Purpose As a promising photodynamic therapy (PDT) agent, Al(III) phthalocyanine chloride tetrasulfonic acid (AlPcS4) provides deep penetration into tissue, high quantum yields, good photostability, and low photobleaching. However, its low delivery efficiency and high binding affinity to serum albumin cause its low penetration into cancer cells, further limiting its PDT effect on gastric cancer. In order to improve AlPcS4/PDT effect, the AlPcS4 delivery sys tems with different drug carriers were synthesized and investigated.Materials and methods Gold nanorods, cationic liposomes, and Pluronic® F127 nanomicellars were used to formulate the AlPcS4 delivery systems. The anticancer effect was evaluated by CCK-8 assay and colony formation assay. The delivery efficiency of AlPcS4 and the binding affinity to serum proteins were determined by fluorescence intensity assay. The apoptosis and necrosis ability, reactive oxygen species and singlet oxygen generation, mitochondrial transmembrane potential and ([Ca2+]i) concentration were further measured to evaluate the mechanism of cell death. Results The series of synthesized AlPcS4 delivery systems with different drug carriers improve the limited PDT effect in varying degrees. In contrast, AlPcS4 complex with gold nanorods has significant anticancer effects because gold nanorods are not only suitable for AlPcS4 delivery, but also exhibit enhanced singlet oxygen generation effect and photothermal effect to induce cell death directly. Moreover, AlPcS4 complex with cationic liposomes shows the potent inhibition effect because of its optimal AlPcS4 delivery efficiency and ability to block serum albumin. In addition, AlPcS4 complex with Pluronic F127 exhibits inferior PDT effect but presents lower cytotoxicity, slower dissociation rate, and longer retention time of incorporated drugs; thus, F127–AlPcS4 is used for prolonged gastric cancer therapy. Conclusion The described AlPcS4 drug delivery systems provide promising agents for gastric cancer therapy.

Although being the first-line treatment of advanced hepatocellular carcinoma (HCC), sorafenib (SOR) outcome is limited due to drug resistance and low tumor accumulation. Herein, with MnO 2 as photothermal agent and chlorine6 (Ce6) as photosensitizer, a tumor-targeting and NIR-triggered multifunctional nanoplatform loading sorafenib (MnO 2 -SOR-Ce6@PDA-PEG-FA, MSCPF) was constructed. Owing to oxygen generator MnO 2 , MSCPF could generate excessive ROS, thus can alleviate tumor hypoxia and improve sorafenib accumulation in cancer cells. Besides, ROS production further strengthens Ce6-mediated PDT and PDA-mediated PTT. By exploiting these features, MSCPF exhibited excellent antitumor effects on HCC in the in vitro and in vivo studies, compared to solo sorafenib or PDT/PTT treatment. Further mechanism experiments suggested that MSCPF could inhibit P-gp expression and induce ferroptosis via deactivation of GPX4 and SLC7A11, which ultimately enhanced the antitumor efficacy of SOR. In summary, our work highlights a promising NIR-triggered and ROS-boosted nanoplatform for enhanced chemo/PDT/PTT synergistic therapy of SOR in HCC treatment.

Organic nanomaterials with efficient ROS generation and high photothermal conversion efficiency have emerged as a new generation therapeutic agent for PDT/PTT synergistic cancer treatment. However, most of the organic nanomaterials exhibited weak emission in water because of the aggregation-caused quenching (ACQ) effect, which seriously hampers their biological application in fluorescence bioimaging. In this study, we design and synthesize a new organic small molecule (T-BDP) with a donor–acceptor–donor (D–A–D) structure by the conjugation of boron-dipyrromethene (BODIPY) and triphenylamine (TPA). Moreover, a 1,8-naphthalenediimide (NI) structure with electron-withdrawing ability is introduced onto the core of BODIPY to further enhance the intramolecular charge transfer, leading to the redshift of absorption to the near-infrared region. T-BDP displays significant aggregation-induced emission (AIE) performance, probably due to the presence of two twisted TPA groups onto the BODIPY core. Accordingly, after self-assembly into nanoparticles, T-BDP NPs exhibit strong near-infrared emission in water. Under single 635 nm laser irradiation, T-BDP NPs could generate reactive oxygen species and heat simultaneously. The photothermal conversion efficiency of T-BDP NPs is determined to be 50.9%. The low dark toxicity and high photocytotoxicity of T-BDP NPs are verified against A549 cells using the MTT assay and the AM/PI staining method. Due to the strong emission of T-BDP NPs, their accumulation and subcellular localization in cancer cells are observed using a laser confocal fluorescence microscope. The results demonstrate that T-BDP NPs are mainly located in the lysosomes of cancer cells. Thus, the as-prepared small molecule-based AIE nanoparticles hold great potential for fluorescence imaging-guided PDT/PTT synergistic tumor therapy.

