Characterisation of microemulsion nanofilms based on Tilapia fish skin gelatine and ZnO nanoparticles incorporated with ginger essential oil: meat packaging application

Le Zhang,Anjun Liu,Wenhang Wang,Ran Ye,Yaowei Liu,Jindong Xiao,Kun Wang

Summary Microemulsion nanofilms based on fish skin gelatine (FSG) and 3% (w/w, gelatine) ZnO nanoparticles (ZnONP) incorporated with ginger essential oil (GEO) at different concentration (10%, 20%, 40%, 80%, w/w, gelatine) were developed and characterised. Film thickness and elongation increased, while film tensile strength and gel strength decreased as GEO level increased ( P < 0.05). The water vapour permeability was reduced ( P < 0.05) by GEO. Microstructural study revealed a dispersion system with GEO lipid droplet embedded into FSG matrix and some aggregation. The developed films showed strong antibacterial activity against food spoilage bacterial of psychrotrophs, mesophiles and lactobacillus spp. (LAB) in situ , and food pathogenic bacteria of Escherichia coli and Listeria monocytogenes in vitro . Together with the antioxidant activity towards lipid oxidation, the release of total volatile bases nitrogen (TVB-N) was subsequently reduced during fresh meat storage. In conclusion, the incorporation of GEO provided the potential to make FSG-ZnONP films more available for meat storage.

Refrigerated Storage-Induced Quality Deterioration of Mandarin Fish (Siniperca chuatsi): Analyzing the Role of Endogenous Enzymes through TMT-Based Quantitative Proteomics

Zhouyi Xiong,Mingqian Zhou,Shan Xiao,Zhongli Zhang,Dan Li

Tandem Mass Tag (TMT)-based proteomics was employed to elucidate the pivotal role of endogenous enzymes in the quality deterioration of Mandarin fish ( Siniperca chuatsi ) during refrigerated storage. Proclean 300 was used to inhibit the growth of psychrophilic microorganisms, emphasizing the contribution of endogenous enzymes. Quality characteristics, including pH, water-holding capacity, color, texture, and myofibrillar fragmentation index (MFI), were compared between the Proclean 300-treated group (P-group) and the control group (C-group). Correlation analysis underscored the collaborative role of calpain and cathepsin B in postmortem degradation. Pearson correlation analysis showed that texture characteristics were significantly negatively correlated with MFI and significantly positively correlated with whiteness and L* in the P-group. Differentially expressed proteins (DEPs) analysis identified 66 DEPs, with 40 exhibiting low abundance, including cytoskeleton proteins and hemoglobin subunits, and 26 showing high abundance, primarily metabolic enzymes. Bioinformatics analysis revealed significant enrichment of DEPs within the glycolysis/gluconeogenesis pathway. Furthermore, protein-protein interaction analysis highlighted key proteins linked to the quality deterioration of Mandarin fish, including beta-enolase, phosphoglycerate kinase 1, and triosephosphate isomerase B. These findings offer valuable insights into the mechanisms behind the quality deterioration of refrigerated Mandarin fish and the intricate relationship with endogenous enzymes.

Six-channel colorimetric sensing of metal ions and advanced molecular information security based on fish scale-derived carbon–gold–silver nanocomposites

Ying Wu,Min Xia Quan,Qing Yu Liu,Zhen Qi Bu,Jie Zhou,Zhi Xin Xie,Jiao Yang Lu,Wei Tao Huang

Due to its high flexibility and expansibility, nanomaterials-based systems have been constructed and applied from sensing, and medicine to molecular information technology. However, the current challenges include tedious and harsh nanomaterials preparation, the need for a wider range of response channels, new information paradigms, higher information density and length. Herein, fish-scale derived carbon nanomaterials (FSCN) were used as reducing and stabilizing agents for direct, green, and facile synthesis of carbon-bimetallic nanocomposites and applications in six-channel colorimetric sensing of Hg 2+ , and multi-combination and multi-round information encoding and protection for long text. First, the reaction sequence of three precursors (FSCN, Au 3+ , Ag + ) was optimized to prepare FSCN–Au–Ag nanocomposites with double plasmonic absorption peaks of Au/Ag nanoparticles (NPs). The addition of Hg 2+ causes the aggregation of nanoparticles and the formation of Ag/Hg amalgam or Au/Hg amalgam, FSCN–Au–Ag NPs were capable of multi-channel sensing (color, absorbance, and peak shift) of Hg 2+ . Especially, the detection limit of Hg 2+ was 8.9 nM based on absorption response in wide wavelength range (456–546 nm), which was at least 6 times more sensitive than that at single wavelength. Moreover, different long text information can be encrypted and hidden in six-channel selective responses of FSCN–Au–Ag NPs-based colorimetric sensing system. By setting the threshold, these selective responses can be abstracted as a series of binary strings and then divided into shorter strings. Utilizing multi-combination and multi-round information coding, these short binary strings can be decrypted and merged into various long text content. This research promotes preparation and multi-purpose application of carbon-bimetallic nanomaterials , which provides new way to improve the sensing sensitivity of nanosystems and also offers new direction for molecular digitization.

