The sustainable recovery of plant-derived polyphenols requires extraction systems that can simultaneously reduce organic solvent use, improve process efficiency, and provide mechanistic clarity. Herein, a computationally guided natural deep eutectic solvent (NADES) strategy was developed for efficient and sustainable polyphenol recovery from Dioscorea cirrhosa Lour. The conductor-like screening model for realistic solvents (COSMO-RS) was first employed to predict solvent-solute affinities among eight NADES candidates, identifying betaine-maleic acid (Bet-MA) as the most promising green solvent due to its favorable polarity distribution and hydrogen-bonding complementarity. The ultrasound-assisted extraction process was subsequently optimized using an artificial neural network coupled with a genetic algorithm. Under identical initial extraction conditions, Bet-MA achieved a 1.63-fold higher TPC yield than 70% ethanol. After ANN-GA optimization, the UABM process reached 38.14 mg GAE/g DW, corresponding to a 1.73-fold increase compared with the conventional UAE control. Mechanistic investigations combining FT-IR, SEM, and 100 ns molecular dynamics simulations revealed a synergistic dual mechanism: ultrasonic cavitation disrupted the compact plant cell wall architecture, while Bet-MA provided stronger thermodynamic stabilization of released phenolics through electrostatic and van der Waals interactions. UPLC-Q Exactive/MS profiling confirmed the enrichment of diverse phenolic acids and flavonoids, and antioxidant assays verified that the intensified process preserved the bioactivity of the recovered extract. The Bet-MA solvent retained approximately 62 and 49% of its initial extraction capacity after the fifth and sixth reuse cycles, respectively, suggesting its reuse potential under the tested conditions. An AGREEprep-based greenness assessment gave a score of 0.80 for the Bet-MA-based process, compared with 0.59 for conventional ethanol extraction, indicating a more favorable greenness profile. This work provides a rational framework for sustainable biomass valorization through computational solvent design, process intensification, molecular-level mechanism elucidation, and solvent reuse.

Achieving a harmonious balance among healability, high mechanical strength, and exceptional toughness remains a persistent and formidable challenge in the development of artificial ligaments. Herein, we report a class of polyphenol-based reversible cross-linked polyurethanes (RCPUs) designed to overcome this long-standing performance trilemma. By synergistically integrating dynamic hydrogen bonds, phenolic urethane bonds, and finely engineered microphase separation structures, the RCPUs achieve an exceptional combination of mechanical robustness, self-healing capability, and antibacterial activity. The optimized RCPU1.8 demonstrates a tensile strength of ∼57.5 MPa and toughness of ∼323.8 MJ m–3, along with a healing efficiency of ∼88.1% after 12 h at 75 °C. Furthermore, a simple prestretching treatment significantly enhances the tensile strength to ∼167.7 MPa while maintaining excellent fatigue resistance over 100 loading cycles. This work provides a scalable and synergistic strategy for developing next-generation multifunctional artificial ligaments with well-balanced performance.

This study aimed to develop a green and efficient technique for extracting rambutan peel polyphenols, while systematically evaluating chemical components, the water solubility, storage stability, in vitro antioxidant activity, and digestive enzyme inhibitory potential of the extracts. Subcritical water (SW) alone and SW followed by enzymatic hydrolysis (SW-EH) were employed to extract rambutan peel polyphenols. The optimal conditions of SW extraction were temperature of 130°C, time of 30 min, and liquid–solid ratio of 80:1 mL/g, which yielded a total phenol content (TPC) of 226.54 mg gallic acid equivalents per gram of dry weight powder. A total of 39 phenolics were identified with ellagic acid as the predominant compound. SW-EH extract maintained similar phenolic profiles but contained less soluble dietary fiber compared to SW extract. The transmittance and polyphenol solubility of SW-EH extract were higher than those of SW extract in the concentration range of 5 to 100 mg/mL. The ellagic acid content in the SW and SW-EH extracts decreased by 19.05% and 1.71%, respectively, after 40 days of storage. Both extracts demonstrated comparable antioxidant activities superior to ascorbic acid, while EH treatment greatly enhanced α-glucosidase inhibitory activity compared to SW extract. With superior water solubility, storage stability, antioxidant activity, and digestive enzyme inhibitory potential, SW-EH extract is a promising product for a wide range of applications.

