiting Lu,Wenge Yang,Yonghong Hu

In this study, the inclusion complexes were prepared using an ultrasound-mediated technique using hydroxypropyl-β-cyclodextrin (HP-β-CD) as the main body and erythritol (Ery) as a functional component. Nanofiber films with antimicrobial and antioxidant activities were prepared by blending metal-organic frameworks (ZIF-8), inclusion complexes with polylactic acid (PLA)/gelatin (Gel)/glycerol (GI), and electrostatic spinning method for active packaging. The infrared spectroscopy and crystallographic analysis results indicate that the HPβCD/Ery inclusion complex was successfully prepared. With the addition of ZIF-8 and inclusion, the diameter of the nanofibers increased from 337.36 nm to 2184.20 nm, which improved the thermal stability, and hydrophilicity, and decreased the crystallinity of the fiber membrane and exhibited a sustained and controllable release of Ery for 240 h, thus delaying the degradation of grapes. This new antimicrobial packaging material with excellent biocompatibility and good slow-release performance will have a broad application prospect in food preservation.

Green extraction of oat saponins based on deep eutectic solvents

Yawen Zhang,Hongli Zhang,Jiangong Ma,Jing Zhao,Huajuan Yin,Xu Jing,Xiaowen Wang

With the advent of new-generation green extraction solvents, deep eutectic solvents (DESs) present significant prospects for the extraction of bioactive substances. In this study, saponins were extracted from oats using an ultrasonic-assisted method with green DESs, replacing conventional toxic organic solvents. Adding sodium chloride markedly reduced solubility, resulting in the separation of two phases from the extract: DES and water. The saponins were separated into the water phase, while the DES was reused in subsequent extractions, supporting extractant recycling and promoting sustainable development. Adding salt offers a more gentle, convenient, and cost-effective alternative to current phase separation methods, such as pH adjustment, temperature variation, and carbon dioxide or nitrogen gas application. Optimizing the extraction and recovery parameters yielded the highest oat saponin extraction efficiency at 18.73 mg g –1 , and the recovery was 86.4 %. When compared with ethanol extracts, DES extracts exhibited superior antioxidant activity. Lastly, the greenness of this extraction method was confirmed through three greenness evaluation programs of greenness. The extraction method based on emerging responsive solvents can be extended to other hydrophilic bioactive substances.

Yu Zhang,Fengrui Li,Honglei Yan,Yan Zhang,Shimiao Feng,Lei Deng,Lixia Zhao,Shuang Gao,Ying Fu,Fei Ye

The development of bio-based active food packaging is urgently needed to address food safety concerns. In this study, nerolidol/hydroxypropyl- β -cyclodextrin inclusion complex nanofibers (Nerolidol/HP β CD-IC-NF) were prepared by electrospinning to improve the thermal stability and water solubility of natural antimicrobial nerolidol. Structural characterization (FTIR, XRD, 1 H NMR) confirmed successful nerolidol encapsulation within HP β CD cavities. The resulting bead-free nanofibers exhibited high surface area and porosity. Nerolidol/HP β CD-IC-NF demonstrated a 3.3-fold increase in free radical scavenging capacity compared to free nerolidol, along with potent antimicrobial activity (88.1 % and 76.0 % inhibition against E. coli and S. aureus , respectively). In strawberry preservation trials, nanofiber membranes reduced B. cinerea lesion diameters by 5.56-fold after 5 days compared to untreated controls, significantly extending shelf life. These findings highlight Nerolidol/HP β CD-IC-NF as a promising solution for preventing microbial contamination and improving food preservation efficacy in active packaging applications.