Constructing cationic hydroxyl-functionalized hypercrosslinked polymer for effectively enriching and detecting phenolic endocrine disruptors in green shrimp, Basa fish and milk

Nuo Xiao,Yanli Dong,Ying Zhao,Qianqian Wang,Qiuhua Wu

Developing sensitive and accurate methods for detecting endocrine-disrupting chemicals (EDCs) is highly significant. Herein, a hydroxy-functionalized cationic hypercrosslinked polymer (OH-iHCP) was synthesized for the first time. The OH-iHCP has good adsorption properties for phenolic EDCs. The adsorption mechanism of EDCs on OH-iHCP mainly includes π-π* stacking, hydrogen bond and electrostatic interaction. A highly viable analytical method for determination of trace EDCs in green shrimp, Basa fish and milk was established by employing OH-iHCP as solid phase extraction adsorbent combined with liquid chromatography-mass spectrometry. The detection limit of the method was 1.45–4.50 ng g −1 , 3.00–7.00 ng g −1 and 0.04–0.15 ng mL −1 for green shrimp, Basa fish and milk, respectively. The recoveries of the method were 80.7 %–118 %, with the relative standard deviation less than 8.0 %. This study demonstrates that the developed OH-iHCP could be served as a valuable adsorbent for adsorption and separation of phenolic organic pollutants.

“Cuckoo in the nest”: L-arginine promoted the migration of Na+ to ameliorate the gel quality of low salt fish balls

Yuanxiu Liu,Tong Shi,Xiyue Cao,Xin Wang,Mengzhe Li,Chenya Lu,Xiaoli Zhang,Ying Tian,Li Yuan,Wengang Jin,Ruichang Gao

This study explored the influences of L-Arg in the ion migration and the quality of low-salt fish balls (L, 0.3 % NaCl). L-Arg significantly increased the weight gain and expansion rate of L by 18.33 % and 13.47 %. Water-holding capacity decreased from the internal to the external part of fish balls, consistent with the looser microstructure. Inductively coupled plasma-mass spectrometry (ICP-MS) revealed that L-Arg promoted Na + migration at setting ( P  < 0.05), decreased the internal concentration from 34.38 to 30.37 mg/L and increased the external one from 25.78 to 29.80 mg/L ( P  < 0.05). Conversely, Cl − concentration was decreased at setting and improved after the two-stage heating ( P  < 0.05), indicating the suppressed Cl − migration. The partial replacement of Na + by L-Arg facilitated the protein unfolding, formation of disulfide bonds and exposure of tryptophan residues. This study enriched the mechanisms of salt reduction and quality maintenance for surimi products regarding ion migration.

Rapid speciation analysis of mercury in seawater and marine fish by cation exchange chromatography hyphenated with inductively coupled plasma mass spectrometry

Xiaopan Chen,Chao Han,Heyong Cheng,Yuanchao Wang,Jinhua Liu,Zigang Xu,Lei Hu

In this work, a hybrid method for the rapid speciation of mercury compounds by cation exchange chromatographic separation and inductively coupled plasma mass spectrometry (ICP-MS) detection is reported. Effective separation of inorganic mercury (Hg 2+ ), methylmercury (MeHg), ethylmercury (EtHg) and phenylmercury (PhHg) within 2–2.5   min was achieved on two consecutive 12.5-mm strong cation exchange guard columns with 2.0   mM l -cysteine or thiourea (pH 2.0) as the mobile phase. This separation met the requirements of green analytical chemistry such as the prevention of toxic waste, safer HPLC mobile phases, and short separation times to reduce operating costs. The detection limits for Hg 2+ , MeHg, EtHg and PhHg were 0.019, 0.027, 0.031 and 0.022   μg   L −1 , each, and the repeatabilities of peak height and peak area (5.0   μg   L −1 for each Hg species) were all lower than 3%. Contents of Hg species and total mercury in certified reference materials of seawater (GBW(E) 080042) and fish tissue (GBW 10029) were in good accordance with the certified values, and satisfactory recoveries (96–102% for GBW(E) 080042 and 94–101% for GBW 10029) validated the developed method. The developed method was applied for the speciation of mercury in five seawater sample and five marine fish samples. The concentrations of mercury species in all analyzed fish samples did not exceed the permissible levels of the National Standard of China.