The transformation of O 2 is primarily facilitated by the catalytic action of redox enzymes, which play a pivotal role in sustaining cellular energy metabolism and redox balance. Bioinspired by O 2 distribution and ROS regulation related to cascade biocatalytic process, a D-peptide NapG D F D F D Y engineered hydrogel has been constructed with encapsulated dual-enzyme superoxide (SOD) and chloroperoxidase (CPO) cascade catalytic circuit and co-assembled photosensitizer of 5-aminolevulinic acid molecules (ALA). Multimodal oxygen modulation has been conducted by the concurrent oxygen generation via SOD-catalyzed ⋅O 2 − dismutation and oxygen consumption for 1 O 2 production by CPO and ALA, achieving self-sustaining enzymatic dynamic therapy (EDT)-photodynamic therapy (PDT) (EDT-PDT) synergy. The endogenous cascade-amplified EDT not only enhances the 1 O 2 efficacy in exogenous PDT therapy, but the intermediate O 2 can also alleviate local neuropathic pain caused by hypoxia for safe PDT treatment. This work pioneers enzyme-mediated dynamic control of tumor redox homeostasis, establishing a new therapeutic axis between biocatalytic amplification and photodynamic processes.

The development of drug-resistant biofilms has resulted in treatment failures for multidrug resistant (MDR) bacteria, presenting a substantial challenge to global public health. Consequently, the development of antibacterial agents has become an urgent priority. To overcome the limitations of poor penetration and inadequate efficacy of single-modal treatments for MDR bacterial biofilm infections, this study developed a Ce6/SFP nanocomposite system utilizing silk fibroin and polydopamine. The nanoparticles, fabricated via a desolvation-in situ polymerization method, demonstrated pH-responsive drug release properties. When subjected to an 808 nm NIR laser, the material exhibited a photothermal conversion efficiency of 29.3%. Furthermore, when paired with singlet oxygen produced through 660 nm excitation, it was highly effective in eliminating MDR Escherichia coli and MDR Staphylococcus aureus. The synergistic effect of photothermal therapy (PTT) and photodynamic therapy (PDT) disrupted the stability of the cell membrane and enhanced the antibacterial efficacy. Experimental results showed that the photothermal effect increased biofilm permeability, significantly improving Ce6 penetration depth. Moreover, in vivo treatments exhibited accelerated wound healing. Biocompatibility assessments revealed a hemolysis rate below 5% and no pathological damage to major organs. This study proposes a synergistic strategy to effectively address the challenges posed by biofilms and bacterial resistance.

At present, cancer has become a major disease threatening human health worldwide. Therefore, developing targeting guided multimode synergetic therapy has become one of the hot spots in current antitumor research and is also a great challenge. Herein, a new Fe 3 O 4 /g-C 3 N 4 @PPy-DOX nanocomposite containing magnetic iron oxide (Fe 3 O 4 ) nanoparticles (NPs), lamellar structure of graphite-like carbon nitride (g-C 3 N 4 ) and polypyrrole (PPy) shell with the loaded anti-tumor drug doxorubicin hydrochloride (DOX) was designed and prepared. The monodisperse Fe 3 O 4 nanoparticles (NPs) with the diameter of 20 nm endowed the nanocomposite with the magnetic targeting ability, reducing damage to normal tissues. It is very interesting that the Fe 3 O 4 NPs also possessed photosensitizer function for photodynamic therapy (PDT). The g-C 3 N 4 sheets as the photocatalysis towards the degradation of water for generating O 2 could effectively improve the hypoxia of solid tumors and increase the efficiency of PDT. In addition, PPy has high light-to-heat conversion efficiency, so was chosen for the cancer photothermal therapy (PTT). Finally, an anticancer drug (DOX) was loaded on the nanocomposite because the presence of mesoporous structure. Thus, the prepared Fe 3 O 4 /g-C 3 N 4 @PPy-DOX nanocomposites exhibit synergetic chemotherapy/PTT/enhanced PDT antitumor effect. This study provides an inspiration for combining targeting and multimodality to improve the anticancer efficiency.

