Fe (III)@TA@IGF-2 microspheres loaded hydrogel for liver injury treatment

Jie Chen,Xicheng Wang,Hua Ye,Zhenyan Yu,Longbao Feng,Jing Zhou,Yajin Che

As one of the most commonly used materials in liver tissue engineering, hydrogel has received much attention in recent years. In this work, we prepared a gelatin methacrylate (GelMA)/oxidized hyaluronic acid (OHA)/galactosylated chitosan (Gal-CS)/Fe (III)@ [email protected] 200 (TA200) hydrogel loaded with insulin-like growth factor 2 (IGF-2) for regeneration of damaged hepatocytes. Fe (III)@TA microspheres served as carrier to achieve sustained release of IGF-2 to promote hepatocytes regeneration. Galactose ligands could bind to the asialoglycoprotein receptor (ASGPR) on the surface of hepatocytes. Galactosylated chitosan could significantly increase the specific function of hepatocytes. The hydrogel we prepared had a storage modulus of 1100 Pa and was suitable for migration of hepatocytes. The release ratio of IGF-2 could reach up to 90% within 14 days. For carbon tetrachloride (CCl 4 ) induced human hepatic stellate cell line LX2 damage, GelMA/OHA/Gal-CS/TA200 hydrogel could significantly improve the survival of LX2 cells. The expression of HNF-4α and transferrin was detected in LX2 cells treated with hydrogel, indicating that the specific function of the liver was also restored. In summary, the GelMA/OHA/Gal-CS/TA200 hydrogels could be used as new tissue engineering scaffolds for the construction of artificial livers.

Promoting vivianite recovery: Crucial role of tightly-bound extracellular polymeric substances

Shu Wang,Nanqi Ren,Danhui Liang,Jifei Chang,Xin Wang,Jiaguo Yan,Xiang Cheng,Lili Dong,Nan Li

Vivianite is an important bio-induced product of dissimilated iron reduction (DIR), which exists widely in activated sludge and digested sludge of wastewater treatment plants (WWTPs). Extracellular polymeric substances (EPS) surrounding dissimilatory iron-reducing bacteria (DIRB) were reported to be transient media for microbial extracellular electron transfer (EET) and DIR. However, what happens when a layer of EPS envelops microbial cells during vivianite recovery? This study tried to assess the relationship between iron reduction and electron transfer by stripping EPS layer-by-layer, then clarified the delamination of different EPS layers on the performance of vivianite recovery. After stripping total EPS (PC-EPS batches) of DIRB, the Fe (III) reduction efficiencies ( R Fe ), Fe (III) reduction rate ( V Fe ), and vivianite formation efficiency ( R V ) decreased by 33%, 37%, and 30%, respectively. Tightly bound-EPS (TB-EPS) was confirmed to play a crucial role in vivianite formation, which contributed 28–51% to vivianite recovery and 34–55% to iron reduction. Electronic shuttle substances were contained in TB-EPS, which enhanced the EET rate and efficiency between cells and Fe (III). The polysaccharide content of TB-EPS was positively related to adhesiveness between cells and mineral, as well as the iron reduction efficiency. Furthermore, redox proteins (PpcA, OmcB) were embedded in EPS, which shortened the distance of electron hopping to across the EPS layer and further accelerated vivianite recovery from wastewater. Long-term operation of anaerobic reactors confirmed that TB-EPS played critical roles in bio-induced vivianite formation.

Determination of Fe(II) and Fe(III) in Paramagnetic Matrices Using NMR Relaxometry

Kaixu Jin,Caipeng Xu,Yufen Zhao,Songsen Fu

Accurately determining the concentration and redox ratio of iron ions is crucial in fields such as biogeochemistry and environmental chemistry. While current NMR relaxometry-based methods are lauded for their simplicity and speed, they face challenges when paramagnetic matrices are present. This study overcomes this limitation by leveraging the redox sensitivity of iron ions. By using appropriate redox reagents and pH conditions, seamless transitions between Fe2+ and Fe3+ are achieved without altering the valence states of other common paramagnetic metals. The significant difference in the relaxometry rates of water protons induced by Fe2+ and Fe3+ enables a strong linear relationship across a concentration range of 0 to 500 μM, with a detection limit of less than 1 μM. The established standard curves were effectively applied to determine the redox ratio and total iron concentration in both artificial and real samples, validated by X-ray photoelectron spectroscopy (XPS) and inductively coupled plasma-optical emission spectrometry (ICP-OES) analyses. Furthermore, the method demonstrates resistance to interference from common ligands, suggesting its promising potential for analyzing biological and food samples.

