The cryoprotective effects of deacetylated konjac glucomannan (DKGM) on gluten and its components in frozen dough remain unclear. This study aimed to investigate the impact of DKGM with varying degrees of deacetylation (DD) on the structural stability of gluten and its components during freeze–thaw (FT) cycles. Compared with konjac glucomannan, DKGM effectively alleviated the gluten structural “depolymerization–aggregation” process during FT cycles, with DK2 (DD, 50.21%) exhibiting the optimal cryoprotective effect. The DK2 group retained higher noncovalent interactions and disulfide bonds during FT cycles, thereby stabilizing the gluten structure. Studies on glutenin and gliadin suggest that glutenin is more susceptible to FT damage. DK2 provided the best protection for glutenin, whereas DK3 (DD, 66.61%) provided the most effective protection for gliadin. These distinct effects were likely attributable to differences in the particle size and steric hindrance of DKGM, as well as the inherent structural characteristics of the protein components.

Endogenous hydrocortisone causes detrimental effects on public health and domestic animal products, but the potential mechanisms remain elusive. Hydrocortisone was detected from seventy-two Guanzhong-Black pigs in three replicates (216 samples) (0.00 ± 46.38 μg kg −1 ), indicating the existence of endogenous hydrocortisone. Herein, we investigated the effects of hydrocortisone on the metabolic signatures in pork via integrative metabolomics and proteomics by UHPLC-Q-Orbitrap HRMS. Animal-derived foods under hydrocortisone-bioaccumulation cause metabolic perturbation by regulating glutamine synthetase expression at the transcriptional level contributed to hydrocortisone-induced toxicity and accelerating the down-regulation of essential amino acids ( l -Histidine 10.74–6.48 mg kg −1 , l -Phenylalanine 3.70–1.57 mg kg −1 ), purine nucleotides (GMP 40.29–5.00 μg kg −1 , etc) that provide nutritional value in a positive feedback loop. Protein-metabolite interactions suggesting that hydrocortisone enhanced nitric oxide synthase and GMP reductase expression (LOQ 3.24–63.39 μg kg −1 ), and affected meat flavor perception, eventually lead to the decline of nutritional value and flesh quality of pork.

This study explored the role of hemocyanin (Hc) in shrimp quality deterioration during storage. Pacific white shrimp ( Litopenaeus vannamei ) subjected to bloodletting and stored at 4 °C showed 22.97 % and 21.51 % reduction in peroxide value and thiobarbituric acid reactive substance (TBARS) at 3 d, respectively, compared to the control. Furthermore, key quality indicators, including K-value, water holding capacity, color, and total volatile basic nitrogen improved significantly. In washed shrimp model with exogenously added Hc and Cu 2+ , the effect of Hc group on promoting lipid oxidation was stronger than Cu 2+ group, and TBARS level increased by 16.42 %. Cu 2+ released from Hc activated adenosine monophosphate deaminase and accelerated inosine monophosphate degradation into hypoxanthine riboside and hypoxanthine, resulting in freshness loss. Hc also activated polyphenol oxidase, leading to shrimp blackening. This is the first study to reveal the impact of Hc and Cu 2+ on the quality deterioration of Pacific white shrimp during storage.

BACKGROUND The gelation properties of surimi gel under various high temperatures (115, 118, and 121 °C) and sterilization intensities ( F 0 values of 3–7 min) were systematically investigated. A kinetic model detailed quality changes during heat treatment through mathematical analysis, elucidating mechanisms for gel quality degradation. RESULTS Increased sterilization intensity significantly reduced the quality characteristics of surimi gel. Compared to the gel without sterilization treatment, when the sterilization intensity was increased to 7 min, the gel strength of the groups treated at 115 °C, 118 °C, and 121 °C decreased by 68.35%, 51.4%, and 51.71%, respectively, and the water-holding capacity decreased by 24.87%, 16.85%, and 22.5%, respectively. The hardness, chewiness, and whiteness of the gel also significantly decreased, and the changes in these indicators all conformed to a first-order kinetic model. Activation energy of 291.52 kJ mol −1 highlighted gel strength as the least heat-resistant. At equivalent sterilization intensities, 115 °C exhibited the poorest gel quality, followed by 121 °C, with 118 °C showing relatively better gel quality. Increased T 22 and decreased PT 22 suggested heightened water mobility and transition of immobilized water within the gel into free water. Protein degradation, weakened disulfide bonds and hydrophobic interaction, and protein conformation changes collectively led to a rough and incoherent gel network structure with large fissures, as verified by the results of scanning electron microscopy. Correlation analysis indicated potential for precise control over surimi gel quality by modulating physicochemical attributes. CONCLUSION The outcomes may be beneficial to improve the production and quality control of ready-to-eat surimi-based products. © 2024 Society of Chemical Industry.

