A SERS-based capillary sensor for the detection of mercury ions in environmental water

Yubin Zhao,Yoshinori Yamaguchi,Yi Ni,Mingda Li,Xiaoming Dou

Mercury ion (Hg 2+ ) is one of the most toxic heavy metal ions which will cause permanent damage to the brain and kidneys. So, it is important to develop a sensitive, simple and reliable approach to detect Hg 2+ . In this work, we report a surface-enhanced Raman scatting (SERS) sensor by decorating the inner wall of capillary with 4,4′-dipyridyl (Dpy) functionalized silver nanoparticles (AgNPs). The main advantage of this sensor is that it can collect samples directly by capillary force and carry out on-site analysis by combining portable Raman spectrometer. In the presence of Hg 2+ , the Dpy molecules would be separated from the surface of AgNPs and coordinated with Hg 2+ , resulting in a decrease in the SERS signal. A linear correlation of Raman intensity with Hg 2+ concentrations from 1 to 100 part-per-billion (ppb) was obtained for quantitative analysis and the limit of detection (LOD) was determined to be 0.1 ppb. The good reproducibility and selectivity of the sensor were also demonstrated. In addition, the sensors were successfully applied to detect Hg 2+ in real environmental water samples, and the sampling process provided operation convenience compared to conventional methods. These results indicated that these capillary sensors had great potential for Hg 2+ detection in practical use.

Interference-Free Automated 96-Well Plate SERS Platform for High-Throughput Detection of Thiamethoxam

Hao Wang,Xinxin Duan,Yuhang Sun,Wenjun Zhang,Wenxi Liu,Mengmeng Yan,Jianqing Wang,Kushiev Khabibjon

Establishing accurate, sensitive, and high-throughput pesticide residue detection technologies is crucial for ensuring food safety. In this study, we developed a quartz plate cover to level the liquid surface and maintain consistent laser focus during SERS acquisition. This design significantly improved signal uniformity and reproducibility across multiple wells. By leveraging the sharp Raman peak at 2155 cm −1 , one-step synthesized gold@Prussian blue nanoparticles (Au@PB NPs) were employed to eliminate spectral interference from complex food matrices and target. In the competitive immunoassay, thiamethoxam (TXM) in the sample competed with TXM antigen-conjugated magnetic beads for binding to the probes. Higher TXM concentrations resulted in reduced binding of SERS probes to the capture substrate and a subsequent decrease in Raman intensity at 2155 cm −1 . Uder optimized conditions, the platform achieved a linear detection range of 1-100 ng/mL and a detection limit of 0.19 ng/mL. The method was validated in pear, leek, and celery samples, showing high accuracy and consistency with HPLC-MS/MS results. This automated SERS platform combines strong anti-interference capability and signal stability with scalable high-throughput performance, providing a robust solution for the trace-level detection of pesticide residues in complex matrices.

Silver nanoparticles deposited inverse opal film as a highly active and uniform SERS substrate

Junhong Zhao,Jian Lin,Xiuhua Li,Guannan Zhao,Wenjun Zhang

Ag-decorated TiO 2 inverse opal films (ATIO) with high surface-enhanced Raman scattering (SERS) enhancement were prepared using an electroless deposition process. The Ag nanoparticles (NPs) are well-dispersed and deposited on the edge of macroporous walls. The structure and optical properties of the sample ATIO have been characterized. The Ag-loading cycles and pore sizes of TiO 2 inverse opal are the key factors determining the magnitude of SERS signal enhancement. The optimized ATIO samples exhibit high SERS signal enhancement ability and reproducibility. The enhancement factor about 10 4 and the detection limit of 10 −10   M for R6G were achieved. The further application in detecting malachite green is demonstrated. A limit of detection approximately 10 −9   M was achieved. The results show that the ATIO nanostructure is promising for using as sensor substrates in SERS applications.

Machine Learning-Driven Multidimensional Tea Profiling from a Single SERS Spectrum: Toward Practical Application

Jincheng Ni,Yanyan Lu,Xuewen Chen,Haoming Bao,Yu Qiao,Shuai Zhang,Daoshuan Ding,Jiamei Jin,Huaiyuan Zhao,Qian Zhao,Jinhua Wang,Hongwen Zhang,Weiping Cai

