Edible tea polyphenol oxidation self-assembled zein films for fruit preservation

Yanqiu Zhang,Mingchuan Yang,Lejia Shi,Biaohua Zhang,Ruixia Dong,Yao Zhang,Wenxi Li,Xiangchun Zhang,Dongxu Wang

EGCG nanoparticles (ENPs) enhance the antibacterial activity by disrupting biofilms and membrane structures. • Zein/ENP composite films show improved mechanical properties and water barrier performance. • The active Zein/ENP films successfully double the shelf life of strawberries and apples. • Biodegradable Zein/ENP films exhibit ideal environmental and biological safety. Tea polyphenols (−)-epigallocatechin-3-gallate (EGCG) oxidation self-assembled nanocomposites (ENP) were synthesized through a one-step self-assembly process without templating or seeding agents and incorporated into zein-based edible films for the purpose of fruit preservation. The ENP nanocomposites maintained the core structural characteristics of EGCG and demonstrated excellent antioxidant activity, and antibacterial properties against E. coli and S. aureus . As a coating, ENP significantly reduced strawberry decay, weight loss, and nutrient degradation during storage. When integrated into zein films, ENP improved structural density, mechanical properties, water vapor permeability, antioxidant capacity, and light transmittance, without affecting the crystallinity of the zein-based films. The zein-based ENP edible films effectively extended the shelf life of strawberries and fresh-cut apples, surpassing the performance of both pure zein and polyethylene films. Mechanistic investigations have demonstrated that ENP compromised bacterial biofilm and structure integrity, leads to the leakage of intracellular contents, and inhibited ATPase activity. Biological safety assessment showed that the ENP nanocomposites exhibited negligible toxicity to zebrafish and seed germination, and the zein-based ENP film has ideal degradation performance. Taken together, zein-based films incorporating ENP offer a sustainable, effective, and safe approach for packaging in fruit preservation. To provide a simple and eco-friendly packaging solution, this study integrated tea polyphenol oxidative self-assembled composites (ENP) into zein-based edible films. This integration enhanced the mechanical strength, moisture resistance, and light transmittance of the films without disrupting the crystallinity of the natural zein. The active zein/ENP films exhibited robust antioxidant and antimicrobial activities, significantly extending the shelf life of strawberries and fresh-cut apples. Spatiotemporally modulated polyphenol–protein coating for accelerated healing of chronic wounds

Linhua Li,Qingyin Li,Chunle Zhang,Zhengjiang Cao,Chang Liu,Rifang Luo,Yunbing Wang,Xiaoxi Zeng,Ping Fu

Chronic wounds are a common and serious complication. Abnormal microenvironments such as excessive oxidative stress, chronic inflammation, bacterial infection, and cellular dysfunction severely inhibit the wound healing process, leading to increased rates of amputation and mortality in diabetic patients. In this study, a novel polyphenol–activated protein coating (PDA/HK@LZM) was successfully constructed via the adsorption of lysozyme on the surface of a polyphenol nanocomposite comprising dopamine and honokiol. The coating effectively maintains the integrity of secondary and tertiary protein structures and preserves up to 91% of the lysozyme activity. Moreover, the PDA/HK@LZM coating achieves the spatiotemporal regulation of the chronic wound microenvironment. In the initial stage, lysozyme dominates the immune response by enhancing macrophage phagocytosis and modulating inflammatory factors, while honokiol synergizes with free radical scavenging and antimicrobial activity; all three synergistically enhance the wound self-cleaning function. In the repair stage, exposed honokiol regulates inflammation, polarizing macrophages from the pro-inflammatory M1-type to the anti-inflammatory M2-type and accelerating wound repair. Moreover, the release of honokiol into the microenvironment prevents the formation of vascular microthrombi, facilitating the delivery of nutrients and oxygen. Full-thickness skin wound experiments confirmed the effectiveness of the PDA/HK@LZM coating dressing in promoting rapid healing of chronic wounds. This study introduces a novel strategy for constructing polyphenol–activated protein coatings with spatiotemporal modulation wound microenvironment, opening new possibilities for the efficient treatment of chronic wounds.