Rui Wu,Yulin Wang,Xi Song,Hairan Zhang

Antibiotic residues in food and environment pose a serious threat to human's health and ecological system. Therefore, it is crucial and urgent challenge to fabricate Surface-Enhanced Raman Scattering (SERS) substrate with high sensitivity to detect pesticide residues. A multifunctional three-layer Fe 3 O 4 /HAP/Ag nanoparticle system was fabricated as an active SERS substrate. The active substrate was characterized by comprehensive techniques, including X-ray diffraction (XRD), transmission electron microscopy (TEM), Scanning electron microscope (SEM), energy dispersive spectroscopy (EDS), mapping, and X-ray photoelectron spectroscopy (XPS). The FHA was not only employed to detect ciprofloxacin (CIP) solution by SERS but also it was successfully applied for detection of CIP in fish body. It showed excellent linear relationship in the range of 1.0 × 10 −8 - 1.0 × 10 −3  mol/L for CIP, with low limit of detection (7.5 × 10 −9  mol/L), satisfactory enhancement factor, stability, and repeatability. A novel approach for detecting analyte in animal's body based on SERS was developed. It could be potential candidate to rapid detection of analyte for animal and it will open new perspective in application of SERS in vivo.

Lysine-assisted ultrasonic treatment of fish myofibrillar proteins conformational rearrangement: A synergistic strategy for enhanced volatile compounds adsorption and emulsion stabilization

Yamei Wu,Zhenxing Li,Yanyan Wu,Yongqiang Zhao,Huan Xiang,Di Wang,Zhiming Hao,Yueqi Wang

This study elucidated the synergistic mechanism of lysine-assisted ultrasonic treatment in mediating sea bass myofibrillar proteins (MPs) conformational rearrangement and optimizing their flavor retention and emulsification dual functionalities. The Lysine-assisted ultrasonic treatment reduced α-helix content to 24.1 %, exposing hydrophobic groups and enhancing volatile compound adsorption. Molecular docking confirmed hexanal, octanal, and heptanal bound to MPs via hydrophobic interactions, van der Waals forces, and hydrogen bonds. Additionally, collaborative treatment decreased the particle size of MPs to 150 nm and increased the absolute value of zeta potential by 23.8 %. These changes not only inhibited MPs aggregation but also promoted covalent cross-linking and interfacial adsorption of MPs. Low-field nuclear magnetic resonance showed treated emulsion transverse relaxation time (T 2 ) rose to 727.29 ms, improving oil droplet fluidity and uniform distribution, thus boosting long-term stability. This work demonstrates potential for developing stable, flavor-controllable emulsified products.

Interactions between curcumin and fish−/bovine-derived (type I and II) collagens: Preparation of nanoparticle and their application in Pickering emulsions

Sili Xiao,Xiaoxia Huang,Xiting He,Zitao Chen,Xiaoyan Li,Xianling Wei,Qiaoyu Liu,Hao Dong,Xiaofang Zeng,Weidong Bai

This study aims to elucidate the interaction mechanisms between curcumin (Cur) and four collagen subtypes (fish type I [FCI], bovine type I [BCI], fish type II [FCII], bovine type II [BCII]), with parallel characterization of the structural and functional attributes of their derived nanoparticles. Type I Collagen/Cur nanoparticles exhibited superior solution stability compared to type II. Cur binding significantly enhanced the surface hydrophobicity, absolute ζ potential, and surface tension of collagen, while reduced dynamic interfacial tension. The binding type of Cur to collagen was static, and binding process was enthalpy-driven exothermic reaction. Molecular dynamics simulations revealed that hydrophobic interactions, hydrogen bonds, and electrostatic forces dominated the binding process. The binding affinity followed the order: FCI/Cur > BCI/Cur > FCII/Cur > BCII/Cur. The binding sites of Cur to type I collagen and type II collagen were around Ser129-Glu135 and Asn179-Ser183 residues. Collagen/Cur nanoparticle stabilized emulsions and improved oxidative stability and storage modulus.

