Organic nanomaterials with efficient ROS generation and high photothermal conversion efficiency have emerged as a new generation therapeutic agent for PDT/PTT synergistic cancer treatment. However, most of the organic nanomaterials exhibited weak emission in water because of the aggregation-caused quenching (ACQ) effect, which seriously hampers their biological application in fluorescence bioimaging. In this study, we design and synthesize a new organic small molecule (T-BDP) with a donor–acceptor–donor (D–A–D) structure by the conjugation of boron-dipyrromethene (BODIPY) and triphenylamine (TPA). Moreover, a 1,8-naphthalenediimide (NI) structure with electron-withdrawing ability is introduced onto the core of BODIPY to further enhance the intramolecular charge transfer, leading to the redshift of absorption to the near-infrared region. T-BDP displays significant aggregation-induced emission (AIE) performance, probably due to the presence of two twisted TPA groups onto the BODIPY core. Accordingly, after self-assembly into nanoparticles, T-BDP NPs exhibit strong near-infrared emission in water. Under single 635 nm laser irradiation, T-BDP NPs could generate reactive oxygen species and heat simultaneously. The photothermal conversion efficiency of T-BDP NPs is determined to be 50.9%. The low dark toxicity and high photocytotoxicity of T-BDP NPs are verified against A549 cells using the MTT assay and the AM/PI staining method. Due to the strong emission of T-BDP NPs, their accumulation and subcellular localization in cancer cells are observed using a laser confocal fluorescence microscope. The results demonstrate that T-BDP NPs are mainly located in the lysosomes of cancer cells. Thus, the as-prepared small molecule-based AIE nanoparticles hold great potential for fluorescence imaging-guided PDT/PTT synergistic tumor therapy.

Bacterial infections considerably limit the success of scaffold transplantation. CuO 2 may generate hydroxyl radicals through a self-cascade catalytic process, which is effective for sterilization. However, reducing CuO 2 -derived Cu + ions to Cu° can lead to a decline in catalytic activity. To overcome these limitations, we anchored CuO 2 onto MoS 2 to establish stable Cu-S-Mo interfacial sites. The CuO 2 /MoS 2 nanozyme was fabricated using a low-temperature co-precipitation technique and introduced into poly- l -lactic acid scaffolds using selective laser sintering. The Cu-S-Mo interfacial sites showed a triad of functions that synergistically enhanced catalytic performance. These sites stabilized the Cu atoms, augmented catalytic activity by facilitating efficient electron transfer through the Mo 4+ /Mo 6+ redox couple, and improved the conduction of heat derived from the photothermal response of MoS 2 to the Cu catalyst. The synergistic action of chemodynamic and photothermal therapy, mediated by the Cu-S-Mo sites, resulted in remarkable antibacterial efficacy, with 92.3 % and 93.5 % inhibition rates against Staphylococcus aureus and Escherichia coli , respectively. Experimental evidence and simulations demonstrated that Cu-S-Mo structure stabilized the Cu atoms and improved electron transfer via interatomic charge transfer. These results highlight the potential of the CuO 2 @MoS 2 /PLLA scaffold in eliminating bacterial infections.

The development of drug-resistant biofilms has resulted in treatment failures for multidrug resistant (MDR) bacteria, presenting a substantial challenge to global public health. Consequently, the development of antibacterial agents has become an urgent priority. To overcome the limitations of poor penetration and inadequate efficacy of single-modal treatments for MDR bacterial biofilm infections, this study developed a Ce6/SFP nanocomposite system utilizing silk fibroin and polydopamine. The nanoparticles, fabricated via a desolvation-in situ polymerization method, demonstrated pH-responsive drug release properties. When subjected to an 808 nm NIR laser, the material exhibited a photothermal conversion efficiency of 29.3%. Furthermore, when paired with singlet oxygen produced through 660 nm excitation, it was highly effective in eliminating MDR Escherichia coli and MDR Staphylococcus aureus. The synergistic effect of photothermal therapy (PTT) and photodynamic therapy (PDT) disrupted the stability of the cell membrane and enhanced the antibacterial efficacy. Experimental results showed that the photothermal effect increased biofilm permeability, significantly improving Ce6 penetration depth. Moreover, in vivo treatments exhibited accelerated wound healing. Biocompatibility assessments revealed a hemolysis rate below 5% and no pathological damage to major organs. This study proposes a synergistic strategy to effectively address the challenges posed by biofilms and bacterial resistance.

