Hydrazone-linked covalent organic framework functionalized with cysteine as a fluorescence sensor and Exploration of paper chip for p-nitrophenol detection

Shan Yang,Xinyue Li,Yifang Liao,Yibing Ji,Ruijun Li

The large use and emission of p -nitrophenol (p-NP) seriously pollute the environment and endanger human health. In this work, a hydrazone-linked fluorescent covalent organic framework (BATHz-COF) was simply synthesized at room temperature and covalently linked N-acetyl-L-cysteine (NALC) via the "thiol-ene" click reaction, where carboxyl groups were introduced to improve dispersion and fluorescence intensity . As a rapid, good selectivity and reusability fluorescence sensor, the obtained COF-NALC has been used for quantitative analysis of p-NP predicated on the internal filtering effect (IFE). Under optimal conditions, COF-NALC enabled quantitative detection of p-NP with a linear range of 5–50 μM and the detection limit was 1.46 μM. The application of COF-NALC to the detection of p-NP in river water samples was successful, and the satisfactory recoveries were 98.0%–109.3%. Furthermore, the fluorescent COF paper chips constructed by in situ growth were combined with a smartphone to build a visual platform for the quick and real-time detection of p-NP, providing an excellent illustration for the development of intelligent fluorescence sensing in environmental analysis .

Design of membrane-based microfluidic chip device based on bionic leaf and adsorption detection of tetracycline antibiotics by specific gel membrane

Yang Yuan,Xu Zhu,Yang Liu,Zihan An,Hui Jia,Jie Wang

In this study, we developed copper alginate-carbon nanotube/copper sulfide-graphene oxide gel membranes based on the design inspired by the surface structure of leaves and plant transpiration in nature and through the modulation of high-density polymer networks. This light-driven water evaporation-specific adsorption gel membrane is capable of using solar energy to drive water evaporation from contaminated surface water to access tetracycline antibiotics for detection. Combined with the designed biomimetic adsorption detection chip, in situ monitoring of tetracycline antibiotics in water bodies was achieved. Compared with the traditional mass spectrometry detection method, it shows more outstanding performance in terms of time, environmental protection and technology. The microscale adsorption mechanism was further explored on the molecular scale by density functional theory (DFT). The results showed that tetracycline antibiotics were adsorbed on the aerogel membrane mainly through π-πEDA interactions in sandwich (S) configuration and parallel displacement (PD) configuration, which ensured high adsorption efficiency. Under the optimized conditions, the tetracycline concentration detected by the sensor showed a good linear relationship with the cyclic voltammetry curve, and the lowest detection limit was 1.59*10 −4  μM. This adsorption detection strategy can reduce the influence of organic matter, inorganic matter, and other classes of antibiotics on the detection of tetracycline antibiotics, and provide a new idea for the high-efficiency photo-thermal actuation and in-situ testing of tetracycline antibiotics.

Microfluidic Chip with Integrated Whole Blood Separation for Simultaneous Accurate Quantification of Multiple Chronic Kidney Disease Markers and Long-Term Monitoring

Jing Yang,Xiaoye Feng,Haiqin Li,Pengtao Chang,Xiaojun Ren,Xiaochun Li

Chronic kidney disease (CKD) has become a public health burden, with a high mortality rate and a trend toward youthfulness. It is imperative to develop a home monitoring system for the early diagnosis and long-term monitoring of CKD serum biomarkers. However, due to the limitations of conventional centrifugation preprocessing of whole blood samples, the real-time detection platform cannot fully meet the requirement of obtaining quantitative results directly from whole blood. Herein, a novel smartphone-assisted microfluidic chip integration of a whole blood separation point-of-care testing (POCT) system was proposed to achieve accurate quantification of CKD biomarkers uric acid (UA), creatinine (CR), and albumin (Alb) based on 30 μL of whole blood (fingertip blood). The whole blood separation module allows rapid separation of plasma from whole blood samples by vertical flow. A smartphone App for automated image recognition and processing was developed, which was applied for at-home routine healthcare. The detection limits of UA, CR, and Alb were 0.1127 mmol/L, 0.2978 μmol/L, and 0.7696 mg/mL, respectively, meeting clinical requirements. Additionally, the quantitative performance of 83 clinical whole blood samples showed high consistency with the clinical analyzer (Beckman AU 5400 automatic biochemical analyzer). This system is simple to operate and user-friendly, providing an accurate, rapid, cost-effective, and reliable detection method for the early screening and real-time monitoring of CKD, which holds significant potential in enhancing home self-management of CKD.