Epidemiological studies indicate that Metabolic Associated Fatty Liver Disease (MAFLD) has become the most prevalent chronic liver disease globally. Identifying bioactive molecules from natural plants that can prevent and ameliorate MAFLD is an effective approach to discovering new MAFLD therapeutics. Liupao tea possesses antioxidant and metabolic regulatory properties. This study aimed to identify the active components of Liupao tea polyphenol extract (PLE), and its potential targets and mechanisms of action in the prevention of MAFLD. We performed experiments on MAFLD mice to establish the prevention efficacy of PLE against MAFLD and determine its optimal dosage. Furthermore, network pharmacology, molecular docking, and SPR were performed to predict that PLE’s targets for preventing MAFLD concentrate on the hepatic EGFR target. Knockout of EGFR in hepatocytes attenuated the effects of PLE. These findings indicate that PLE inhibits lipid accumulation by downregulating the EGFR/pEGFR-AKT/pAKT-SREBP-1-ACC1 pathway through binding to the EGFR receptor, thereby preventing MAFLD.

Lignin-derived bio-oils contain abundant methoxyphenols exhibiting limited antioxidant activity, which requires their conversion to high-value polyphenols. This study develops titanium-loaded Hβ zeolite catalysts (xTi/Hβ) for the selective O-demethylation of methoxyphenols to catechols under subcritical aqueous conditions. Comprehensive characterization (XRD, XPS, STEM, NH₃-TPD, Py-FTIR) reveals that Ti exists as both framework-incorporated tetrahedral Ti⁴⁺ species (via Al³⁺ substitution) and surface-anchored anatase TiO 2 nanoparticles (NPs). The optimal 5Ti/Hβ catalyst maximizes synergistic effects: framework Ti sites enhance Brønsted acidity, while Ti NPs facilitate H 2 O enrichment near active sites. This achieves 99.46% conversion of 4-propylguaiacol to 4-propylcatechol. Applied to distilled bio-oil, 5Ti/Hβ reduces methoxyphenol content from 48.50 Area.% to 20.25 Area.% and increases total polyphenols from 25.39 Area.% to 43.64 Area.%. Catalyst stability challenges under prolonged hydrothermal conditions are identified for future optimization.

Ethnopharmacological relevance Salvia miltiorrhiza (the roots of S. miltiorrhiza Bunge, Danshen in Chinese), a traditional Chinese medicine , has been clinically used to prevent and treat various diseases, such as cardiovascular and cerebrovascular diseases , diabetes, and hepatitis B, in China and some other Asian countries. Lithospermic acid (LA), a polyphenol derived from S . miltiorrhiza , has been reported to exhibit multiple pharmacological properties, such as anti-inflammatory, anti-HIV, and anti-carbon tetrachloride-induced liver injury activities. However, little is known about the anti-hepatitis B virus (HBV) activity of LA. Aim of the study The study was projected to investigate the anti-HBV activity of LA in vitro (HepG2.2.15 and pHBV1.3-transfected HepG2 cells) and in vivo (pAAV-HBV1.2 hydrodynamic injection [HBV-HDI] mice) and explore the potential mechanism as well. Materials and methods Hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) contents were detected by ELISA kits. HBV DNA and hepatitis B core antigen (HBcAg) levels were evaluated by quantitative real-time polymerase chain reaction and immunohistochemistry assay, respectively. The proteins in autophagy process, lysosomal acidic function, and autophagy-related signaling pathways were examined by Western blot . Transmission electron microscopy was used to observe the number of autophagosomes and autolysosomes. Confocal microscopy was applied to analyze the autophagic flux and lysosomal acidification, using mCherry-enhanced green fluorescent protein (EGFP)-microtubule-associated protein light chain (LC)3 and lysosomal probes, respectively. Results LA exhibited anti-HBV activity by inhibiting HBV DNA replication in HepG2.2.15 and pHBV-transfected HepG2 cells in dose- and time-dependent manners and hampering HBsAg and HBeAg levels in HepG2.2.15 cells to a certain extent. LA reduced HBV DNA, HBsAg/HBeAg, and HBcAg levels in the serum/liver tissues of HBV-HDI C57BL/6 mice during the 3-week treatment and suppressed the withdrawal rebound of HBV DNA and HBsAg in the mice serum. LA increased LC3-II protein expression and the number of autolysosomes/autophagosomes and promoted the degradation of sequestosome 1(p62) protein in vitro and in vivo . LA enhanced the co-localization of LC3 protein with autolysosomes, further confirming the ability of LA to induce a complete autophagy. Knockdown of autophagy-related gene (Atg) 7 or 5 in vitro and administration of 3-methyladenine (an autophagic inhibitor) in vivo disabled the inhibitory efficacy of LA on HBV DNA replication, suggesting that the anti-HBV efficacy of LA depended on its ability of inducing autophagy. LA could enhance lysosomal acidification and improve the function of lysosomes by promoting the protein expression of lysosomal-associated membrane protein (LAMP)-1, LAMP-2, and mature cathepsin D , which may contribute to the autophagic induction of LA. LA inhibited the activation of AKT and mammalian target of rapamycin (mTOR) induced by HBV, which was reversed by IGF-1 (an agonist of the PI3K/AKT/mTOR signaling pathway), indicating that LA elicited autophagy through hampering the PI3K/AKT/mTOR signaling pathway. Conclusion We revealed the anti-HBV activity and mechanism of LA in vitro and in vivo . This study facilitates a new understanding of the anti-HBV potent components of S. miltiorrhiza and sheds light on LA for further development as an active constituent or candidate used in the therapy against HBV infection.

