A Bilayer Polyurethane Patch with Sustained Growth Factor Release and Antibacteria for Re-epithelization of Large-Scale Oral Mucosal Defects

Qiao Zhang,Jinlin Chen,Tianyu Zhang,Dan Liu,Xirui Long,Jiehua Li,Lu Jiang,Yanchao Wang,Hong Tan

In the field of oral and maxillofacial surgery, extensive oral soft-tissue injuries occur repeatedly in clinical practice; however, effective restorative materials are lacking. In this study, a biodegradable waterborne polyurethane patch featuring a mucosa bionic bilayer structure is presented. This patch consists of a porous scaffold layer that faces the lesion, incorporating a polydopamine coating to achieve sustained release of epidermal growth factors (EGFs) for mucosal defect reconstruction. Additionally, there is a dense barrier layer toward the oral cavity loaded with silver nanoparticles, which prevents bacteria from entering the wound and simultaneously acts as a physical barrier. This patch can sustainably release EGF in vitro for 2 weeks, thereby facilitating the proliferation and migration of HaCaT and L929 cells, while effectively killing common oral cavity bacteria. In a rabbit buccal mucosal full-thickness defect model, the patch demonstrates better efficacy than the clinical benchmark, decellularized extracellular matrix (dECM). It effectively reduces wound inflammation and significantly upregulates gene expression associated with epithelialization by activating the EGF/epidermal growth factor receptor (EGFR) pathway. These mechanisms promote the proliferation, differentiation, and migration of epithelial/keratinocyte cells, ultimately expediting mucosal defect healing and wound closure.

Zwitterionic polymer coating on medical sponges via a silane coupling agent for inhibiting bacterial and fibroblast adhesion

Shizhen Jia,Hui Cao,Xiangchao Liang,Shuang Wu,Rong Zeng,Mei Tu

Medical sponges are widely used in wound management, however the adhesion of bacterial or wound tissue to them may cause infection or secondary damage to patients. In this work, anti-bioadhesive polyvinyl alcohol-formaldehyde sponges with zwitterionic coating were prepared by the dip-coating process using the reactive zwitterionic phosphorylcholineated polyvinylamine (PVAm-PC) and commercial silane coupling agent KH560 at different mass ratios, and their structure, physicochemical, biocompatible and anti-bioadhesive properties were investigated. As compared to pristine sponges, the PVAm-PC-KH560 (PPK) modified sponges with the interconnected pore structure had almost unchanged water vapor transmission rate but decreased swelling degree to a certain extent possibly due to the formation of rigid Si– O –Si networks in the sponges. In vitro biological evaluation confirmed that PVAm-PC-KH560 modified sponges were not cytotoxic and had good resistance to bacteria adhesion with only ∼16% and ∼11% relative bacterial adhesion rate for E. coli and S. aureus respectively. And the optimized PPK-2 sponge showed the best comprehensive bacterial adhesion inhibition properties. Further adhesion assay of L929 fibroblasts showed that PPK-2 sponge could significantly reduce the adhesion of L929 fibroblasts. The results strongly suggested that dip-coating with zwitterionic PVAm-PC and KH560 may provide a promising and facile method to construct an anti-bioadhesive surface for biomedical applications.

A composite hydrogel with co-delivery of antimicrobial peptides and platelet-rich plasma to enhance healing of infected wounds in diabetes

Shikun Wei,Pengcheng Xu,Zexin Yao,Xiao Cui,Xiaoxuan Lei,Linlin Li,Yunqing Dong,Weidong Zhu,Rui Guo,Biao Cheng

Diabetic wound healing remains a major challenge due to its vulnerability to bacterial infection, as well as the less vascularization and prolonged inflammatory phase. In this study, we developed a hydrogel system for the treatment of chronic infected wounds, which can regulate inflammatory (through the use of antimicrobial peptides) and enhance collagen deposition and angiogenesis (through the addition of platelet-rich plasma (PRP)). Based on the formation of Schiff base linkage, the ODEX/HA-AMP/PRP hydrogel was prepared by mixing oxidized dextran (ODEX), antimicrobial peptide-modified hyaluronic acid (HA-AMP) and PRP under physiological conditions, which exhibited obvious inhibition zones against three pathogenic bacterial strains ( E. coli, S. aureus and P. aeruginosa ) and slow release ability of antimicrobials and growth factors. Moreover, CCK-8, live/dead fluorescent staining and scratch test confirmed that ODEX/HA-AMP/PRP hydrogel could facilitate the proliferation and migration of L929 fibroblast cells. More importantly, in vivo experiments further demonstrated that the prepared hydrogels could significantly improve wound healing in a diabetic mouse infection by regulating inflammation, accelerating collagen deposition and angiogenesis. In addition, prepared hydrogel showed a significant antibacterial activity against S. aureus and P. aeruginosa, inhibited pro-inflammatory factors (TNF-α, IL-1β and IL-6), enhanced anti-inflammatory factors (TGF-β1) and vascular endothelial growth factor (VEGF) production. The findings of this study suggested that the composite hydrogel with AMP and PRP controlled release ability could be used as a promising candidate for chronic wound healing and infection-related wound healing.