At present, cancer has become a major disease threatening human health worldwide. Therefore, developing targeting guided multimode synergetic therapy has become one of the hot spots in current antitumor research and is also a great challenge. Herein, a new Fe 3 O 4 /g-C 3 N 4 @PPy-DOX nanocomposite containing magnetic iron oxide (Fe 3 O 4 ) nanoparticles (NPs), lamellar structure of graphite-like carbon nitride (g-C 3 N 4 ) and polypyrrole (PPy) shell with the loaded anti-tumor drug doxorubicin hydrochloride (DOX) was designed and prepared. The monodisperse Fe 3 O 4 nanoparticles (NPs) with the diameter of 20 nm endowed the nanocomposite with the magnetic targeting ability, reducing damage to normal tissues. It is very interesting that the Fe 3 O 4 NPs also possessed photosensitizer function for photodynamic therapy (PDT). The g-C 3 N 4 sheets as the photocatalysis towards the degradation of water for generating O 2 could effectively improve the hypoxia of solid tumors and increase the efficiency of PDT. In addition, PPy has high light-to-heat conversion efficiency, so was chosen for the cancer photothermal therapy (PTT). Finally, an anticancer drug (DOX) was loaded on the nanocomposite because the presence of mesoporous structure. Thus, the prepared Fe 3 O 4 /g-C 3 N 4 @PPy-DOX nanocomposites exhibit synergetic chemotherapy/PTT/enhanced PDT antitumor effect. This study provides an inspiration for combining targeting and multimodality to improve the anticancer efficiency.

Chemodynamic therapy (CDT) can effectively induce immunogenic cell death (ICD) in tumours and is thus a promising strategy for boosting the efficacy of immunotherapy. However, the mechanism by which CDT enhances ICD and lowers ICD efficiency is unknown and this restricts its clinical application. In this study, a second near-infrared (NIR-II) window irradiation-triggered hydrogen peroxide (H 2 O 2 ) self-supplying nanocomposite ((Cu 2 Se-CaO 2 )@LA) was constructed. The modified lauric acid was melted by the heat energy of the NIR-II irradiation, to expose the CaO 2 nanoparticles, and they then reacted with water to produce H 2 O 2 and Ca 2+ . H 2 O 2 was then converted to hydroxyl radicals by the photothermal-enhanced CDT process of the Cu 2 Se nanocubes. Notably, the CDT and Ca 2+ overload was found to induce sequential damage to the mitochondria and endoplasmic reticulum (ER), which upregulated the PERK-mediated eIF2α phosphorylation pathway and caused subsequent ICD. NIR-II irradiation of the (Cu 2 Se-CaO 2 )@LA also increased reactive oxygen species (ROS) formation and this was sufficient to increase dendritic cell maturation, attracting cytotoxic T lymphocytes, and suppressing tumour growth in vivo. Overall, we demonstrated that an enhanced CDT strategy under NIR-II exposure and H 2 O 2 self-supply can induce extensive ICD by inducing mitochondria-associated ER stress, which represents a highly effective and promising strategy for ICD amplification and tumour immunotherapy. Statement of significance In this study, a second near-infrared window (NIR-II) irradiation-triggered and H 2 O 2 self-supplying nanocomposite (named (Cu 2 Se-CaO 2 )@LA) was constructed and tested both in vitro and in vivo . These nanoparticles demonstrated promising antitumor activity as designed. Mechanistically, the nanoparticles could damage mitochondria and upregulate the PERK-mediated eIF2αphosphorylation pathway, further causing endoplasmic reticulum stress, and inducing immunogenic cell death through dendritic cell maturation and cytotoxic T lymphocyte recruitment augmented activity. This system represents a highly effective and promising strategy for enhancing tumor immunotherapy and provides new insights for future studies and design refinements.

Titanium-based materials hold great biomedical potential owing to excellent biocompatibility. But titanium diboride (TiB 2 ), an emerging metal boride, is rarely explored for antitumor use, and developing multifunctional TiB 2 -based nanoplatforms with enhanced catalytic activity is urgently needed. Herein, we constructed a nanoplatform (TiB 2 -PEG-DOX) by modifying TiB 2 nanosheets (TiB 2 NSs) with amino polyethylene glycol (NH 2 -PEG-NH 2 ) and loading doxorubicin (DOX) via electrostatic interaction. We first clarified the boron (B)-mediated Ti valence cycle: B regulates Ti electron transfer, promotes Ti 4+ reduction to Ti 2+ , enhancing H 2 O 2 -rich tumor microenvironment Fenton-like reaction to generate cytotoxic •OH for chemodynamic therapy (CDT). Meanwhile, TiB 2 -PEG-DOX shows 53.53% photothermal conversion efficiency under 808 nm near-infrared (NIR) irradiation for efficient photothermal therapy (PTT), and NIR further triggers on-demand DOX release. DOX accumulates at tumor sites via the enhanced permeability and retention (EPR) effect to exert chemotherapy (CT). In vitro, it eliminates 86.32% of HCT-116 colorectal cancer cells via CDT/PTT/CT synergy; in vivo, it achieves nearly complete tumor ablation in mice with excellent biocompatibility. This work provides a high-performance synergistic antitumor platform and establishes a B-modulated metal valence cycling paradigm, paving the way for titanium-based nanomedicines.