Rapid, sensitive, and non-destructive on-site quantitative detection of nanoplastics in aquatic environments using laser-backscattered fiber-embedded optofluidic chip

Yongkai Lu,Tianxiang Ji,Wenjuan Xu,Dan Chen,Ping Gui,Feng Long

A definitive link between the micro- and nano-plastics (NPLs) and human health has been firmly established, emphasizing the higher risks posed by NPLs. The urgent need for a rapid, non-destructive, and reliable method to quantify NPLs remains unmet with current detection techniques. To address this gap, a novel laser-backscattered fiber-embedded optofluidic chip (LFOC) was constructed for the rapid, sensitive, and non-destructive on-site quantitation of NPLs based on 180º laser-backscattered mechanism. Our theoretical and experimental findings reveal that the 180º laser-backscattered intensities of NPLs were directly proportional to their mass and particle number concentration. Using the LFOC, we have successfully detected polystyrene (PS) NPLSs of varying sizes, with a minimum detection limit of 0.23 μg/mL (equivalent to 5.23 ×10 7 particles/mL). Moreover, PS NPLs of different sizes can be readily differentiated through a simple membrane-filtering method. The LFOC also demonstrates high sensitivity in detecting other NPLs, such as polyethylene, polyethylene terephthalate, polypropylene, and polymethylmethacrylate. To validate its practical application, the LFOC was used to detect PS NPLs in various aquatic environments, exhibiting excellent accuracy, reproducibility, and reliability. The LFOC provides a simple, versatile, and efficient tool for direct, on-site, quantitative detection of NPLs in aquatic environments.

Full color-emitting (Y,Tb,Eu)NbO4 nanophosphors: calcination-assisted hydrothermal synthesis, energy interaction, and application in deep UV chip-based WLEDs

Zhihao Wang,Qinghong Meng,Chunyun Wang,Dianyuan Fan,Yu Wang

Phosphor-converted white light-emitting diodes (pc-WLEDs) integrated with deep UV chips and single-component white light-emitting phosphors have attracted widespread research interest recently. (Y,Tb,Eu)NbO4 nanophosphors with full-color emission properties were achieved in this work by a facile calcination-assisted hydrothermal method, and the elaborately designed white light-emitting (Y0.988Tb0.010Eu0.002)NbO4 phosphor was successfully applied to a 275 nm chip excited pc-WLED with a relatively low correlated color temperature (CCT) of 5517 K and a high color-rendering index (CRI) of 86.7. Through a comprehensive characterization by XRD, elemental analysis, FE-SEM, TEM, TG, FT-IR, UV-vis, and optical spectroscopy, the YNbO4 nanophosphor with optimal PL properties was evidenced to be obtainable through calcining the hydrothermally crystallized precursor at 1000 °C. The quenching concentrations of Tb3+ and Eu3+ in YNbO4 were determined to be ∼4 and 5 at%, respectively, and the efficiency of the NbO43− → Tb3+ energy transfer was calculated to be ∼47.93%. Upon excitation at 260 nm, the ternary (Y,Tb,Eu)NbO4 phosphor simultaneously displays blue NbO43−, green Tb3+, and red Eu3+ emissions. Benefiting from the NbO43− → Tb3+ → Eu3+ energy transfer, full color emission was achieved via adjusting the Tb3+ and Eu3+ concentrations in (Y,Tb,Eu)NbO4. The white light-emitting (Y0.988Tb0.010Eu0.002)NbO4 phosphor with color coordinates of (0.336, 0.334) can maintain its emission color and ∼67% of its overall emission intensity at 150 °C, indicating that the phosphor is highly promising for application in deep UV chip-based pc-WLEDs.

An amplification-free and in situ detection system for miRNA-451 with single-nanoparticle LSPR biosensing on a multi-channel microfluidic chip

Kuilin Liu,Xingyu Zi,Jianqi Wang,Zhijia Bao,Haiying Chen,Xinyu Gao,Guohua Liu,Mingqing Zhang

Early screening for colorectal cancer (CRC) is crucial for reducing its mortality. This study developed a amplification-free, highly sensitive sensing system based on localized surface plasmon resonance (LSPR) technology integrated with a multi-channel microfluidic chip platform for the in situ detection of the key CRC biomarker microRNA-451 (miRNA-451) using gold nanoparticles (AuNPs), achieving a limit of detection (LOD) of 19.2 fM. The core of this system is a multi-channel microfluidic chip serving as the detection substrate, where single-stranded DNA (ssDNA) probes are immobilized on the AuNPs surfaces to construct the sensor. In the presence of the target miRNA-451, it hybridizes specifically with two types of ssDNA probes, inducing the formation of AuNPs dimers and consequently causing a red shift in their LSPR scattering spectra. Unlike conventional methods, this study employed a precise single-nanoparticle tracking strategy, quantifying the target by statistically analyzing the proportion of AuNPs exhibiting significant spectral red shifts within the same field of view (FOV). This approach effectively mitigates spatial sampling errors arising from FOV inconsistencies. Furthermore, the system decouples the "hybridization" and "detection" functions, significantly enhancing detection efficiency and accuracy. This work provides a robust technical platform for the amplification-free and highly sensitive detection of miRNA-451.

