Hydrogel microspheres are considered ideal carriers with broad applications in 3D cell culture, drug delivery, and microtissue construction. Although multiple methods have been developed for generating hydrogel microspheres, there is still a lack of a universal approach that combines operability, stability, cost-effectiveness, and biocompatibility. In this work, a novel rotating microfluidic system (RMS) is proposed, which can rapidly fabricate diverse poly(ethylene glycol) diacrylate/sodium alginate (PEGDA/SA) hydrogel microspheres by motor-driven rotation of the oil phase to form a special T-shaped structure with the needle. The main part of the system consists of commercially available motors, a beaker, and needles that do not require precision machining and are user-friendly with low cost. Moreover, by adjusting system parameters such as the needle structure, flow rate, and rotational speed, the platform enables rapid fabrication of hydrogel microspheres with different sizes and diverse cores, including crescent, thick wavy, oval, and spherical. Furthermore, tumor cell-laden hyaluronic acid methacrylate/sodium alginate (HAMA/SA) hydrogel microspheres were fabricated by using this system, which demonstrated good cell viability and proliferation in the subsequent 3D culture. In vitro drug evaluation of tumor models using cisplatin revealed the potential of this system for drug evaluation. These results indicated that RMS has good potential in other 3D cell culture-based biomedical applications.

PurposeThis study reports on the synthesis and characterization of biodegradable dextran–allyl isocyanate–ethylamine (Dex-AE)/polyethylene glycol–diacrylate (PEGDA) hydrogels for the controlled release of gentamycin sulphate (GS) and in vitro inhibition of organisms.MethodsThe Dex-AE precursor was prepared through a 2-step chemical modification and characterized by Fourier transform infrared spectroscopy (FTIR).ResultsScanning electron microscopy (SEM) results revealed that an increase in Dex-AE content led to an initial decrease in pore size of the Dex-AE/PEGDA hydrogels, but a further increase in Dex-AE content resulted in a slightly increase of pore size. The swelling data indicated that the swelling ratio depended on the precursor feed ratio. GS was incorporated into the hydrogels through 2 different methods, i.e., immersed and crosslinked. The crosslinked GS-Dex-AE/PEGDA hydorgels exhibited stronger antimicrobial activities against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. Finally, the viscoelastic properties of crosslinked GS-Dex-AE/PEGDA hydorgels were investigated.

The manipulation of micron-scale semicircular tubular structures has wide applications in micro-nano processing, device manufacturing, biomedicine, and micron sensing and measurement. Here, we propose a method to fabricate and manipulate semicircular tubular structures based on optically induced dielectrophoresis (ODEP). First, electric field intensity simulations are performed for polyethylene glycol diacrylate (PEGDA) semicircular tubular structures with different conductivities and of different heights. In addition, the polarization model based on slender rods reveals that the semicircular tubular structure is subject to a negative dielectrophoretic force and tends to move along the vertical direction of the central axis. Finally, according to the maximum movement speed of the semicircular tubular structure, the resistance and dielectrophoretic force it receives are characterized. This allows for the realization of the translation and rotation operations of semicircular tubular structures of different lengths, and the assembly of multiple structures into different shapes. This assembly method holds significant promise for applications in biomedicine and the manufacturing and processing of micro-nano devices.

Clinical work and research on diabetic wound repair remain challenging globally. Although various conventional wound dressings have been continuously developed, the efficacy is unsatisfactory. The effect of drug delivery is limited by the depth of penetration. The sustained release of biomolecules from biological wound dressings is a promising treatment approach to wound healing. An assortment of cell-derived exosomes (exos) have been proved to be instrumental in tissue regeneration, and researchers are dedicated to developing biomolecules carriers with unique properties. Herein, we reported a methacrylate gelatin (GelMA) microneedles (MNs) patch to achieve transdermal and controlled release of exos and tazarotene. Our MNs patch comprising GelMA/PEGDA hydrogel has distinctive biological features that maintain the biological activity of exos and drugs in vitro. Additionally, its unique physical structure prevents it from being tightly attached to the skin of the wound, it promotes cell migration, angiogenesis by slowly releasing exos and tazarotene in the deep layer of the skin. The full-thickness cutaneous wound on a diabetic mouse model was carried out to demonstrate the therapeutic effects of GelMA/PEGDA@T + exos MNs patch. As a result, our GelMA/PEGDA@T + exos MNs patch presents a potentially valuable method for repairing diabetic wound in clinical applications. Graphic Abstract

This paper studies the influences of fabrication parameters on the optical properties of poly(ethylene glycol) diacrylate(PEGDA) hydrogels during polymerization, including the irradiation intensity, irradiation time, photoinitiator concentration, and water content. The refractive index of PEGDA hydrogels polymerized under various conditions is measured, with the results shown to be valuable for future research applying PEGDA hydrogels as optical materials. In addition, it is found that the photoinitiator concentration used can be as low as 1.0 wt%, which is severalfold lower than that previously reported, making PEGDA hydrogels more desirable for bioapplications.