BODIPY dyes have recently been used for photothermal and photodynamic therapy of tumors. However, complex multi-material systems, multiple excitation wavelengths and the unclear relationship between BODIPY structures and their PTT/PDT efficiency are still major issues. In our study, nine novel BODIPY near-infrared dyes were designed and successfully synthesized and then, the relationships between BODIPY structures and their PTT/PDT efficiency were investigated in detail. The results showed that modifications at position 3,5 of the BODIPY core with conjugated structures have better effects on photothermal and photodynamic efficiency than the modifications at position 2,6 with halogen atoms. Density functional theory (DFT) calculations showed that this is mainly due to the extension of the conjugated chain and the photoinduced electron transfer (PET) effect. By encapsulating BDPX-M with amphiphilic DSPE-PEG2000-RGD and lecithin, the obtained NPs not only show good water solubility and biological stability, but also could act as superior agents for photothermal and photodynamic synergistic therapy of tumors. Finally, we obtained BODIPY NPs that exhibited excellent photothermal and photodynamic effects at the same time under single irradiation with an 808 nm laser (photothermal conversion efficiency: 42.76%, A/A0: ∼0.05). In conclusion, this work provides a direction to design and construct phototherapeutic nanoparticles based on BODIPY dyes for tumor treatment.

Introduction Photoimmunotherapy is a breakthrough treatment for malignant tumors. Its uniqueness is that it uses antibody mediated targeted delivery to achieve high tumor specificity and uses laser-activated biophysical mechanism to accurately induce the rapid death of cancer cells and avoid damaging the surrounding normal tissues. Methods In this paper, an iron-based micelle was designed to encapsulate the photothermal agent indocyanine green (ICG) and a cyclic tripeptide of arginine-glycine-aspartic acid (cRGD) as targeted multifunctional ICG@SANPs-cRGD nanoparticles for combined photothermal/photodynamic/immune therapy of breast cancer. Results The experimental results show that ICG@SANPs-cRGD nanoparticles have good biocompatibility and photothermal conversion ability. Photothermal therapy (PTT) and photodynamic therapy (PDT) based on ICG@SANPs-cRGD can not only inhibit the proliferation, invasion and migration of tumor cells, but also directly kill tumor cells by inducing apoptosis or necrosis. Dual-mode fluorescence light (FL) and magnetic resonance imaging (MRI) imaging in mice confirmed the selective accumulation at tumor sites and imaging ability of ICG@SANPs-cRGD. PTT/PDT combined with Anti-PD-L1 immunotherapy based on ICG@SANPs-cRGD mediated the immunogenic cell death (ICD) of tumor cells by regulating the expression of immune-related indicators and activated the body's immune response mechanism, which enhanced the immunotherapy effect of immune checkpoint block (ICB). PTT/PDT combined with Anti-PD-L1 therapy not only prevented the progression of the primary tumor but also inhibited the distant metastasis of the tumor. Discussion This study explores the biomedical application of PTT/PDT combined with Anti-PD-L1 based on ICG@SANPs-cRGD nanomaterials for breast cancer treatment and demonstrates the potential of ICG@SANPs-cRGD as a multifunctional therapeutic platform for future cancer therapy.