The quality of yellowfin tuna is highly vulnerable to microbial proliferation, which accelerates quality deterioration and spoilage throughout storage. In this study, the dynamic changes in biogenic amines (BAs), total volatile basic nitrogen (TVB-N), and total plate count (TPC) across a different storage temperature range of 0 to 25 °C were studied, followed by a shelf-life prediction based on kinetic models. The TPC, TVB-N, and most BAs increased significantly with increasing storage time and temperature. Correlation analysis was used as an exploratory approach and showed that only histamine and cadaverine exhibited significantly positive correlations (|r| ≥ 0.6,p< 0.05) with TPC and TVB-N within the present dataset. Histamine was selected as a candidate BA indicator for kinetic modeling because only this BA changed significantly during storage at different temperatures. Utilizing histamine, TPC, and TVB-N, preliminary kinetic equations for shelf-life estimation were developed using first-order kinetics and the Arrhenius equation. Among the tested indicators, TPC showed relatively greater temperature sensitivity and preliminary within-batch agreement. These results provide preliminary reference data for further validation of tuna during the refrigeration process.

Introduction Cerebral Venous Sinus Thrombosis (CVST) is a disease with a low incidence rate but is prone to progression and Early Deterioration of Parenchymal Lesions (EDPL) leading to severe neurological impairment. Exploring the potential molecular mechanisms and key genes of EDPL to achieve early clinical intervention and treatment is of vital importance. This study aims to investigate the potential molecular mechanisms and key genes of EDPL in CVST. Methods The CVST patients were prospectively recruited and their peripheral venous blood was collected. The gene expression matrices of three EDPL patients and three non-EDPL patients were obtained by transcriptome sequencing technology differentially expressed genes were analyzed a competing endogenous RNA (ceRNA) network for lncRNA-miRNA-mRNA was constructed and the target genes of the ceRNA network were subjected to GO and KEGG analyses. The Hub genes of differentially expressed mRNAs (DEmRNAs) were screened using Cytoscape. Subsequently the key genes were obtained by intersecting the Hub genes with the target genes. Results In the present study 172 differentially expressed lncRNAs (DElncRNAs 145 up-regulated and 27 down-regulated) and 107 DEmRNAs (53 up-regulated and 54 down-regulated) were obtained between the EDPL group and the non-EDPL group. We constructed a ceRNA network of EDPL containing 75 DElncRNAs 34 miRNAs and 46 DEmRNAs and explored the functional enrichment of the target genes in the ceRNA network. It was shown that the target genes were mainly concentrated in the focal adhesion and the ECM-receptor interaction. Subsequently the target genes of the ceRNA network were intersected with the Hub genes and four key genes (RELN CAV1 CCNB1 NME1-NME2) were screened. Discussion This study constructed the ceRNA regulatory network of EDPL to analyze the potential mechanisms and screen key genes but there are still certain limitations such as a small sample size and a lack of in vivo and in vitro experimental verification. Large-scale multi-center and prospective studies are needed to further verify these results. Conclusion It was shown that focal adhesion and ECM-receptor interaction may be involved in the mechanisms of EDPL and RELN CAV1 CCNB1 and NME1-NME2 may be promising intervention targets for EDPL.