MODELS FOR LIPOXYGENASE WITH TWO ALKOXIDE–Fe(III) CENTERS: SYNTHESIS, STRUCTURES, AND PROPERTIES

Wu G. L.,Zheng C. Y.

Abstract [Fe III (OMe)(Hedtb)](ClO 4 ) 3 ·6.5CH 3 OH ( 1 ) and [Fe III (O n- Bu)(Hedtb)](ClO 4 ) 3 ·3 n -BuOH ( 2 ) (edtb =  N,N,N′,N′-tetrakis[(2-benzimidazolyl)methyl]-1,2-ethanediamine) were prepared and display lipoxygenase activity. X-ray crystallography reveals that the Fe III centers in complexes 1 and 2 are ligated to three nitrogens of benzimidazole and two alkylamine nitrogen atoms from the edtb ligand, with a terminal alkyoxyl group occupying the sixth position. Complexes 1 and 2 both possess a Fe III –O R center ( R  = Me, 1 ; n -Bu, 2 ) with Fe–O bond distances of 1.799(3) Å and 1.789(5) Å, respectively. The UV-Vis spectra and electrochemical properties of the complexes were recorded. Both complexes have strong absorption peaks at around 375 nm and display lipoxygenase activity.

Improvement of Sulfamethazine photodegradation by Fe(III) assisted MIL-53(Fe)/percarbonate system

Ruobai Li,Zhiming Chen,Meixuan Cai,Jiaxing Huang,Ping Chen,Guoguang Liu,Wenying Lv

Metal-organic frameworks (MOFs) have attracted tremendous interest as photocatalysis for the destruction of organic contaminants. Herein, we synthesize a Fe-based MOF namely MIL-53(Fe), combined with Fe(III) and percarbonate, exhibits photocatalytic activity for the destruction of Sulfamethazine (SMT) under visible LED light irradiation. The morphology and physicochemical properties of the MIL-53(Fe) were determined using X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-ray spectra (EDS), Fourier transform infrared spectroscopy (FTIR) and photoluminescence spectroscopy (PL). The results indicated that the photocatalytic performance of MIL-53(Fe) with unsatisfactory efficiency resulted from the rapid recombination of photogenerated electrons and holes. This may be suppressed through the introduction of an external electron acceptor (e.g., Fe(III) and percarbonate) to the catalytic process. It could be seen that the SMT degradation efficiency in the system of Vis/MIL-53(Fe)/Fe(III)/SPC attained nearly 90% within 60 min, while only 12% achieved in the system of Vis/MIL-53(Fe). The quenching experiments and EPR analysis were employed to illuminate the predominant reactive species in the system of Vis/MIL-53(Fe)/Fe(III)/SPC. It must be mentioned that the higher separation efficiency of photoinduced electron- hole pairs was achieved via the addition of Fe(III). The accelerated photocatalytic degradation of SMT could be attributed to the subsequent formation of reactive species, which were generated from the activation of SPS by Fe(II) reduced from Fe(III) in the Vis/MIL-53(Fe)/Fe(III)/SPC system. Furthermore, the mineralization of SMT was systematically investigated, as well as the effects of operating parameters (e.g., the dose of catalyst, the concentration of Fe(III), and pH). The work has expanded the possibility of MOFs as visible light responsive photocatalysts together with percarbonate for the remediation of contaminated water.