Phototheranostics has attracted considerable attention in the fields of cancer diagnosis and treatment. However, the complete eradication of solid tumors using traditional phototheranostics is difficult because of the limited depth and range of laser irradiation. New phototheranostics enabling precise phototherapy and post-treatment imaging-guided programmed therapy for residual tumors is urgently required. Accordingly, this study developed a novel transformable phototheranostics by assembling hyaluronic acid (HA) with copper-nitrogen-coordinated carbon dots (CDs). In this transformable nanoplatform, named copper-nitrogen-CDs@HA, the HA component enables the specific targeting of cluster determinant (CD) 44-overexpressing tumor cells. In the tumor cells, redox glutathione converts Cu(II) (cupric ions) into Cu(I) (cuprous ions), which confers the novel transformable functionality to phototheranostics. Both in vitro and in vivo results reveal that the near-infrared-light-photoactivated CuII-N-CDs@HA could target CD44-overexpressing tumor cells for precise synergistic photothermal therapy and photodynamic therapy. This study is the first to observe that CuII-N-CDs@HA could escape from lysosomes and be transformed in situ into CuI-N-CDs@HA in tumor cells, with the d9 electronic configuration of Cu(II) changing to the d10 electronic configuration of Cu(I), which turns on their fluorescence and turns off their photothermal properties. This transformable phototheranostics could be used for post-treatment imaging-guided photodynamic therapy on residual tumor cells. Thus, the rationally designed copper-nitrogen-coordinated CDs offer a simple in situ transformation strategy for using multiple-stimulus-responsive precise phototheranostics in post-treatment monitoring of residual tumor cells and imaging-guided programmed therapy.

A limited infiltration and the subsequent low effective drug concentration result in poor chemotherapeutic outcomes against tumors, and even further promote tumor resistance and metastatic. Herein, iRGD-modified graphene oxide (GO) nanosheets (IPHG) are developed for the intensive treatment of metastatic tumors using focus-specific penetrated delivery together with photothermal therapy-mediated chemosensitization and photothermal therapy-strengthened integrin targeting-based antimigration. In vitro and in vivo data verified the mechanism of the tumor-selective infiltration of IPHG is based on a rigid 2D structure-associated advantage regarding hemodynamics and endothelial contact, followed by iRGD-endowed transendothelial and intratumoral transport. Once IPHG-DOX-penetrated 4T1 tumors are exposed to near-infrared irradiation, hyperthermia stress and photothermal therapy-elevated effective drug concentrations result in chemosensitization and prominent tumor suppression. Meanwhile, the specific binding of iRGD to integrins and photothermal therapy leads to the synergistic perturbation of cytoskeleton remodeling and subsequent impairment of cell motility and metastasis. The tailored design of IPHG validates a promising paradigm for drug delivery to combat tumor resistance and metastasis resulting from poor target access for single chemotherapy.

The combination of photothermal therapy (PTT) and reactive oxygen species (ROS)-involved photodynamic therapy (PDT) holds great promise in enhancing anticancer effect. Herein, a MoS 2 -based multifunctional biomimetic nanotherapeutic platform for synergistic PTT and PDT anti-tumor therapy and real-time self-evaluation of therapeutic effect was developed. MoS 2 hollow nanospheres and fluorescence TAPP-Ma was cloaked with a 4T1 cell-activated fibroblast cell (CM-AF) hybrid membrane to give nanosystem CM-AF-MoS 2 @TAPP-Ma. The hybrid membrane as a delivery system not only offers the nanotherapeutic platform with homotypic targeting ability, but also effectively eliminates the barrier effect of tumor tissue microenvironment. Caspase 3 (Casp3) inside cancer cells activated by the PTT effect of MoS 2 hollow nanospheres broken down TAPP-Ma, triggering the PDT of TAPP. At the same time, the fluoresce “turn-off” of TAPP-Ma provided the base for the self-evaluation of therapeutic effect. The in vitro and in vivo outcomes suggested that the MoS 2 -based multifunctional biomimetic nanotherapeutic platform exhibited synergistic cascade effect for cancer therapy, making it an promising anti-cancer candidate in biomedical clinical application.

Metal–organic frameworks (MOFs) derivatives had been widely explored in electronic and environmental fields, but rarely evaluated in the biomedical applications. Herein, Fe–N codoped carbon (FeNC) nanoparticles were synthesized and characterized via facile pyrolysis of precursor ZIF-8 (Fe/Zn) nanoparticles, and their potential applications in tumor therapy were assessed in this investigation both in vitro and in vivo. After PAA (sodium polyacrylate) modification, the FeNC@PAA nanoparticles were able to initiate a Fe-based Fenton-like reaction to generate ·OH and O2 for chemodynamic therapy (CDT) and O2 evolution. Meanwhile, the porphyrin-like metal center in the FeNC@PAA nanoparticles could be used as a photosensitizer for photodynamic therapy (PDT) of tumors, which could be enhanced by O2 generated in CDT. Furthermore, the FeNC@PAA nanoparticles were also found to be effective in photothermal therapy (PTT) with a photothermal conversion efficiency of 29.15%, owing to a high absorbance in the near-infrared region (NIR). In conclusion, the synthesized FeNC@PAA nanoparticles exhibited promising applications in O2 evolution and CDT/PDT/PTT synergistic treatment of tumors.