Antibiotic therapy is a key strategy for treating infections associated with orthopedic implants, yet its limited effectiveness and potential to disrupt bone healing highlight the need for innovative approaches. Herein, a TiO 2 -Graphene (TiO 2 -G) metastructure is developed on Ti implant surface using a hydrothermal method coupled with plasma-enhanced chemical vapor deposition (PECVD), showing strong near-infrared light (NIR) absorption. The antibiotic doxycycline (DOX) is successfully loaded onto TiO 2 -G and exhibited enhanced NIR release. The promising antibacterial efficacy is proven by both in vitro and in vivo tests with NIR irradiation for 5 min, which is ascribed to the synergistic photocatalytic activity of TiO 2 -G and NIR-responsive release of DOX. Interestingly, after 5 min of NIR irradiation, the TiO 2 -G metasurface neutralized the immediate negative effects of photodynamics and even upregulated the expression of osteogenic genes (osteocalcin (OPN), osteopontin (BSP), and bone sialoprotein (OCN)), with particularly enhanced effects observed on day 14. Moreover, the sustained release function of TiO 2 -G significantly mitigated the cytotoxicity of free antibiotics with antibacterial capabilities comparable to those of TiO 2 -G/DOX under NIR irradiation for 5 min. Consequently, the in vivo studies proved that the TiO 2 -G metastructure enhanced the osteointegration of the implant even in the absence of infection when loaded with DOX.

Photodynamic therapy (PDT) is a promising approach for tumor treatment, however, the therapeutic resistance of cancer stem cells (CSCs) severely limits its efficacy and easily lead to recurrence. Herein, we develop a hyaluronic acid (HA)-Ce6-Olaparib (OLA) micelle (HCCO), which combines the CSC targeting of HA, the PDT effect of Ce6, and the DNA damage repair inhibition of OLA. More importantly, HCCO induces immunogenic cell death (ICD) effects, promotes dendritic cells maturation, and alleviates myeloid-derived suppressor cells (MDSCs) infiltration to reverse CSC resistance. As a result, HCCO not only significantly inhibits the growth of 4T1 breast cancer cells and CSCs in vitro, but also effectively inhibits tumor recurrence and metastasis in vivo. Our study provides a novel strategy for preventing tumor recurrence and metastasis by the combination of inhibiting DNA damage repair, reversing CSC resistance, and enhancing PDT. BODIPY dyes have recently been used for photothermal and photodynamic therapy of tumors. However, complex multi-material systems, multiple excitation wavelengths and the unclear relationship between BODIPY structures and their PTT/PDT efficiency are still major issues. In our study, nine novel BODIPY near-infrared dyes were designed and successfully synthesized and then, the relationships between BODIPY structures and their PTT/PDT efficiency were investigated in detail. The results showed that modifications at position 3,5 of the BODIPY core with conjugated structures have better effects on photothermal and photodynamic efficiency than the modifications at position 2,6 with halogen atoms. Density functional theory (DFT) calculations showed that this is mainly due to the extension of the conjugated chain and the photoinduced electron transfer (PET) effect. By encapsulating BDPX-M with amphiphilic DSPE-PEG2000-RGD and lecithin, the obtained NPs not only show good water solubility and biological stability, but also could act as superior agents for photothermal and photodynamic synergistic therapy of tumors. Finally, we obtained BODIPY NPs that exhibited excellent photothermal and photodynamic effects at the same time under single irradiation with an 808 nm laser (photothermal conversion efficiency: 42.76%, A/A0: ∼0.05). In conclusion, this work provides a direction to design and construct phototherapeutic nanoparticles based on BODIPY dyes for tumor treatment.