Polylactic acid (PLA) is one of the biodegradable materials that has been used in the areas of surgical healing lines, cancer treatment, and wound healing. However, the application of PLA is still rather limited due to its high hydrophobicity and poor antibacterial activity. In order to enhance the antifouling and antibacterial performances of PLA, here we modified the surface of PLA with various sizes of hydrogel micropatterns in negative or positive mode using plasma treatment, the photomask technique, and UV-graft polymerization. The hydrogel micropatterns consist of poly(ethylene glycol) diacrylate (PEGDA), poly(2-methacryloyloxyethylphosphorylcholine) (PMPC), and poly(methacryloyloxyethyltrimethylammonium chloride) (PDMC). Compared to PLA, the patterned PLA (PLA–PMPC/PDMC/PEGDA) shows obviously enhanced antifouling and antibacterial activities. For PLA–PMPC/PDMC/PEGDA with either positive or negative micropatterns, the antifouling and antibacterial properties are gradually increasing with decreasing the size of micropatterns. Compared with PLA–PMPC/PDMC/PEGDA bearing positive and negative micropatterns in the same size, the PLA–PMPC/PDMC/PEGDA with negative micropatterns exhibits slightly better biological activity and the PLA–PMPC/PDMC/PEGDA with 3 μm negative hydrogel micropatterns shows the best hydrophilicity, antifouling, and antibacterial properties. Combining the in vitro hemolysis assay, cytotoxicity, water absorption test, and degradation test results, it is suggested that the fabrication of hydrogel micropatterns onto the PLA surface could significantly improve biological activities of PLA. We expect that this work would provide a new strategy to potentially develop PLA as a promising wound dressing.

Polymeric microneedles have attracted increasing attention as a minimally invasive platform for delivering drugs or vaccines in a more patient-friendly manner. However, traditional microfabrication techniques using negative molds with needle-shaped cavities usually require cumbersome centrifugation and vacuum degassing processes, which have restricted the scaled-up mass production of polymeric microneedles. Herein, a novel polydimethylsiloxane (PDMS)-based negative mold with cavities packed with silk fibroin scaffold is developed for rapid fabrication of polymeric microneedles, which comprise primarily the composition of poly(ethylene glycol) diacrylate (PEGDA) and sucrose as the needle matrix. Fibroin scaffolds can instantly adsorb prepolymer solution due to capillary force, and subsequently initiate the formation of microneedles via photoinduced polymerization. Based on three types of model drugs, including Rhodamine B (RhB), indocyanine green (ICG), and doxorubicin (DOX), the fabricated PEGDA/sucrose microneedles can realize effective transdermal delivery and controllable release of therapeutic molecules by regulating the sucrose content. The presented method provides a simple strategy for quick fabrication of polymeric microneedles toward transdermal drug delivery applications.

Type 2 diabetes is one of the major challenges that the world is facing today. However, metformin (MET) as most type 2 diabetics’ first-line oral hypoglycemic drug may cause serious side effects such as gastrointestinal irritation and nausea which reduce the patients’ medication compliance. Therefore, the aim of the study was to design a safe and effective self-treatment device for the delivery of MET. Here, a kind of coated microneedle (MN) patches based on poly(ethylene glycol)diacrylate (PEGDA) were prepared by a two-step casting method and photopolymerization process for transdermal administration of MET. The needles wrapped with drug-loaded hyaluronic acid (HA) coating showed promising mechanical properties and drug delivery ability that allowed them to penetrate the skin barrier for rapid drug delivery, and they had no skin irritancy. The in vivo experiment of type 2 diabetic rats showed a satisfying hypoglycemic effect of the coated MN patches. The study shows that the prepared MN patches will be a potential method for the treatment of type 2 diabetes in the future. Graphical Abstract

Chitosan (CS)-based nanocomposites have been studied in various fields, requiring a more facile and efficient technique to fabricate nanoparticles with customized structures. In this study, Ag@methacrylamide CS/poly(ethylene glycol) diacrylate (Ag@MP) micropatterns are successfully fabricated by femtosecond laser maskless optical projection lithography (Fs-MOPL) for the first time. The formation mechanism of core-shell nanomaterial is demonstrated by the local surface plasmon resonances and the nucleation and growth theory. Amino and hydroxyl groups greatly affect the number of Ag@MP nanocomposites, which is further verified by replacing MCS with methacrylated bovine serum albumin and hyaluronic acid methacryloyl, respectively. Besides, the performance of the surface-enhanced Raman scattering, cytotoxicity, cell proliferation, and antibacterial was investigated on Ag@MP micropatterns. Therefore, the proposed protocol to prepare hydrogel core-shell micropattern by the home-built Fs-MOPL technique is prospective for potential applications in the biomedical and biotechnological fields, such as biosensors, cell imaging, and antimicrobial.