Multidimensional tea profiling is crucial for both consumer experience, brand reputation, and market integrity. Here, we introduce a rapid and systematic strategy for multidimensional tea authentication based on a single surface-enhanced Raman spectroscopy (SERS) measurement integrated with machine learning. We first demonstrated, through machine learning analyses of individual variables, that a single SERS spectrum inherently encodes multidimensional information encompassing tea category, grade, storage quality, and pesticide residue levels.Partial least squares discriminant analysis enabled highly accurate classification of tea categories and grades (accuracies ≥ 98.9%), while support vector machine regression achieved precise prediction of storage quality and pesticide residue levels (R 2 > 0.99). Building upon these results, we developed a two-tier framework that integrates these multidimensional predictions from a single spectrum, enabling comprehensive authentication in one step. This framework was further implemented in a user-friendly application for real-time, singlespectrum analysis, achieving an overall accuracy of 98.2%. Collectively, this work demonstrates that SERS coupled with machine learning provides an efficient, cost-effective, and scalable approach for multidimensional tea authentication, offering a foundation for collaborative improvement and data sharing across the tea industry.

Multi-phase SERS detection of SARS-CoV-2 from liquids to aerosols and pseudovirus

Zhenle Qin,Xiaohui Fang,Yansheng Liu,Guofu Wang,Yang Li,Xinping Zhang

Rapid, ultrasensitive, and specific detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains challenging, particularly for micro-volume samples. This study presents a capillary-based three-dimensional Ag/ZnO nanorod surface-enhanced Raman spectroscopy (SERS) biosensing platform that enables dual-mode (liquid and aerosol) viral detection within the same device. The capillary configuration inherently provides a large surface-to-volume ratio and acts as a confined reaction chamber, eliminating the need for external liquid or gas cells while minimizing sample consumption. Vertically aligned Ag/ZnO nanorod arrays grown on the inner wall further increase the density of electromagnetic hotspots, achieving an enhancement factor of 1.07 × 10 ¹ ¹ with excellent signal uniformity (RSD = 4.65%). After ACE2 functionalization, the biosensor enables label-free, direct detection of the 2019-nCoV antigen S protein in phosphate-buffered saline (PBS) and pharyngeal swabs solution (PSS), with a limit of detection (LOD) of 1 fg/mL. In addition, a sandwich-structured sensing configuration was designed to further amplify the Raman response. Importantly, without additional instrumentation, the same capillary SERS platform also enables direct detection of the 2019-nCoV antigen S protein aerosol with a LOD of 10 fg/mL and the SARS-CoV-2 Spike Pseudovirus at 100 fg/mL. This biosensor integrates sampling, enrichment, and readout in a low-volume device, reduces the risk of cross-contamination, and maintains high analytical performance.

Immunochromatographic SERS sensor-deep learning combined strategy for intelligent diagnosis of chronic kidney disease

Yu Zengshan,Zhang Zhibin,Guo Shan,Li Zelong,Chen Hao,Gai Jichuan,Wei Jiyuan,Xu Shiqi,Wang Mingli,Shi Guochao

A combined “immunochromatographic surface-enhanced Raman scattering (SERS) sensor–deep learning” detection strategy is proposed. Specifically, we first prepared an immunochromatographic SERS sensor with excellent surface enhancement capability for the specific recognition of neutrophil gelatinase-associated lipocalin (NGAL) molecules in serum, significantly enhancing the detection sensitivity of the target molecule. Subsequently, we integrated deep learning algorithms to perform preprocessing, feature extraction, and pattern recognition analysis on complex SERS spectral signals, overcoming the limitations of traditional algorithms in handling high-dimensional nonlinear data. This approach enables rapid, sensitive, and highly specific detection of NGAL. The platform achieves a detection limit as low as 0.1059 ng/mL with a broad linear detection range. For clinical validation, serum samples from 14 volunteers were analyzed. Six deep learning models were employed to classify the acquired clinical spectral data, with the Residual Network (ResNet) model achieving a classification accuracy of 98.81%, a loss value of only 0.0877, and an area under the receiver operating characteristic curve (AUC) of 99.97%. In addition, in the analysis of new samples, the classification and prediction of data from patients and normal individuals were successfully achieved. The proposed “immunoassay surface-enhanced Raman scattering sensor - deep learning” combined detection strategy has demonstrated extraordinary potential in the rapid, sensitive and specific detection of NGAL, providing crucial technical support for the intelligent discrimination of chronic kidney disease. SERS spectral evolution of azo-reactions mediated by plasmonic Au@Ag core–shell nanorods