Cancer remains a leading cause of global mortality, with nearly 10 million annual deaths. High intracellular glutathione (GSH) levels and tumor hypoxia critically limit chemotherapy efficacy and photodynamic therapy (PDT) effectiveness. Conventional nanocatalytic materials, particularly blue/green-emitting nanomaterials, face limited deep-tissue penetration, hindering their utility in bioimaging and targeted therapy. Herein, we report a microwave-assisted one-pot synthesis of copper-doped red-emissive carbon dots (Cu-CDs) using dicyandiamide, 4′-aminoacetanilide, and anhydrous copper sulfate as precursors. The synthesized Cu-CDs exhibit excellent optical properties ( λ ex / λ em = 525/595 nm) with a quantum yield of 12.7%, enabling nuclear fluorescent imaging for precise tumor localization. Notably, Cu-CDs demonstrate triple-enzyme (peroxidase, catalase, oxidase) mimetic activities, which synergistically deplete GSH, alleviate tumor hypoxia, and enhance reactive oxygen species (ROS) generation. In vitro studies revealed that Cu-CDs-mediated chemodynamic therapy (CDT) alone achieved 60.4% tumor cell apoptosis, while combination with PDT elevated the rate to 92.96%. Mechanistically, Cu-CDs exploit endogenous H 2 O 2 overexpression in tumors to produce cytotoxic ‧OH radicals via peroxidase activity, while catalase activity mitigates hypoxia by decomposing H 2 O 2 into O 2 . This multi-enzyme synergy reduces ROS scavenging by GSH, amplifying PDT efficacy. Biocompatibility assessments confirmed low cytotoxicity and efficient cellular uptake of Cu-CDs. The red-emissive feature enables deep-tissue penetration and nuclear targeting, minimizing off-target damage. This work establishes a paradigm for metal-doped CDs in cancer theranostics, overcoming single-enzyme limitations and advancing synergistic enzyme-mimetic therapy. The strategy holds significant promise for biomedical applications, offering a novel platform for precision tumor treatment.

Introduction Photoimmunotherapy is a breakthrough treatment for malignant tumors. Its uniqueness is that it uses antibody mediated targeted delivery to achieve high tumor specificity and uses laser-activated biophysical mechanism to accurately induce the rapid death of cancer cells and avoid damaging the surrounding normal tissues. Methods In this paper, an iron-based micelle was designed to encapsulate the photothermal agent indocyanine green (ICG) and a cyclic tripeptide of arginine-glycine-aspartic acid (cRGD) as targeted multifunctional ICG@SANPs-cRGD nanoparticles for combined photothermal/photodynamic/immune therapy of breast cancer. Results The experimental results show that ICG@SANPs-cRGD nanoparticles have good biocompatibility and photothermal conversion ability. Photothermal therapy (PTT) and photodynamic therapy (PDT) based on ICG@SANPs-cRGD can not only inhibit the proliferation, invasion and migration of tumor cells, but also directly kill tumor cells by inducing apoptosis or necrosis. Dual-mode fluorescence light (FL) and magnetic resonance imaging (MRI) imaging in mice confirmed the selective accumulation at tumor sites and imaging ability of ICG@SANPs-cRGD. PTT/PDT combined with Anti-PD-L1 immunotherapy based on ICG@SANPs-cRGD mediated the immunogenic cell death (ICD) of tumor cells by regulating the expression of immune-related indicators and activated the body's immune response mechanism, which enhanced the immunotherapy effect of immune checkpoint block (ICB). PTT/PDT combined with Anti-PD-L1 therapy not only prevented the progression of the primary tumor but also inhibited the distant metastasis of the tumor. Discussion This study explores the biomedical application of PTT/PDT combined with Anti-PD-L1 based on ICG@SANPs-cRGD nanomaterials for breast cancer treatment and demonstrates the potential of ICG@SANPs-cRGD as a multifunctional therapeutic platform for future cancer therapy.

The combination of photothermal therapy (PTT) and reactive oxygen species (ROS)-involved photodynamic therapy (PDT) holds great promise in enhancing anticancer effect. Herein, a MoS 2 -based multifunctional biomimetic nanotherapeutic platform for synergistic PTT and PDT anti-tumor therapy and real-time self-evaluation of therapeutic effect was developed. MoS 2 hollow nanospheres and fluorescence TAPP-Ma was cloaked with a 4T1 cell-activated fibroblast cell (CM-AF) hybrid membrane to give nanosystem CM-AF-MoS 2 @TAPP-Ma. The hybrid membrane as a delivery system not only offers the nanotherapeutic platform with homotypic targeting ability, but also effectively eliminates the barrier effect of tumor tissue microenvironment. Caspase 3 (Casp3) inside cancer cells activated by the PTT effect of MoS 2 hollow nanospheres broken down TAPP-Ma, triggering the PDT of TAPP. At the same time, the fluoresce “turn-off” of TAPP-Ma provided the base for the self-evaluation of therapeutic effect. The in vitro and in vivo outcomes suggested that the MoS 2 -based multifunctional biomimetic nanotherapeutic platform exhibited synergistic cascade effect for cancer therapy, making it an promising anti-cancer candidate in biomedical clinical application.