Biomimetic MOF/MIPs carbohydrate microfluidic paper chip: A versatile sialic acid detection platform from point-of-care screening to food rapid analysis

Zhi Sun,Donghui Wang,Nan Wang,Juan Jin,Yiming Zhang

The development of new technologies for at-home self-testing holds promising potential for chronic disease management and monitoring. Recently, microfluidic paper-based analytical devices (μPADs) have demonstrated versatile capabilities for point-of-care testing (POCT). Sialic acid (SA), closely linked to the modification of glycoproteins in vivo, serves as an important biomarker for home-based self-testing. In this study, a novel paper-based microfluidic biomimetic chip (FP@Zr-BA@MIP) was successfully developed using boronate affinity surface imprinting technology. This chip exhibited a high adsorption capacity (Q e  = 352.83 mg g −1 ), good selectivity (imprinting factor, IF = 3.67), rapid detection efficiency (within 30 min), and excellent anti-interference capability against sialic acid. Additionally, a visual sensing system was integrated into the platform, enabling rapid, visual, and quantitative analysis of SA. A linear range of 0.1–25 mg L −1 was achieved, with the limit of detection (LOD) and limit of quantitation (LOQ) as low as 0.066 mg L −1 and 0.21 mg L −1 , respectively. The synergistic combination of the simple operational features inherent to paper-based microfluidics and the intuitive readout advantage of visual sensing significantly enhanced the simplicity and overall efficiency of the detection process. This design offers a novel approach for realizing a POCT platform for non-invasive analysis of SA.

Temperature-Responsive Agarose-Based Digital Microfluidics: An In-Chip Sample Preparation To Mass Spectrometry Analysis for Trace Cells and Single-Cell Proteomics

Ling Yan,Jiacheng Fang,Huimin Zhang,Yali Lin,Dan Li,Yingyan Zhou,Lin Zhu,Zhi Zhu,Pengyuan Yang,Zongwei Cai

Mass spectrometry (MS)-based single- and trace-cell proteomics provides critical insights into cellular phenotypes, but widespread use is limited by the cost and complexity of advanced MS systems. We present a cost-effective, accessible workflow compatible with standard MS platforms and scalable for multiomics. The temperature-responsive agarose-based digital microfluidics (TRA-DMF) platform enables one-step sample processing, including lysis, reduction/alkylation, and digestion, in a parallel four-channel format. Unlike conventional droplet-based microfluidics or fluorescence-activated cell sorting (FACS) approaches, our DMF system ensures real-time visualization and confirmation of single-cell capture and 98.3% sample recovery, minimizing losses through nonpipetting transfer. The TRA-DMF system also overcomes the MS-incompatibility of oil-phase microfluidics, allowing high-efficiency droplet transfer to MS vials. The entire TRA-DMF for single cell proteomics (TRA-DMF-SCP) workflow is completed in ∼3 h (including single-cell capture), with seamless sample introduction into standard LC-MS/MS systems. Using a regular benchtop MS instrument Orbitrap Fusion, we identified over 4000 protein groups (PGs) from 50 293T cells with excellent reproducibility and robustness. This system offers a practical and scalable solution for trace- and single-cell proteomics and holds strong potential for integration into routine multiomics workflows.

Microarray chip based on surface-enhanced raman scattering-chiral anisotropy for ultrasensitive detection of Aβ42 as an Alzheimer’s disease-associated serum biomarker

Wang Zixun,Wang Yinan,Bi Caili,Zhou Yangfan,Zhu Tao,Jin Xing

In this study, a highly sensitive surface-enhanced Raman scattering-chiral anisotropy (SERS-ChA)-based microarray chip was fabricated for the quantitative analysis of β-amyloid protein (Aβ42) in Alzheimer’s disease (AD). To construct the chip, D-type chiral gold nanoparticles (CG NPs) were employed as the plasmonic substrate, creating uniformly distributed and densely packed hotspots. The localized chiral plasmonic electromagnetic fields generated at the chiral hotspots provide a chirality-dependent enhancement preference for Aβ42 molecules, leading to significant enhancement of their characteristic Raman signals and thereby improving the sensitivity and specificity of detection. Within the linear range of 100 fg/mL to 10 ng/mL, the logarithmic concentration of Aβ42 showed a strong linear correlation with the SERS signal intensity at 1322 cm − 1 . The linear regression equation was y = 1529.72x + 2748.38 (R 2  = 0.9871), and the limit of detection reached the fg/mL level. This microarray chip features low sample consumption, high sensitivity, and high throughput, demonstrating considerable potential for non-invasive early diagnosis of AD. Graphical