Mengen Hu,Zhulin Huang,Rui Liu,Ningning Zhou,Haibin Tang,Guowen Meng

The mechanism and application of localized surface plasmon resonance induced photocatalytic reactions remain an issue of interest. In this work, we used Au@Ag core–shell nanorods as a platform for plasmon-driven photocatalysis, which was in situ investigated by surface-enhanced Raman scattering (SERS) spectroscopy. The para-aminothiophenol (PATP) and para-nitrothiophenol (PNTP) adsorbed on the nanorods were irradiated with different excitation wavelengths (633 nm, 785 nm) and transformed into 4,4′-dimercaptoazobenzene (DMAB) as evidenced by the emerging Raman peaks at 1142 cm−1, 1390 cm−1, 1440 cm−1, and 1477 cm−1, corresponding to hot carrier dominated oxidation of PATP and reduction of PNTP. Preliminary azo-reaction kinetics and in situ SERS measurements were conducted by comparing the relative intensity ratio of SERS peaks at 1440 cm−1 (DMAB stretching of NN) and 1080 cm−1 (C–S stretching of PATP and PNTP). These results indicate that the catalytic efficiency was dominated by the excitation wavelength as well as the resonance condition between the plasmon band of the nanorods and the excitation line. As a proof of concept, the Au@Ag core–shell nanorods were used to catalyze 4-nitrophenol molecules, and 4-hydroxyazobenzene molecules as the product were confirmed by in situ SERS spectra as well theoretical predictions, showing potential in plasmon driven catalysis and degradation of organic molecules.

Cheng Peng,Lili Jin,Feng Wang,Haifeng Yang,Hongyan He

It is a significant challenge to perform painless invasive detection of inflammation progression in relation to the evolution of pH, redox potential , and reactive oxygen species (ROS) levels in an in situ and real-time manner. In this work, polydopamine-modified, silver nanoparticle-decorated poly (methyl methacrylate) microneedles ( [email protected] @MNs) have been developed as a multiplexed surface enhanced Raman scattering (SERS) diagnostic platform. Using rhodamine 6G as the Raman signal molecule, the [email protected] @MNs demonstrated a significant enhancement with reasonable linearity in the range of 10 −3 -10 −9  mol/L and the limit of detection is 1.0 × 10 −10  mol/L 4-mercaptobenzoic acid, anthraquinone-2-carboxylic acid and para-aminothiophenol were covalently anchored on [email protected] @MNs SERS substrate. I 1143 /I 1183 , I 1606 /I 1667 and I 1143 /I 1077 were used as assay standards for pH, redox potential and ROS level detection, respectively. The SERS multiplexed transparent microneedles (SERS mtMNs) linearly responded to pH in the range of 4.0–8.0, redox potential in the range of 417.0–599.8 mV, and ROS levels in the range of 0–480 ng/mL, demonstrating a significant ability to detect complex inflammation in vivo , in situ and in real-time.

Au@Ag-Au core@double shell SERS bioprobes for high-resolution tumor cells imaging

Gul AWIAZ,Xiaoxia WU,Chenguang ZHANG,Ting PAN,Xiawei XU,Jie LIN,Aiguo WU

Surface-enhanced Raman spectroscopy (SERS) technique with high sensitivity and specificity, exhibits huge application potential in tumor cells imaging. In this work, Au@Ag-Au core@double shell bimetallic nanoparticles (Au@Ag-Au NPs) with excellent biocompatibility and good SERS activity were successfully designed and prepared, and Au@Ag-Au NPs showed uniform size and shape. The results of UV-vis spectra revealed that surface plasmon resonance (SPR) peak of Au@Ag-Au NPs generated bathochromic shift compared to that of Au@Ag NPs, inducing high SERS activity under 532 nm illumination. The limit of detection (LOD) for 4-mercaptobenzoic acid (4MBA) molecules adsorbed on Au@Ag-Au NPs was 5 × 10 −9 M, indicating this alloy nanostructure with good SERS detection sensitivity. Moreover, Au@Ag-Au NPs SERS substrate exhibited almost none toxicity from the methyl thiazolyl tetrazolium (MTT) assay results. Au@Ag-Au@4MBA SERS bioprobe was grafted with reductive bovine serum albumin-folic acid (rBSA-FA), which improving SERS bioprobe stability, and providing specific recognition ability for tumor cells. Importantly, SERS imaging results of HepG2 cancer cells targeted with Au@Ag-Au@4MBA@rBSA-FA bioprobes demonstrated Au@Ag-Au NPs-based SERS bioprobe could effectively recognize tumor cells. This work indicated that high-resolution SERS imaging mode would play an important role in tumor imaging for cancer diagnosis.