Comparative Study of Atenolol Photodegradation by Fe(III)-Complex Activated Peroxydisulfate/Peroxymonosulfate Systems

Yanlin Wu,Lanhua Luo,Yuan Li,Shanghua Shi,Xiaoning Wang,Wenbo Dong,Gilles Mailhot

Over the past 20 years, the iron-activated persulfate systems have been widely used for removing pharmaceuticals and personal care products (PPCPs) from water. However, slow Fe(III)/Fe(II) redox cycling and precipitation of iron, unless in very acidic conditions, were the main limitations. Thus, two ligand-assisted Fe(III)/persulfate systems, Fe(III)-acetohydroxamic acid (AHA)/peroxydisulfate (PDS) and Fe(III)-nitrilotriacetic acid (NTA)/peroxymonosulfate (PMS), were comparatively investigated for the degradation of atenolol (ATL) in this study. The experimental results showed that the Fe(III)-NTA/PMS system worked much better than the AHA system. However, the cost of PMS is higher than that of PDS, which should be considered. The primary advantage of the NTA system was its ability to overcome the pH limitations. It worked well over a wide pH range (3.0–10.0), whereas the AHA system could only be used in a narrower pH window (pH 2.4 to 6.5). The investigation of radicals that contributed to ATL degradation revealed that sulfate radicals (SO4•−) were dominant in the NTA system, while hydroxyl radicals (•OH) and SO4•−were the primary and secondary radicals in the AHA system. These results provided useful insight into the comparative behavior of two ligand-assisted Fe(III)/persulfate systems for ATL degradation, with the Fe(III)-NTA/PMS system showing clear potential under neutral or near-neutral conditions, while Fe(III)-AHA/PDS may still represent a lower-cost option under acidic conditions.

Fe(III) mediated tetracycline degradation by UVA/periodate: Self- coordination driven degradation process

Longji Li,Mengting Wang,Yanru Zhu,Ziyu Zou,Pin Gao,Xiaoran Li,Yajie Qian,Gang Xue

Ultraviolet A (UVA) as the promising light source in disinfection and photochemical applications is less effective in activating peroxides. Iron is wide spread in natural waters, while Fe(III) is less reactive in degrading organics. In this study, Fe(III) was found to be efficient in promoting the effectiveness of UVA/periodate (PI) in degrading various micropollutants, including carbamazepine (CBZ), sulfamethoxazole (SMX), nitrobenzene (NB), benzoic acid (BA) and tetracycline hydrochloride (TC). Among them, TC degraded with the highest rate constants, which was further confirmed to be driven by the generation of Fe(III)-TC complex. Fe(III)-TC complex facilitated the reduction of Fe(III) to Fe(II), and this reduction would be further accelerated by UVA. The generated Fe(II) thereafter enhanced the efficiency of PI activation. Scavenger combined with probing experiments preliminarily demonstrated TC degradation was based on • OH •OH , IO 3 • IO3• dominated radical oxidation and 1 O 2 , Fe(IV) dominated nonradical oxidation. Electron paramagnetic resonance (EPR) combined with probing test further confirmed the • OH •OH , IO 3 • IO3• , and 1 O 2 , Fe(IV) involved TC oxidation process. The degradation process was little influenced by water matrices. Cl − and NO 3 − even slightly promoted TC degradation through reduction the solution pH, since acidic condition was more favorable by TC degradation. Excessive humic acid (HA) would hinder TC degradation, as UVA transparency would be reduced by HA. The degradation of TC in the Fe(III)/UVA/PI system proceeded through multiple pathways involving oxidation, deamination, demethylation, ring-opening. Ecotoxicity analysis targeting on E. coli . Found the toxicity of TC intermediates increased, while the end products were nontoxic. Fe(III)/UVA/PI was efficient in inactivating E. coli. , which highlighted the application in water purification as the dual roles of oxidation and disinfection.