Optical Weak Measurements for Protein-Polyphenol Interactions: A Novel Detection Approach

Yang Xu,Cuixia Guo,Yuxuan Yang,Chongqi Zhou,Lizhong Zhang,Han Li,Zishuo Song,Bei Wang,Yonghong He,Ke Cao

Protein-polyphenol interactions are crucial in food chemistry, yet current detection methods have limitations. This study innovatively uses optical weak measurement. Key variables like central wavelength shifts were measured in systems with chlorogenic acid, tea polyphenols, and various proteins. A linear relationship between polyphenol concentration and wavelength shift was found, and a new calculation strategy for binding constants and sites was developed, showing consistent trends with fluorescence quenching. The method is label-free and applicable to non-immobilized biomolecules, offering cost-effective and accurate detection. However, it's sensitive to solution fluctuations and complex samples. Future research should optimize the device and detection techniques to improve stability and capture interaction dynamics. This study provides a valuable tool for food chemistry research.

Polyphenol-based coacervate droplets as building blocks for protocells and prototissues

Qi Xie,Wei Chen,Zijian Huang,Yuan Gao,Haoyu Zhang,Zhanhao Zhou,Samira Batur,Chuansheng Fu,Jiao Zhang,Conglian Yang,Junjun Liu,Li Kong,Zhiping Zhang

Despite the nascent stage of research into protocells and prototissues, their substantial potential has increasingly positioned them as a focal point in recent years. The bottom-up reconstruction of life systems is gradually garnering widespread attention. In this study, a type of protocells and corresponding prototissues based on polyphenol coacervate droplets were developed via hydrogen bond interactions. The droplets exhibited properties reminiscent of cytoplasm, enriching a wide range of substances. Upon further modification with lipid bilayers, the protocells not only achieved a structural resemblance to natural cells, but also effectively maintained the stability. Prototissues formed by the orderly aggregation of protocells can not only encapsulate living cells, but also maintain the structural integrity and function of both artificial and living cells. At the macroscopic level, the prototissues were injectable, customizable in shape, bioadhesive, and capable of long-term retention in vivo. Moreover, the prototissues showed no obvious inflammatory responses. Polyphenol-based protocells and tissues with unique characteristics may offer new insights for research and applications in organoids, transplantation, and medical devices.

Metal-polyphenol Multistage Competitive Coordination System for Colorimetric Monitoring Meat Freshness

Jun Cheng,Yao Shen,Yulu Gu,Tongyue Xiang,Hui Shen,Yi Wang,Zhenyang Hu,Zhen Zheng,Zhilong Yu,Qin Wu,Yinghui Wang,Tiancong Zhao,Yunfei Xie

A low-cost, high-precision, and secure real-time system for monitoring food freshness can significantly improve spoilage issues, yet traditional colorimetric sensor arrays often suffer from chemical dyes’ high toxicity and limited color changes. Here, a metal-polyphenol network colorimetric sensor array (MPN-CSA) is built for detecting total volatile base nitrogen (TVB-N) markers of meat freshness. The multi-level competitive coordination process between the metal-polyphenol system and amine substances endows the system with color changes far beyond those of traditional dyes (reaching a detection limit of 300 ppb). By integrating convolutional neural network (CNN) technology, an online platform is developed for monitoring meat freshness, achieving an overall detection accuracy rate of 99.83%. This environmentally friendly, economically viable MPN-CSA that monitors the freshness of meat in complex storage environments can be incorporated into food packaging boxes, enabling consumers and suppliers to assess the freshness of meat in real-time, thus helping to reduce food waste and prevent foodborne illnesses.

Copper-Polyphenol Nanocomplexes with Peroxidase-Like Activity for Enhanced Fungal Eradication

Fang Liu,Yongcheng Chen,Yue Huang,Qiao Jin

The increasing severity of fungal infections globally and the limited availability of effective treatments necessitate novel therapeutic strategies. In this study, we developed a copper-polyphenol nanocomplexes (CPE NPs) through a one-step synthetic process by the coordination self-assembly of copper sulfate (CuSO4), protocatechuic acid (PA), and ε-Poly-L-lysine (EPL). CPE NPs demonstrated peroxidase-like activities, which could catalyze H2O2 to generate hydroxyl radicals (•OH) to effectively eradicate fungi. CPE NPs were stable in physiological neutral environment. However, since metal-phenolic coordination interaction was unstable in acidic pH, CPE NPs could be dissociated in acidic environments, such as fungal infection sites, leading to effective release of Cu2+ and EPL, thereby exerting antifungal effects. CPE NPs exhibited excellent fungicidal ability at both pH 7.4 and 6.0 against Candida albicans and Aspergillus flavus. More importantly, CPE NPs could effectively eradicate fungi in 20% Sabouraud dextrose broth (SDB) medium containing rich inorganic salts and proteins, in which dissociative Cu2+ completely lost its fungicidal ability. In summary, CPE NPs could effectively deliver Cu2+ to fungal surface for enhanced fungal eradication by generating •OH, highlighting their great potential as a promising nano-therapeutic agent for the treatment of fungal infections.