Commercial dentin adhesive systems are applied to restorations due to their resistant bonding properties, but they suffer from the lack of bioactivity and are prone to hydrolysis. Therefore, to overcome these limitations, an eco-friendly natural monomer, urushiol, was adopted to be a primer in dentin bonding due to its interaction with collagen and antibacterial activity, preventing further hydrolysis development. First, urushiol was determined to be capable of improving the biological stability of dentin collagen through cross-linking. Using high-fidelity analytical chemistry techniques, such as Fourier transform infrared spectroscopy, we quantified the effects of urushiol on collagen molecules. It could also effectively decrease weight loss after collagenase ingestion by improving the stability of dentin. Moreover, urushiol inhibited Streptococcus mutans growth as well as its biofilm formation. Finally, we demonstrated that the urushiol primer could improve the bonding strength, particularly after aging. The cross-linking and antibacterial functions of urushiol have provided promising developmental prospects for biomaterials in dentin adhesion.

In order to improve the hydrophobicity of traditional polyethersulfone (PES) membranes, this study combined the reverse thermally induced phase separation (RTIPS) method with the constructed bimetallic polyphenol networks (BMPNs) to prepare hydrophilic anti-fouling membranes. As for BMPNs, tannic acid (TA) was served as an intermediate to construct both the inner and surface hydrophilic layers of the PES membranes. On the one hand, etching Zeolitic imidazolate framework-8 (EZIF-8) with synergistic etching and surface functionalization via TA not only retained the high pore structure of MOFs, but also had good hydrophilicity. On the other hand, the MPN hydrophilic layer was formed on the membrane surface by the combination of TA from the surface of EZIF-8 and iron ions in the coagulation bath. Therefore, BMPNs structure penetrated the interior and surface of PES membrane, which greatly improved the hydrophilic properties. In addition, the membrane with porous surfaces and spongy cross sections by RTIPS method improved the permeability and mechanical properties of the membrane by several times compared with the membrane via NIPS method. The obtained membranes in this experiment showed excellent permeability, just like pure water flux reached 1662.16 L/m 2 h, while BSA rejection rate remained at 92.78%. Compared with pure membrane, it showed a better flux recovery rate (FRR = 83.33%) after cleaning, and the reduction of irreversible (R ir = 16.67%) fouling indexes indicated that the adsorption of protein was inhibited. These results suggested that the hydrophilic anti-fouling PES membranes prepared by this method possessed great application potential in membrane separation technology.

To avoid catastrophic bacterial infection in prosthesis failure, ultrahigh molecular weight polyethylene (UHMWPE), a common bearing material of artificial joints, has been formulated with antibiotics to eliminate bacteria locally at the implant site. However, the pressing issues regarding cytotoxic effects and evolution of drug resistant bacteria necessitates the development of bio-friendly bacteriostat with long bacteriostatic efficacy. Herein, tea polyphenol extracted from nature source was introduced in UHMWPE as a biogenic antimicrobial. Controlled antimicrobial activity was achieved by chemical crosslinking to regulate the release of the tea polyphenol. In addition, the crosslinking efficiency of UHMWPE blends with high loaded tea polyphenol was significantly improved in comparison to radiation crosslinking. The immobilized tea polyphenols also enhanced the oxidation stability of the UHMWPE, which is essential to prolong the service life in vivo and the storage time in vitro. The blends presented good biocompatibility, despite cell repellent on the highly crosslinked surface. Chemically crosslinked tea polyphenol/UHMWPE exhibited feasible properties for total joint implants, which is promising for clinical application.