Exploring the Mechanism of Directional Production of Characteristic Compounds by Interaction between Fish Myofibrillar Protein and Key Lipids with Different Saturations

Huilin Zhao,Sijia Lin,Jianbin Zhang,Ayong Wang,Lei Qin,Xuhui Huang

Lipid saturation and protein oxidation influence the types and amounts of volatile and nonvolatile compounds by affecting lipid oxidation. This study examined three reaction modes: systems containing only fatty acids with three different saturations, only myofibrillar proteins (MPs), or their combinations. Sensomics (including volatilomics) and metabolomics were used to analyze these systems. Results showed that MPs enhanced saturated fatty acid oxidation, suppressed unsaturated fatty acid double-bond oxidation, and adsorbed volatiles from unsaturated fatty acid thermal oxidation. MPs promoted secondary oxidation to generate short-chain volatiles. The volatile products derived from stearic acid (SA) were mainly dominated by C6–C8 compounds, while oleic acid (OA) was mainly dominated by C7–C10 compounds, and linoleic acid (LA) was mainly dominated by C5–C8 compounds. The concentrations of volatile compounds followed the order: LA (14,061.66 ng/g) > OA (11,943.52 ng/g) > MPs-LA (2,224.45 ng/g) > MPs-OA (154.63 ng/g) > MPs-SA (28.00 ng/g) > SA (24.11 ng/g) > MPs (0.73 ng/g). Through multivariate statistical analysis, hexamethylenetetramine was identified as a differential metabolite specifically associated with the reaction between SA and MPs. This compound may act as an intermediate, potentially playing a regulatory role in flavor formation. Additionally, α-methyl-dl-phenylalanine and triethylene glycol were found to be differential metabolites in the reaction between LA and MPs. This study systematically explained how characteristic volatile and nonvolatile compounds were formed and transformed. The research findings provided important theoretical support for the flavor regulation of fish products, promoting innovative development in the food industry regarding flavor quality enhancement.

ZnWO4/f-MWCNTs Nanocomposite for H2S Gas Sensor with Fast Response and Recovery: Mechanism Study and Application for Detecting Fish Spoilage

Zhihua Zhao,Shixin Ma,Yao Wang

Hydrogen sulfide (H2S), a characteristic byproduct of sulfur-containing amino acid decomposition during fish spoilage, serves as a critical freshness indicator and poses substantial food safety risks. This study develops a novel H2S gas sensor using a ZnWO4/f-MWCNTs nanocomposite synthesized via one-step hydrothermal method. The incorporated functionalized multiwalled carbon nanotubes (f-MWCNTs) improve the sensing material’s conductivity, dispersion stability, and gas adsorption capacity. XRD, XPS, FTIR, SEM, TEM, and BET analyses verify the composite’s successful synthesis and optimal physicochemical properties. The ZnWO4/f-MWCNTs sensor demonstrates exceptional H2S detection at 120 °C, achieving a response value of 18.67 (Ra/Rg) to 50 ppm of H2S with fast response/recovery times (14 s/30 s) and excellent selectivity against interferents. Real-time monitoring of fish spoilage further confirms its practical utility, highlighting the sensor’s potential for meat freshness assessment and food safety applications.

Efficient quenching activity and thermostability of N-acyl homoserine lactonase AiiA against fish spoilage mediated by Pseudomonas fluorescens via rational design

Jiashi Li,Zhong Lu,Xiaoli Zeng,Hanxiao Zhang,Haixia Lu,Lifang Feng,Junli Zhu

Quorum quenching through natural and nontoxic N -acyl homoserine lactonase (AHL-lactonase) has been regarded as a promising approach for inactivating QS-mediated bacterial spoilage. However, their poor catalytic performance remains a major challenge for practical application. To address these limitations, we developed an efficient rational design strategy for AHL lactonase AiiA to obtain a variant with high catalytic efficiency and a broad substrate spectrum, based on computer-aided structural analysis, molecular dynamics simulation, and energy calculation. Results showed that three highly flexible regions in AiiA were identified as vital sites to recognize and position different substrates, possessing high engineering potential. Among the obtained 10 mutants, AiiA A130T exhibited the strongest degradation against short-chain AHL (C 4 -HSL and C 6 -HSL), along with improved thermostability, compared to wild-type enzyme (AiiA WT ). In Pseudomonas fluorescens PF07, a majority of AHLs were quenched to undetectable levels by AiiA A130T , and the lactone ring of C 4 -HSL was verified to hydrolyze and produce form butyryl-L-homoserine. Its biofilm biomass, exopolysaccharides, exoprotein contents, and exoprotease activity were effectively repressed by 64.63 %, 48.86 %, 51.85 %, 61.50 % by AiiA A130T , respectively, in contrast with 46.51 %, 29.08 %, 23.55 %, 19.5 % by AiiA WT , without affecting bacterial growth. Moreover, the transcription levels of 16 crucial QS and biofilm-related genes in PF07 were dramatically downregulated ( P  < 0.05), as evidenced by qRT-PCR. In contrast with the slight preservation for AiiA WT , AiiA A130T effectively retarded the fillet deterioration of large yellow croaker and extended their shelf life by 2 days. Overall, this study presents an efficient strategy for modifying AHL lactonase and yields a high-performance mutant, offering the application potential of AHL lactonase for controlling QS-mediated microbial spoilage of seafood.