Due to their internal continuity and smooth surface geometry, three-period minimal surfaces (TPMS) have been widely applied in the structural design and fabrication of bone tissue engineering scaffolds. To achieve a biomimetic simulation of natural bone tissue structure and enhance the antimicrobial and anti-inflammatory properties of the implant site, this study innovatively proposes a biomimetic gradient TPMS scaffold with antimicrobial and anti-inflammatory functions. First, based on the hierarchical structural characteristics of compact bone and cancellous bone in natural bone, four TPMS gradient structures were parametrically designed. These four structures were then subjected to comprehensive testing to identify the one with the optimal performance. Furthermore, to address the limitation of single-function traditional bone repair scaffolds, this study used polyethylene glycol diacrylate (PEGDA) as the matrix and prepared a multifunctional composite slurry by incorporating nano-hydroxyapatite (HA), nano-magnesium oxide (MgO), and tannic acid (TA). A series of test results indicate that the I-WP scaffold exhibits the optimal combination of mechanical and permeability properties, while also demonstrating excellent in vitro osteogenic differentiation capacity and antibacterial and anti-inflammatory effects. In summary, the gradient-structured composite scaffold prepared in this study achieves an integrated structure that enhances mechanical properties, boosts osteogenic mineralization activity, and provides synergistic antibacterial and anti-inflammatory functions. It offers new insights for the structural design and functional optimization of bone tissue engineering scaffolds and holds promising application prospects.

Bile duct injury is a common complication of hepatobiliary surgeries in clinical practice with poorly satisfactory treatment outcomes. Implanting a tissue-engineered tubular scaffold for supporting and replacing defected native bile duct is an important treatment option for bile duct injury. Here a dual-layer tubular scaffold with poly (lactic-co-glycolic acid) (PLGA) inner layer and gelatin methacrylate (GelMA)/poly (ethylene glycol) diacrylate (PEGDA) outer layer was fabricated using a two-stage molding method for bile duct regeneration. Further, induced mesenchymal stem cells (MSC)-derived cholangiocyte-like cells were embedded in the outer layer during fabrication. The inner layer of PLGA offered adequate mechanical strength for the scaffold, and the outer layer of GelMA/PEGDA provided excellent biocompatibility and conditions for the MSCs and their induced differentiation. Moreover, cholic acid and its derivates are believed to induce MSC-cholangiocyte differentiation in two-dimensional culture and could affect the MSCs transforming to cholangiocyte-like cells in the hydrogel. Induced-MSC-laden GelMA/PEGDA-PLGA dual-layer tubular scaffold and PLGA single-layer scaffold were transplanted into animal models for assessing the repair effect on bile duct injury. After 12 wk, induced-MSC-laden dual-layer tubular scaffold promoted bile duct repairing and enhanced more intact biliary epithelium regenerating compared to PLGA single-layer scaffold. Therefore, the induced-MSC-laden dual-layer tubular scaffold with superior biocompatibility and distinct function of inducing differentiation exerted a satisfactory effect on bile duct regeneration and could provide an ideal option for the clinical treatment of bile duct injury in the future.

This paper proposes a dissolved oxygen sensing system based on PMMA plastic optical fiber covered at one end with biocompatible PEGDA (Polyethylene glycol diacrylate) hydrogel film. PEGDA, 2-hydroxy-2-methyl-propiophenone and DI water mixture was used as hydrogel precursors, and a certain concentration of oxygen-sensitive fluorescent indicator was incorporated into it. Fluorescence quenching method was used to detect dissolved oxygen in water, the doping concentration of fluorescent indicator in the hydrogel film was optimized. The experimental results indicate that the plastic optical fiber coated with hydrogel sensing film has good reproducibility, and the response time is within 10 s. The sensing characteristics of dissolved oxygen at 30 °C, 35 °C, and 40 °C were tested respectively, and the results showed that the linearity of the system is best at 35 °C. Within the range of pH 2–12, the fluorescence intensity of the fluorescent indicator is very stable. The sensor with biocompatible material is expected to be applied to the monitor of dissolved oxygen in the domain of biomedicine.