This study aimed to determine the anti-obesity effects and mechanisms of Cerasus humilis polyphenol (CHP) in C57BL/6 obese mice and 3T3-L1 cells. High-performance liquid chromatography–electrospray ionization-tandem mass spectrometry was used for the qualitative and quantitative identification of CHP components. The obese mice, induced by feeding high-fat diet (HFD), were treated with CHP (250 mg/kg/day) by gavage for 12 weeks. Orlistat was gavaged at 15.6 mg/kg bw/day, as a positive control group. The analysis revealed that the main components of CHP were procyanidin B2, cyanidin-3-glucoside, and pelargonidin-3-glucoside. CHP dietary supplementation significantly reduced body weight and improved blood lipid measurements in HFD-fed mice (p < 0.01). Moreover, it inhibited mRNA expression of miR-122, Srebp-1c, and Cpt1a (p < 0.01) and reduced hepatic lipid deposition, as seen by hematoxylin and eosin staining. CHP downregulated the protein expression of PPARγ and C/EBPα in HFD-induced obese mice and inhibited adipocyte differentiation (p < 0.01). Compared with the HFD group, CHP supplementation had an obvious anti-inflammatory effect (decreased protein expression, such as TNF-α, IL-6, and MCP1), reducing leptin levels and TNF-α secretion in serum and cells (p < 0.01). CHP significantly inhibited the expression of miR-27a/b (53.3 and 29.9%, p < 0.01) in mice retroperitoneal white adipocytes, enhancing the expression of the target gene Prdm16 and significantly upregulating Sirt1 (105.5%, p < 0.01) compared with the HFD group. Moreover, CHP supplementation effectively improved oxidative stress (ROS, T-AOC, SOD, CAT, and GSH-Px) induced by HFD in obese mice (p < 0.01). Thus, CHP mitigates adipocyte differentiation, browning of white adipocytes, and reduction of inflammation and antioxidant activity to reduce obesity. Consequently, these results provide novel insights into the anti-obesity roles of CHP in HFD-induced obesity.

An abundance of low-salt sausages had emerged in the market as high salt intake may pose health risks to humans while their limited shelf-life becomes an urgent issue. In this study, tea-polyphenol/β-cyclodextrin/NaCl inclusion complexes (ICs) with thermal stability and better antioxidant activity were successfully prepared, their effects on the quality of low-salt sausages were investigated. The sausages were manufactured with the addition of ICs and NaCl with or without tea-polyphenols respectively, and they were stored at 25 °C for 28 days. IC group exhibited significantly lower oxidation of lipids and protein as the tea-polyphenols in the ICs could exert better antioxidant properties because of the special encapsulation structure. Besides, ICs could inhibit the growth of pathogens preventing the off-flavor. Also, ICs promoted the growth of P. pentosaceus with a strong antioxidant capacity in the sausage model, which maintained the existing pleasant flavor by preventing the oxidation of phenol and alcohol compounds. Meanwhile, ICs also accelerated the growth of Tetragenococcus , which contributed to flavor development at the later stages of fermentation by producing more esters. Hence, it was suggested that ICs could be utilized as a promising salt replacement to improve the quality of traditional Sichuan-style sausages.

Chinese chestnut shell is a by-product of chestnut food processing and is rich in polyphenols. This study sought to investigate the effect of chestnut shell polyphenol extract (CSP) on weight loss and lipid reduction in a 12-week high-fat diet (HFD)-induced murine obesity model. CSP (300 mg per kg body weight) was administered intragastrically daily. AG490, a JAK2 protein tyrosine kinase inhibitor, was also intraperitoneally injected. The results showed that an HFD induced leptin resistance (LR). Compared to corresponding values in the HFD group, CSP treatment improved blood lipid levels, weight, and leptin levels in obese mice (p < 0.01). Additionally, CSP treatment enhanced enzyme activity by improving total antioxidant capacity, attenuating oxidative stress, and reducing fat droplet accumulation and inflammation in the liver, epididymal, and retroperitoneal adipose tissue. CSP also activated the LEPR-JAK2/STAT3-PTP1B-SOCS-3 signal transduction pathway in hypothalamus tissue and improved LR while regulating the expression of proteins related to lipid metabolism (PPARγ, FAS, and LPL) in white adipose tissue in the retroperitoneal cavity. However, the amelioration of lipid metabolism by CSP was dependent on JAK2. Molecular docking simulation further demonstrated the strong binding affinity of procyanidin C1 (−10.3983297 kcal mol−1) and procyanidin B1 (−9.12686729 kcal mol−1) to the crystal structure of JAK2. These results suggest that CSP may be used to reduce HFD-induced obesity with potential application as a functional food additive.