Fe(III)-Doped Polyaminopyrrole Nanoparticle for Imaging-Guided Photothermal Therapy of Bladder Cancer

Heng Liu,Mengsi Zhang,Hao Jin,Kepeng Tao,Chao Tang,Yanpeng Fan,Shuwei Liu,Yi Liu,Yuchuan Hou,Hao Zhang

Clinically, the surgical treatment of bladder cancer often faces the problem of tumor recurrence, and the surgical treatment combined with postoperative chemotherapy to inhibit tumor recurrence also faces high toxicity and side effects. Therefore, the need for innovative bladder cancer treatments is urgent. For the past few years, with the development of nano science and technology, imaging-guided therapy using nanomaterials with both imaging and therapy functions has shown great advantages and can not only identify the locations of the tumors but also exhibit biodistributions of nanomaterials in the tumors, significantly improving the accuracy and efficacy of treatment. In this work, we synthesized Fe(III)-doped polyaminopyrrole nanoparticles (FePPy-NH2 NPs). With low cytotoxicity and a blood circulation half-life of 7.59 h, high levels of FePPy-NH2 NPs accumulated in bladder tumors, with an accumulation rate of up to 5.07%ID/g. The coordination of Fe(III) and the amino group in the structure can be used for magnetic resonance imaging (MRI), whereas absorption in the near-infrared region can be applied to photoacoustic imaging (PAI) and photothermal therapy (PTT). MRI and PAI accurately identified the location of the tumor, and based on the imaging data, laser irradiation was employed accurately. With a high photothermal conversion efficiency of 44.3%, the bladder tumor was completely resected without recurrence. Hematological analysis and histopathological analysis jointly confirmed the high level of safety of the experiment.

Enhanced transformation capability towards benzo(a)pyrene by Fe(III)-modified manganese oxides

Pengfei Cheng,Zhipeng Lin,Xuqiang Zhao,Michael Gatheru Waigi,Galina K. Vasilyeva,Yanzheng Gao

Manganese oxides (Mn oxides) are ubiquitous and may coexist with Fe(III) ions in soil environments. In this study, acid birnessite , alkaline birnessite, cryptomelane, pyrolusite, manganite , and their Fe(III)-modified analogues were synthesized and used for benzo(a)pyrene transformation. Fe-modified Mn oxides show a markedly enhanced transformation capability towards benzo(a)pyrene. Specifically, the benzo(a)pyrene transformation rate constants k for Bir-H, Bir-OH, Cry, Pyr, and Man were 0.49, 0.080, 0.0071, 0.0055, and 0.0022 h −1 , respectively. After Fe(III) modification, the transformation rate constants were increased to 22, 2.7, 0.25, 0.0072 and 0.0098 h −1 , respectively. Fe(III)-modified layered birnessites exhibited better activity than Fe(III)-modified tunnel Mn oxides, which was attributed to their high Fe(III) contents and abundant active free radicals. Fe(III) was found to accept electrons from benzo(a)pyrene, thereby accelerating the benzo(a)pyrene transformation. Moreover, modification with Fe(III) increased the surface adsorbed water and oxygen, and promoted the generation of active free radicals. Finally, the physicochemical and biochemical properties of transformation products showed the environmental benefits of this process. Overall, the results indicate that the occurrence of Fe(III) ions could promote the removal of PAHs in Mn oxides-rich soils, and this study provides a credible understanding of PAH fates in natural soils.

Enhancing Tumor Therapy of Fe(III)-Shikonin Supramolecular Nanomedicine via Triple Ferroptosis Amplification

Wenjie Feng,Wanrui Shi,Ze Wang,Yanqi Cui,Xinxin Shao,Shuwei Liu,Li Rong,Yi Liu,Hao Zhang