Biomimetic Design of Underwater Adhesives Based on Tea Polyphenol-Modified Gelatin

Ziwei Wu,Zhipeng Li,Yixiao Li,Haoyu Wang,Jiang Yue,Tieling Xing

Although many tissue adhesives with good biocompatibility are currently available, their lack of wet adhesion capacity significantly hinders their clinical application. Therefore, further development and exploration of new medical adhesives are necessary. Inspired by the adhesion mechanism of marine mussels, through modifying gelatin protein with gallic acid (GA) for wet adhesion and cross-linking gelatin (Gel) molecular chains with tea polyphenols (TP), the adhesive TP-GA/Gel was developed. The adhesive exhibited an adhesion strength of up to 130.47 kPa to porcine skin tissues and maintained a high adhesion state in various aqueous environments, demonstrating excellent and reproducible adhesion. Additionally, TP-GA/Gel possessed outstanding antimicrobial, antioxidant, and biocompatibility properties. In an in vivo wound healing study with SD rats, the wound area treated with TP-GA/Gel adhesive decreased from 10.3 mm2to 0.9 mm2after 15 days, promoting effective and scarless wound healing. These results highlight the promising clinical potential of TP-GA/Gel as a medical adhesive.

Carbon nanotube incorporated polyphenol electro-responsive ultrafiltration membranes toward dye separation

Xinyi Wang,Zezhen Zhang,Weishan Deng,Haolan Xiao,Liming Xia,Lili Wu

Carbon nanotube (CNT) is a promising raw material for membrane fabrication due to its good electrical conductivity, great specific surface area, and special piping structure. In this report, tannin (TA), CNT, and D-aspartic acid (DAS) were co -deposited on the surface of polyethersulfone (PES) ultrafiltration membrane to construct an electrically responsive coating. The membrane can maintain the high electrical conductivity while avoiding the membrane defects due to the accumulation of CNTs and the different lengths of chain segments in the process of CNT co-blending to form the membrane. The results show that the average pore size of the membrane is reduced from 19.55 nm to 8.165 nm, and the retention capacity is greatly improved, showing good separation efficiency (99.5 %) and hydrophilicity (water contact angle of 13°). Through adsorption and electric field deflection, CNT endows the membrane with the ability to selectively separate dyestuffs with different electrically charged properties. Long-term stability tests show that the modified membrane still maintains a high level of pure water flux and conductivity after seven days of operation. This paper is of exploratory significance in the field of electrically responsive smart membranes for dye separation. The membrane developed in this study exhibits unique selective separation performance and can dynamically adjust its separation efficiency by externally controlling the electric field strength. This demonstrates its excellent intelligence, making it highly promising for applications in electrically responsive intelligent separation membranes. Additionally, it offers a novel approach for treating dye-containing wastewater using electrically charged functional membranes.

Polyphenol-Reinforced Glycocalyx-Like Hydrogel Coating Induced Myocardial Regeneration and Immunomodulation

Huining Wan,Xiaohui Yang,Yutong Zhang,Xiyu Liu,Yanyan Li,Yumei Qin,Hui Yan,Lan Gui,Ke Li,Longjian Zhang,Li Yang,Bo Zhang,Yunbing Wang

Although minimally invasive interventional occluders can effectively seal heart defect tissue, they still have some limitations, including poor endothelial healing, intense inflammatory response, and thrombosis formation. Herein, a polyphenol-reinforced medicine/peptide glycocalyx-like coating was prepared on cardiac occluders. A coating consisting of carboxylated chitosan, epigallocatechin-3-gallate (EGCG), tanshinone IIA sulfonic sodium (TSS), and hyaluronic acid grafted with 3-aminophenylboronic acid was prepared. Subsequently, the mercaptopropionic acid-GGGGG-Arg-Glu-Asp-Val peptide was grafted by the thiol–ene “click” reaction. The coating showed good hydrophilicity and free radical-scavenging ability and could release EGCG–TSS. The results of biological experiments suggested that the coating could reduce thrombosis by promoting endothelialization, and promote myocardial repair by regulating the inflammatory response. The functions of regulating cardiomyocyte apoptosis and metabolism were confirmed, and the inflammatory regulatory functions of the coating were mainly dependent on the NF-kappa B and TNF signaling pathway.