Sensitive determination of Patulin by aptamer functionalized magnetic surface enhanced Raman spectroscopy (SERS) sensor

Zhiming Guo,Lingbo Gao,Shuiquan Jiang,Hesham R. El-Seedi,Islam M. El-Garawani,Xiaobo Zou

Food pollution caused by Patulin (PAT) seriously threatens the safety of human diets and has attracted extensive attention. Early and accurate detections of PAT are essential to prevent further toxin spreading and contamination. A surface enhanced Raman scattering (SERS) aptasensor was fabricated by combining a gold-silver core-shell structure containing signal molecule (ADANRs) and chitosan modified magnetic nanoparticles (CS-Fe 3 O 4 ). The modified ADANRs with the complementary chain of the aptamer (SH-cDNA-ADANRs) which were served as the signal probes and the aptamer modified CS-Fe3O4 (NH 2 -apt-CS-Fe 3 O 4 ) were served as the capture probes. In addition to the ability to recognize the target PAT, the capture probe also showed a strong enrichment ability under the action of external magnetic force. The intraparticle plasma coupling between the inner gold core and the outer silver shell can greatly improve the SERS activity of the signal molecules. SERS aptasensor was used to collect the spectra of actual apple samples spiked with different PAT content. The minimum detection limit of SERS aptasensor for detecting PAT in actual samples was 0.0384 ng/mL and the recovery rate range was from 96.3% to 108%. In conclusion, the sensitive and specific SERS aptasensor detection of PAT based on aptamer functionalized nanoparticles exhibited great potential for practical application in mycotoxin detection and analysis.

Quantitative SERS Detection of TBBPA in Electronic Plastic Based on Hydrophobic Cu-Ag Chips

Pei Dai,Xianzhi Huang,Yaqian Cui,Lihua Zhu

Tetrabromobisphenol A (TBBPA) was one of the most widely used brominated flame retardants. However, it easily contaminates nature and harms the environment and human health during its production and use. Therefore, it is necessary to strictly control the content of TBBPA in electronics. Surface-enhanced Raman spectroscopy has the advantages of being fast and sensitive, but it is difficult to obtain the SERS spectra of TBBPA because the hydrophobic TBBPA molecule is difficult to approach with the hydrophilic surface of common noble metal SERS substrates. In the present work, a hydrophobic Cu-Ag chip was developed for the SERS detection of TBBPA. The integration of the hydrophobic interaction and the Ag-Br bonding promoted the adsorption of TBBPA on the Cu-Ag chip, allowing for SERS detection. It was observed that both the hydrophobicity and bimetallic composition of the Cu-Ag chip played important roles in the SERS detection of TBBPA. Under the optimized conditions, the low limit of detection of the established SERS method for TBBPA was 0.01 mg L−1, within a linear range of 0.1–10 mg L−1. Combined with ultrasonic-assisted extraction, the substrate could be used for the quantitative determination of TBBPA in electronic products. Compared with the HPLC-UV method used as a national standard, the relative error of the SERS method for quantifying the TBBPA content in a mouse cable and shell was ±3% and ±7.7%, respectively. According to the SERS results, the recovery of TBBPA in the spiked mouse shell was 95.6%.Keywords:tetrabromobisphenol A;surface-enhanced Raman scattering;hydrophobic interaction;bi-metal chips;synergistic coupling

An acousto-assisted liquid-marble-based microreactor for quantitative SERS detection of alkaline phosphatase

Weiming Lin,Shuxia Lai,Dechan Lu,Qun Zhang,Xueliang Lin,Juqiang Lin,Jing Wang,Zufang Huang

Liquid-phase-based direct surface-enhanced Raman spectroscopy (SERS) has extensive applications in the field of (bio)chemical analyses. Yet, existing techniques are generally constrained by signal fluctuations caused by the evaporation effect. Liquid marbles, acting as enclosed microreactors , have been proposed to minimize the evaporation effect and have shown great application potentials in microfluidics and sensors. Herein, an acousto-assisted liquid-marble-based SERS platform was proposed to achieve non-destructive and rapid coalescence of liquid marbles and the subsequent high-sensitivity SERS detection. To evaluate the feasibility of our platform, alkaline phosphatase (ALP)—an important diagnostic indicator for specific human diseases (e.g., osteoporosis, lymphoma, leukemia, and hepatitis)—was selected. Our proposed method achieved a remarkable sensitivity of detecting the ALP activity, with a limit of detection of 0.33 UL −1 . We also determined the ALP activity in ten complex human serum samples, showing excellent consistency with the ones obtained by the commercial ALP assay kit. We envision that our acousto-assisted liquid-marble-based SERS platform could be further applied as a versatile and reliable technique for liquid-phase biomedical and chemical applications, with the advantages of negligible evaporation and fully automatic mixing operations.
