Pu’ er raw tea extract alleviates DSS-induced colitis in mice by restoring intestinal barrier function and maintaining gut microbiota homeostasis

Su Zhou,Jiaying Yang,Yani Pan,Xinyu Feng,Hao Hu,Shicheng Ma,Cansong Ou,Fangyuan Fan,Shuying Gong,Yuefei Wang,Ping Chen,Qiang Chu

Numerous investigations have uncovered the outstanding therapeutic effect of tea extract on ulcerative colitis (UC). However, the therapeutic potential on UC of Pu’ er raw tea extract (PRTE) remains debated and need to be investigated. In our study, PRTE (100 and 300 mg/kg) was applied to the DSS-induced colitis mice via intragastric administration. Results have shown that PRTE treatment significantly ameliorated DSS-induced weight loss, loose stool, hematochezia, colon atrophy, splenomegaly and colon pathological injury. It could also inhibit UC by down-regulating pro-inflammatory cytokines (IL-1β, IL-6 and TNF-α) and increasing the expression of tight junction protein (zonula occludens-1, ZO-1) to reinforce intestinal barrier. Fecal microbiota transplantation has further confirmed that PRTE could maintain intestinal homeostasis and relieve UC by improve the abundance of beneficial bacteria such as Lactobacillus , Muribaculum , Lachnospiraceae and Rikenellaceae , and reduce the abundance of harmful bacteria such as Turicibacter and Romboutsia . This study suggested that PRTE exhibited a noteworthy therapeutic effect on UC via restoring intestinal barrier function and maintaining gut microbiota homeostasis, which develops a safe and efficient novel strategy for UC prophylaxis and therapy, and provides theoretical support for medical application of Pu’ er raw tea in the future.

Chemodynamic therapy agents Cu(II) complexes of quinoline derivatives induced ER stress and mitochondria-mediated apoptosis in SK-OV-3 cells

Wen-Ying Shen,Chun-Peng Jia,An-Na Mo,Hong Liang,Zhen-Feng Chen

Three Cu(II) complexes of quinoline derivatives as cancer chemodynamic therapy agents were synthesized and characterized. These complexes were heavily taken up by cells and reacted with cellular glutathione (GSH) to reduce Cu 2+ to Fenton-like Cu + , which catalyzed endogenous H 2 O 2 to produce the highly toxic hydroxyl radicals (•OH) to kill cancer cells. Cu1 and Cu2 initiated CAT activity declines, mitochondrial membrane potential and ATP concentration decrease, mitochondrial Ca 2+ overload and ER stress response, leading to cell cycle arrest in sub-G1 and cancer cell caspase-dependent apoptosis . On account of the high GSH and H 2 O 2 specific properties of the tumor microenvironment , Cu1 and Cu2 exhibited higher in vitro anticancer activity and lower toxicity to normal cells. Cu1 and Cu2 efficiently inhibited tumor growth in the SK-OV-3 xenograft mouse model without obvious systemic toxicity.

Hispidulin exhibits potent anticancer activity in vitro and in vivo through activating ER stress in non‑small‑cell lung cancer cells

Li Lv,Wenhui Zhang,Tingting Li,Lifeng Jiang,Xinyan Lu,Jie Lin

Hispidulin is a medicinal natural compound isolated from S. involucrata, which exhibits potent anticancer properties. However, there are few reports on its effects on lung cancer cells. Therefore, the current study investigated the effects of hispidulin on cell viability and apoptosis in human non‑small‑cell lung cancer (NSCLC) cell lines NCI‑H460 and A549 in vitro and in vivo. Methyl thiazolyl tetrazolium, colony formation assay, Hoechst 33342 staining, flow cytometry and western blotting were performed on Human NCI‑H460 and A549 cells. A mouse xenograft model was also established using NCI‑H460 cells. The results showed that the growth of NCI‑H460 and A549 cells was inhibited, while apoptosis was promoted by hispidulin via increased generation of reactive oxygen species (ROS) in a dose‑dependent manner. Furthermore, hispidulin triggered apoptosis in NSCLC cells through upregulating the expression of cleaved caspase‑3 and cleaved poly [ADP‑ribose] polymerase. All these effects were reversed upon pretreatment with glutathione, a selective ROS inhibitor. In addition, endoplasmic reticulum stress (ER stress) in NCI‑H460 cells was activated by hispidulin. Pretreatment with tauroursodeoxycholic acid, a specific ER stress inhibitor, effectively reduced the cell apoptosis induced by hispidulin. In conclusion, hispidulin induces ROS‑mediated apoptosis via activating the ER stress pathway. The current study provides theoretical basis for the antitumor effect of hispidulin in NSCLC.