Tea polyphenol-derived nanomedicine for targeted photothermal thrombolysis and inflammation suppression

Wang Hui,Tang Cui,Xiang Yuxia,Zou Chan,Hu Jianming,Yang Guoping,Zhou Wenhu

Thrombotic diseases impose a significant global health burden, and conventional drug-based thrombolytic therapies are encumbered by the risk of bleeding complications. In this study, we introduce a novel drug-free nanomedicine founded on tea polyphenols nanoparticles (TPNs), which exhibits multifaceted capabilities for localized photothermal thrombolysis. TPNs were synthesized through a one-pot process under mild conditions, deriving from the monomeric epigallocatechin-3-gallate (EGCG). Within this process, indocyanine green (ICG) was effectively encapsulated, exploiting multiple intermolecular interactions between EGCG and ICG. While both TPNs and ICG inherently possessed photothermal potential, their synergy significantly enhanced photothermal conversion and stability. Furthermore, the nanomedicine was functionalized with cRGD for targeted delivery to activated platelets within thrombus sites, eliciting robust thrombolysis upon laser irradiation across diverse thrombus types. Importantly, the nanomedicine’s potent free radical scavenging abilities concurrently mitigated vascular inflammation, thus diminishing the risk of disease recurrence. In summary, this highly biocompatible multifunctional nanomaterial holds promise as a comprehensive approach that combines thrombolysis with anti-inflammatory actions, offering precision in thrombosis treatment.

Starch-nanoencapsulated polyphenol-induced polysaccharide gel coatings with efficient preservation capability

Changsheng Luo,Fang Xie,Qianqian Chen,Jiaojiao Zhou,Zhijun He,Jie Cai

Here, a facile self-assembly strategy was used to fabricate octenyl succinic anhydride starch (OSAS) nano micelles for encapsulation of K-carrageenan comprising curcumin (Cur) (KC/Cur-OSAS). KC/Cur-OSAS was used as a multipurpose edible food packaging coating on grapes. The characteristics, storage stability, photoactivated antibacterial properties and antimicrobial mechanisms of KC/Cur-OSAS were investigated. The experimental results demonstrated the potential photoactivated antibacterial performance of KC/Cur-OSAS gel coating, with an antibacterial efficacy of more than of 99 % against S. aureus and E. coli . Moreover, the coating showed good biocompatibility and 3D printing potential, and could reduce dewatering, prevent nutrient degradation, inhibit bacterial proliferation, increase nutritional content, maintain food quality, and extend the shelf life of food by more than 3 days, when conserved at a temperature of 25 °C. Hence, the light-driven KC/Cur-OSAS coatings prepared in this has great potential for improving nutrient delivery and extending the shelf life of perishable food and can be used for developing food packaging and preservation materials.

Metal-polyphenol network coated photothermal nanocarriers for pH-activated drug delivery

Qiong Wang,Jing Chen,Junhong Ling,Han Zhao,Xiao-kun Ouyang,Nan Wang

The combination of photothermal and chemotherapy is a promising strategy for the synergistic inhibition of tumor cells. By exploiting the tumor microenvironment, chemotherapeutic drugs can target the tumor site, thereby enhancing their efficacy. This study focused on the development of nanodrugs, aiming to harness the synergistic photothermal and chemotherapy capabilities for enhanced anti-tumor effects. The core material was made of copper sulfide (CuS), which possesses photothermal conversion properties, while doxorubicin (DOX) was loaded on mesoporous polydopamine (MPDA). The drug-loaded nanoparticles were then encapsulated in acid-sensitive tannic acid and an Fe complex (TA-Fe). The photothermal conversion properties, drug loading, and release of DOX from CuS-MPDA-DOX@TA-Fe were then investigated. The results showed that DOX had an encapsulation ratio of 82.2 % and a release ratio of 43.49 % in an acidic environment (pH 5.2). In vitro studies showed that CuS-MPDA-DOX@TA-Fe triggered by NIR effectively inhibited tumor cell proliferation and that the tumor cells showed suitable levels of nanoparticle uptake. The results thus show that CuS-MPDA-DOX@TA-Fe nanodrugs could be utilized as a promising therapeutic strategy for cancer treatment with synergistic photothermal/chemotherapeutic anti-tumor effects.