Ferroptosis has been considered as a promising pathway to overcome apoptosis-induced tumor chemoresistance. However, the antitumor efficacy of ferroptosis-inducing agents is still limited because of the complexity and diversity of tumor microenvironments. Herein, we demonstrate a triple ferroptosis amplification strategy for tumor therapy by associating iron-based nanocarriers, ferroptosis molecular drugs, and H2O2-producing enzymes. Fe(III)-Shikonin (FeShik) metal-polyphenol-coordinated networks are employed to load a ferroptosis inducer of sorafenib (SRF) inside and glucose oxidase (GOx) outside, thus producing SRF@FeShik-GOx supramolecular nanomedicines (SNs). After delivering into glutathione (GSH)-overexpressed tumor cells, FeShik will disassemble and release Fe2+ to induce cell death via ferroptosis. At the same time, GOx executes its catalytic activity to produce an acid environment and plenty of H2O2 for stimulating •OH generation via the Fenton reaction. Moreover, SRF will suppress the biosynthesis of GSH by inhibiting system Xc–, further deactivating the enzymatic activity of glutathione peroxidase 4 (GPX4). Up-regulation of the oxidative stress level and down-regulation of GPX4 expression can dramatically accelerate the accumulation of lethal lipid peroxides, leading to ferroptosis amplification of tumor cells. The current strategy that utilizes ferroptosis-inducing agents as both nanocarriers and cargoes provides a pathway to enhance the efficacy of ferroptosis-based tumor therapy.

Photooxidation of Methacrolein in Fe(III)-Oxalate Aqueous System and Its Atmospheric Implication

Wang Yu,Zhao Jie,Liu Huihui,Li Yuan,Dong Wenbo,Wu Yanlin

Iron and oxalic acids are widely distributed in the atmosphere and easily form ferric oxalate complex (Fe(III)-Ox). The tropospheric aqueous-phase could provide a medium to enable the photo-Fenton reaction with Fe(III)-Ox under solar irradiation. Although the photolysis mechanisms of Fe(III)-Ox have been investigated extensively, information about the oxidation of volatile organic compounds (VOC), specifically the potential for Secondary Organic Aerosol (SOA) formation in the Fe(III)-Ox system, is lacking. In this study, a ubiquitous VOC methacrolein (MACR) is chosen as a model VOC, and the oxidation of MACR with Fe(III)-Ox is investigated under typical atmospheric water conditions. The effects of oxalate concentration, Fe(III) concentration, MACR concentration, and pH on the oxidation of MACR are studied in detail. Results show that the oxidation rate of MACR greatly accelerates in the presence of oxalate when compared with only Fe(III). The oxidation rate of MACR also accelerates with increasing concentration of oxalate. The effect of Fe(III) is found to be more complicated. The oxidation rate of MACR first increases and then decreases with increasing Fe(III) concentration. The oxidation rate of MACR increases monotonically with decreasing pH in the common atmospheric water pH range or with decreasing MACR concentration. The production of ferrous and hydrogen peroxide, pH, and aqueous absorbance are monitored throughout the reaction process. The quenching experiments verify that · OH and \(O_2^{\bar .}\) are both responsible for the oxidation of MACR. MACR is found to rapidly oxidize into small organic acids with higher boiling points and oligomers with higher molecular weight, which contributes to the yield of SOA. These results suggest that Fe(III)-Ox plays an important role in atmospheric oxidation.

Fe(III)-mediated reversible catalytic activity of MoS2 nanozymes for bisphosphonate drug sensing

Pu Zhang,Wanqiang Xia,Ping Deng,Yuanhong Min,Juan Tan,Yi Wang,Wensheng Fu

Peroxidase-like activity of MoS 2 quantum dots (QDs) can be reversibly regulated by means of Fe 3+ /alendronate sodium (ALDS)-induced aggregation/disaggregation of the QDs in solution. Specifically, Fe 3+ can selectively aggregate the MoS 2 QDs and thus greatly enhance their peroxidase-like activity, while such enhancement can be inhibited in the presence of ALDS owing to the competitive coordination of ALDS with Fe 3+ . By regulating the enzyme-like activity of MoS 2 QDs, different colorimetric signal of a typical substrate of horseradish peroxidase , 3,3΄,5,5΄-tetramethylbenzidine, can be measured in the presence of H 2 O 2 . Based on this mechanism, we develop a colorimetric approach for the determination of ALDS and further applied in quality control of pharmaceutical products, utilizing either smartphone or UV–vis spectrometer as a readout. This detection method is rapid and selective, where derivatization of ALDS before detection is not needed. Such a smartphone-based colorimetric detection platform is promising to be applied in point-of-care testing at home, small clinics, or underdeveloped regions.