An electrochemiluminescence cytosensor for sensitive detection of HeLa cells based on a signal amplification strategy of Au-NaYF4:Yb,Er nanocomposites

Jinzha Zhang,Yang Shen,Yuhong Liu,Zaiying Hou,Yuanqing Gu,Wenbo Zhao

A novel electrochemiluminescence (ECL) cytosensor was proposed for the quantitative detection of HeLa cells (human cervical cancer cells) with the help of a signal amplification strategy. Firstly, the Au-NaYF4:Yb,Er nanocomposites were prepared by a simple in situ hydrothermal method and characterized by transmission electron microscopy (TEM) images, X-ray diffraction (XRD) patterns, UV-vis spectra and Fourier transform infrared (FTIR) spectra. Compared with the bare NaYF4:Yb,Er nanocomposites, the ECL intensity of Au-NaYF4:Yb,Er nanocomposites was greatly enhanced by about 4.2-fold which can be attributed to the good conductivity of gold nanoparticles (Au NPs). The nanocomposites showed high and stable ECL emission, fast response and superior conductivity, all of which were advantageous to the ECL detection. Furthermore, HeLa cells were immobilized on the modified electrode via the interaction between folic acid and a folate receptor present on the cell surface. The ECL cytosensor showed satisfactory sensitive response to HeLa cells in a linear range of 4.25 × 102–4.25 × 105 cells per mL with a low detection limit of 326 cells per mL. The proposed cytosensor had good sensitivity and stability, which can offer a great potential platform for bioassay analysis.

TPGS-stabilized NaYbF4:Er upconversion nanoparticles for dual-modal fluorescent/CT imaging and anticancer drug delivery to overcome multi-drug resistance

Gan Tian,Xiaopeng Zheng,Xiao Zhang,Wenyan Yin,Jie Yu,Dongliang Wang,Zhiping Zhang,Xiangliang Yang,Zhanjun Gu,Yuliang Zhao

Multi-drug resistance (MDR) is a major cause of failure in cancer chemotherapy. Tocopheryl polyethylene glycol 1000 succinate (TPGS) has been extensively investigated for overcoming MDR in cancer therapy because of its ability to inhibit P-glycoprotein (P-gp). In this work, TPGS was for the first time used as a new surface modifier to functionalize NaYbF 4 :Er upconversion nanoparticles (UNCPs) and endowed the as-prepared products (TPGS-UCNPs) with excellent water-solubility, P-gp inhibition capability and imaging-guided drug delivery property. After the chemotherapeutic drug (doxorubicin, DOX) loading, the as-formed composites (TPGS-UCNPs-DOX) exhibited potent killing ability for DOX-resistant MCF-7 cells. Flow-cytometric assessment and Western blot assay showed that the TPGS-UCNPs could potently decrease the P-gp expression and facilitate the intracellular drug accumulation, thus achieving MDR reversal. Moreover, considering that UCNPs process efficient upconversion emission and Yb element contained in UCNPs has strong X-ray attenuation ability, the as-obtained composite could also serve as a dual-modal probe for upconversion luminescence (UCL) imaging and X-ray computed tomography (CT) imaging, making them promising for imaging-guided cancer therapy.

Linlin Zhao,Juan Li,Yuhong Liu,Yuan Wei,Jin Zhang,Jinzha Zhang,Qing Xia,Qiang Zhang,Wenbo Zhao,Xiaoqiang Chen

We reported a novel ECL immunosensor for detection of carcinoembryonic antigen (CEA) based on the reduced graphene Oxide-BaYF 5 :Yb, Er (rGO-BaYF 5 :Yb, Er) nanocomposites. The rGO-BaYF 5 :Yb, Er nanocomposites, used as electrode luminescence materials, were prepared by in situ hydrothermal method, and characterized by TEM, XRD, UV–vis spectra, and upconversion fluorescence spectra. Further, poly (diallyldimethylammonium chloride) (PDDA), Au nanoparticles (AuNPs), and Anti-CEA were successively assembled on the surface of rGO-BaYF 5 :Yb, Er nanocomposites modified-electrode for fabricating ECL immunosensor. The results of the ECL measurements of CEA indicated that this novel ECL immunosensor exhibited high sensitive response to CEA in a linear range of 0.001–80   ng   mL −1 with a low detection limit of 0.87   pg   mL −1 , good selectivity and satisfactory stability.

Core–shell–shell heterostructures of α-NaLuF4:Yb/Er@NaLuF4:Yb@MF2 (M = Ca, Sr, Ba) with remarkably enhanced upconversion luminescence

Yue Su,Xiuling Liu,Pengpeng Lei,Xia Xu,Lile Dong,Xianmin Guo,Xingxu Yan,Peng Wang,Shuyan Song,Jing Feng,Hongjie Zhang

Core–shell–shell heterostructures of α-NaLuF4:Yb/Er@NaLuF4:Yb@MF2 (M = Ca, Sr, Ba) have been successfully fabricated via the thermal decomposition method. Upconversion nanoparticles (UCNPs) were characterized by powder X-ray diffraction (XRD), transmission electron microscopy (TEM), upconversion luminescence (UCL) spectroscopy, etc. Under 980 nm excitation, the emission intensities of the UCNPs are remarkably enhanced after coating the MF2 (M = Ca, Sr, and Ba) shell. Among these samples, CaF2 coated UCNPs show the strongest overall emission, while BaF2 coated UCNPs exhibit the longest lifetime. These results demonstrate that alkaline earth metal fluorides are ideal materials to improve the UCL properties. Meanwhile, although the lattice mismatch between the ternary NaREF4 core and the binary MF2 (M = Sr and Ba) shell is relatively large, the successfully synthesized NaLuF4:Yb/Er@NaLuF4:Yb@MF2 indicates a new outlook on the fabrication of heterostructural core–shell UCNPs.