Abstract Hydrophobic emulsions (HEs) are widely used to improve the moisture resistance of cement-based materials, but their adsorption on cement particle surfaces can form a physical barrier that retards hydration and compromises mechanical performance. Herein, a polycarboxylate superplasticizer (PCE) was introduced into hydrophobic emulsion-modified calcium sulfoaluminate (CSA) cement mortars to regulate the interfacial adsorption behavior between HE droplets and CSA cement particles. The coupled effects of PCE and HE were systematically investigated through fluidity, mechanical strength, rheology, hydration heat, pore structure, water absorption, adsorption capacity, and ζ-potential analyses. The results show that 1% PCE provided the best overall balance between the performance recovery and hydrophobic functionality under the investigated conditions. Compared with the hydrophobic mortar without the PCE, the addition of 1% PCE increased fluidity by 25% and enhanced the 28 day compressive strength by 129% while maintaining the inherently low water absorption. Adsorption capacity and ζ-potential analyses further supported competitive adsorption between the PCE and HE at the CSA cement interface. The incorporation of the PCE alleviated the hydration retardation induced by HE and improved the pore structure without compromising the hydrophobic functionality of the mortar. This work provides an interfacial regulation strategy for balancing workability, hydration, mechanical performance, and moisture resistance in hydrophobic CSA cement mortars.

BaCO 3 -shelled phase change microcapsules improve sulfate resistance of cement paste. • The BaCO 3 shell immobilizes sulfate ions through in-situ ion exchange. • BMPCM regulates sulfate-attack products and pore-network deterioration. • Optimal BMPCM dosage reduces eroded area by 69.9% and enhances strength. Sulfate attack is one of the major degradation mechanisms responsible for expansion cracking, surface spalling, and long-term performance deterioration of cementitious materials. To improve the durability of cementitious materials in sulfate environments, a functional microcapsule (BMPCM) with a BaCO 3 shell and tetradecane core was prepared, and its effect on sulfate resistance was systematically evaluated. Cement paste specimens incorporating different BMPCM dosages were separately immersed in deionized water and MgSO 4 solution for up to 360 d. The deterioration behavior and underlying mechanisms of cement paste incorporating BMPCM were elucidated at multiple scales through macroscopic damage characterization, microstructural observation, hydration/sulfate-attack product analysis, pore connectivity assessment, and compressive strength testing. Results showed that the BaCO 3 shell reacted with invading SO 4 2− to form insoluble BaSO 4 , thereby chemically immobilizing sulfate ions. This process reduced the effective sulfate activity within the matrix, suppressed the formation of expansive products such as gypsum and ettringite, and mitigated the leaching of soluble hydration products such as Ca(OH) 2 . Compared with controls, specimens with an appropriate BMPCM dosage exhibited a 69.9% reduction in the severely deteriorated area, a 24.4% decrease in interconnected porosity, and a 44.9% increase in compressive strength after 360 d. In contrast, excessive BMPCM incorporation promoted microcapsule agglomeration and introduced initial defects, thereby weakening sulfate resistance. In addition, thermal analysis confirmed that BMPCM retained its phase-change response near the freezing point, suggesting its potential for further application in sulfate-resistant cementitious materials exposed to complex temperature-fluctuation service environments.

This research evaluated the effects of three specific spoilage organisms (SSOs)- Shewanella baltica , Pseudomonas brenneri and Pseudomonas psychrophila - on quality deterioration and myofibrillar proteins (MPs) degradation in chilled large yellow croaker fillets. The SSOs were found to expedite the conversion of umami/sweet amino acids to biogenic amines (BAs), reduced water holding capacity and textural deterioration. This altered taste activity value (TAV) of representative amino acids and increased the equivalent umami concentration (EUC). SSOs exacerbated MP conformational changes from ordered rigid to disordered flexible structures, enhancing protein solubility and tyrosine content. Findings from SDS-PAGE and TEM indicated significant degradation of myosin, actin, and tropomyosin, and disruption of sarcomere structure. Correlations between quality traits and MPs stability were established. Spoilage impact ranked: Shewanella baltica > Pseudomonas brenneri > Pseudomonas psychrophila . This study highlights the importance of preventing SSOs colonization in delaying quality deterioration and provides comprehensive insights into MPs degradation in SSO-induced spoilage.