Additive-Free Super-reactive metastale intermixed C-doped Al/Co3O4 coating with excellent Structural, exothermic and hydrophobic stability for a Transient-chip

Xiaogang Guo,Taotao Liang,Huisheng Huang,Binfang Yuan,Jing Wang

In 21st century, the microchip plays an essential role in data and information security, and preventing information leakage stored in chips becomes more challenging. This paper firstly introduced electrophoretic assembly technique and surface functionalization to design the C-doped nano-Al/Co 3 O 4 functional energetic material (nFEM) coating with triple stability of anti-wetting, micro-structure and energy-release for at least one year, and the energetic film was integrated with copper based-chip to construct the C-doped Al/Co 3 O 4 nFEM-chip to realize its self-destruction. Product shows outstanding anti-wetting properties with high water contact angle of 168 ± 1° and extremely low sliding angle of ∼ 1°, and well weather resistance in variable environments. Impressively, the output of heat (Q) of product can reach up to 3598 J/g nearly 85% of theoretical value with a relatively low activation energy of 235.46 kJ/mol, and keeps almost thermopositive stability even for immersed samples for fifty cycles after one year exposure test. In addition, C-doped Al/Co 3 O 4 nFEM-chip after immersion and exposure tests can also be effectively destroyed or ignited with a dazzling flame in an ultrashort time (<1ms), demonstrating the great advantage and huge application potentials for the design of other functional transient-chips with nano/micro-scale high-energy systems applied in different environments, even underwater.

In-situ and ultrasensitive detection of mercury (II) ions (Hg2+) using the localized surface plasmon resonance (LSPR) nanosensor and the microfluidic chip

Wenjia Zhang,Guohua Liu,Jinqiang Bi,Kexin Bao,Peiren Wang

Mercury is one of the most dangerous heavy metals due to its extreme toxicity to both humans and the biosphere. In this work, an in-situ and ultrasensitive method for the detection of mercury (II) ions (Hg 2+ ) using the dark-field microspectroscopy-based localized surface plasmon resonance (LSPR) nanosensor is demonstrated. In our homemade high-throughput microfluidic chip, the fixed nanorods and the free small gold nanospheres combine to form the core-satellite structures with the help of T- Hg 2+ -T chemical coordination. Such a process results in a spectral red shift due to plasmonic coupling, which allows us to reach a picomolar detection limit and a linear response range from 10 pM to 10 µM. In addition, our LSPR nanosensor has excellent selectivity and no interference for the detection of Hg 2+ , which was evaluated by monitoring responses to different heavy metal ions (Pb 2+ , Cd 2+ , Cr 2+ , Ni 2+ , and Cu 2+ ) and their mixed samples. Our sensor platform's effectiveness offers a practical method for detecting traces of Hg 2+ in real-life water samples.

Simultaneously responsive microfluidic chip aptasensor for determination of kanamycin, aflatoxin M1, and 17β-estradiol based on magnetic tripartite DNA assembly nanostructure probes

He Liyong,Shen Zhipeng,Wang Jiaqi,Zeng Jin,Wang Wenhai,Wu Huihui,Wang Qiqin,Gan Ning

The authors describe a microfluidic chip-based aptasensor platform combined with magnetic tripartite DNA structure-functionalized nanocomposites to achieve simultaneous determination of kanamycin (KANA), aflatoxin M1 (AFM1), and 17β-estradiol (E2) in milk. The two-duplex tripartite DNA nanostructure was first assembled on the surface of magnetic beads. When the aptamer on the probes recognized the specific target, the aptamer-target would be released into the supernatant. The pre-primer@circular DNA template structure initiates rolling circle amplification (RCA) by phi29 polymerase. After magnetic separation, the magnetic nanocomposites were added into a solution containing three different lengths of complementary strands to the RCA products. The number of complementary strands significantly decrease, and this can be quantitated by the microfluidic chip. Further, the employment of magnetic nanocomposites and microfluidic chip not only resolve the complex matrix interference, but also dramatically enhances the determination selectivity and sensitivity. This aptasensor allows for determination of KANA, AFM1, and E2 with limits of detection as low as 0.32 pg mL −1 , 0.95 pg mL −1 , and 6.8 pg mL −1 , respectively. This novel method exhibits the advantages of excellent stability and fast response time (< 3 min on microfluidic chip platform) for simultaneous determination of KANA, AFM1, and E2 in milk samples and ensures food safety.