Calcination-assisted hydrothermal crystallization and comparative up-/down-conversion luminescence of RE3+ (RE=Ho, Er and Tm)-doped YbNbO4 and Yb3NbO7

Bingxin Yuan,Meiting Li,Chi Zhang

The RE 3+ doped YbNbO 4 and Yb 3 NbO 7 nanophosphors (RE=Ho, Er and Tm) were synthesized via a calcination-assisted hydrothermal procedure. YbNbO 4 intensified almost the whole up-conversion (UC)/down-conversion (DC) emissions of RE 3+ by ∼2 times when compared with Yb 3 NbO 7 components, indicating that a lower symmetry environment in the former case is more favorable for photoluminescence. UC luminescence in each case was dominated by a green band at ∼539 nm for Ho 3+ and a NIR band at ∼803 nm for Tm 3+ , while doping YbNbO 4 and Yb 3 NbO 7 with Er 3+ separately yielded the slightly stronger green 556 nm and red 658 nm emissions. The UC/DC luminescence mechanisms were elaborated via Yb 3+ → RE 3+ forward/backward energy transfer and cross relaxation.

Multispectral luminescence of core-shell rare-earth NaLuF4:Yb,Er@NaLuF4:Yb,Tm@NaLuF4 upconversion nanomaterials for ratiometric optical temperature sensing

Wei Zhou

This paper focuses on the study of core-shell rare earth NaLuF 4 :Yb,Er@NaLuF 4 :Yb,Tm@NaLuF 4 upconversion nanoparticles (UCNPs) with multispectral emission for applications in the field of ratiometric temperature sensors. This approach lays the foundation for exploring more luminous intensity ratios (LIRs) between different spectra for temperature sensing. Additionally, it facilitates the use of different LIRs for various temperatures, yielding higher sensitivity. Under 980 nm laser excitation, these UCNPs produce multispectral luminescence covering blue, green, red, and infrared luminescence intervals. Within the temperature range of 293–473 K, we measured the luminescence intensity of different emission peaks and observed that the 803 nm peak exhibits an anomalous thermal enhancement effect. The LIR between the luminescence intensities of different emission peaks is fitted using formulas to calculate the absolute sensitivity ( S a ), relative sensitivity ( S r ), uncertainty ( ΔT ), and repeatability ( R ) of the corresponding LIR. Comparisons revealed that LIR(I 803 /I 540 ) demonstrates greater S a compared to the other LIRs. Furthermore, the S r of LIR(I 803 /I 540 ) is higher in the elevated temperature range, whereas LIR(I 522 /I 540 ) shows greater values in the lower temperature range. By combining the advantages of these two LIRs at different temperatures, LIR(I 522 /I 540 ) is utilized at low temperatures and LIR(I 803 /I 540 ) at high temperatures, achieving a combined S r of 0.67–1.06 %K −1 and a combined ΔT of less than 0.045. The R of LIR(I 803 /I 540 ) is tested and found to be greater than 99 % averaged over five cycles. In summary, the NaLuF 4 :Yb,Er@NaLuF 4 :Yb,Tm@NaLuF 4 UCNPs proposed in this paper demonstrate potential applications in the field of temperature sensing.

Morphology control and upconversion luminescence enhancement of Na1-xLixY.78-yLuyF4:Er.02Yb.2 microcrystals by doping ions with different valences

Dandan Ju,Feng Song,Yingdong Han,Wenjing Cui,Feifei Song,Lisa Liu,Aihua Zhou,Xueqin Wang,Shujing Liu

Trivalent lanthanide ion (Ln 3+ ) doped UC materials can emit visible light under near-infrared (NIR) excitation due to anti-stokes shift and have received wide attention. In this paper, we presented a new strategy to synthesize the uniform and smooth microstructures (rodlike and tubular) based on hexagonal phase NaYF 4 microcrystals . Moreover, we systematically discussed the method of enhancing the upconversion (UC) luminescence intensity via co-doping the ions with different valences (Li + and Lu 3+ ) in order to tailor the environment around lanthanide ions. The UC intensities in the two microstructures were significantly improved. The results suggested the great application potential of tubes and rods of β-NaYF 4 microcrystals in anti-counterfeiting, laser media, photonic crystals , and waveguides .