Pure or highly concentrated ionic liquids (ILs, CIL > 40% v/v) have been widely applied for the preparation of value-added products from polysaccharides by breaking their hydrogen bonds. Reducing the dosage of ILs would benefit the establishment of a greener process. However, the effect of low-concentration ILs on the pretreatment of polysaccharides, especially water-soluble recalcitrant polysaccharides, has not been fully investigated. In this study, aqueous [Bmim]Cl (1-butyl-3-methylimidazolium chloride) with different dilution ratios was applied to treat xanthan gum (XG) and assist XG hydrolysis. Structural characterization and hydrolysate analyses showed that 1% (v/v) [Bmim]Cl could effectively break intramolecular hydrogen bonds of XG through the binding of [Bmim]+ to XG side chains, disrupting the XG conformation. Such changes increased the accessibility of XG for chemical reagents. Moreover, [Bmim]Cl could significantly increase the content of hydroxyl and superoxide radicals (from H2O2) in the reaction system. These interactions strengthened the acidic oxidative hydrolysis of XG. Notably, a low concentration of [Bmim]Cl was proved to inhibit the Maillard browning of the acidic oxidative hydrolysis system, which also contributed to XG degradation. Under hydrolysis conditions of 0.4 M HCl, 0.75% (w/v) H2O2 and 1% (v/v) [Bmim]Cl for 4 days at 80 °C, XG could be hydrolyzed into a mixture with a high dextrose equivalent value of 29.16% containing 25 types of oligosaccharides. Meanwhile, the XG hydrolysates showed good antioxidant ability and elicitor activity against soybean cotyledon. Our work should be valuable for establishing a green and economic platform for valorizing water-soluble recalcitrant polysaccharides.

Osteoporosis (OP) has been a global health concern, and calcium supplements are still an effective method to relieve the disease. In this study, we investigated the pharmaceutical properties, antiosteoporosis, and adverse effects of a novel calcium supplement named compound calcium carbonate granule (CCCG). The dissolution rate, stability of vitamin D3, and drug permeability of the calcium supplements were explored. And the osteoporosis was constructed in mice with retinoic acid (100 mg/kg/d) for 21 days, and orally treated with CCCG (0.34, 0.68, and 1.36 g/kg/d) for 36 days to evaluate the efficacy against osteoporosis and the occurrence of kidney stones. The results showed that CCCG exhibited higher dissolution rate and better vitamin D3 stability than the standard calcium carbonate granule. CCCG also displayed more absorption in Caco-2 cell model. On the other hand, CCCG alleviated the deterioration in femur microstructure and oxidative stress in the liver and kidney, and increased the expression of bone metabolic markers. CCCG also exhibited lower risk of nephrolithiasis than the commercial calcium carbonate granule. In summary, from the perspectives of both pharmaceutics and efficacy, we illustrated the profiles of an intriguing calcium supplement, which could possibly be an alternative of commonly used ones. Practical Application. Calcium carbonate is widely used to supplement calcium, primarily because of its high calcium load, low cost, and good efficacy. Now, we provided a new calcium supplement named CCCG, which was prepared by inclusion and complexation techniques. Vitamin D3 in CCCG was made into vitamin D-β-cyclodextrin inclusion complex by inclusion technology, which improved the stability of vitamin D3. And calcium carbonate was converted to a water-soluble calcium citrate complex via complexation techniques to accelerate the release of calcium ions and minimize the risk of kidney stone formation. From the perspective of both pharmaceutics and efficacy, we illustrated the profiles of an intriguing calcium supplement, which could possibly be an alternative for the supplemental and preventive treatment of osteoporosis.

The study created an electrospun fiber membrane with polyacrylonitrile, nano silver, and oregano oil using the electrospinning technique. After examining the preparation parameters, an appropriate formula for the composite film spinning solution and electrospinning parameters were found: 11 wt percent polyacrylonitrile, 6 wt percent natural oregano essential oil, 15 kV of spinning voltage, 0.6 mL/h of advancing rate, and 20 cm of spinning distance. The composite nanofiber membrane demonstrated improved shape and structure, a more balanced antibacterial effect, and persistence against microbes when subjected to the prescribed formula and circumstances. In the end, the strong antimicrobial qualities of the nano-fiber material were validated by the Feilaifeng field testing. This study showed that when combined with natural plant essential oil, the composite nanofiber membrane has potential for use as a green microbial control material for Grottoes historic heritage. By preventing an intense direct contact between antimirobial agents and the cultural relics of the grottoes, the composite barrier reduced the potential harm that could be caused by the solution soaking into the rocks. Moreover, the composite nanofiber membrane provides the advantages of large-scale antibacterial activity from natural essential oil and long-lasting antimicrobial capabilities from nanosilver. It also has no effect on the cultural relics of the Grotto, is simple to remove, and leaves little trace.