Radially Electrospun Fibrous Membrane Incorporated with Copper Peroxide Nanodots Capable of Self-Catalyzed Chemodynamic Therapy for Angiogenesis and Healing Acceleration of Diabetic Wounds

Yong Huang,Lin Qi,Zheng Liu,Yulin Jiang,Jing Wang,Limin Liu,Yubao Li,Li Zhang,Ganjun Feng

Vascular dysfunction severely hinders the healing process of diabetic wounds. Therefore, a radially structured fibrous membrane was fabricated through electrospinning by using a polycaprolactone (PCL) and polyvinylpyrrolidone (PVP) mixed solution containing copper peroxide nanoparticles (CPs) as the chemodynamic therapy (CDT) agents, aiming to simultaneously accelerate tissue regeneration and angiogenesis. The fabricated membrane allowed for the in situ H2O2 generation activated by the acidic diabetic microenvironment and the subsequent Fenton-type reactions to realize 99.4% elimination against Staphylococcus aureus. Besides, the released Cu2+ ions significantly enhanced the expression of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in human umbilical vein endothelial cells (HUVECs), and they showed enhanced in vitro angiogenesis. Interestingly, the CP-embedded membrane also guided cell spreading and orientated migration of L929 fibroblasts along the fiber distribution through the radially aligned topology. The in vivo implantation indicated that the raidally structured membrane modified by CPs not only dramatically accelerated wound healing of diabetic Sprague–Dawley (SD) rats in 14 days but also promoted angiogenesis in wound sites. The combination of the in situ CDT with the radially structured morphology of the functional membrane is highly promising in applications to promote diabetic wound healing through anti-infection and revascularization.

Construction of bio-inspired multifunctional triple-layered nanocomposite enhances the cell adhesion and treatment of wound repair

Hua Chao,Lyu Guozhong

A bio-inspired multifunctional nanocomposite comprises a triple-layer keratin (KER)/collagen (COG)/bioactive glass (BGN) matrix. A fibrous material was developed by combining lyophilized methods. The material consists of three layers: a fibrous layer on top, a spongy layer, and a porous layer at the bottom. The design aims to enhance the physicochemical and biological properties of the nanocomposites through synergistic effects. FTIR and SEM were employed to evaluate the physicochemical characteristics of the final material. The triple-layer nanocomposites include a porous structure and fibrous with excellent stability, swelling, and porosity, which are crucial for tissue engineering. Fibroblast L929 and keratinocyte HaCaT cells were utilized to study the nanocomposites’ in vitro biological and fluorescence properties. L929 cell migration was enhanced at 100 μg/mL concentrations of the samples after 24 h of damage in a wound scratch assay. The results demonstrated strong cell adhesion and growth throughout the composite layers. The antimicrobial was examined using Staphylococcus aureus (Gram +Ve) and Escherichia coli (Gram −Ve) pathogens, and the results reveal that KER/COG/BGN triple-layered nanocomposite effectively kills the bacteria. Combining fibrous material implantation on collagen with keratin and bioglass has successfully constructed a triple-layer matrix for wound-repairing tissue engineering.

A cochlear implant loaded with dexamethasone and coated with hyaluronic acid to inhibit fibroblast adhesion and proliferation

Kunhong Yu,Jingwen Hou,Zhu Jin,Keqin Wu,Shaohui Xu,Ning Yang,Yuanyuan Shen,Tingting Tang,Shengrong Guo

Inflammation and fibrosis caused by inserting cochlear implants into the inner ear are of great concern in clinical treatment. To address these issues, we studied new cochlear implant electrode analogs, polydimethylsiloxane (PDMS) filaments loaded with anti-inflammation/fibrosis dexamethasone (Dex) and coated with hyaluronic acid (HA) as a surface modifier. The PDMS filaments were prepared by incorporating Dex into PDMS that contained different amounts of poloxamer 188 (P188) as a drug release enhancer. HA coatings were performed to prevent protein adsorption by treating the surfaces of the PDMS filaments with O 2 plasma, 3-aminopropyltriethoxysilane and HA in sequence. The prepared PDMS filaments were investigated by attenuated total reflection Fourier transform infrared spectroscopy , contact angle measurement and atomic force microscopy . Their protein adsorption and drug release behaviors were examined as well. L929 mouse fibroblast cells were used to evaluate the anti-cell adhesion/proliferation effects of the PDMS filaments. The results showed that the prepared PDMS filaments had the expected structure, good surface properties and excellent anti-fibroblast cell proliferation effects. The HA coatings provided filament surfaces that were more hydrophilic, smoother and more protein-repelling. Dex release was significantly facilitated by adding P188 and slightly slowed down by the HA coatings. The PDMS filaments that contained 5% Dex and 5% P188 and that were coated with HA showed a significant decrease in the fibroblast cell number by approximately 51%, with obviously reduced cell adhesion on the surfaces.

A multi-functional double cross-linked chitosan hydrogel with tunable mechanical and antibacterial properties for skin wound dressing

Fengling Liu,Lu Wang,Xinrang Zhai,Shunxian Ji,Jingjia Ye,Zhiqiang Zhu,Chong Teng,Wei Dong,Wei Wei

Chitosan hydrogels with essential antibacterial properties and biocompatibility have great potential in tissue engineering and regeneration medicine. However, pure chitosan hydrogel could be limited by insufficient mechanical properties. In this work, we designed a multi-functional chitosan hydrogel based on the combination of chitosan methacrylate (CTSMA) and sulfhydrated chitosan (CTSSH), which is cross-linked simultaneously by free-radical polymerization reaction and Thiol-ene reaction. The CTSMA/CTSSH (CMS) hydrogels displayed superior tissue adhesive and mechanical properties when compared to pure CTSMA hydrogel. Additionally, the resulting hydrogels exhibited potent antimicrobial effects against both E. coli and S. aureus . Besides, the CMS hydrogels exhibited good biocompatibility as demonstrated by cytotoxicity and cell proliferation experiments using fibroblasts cells (L929) and adipose-derived stem cells (ADSCs). In vivo experiment, the repairing effect of hydrogels on full-thickness skin defect model in rats was studied. Histological and immunohistochemical staining results showed that CMS hydrogels promoted angiogenesis , dermal repair and epidermal regeneration. Overall, the study highlights the potential of the CMS hydrogels as a promising biomaterial in wound healing applications.
