Betaine improves appetite regulation and glucose-lipid metabolism in mandarin fish (Siniperca chuatsi) fed a high-carbohydrate-diet by regulating the AMPK/mTOR signaling

Hongyan Li,Yanzhi Zeng,Guangjun Wang,Kai Zhang,Wangbao Gong,Zhifei Li,Jingjing Tian,Yun Xia,Wenping Xie,Jun Xie,Shouqi Xie,Ermeng Yu

Diets with high carbohydrate (HC) was reported to have influence on appetite and intermediary metabolism in fish. To illustrate whether betaine could improve appetite and glucose-lipid metabolism in aquatic animals, mandarin fish ( Siniperca chuatsi ) were fed with the HC diets with or without betaine for 8 weeks. The results suggested that betaine enhanced feed intake by regulating the hypothalamic appetite genes. The HC diet-induced downregulation of AMPK and appetite genes was also positively correlated with the decreased autophagy genes, suggesting a possible mechanism that AMPK/mTOR signaling might regulate appetite through autophagy. The HC diet remarkably elevated transcriptional levels of genes related to lipogenesis, while betaine alleviated the HC-induced hepatic lipid deposition. Additionally, betaine supplementation tended to store the energy storage as hepatic glycogen. Our findings proposed the possible mechanism for appetite regulation through autophagy via AMPK/mTOR, and demonstrated the feasibility of betaine as an aquafeed additive to regulate appetite and intermediary metabolism in fish.

An efficient hydrophobic modification strategy for enhancing collagen stability: Effect of lauric acidic N-hydroxysulfosuccinimide ester on the structure and properties of fish collagen

Cong Bai,Hao Jiang,Minggao Deng,Qingtian Gong,Peiyu Li,Wentao Liu

The hydrophobic modifications of collagen have emerged as effective approaches to endow collagen-based materials with enhanced structural stability and advanced functions, but the existing hydrophobic modification strategies are relatively inefficient and environmentally unfriendly due to the use of large quantities of organic solvents. It is highly appealing to develop a greener and more efficient strategy to prepare hydrophobically modified collagens (HMCs). Herein, the ester of N -hydroxysulfosuccinimide activated lauric acid (NHSS-LA) was prepared as a promising alternative hydrophobic modifier, which could be efficiently used in aqueous solutions for enhancing the structure and properties of HMCs and hydrophobically modified collagen fiber membranes (HMCFMs). The performance of HMCs and HMCFMs could be tuned through adjusting the dosages of NHSS-LA. Besides maintaining the triple helical structure and cytocompatibility of HMCs, the hydrophobic modification could improve the thermal stability of HMCs owing to the enhancement of hydrophobic associations between collagen molecules, which was closely related to collagen concentrations and modification rates. Furthermore, the water resistance, thermal stability, and enzymatic degradation stability of HMCFMs were significantly improved. This work provides an efficient strategy for modifying fish collagen and fabricating high-performance collagen-based materials.

A ray-inspired flexible robot fish with stable controllability actuated by photo -responsive composite hydrogels with poly(N-isopropylacrylamide) and multi-walled carbon nanotubes

Wenguang Yang,Qinghao Guo,Xiangyu Teng,Zezheng Qiao,Haibo Yu

Drawing inspiration from the movement behaviour of rays in nature, we have successfully developed a ray-inspired flexible robot fish driven by visible light. A composite made of poly(N-isopropylacrylamide) (PNIPAM) and multi-walled carbon nanotubes (MWCNTs) has excellent biocompatibility and photo-responsive deformability . The PNIPAM hydrogel flexible material of the robot fish was prepared by light curing printing technology. Under the control of visible light, the robot fish swims in a straight line at an average speed of about 0.42 mm/s. The photoinduced asymmetric deformation mechanism is used to control the deformation of the fins on both sides of the robot fish, so that the robot fish can turn and realize directional swimming control. These findings have important implications for the development of flexible bionic robots and provide valuable insights for the research of bionic robots driven by visible light.

Vortex-Assisted Liquid-Liquid Micro-extraction Followed by Head Space Solid Phase Micro-extraction for the Determination of Eugenol in Fish Using GC-MS

Liang Xu,Feng Ting-Ting,Wu Jian-Hai,Du Ming,Qin Lei,Wang Zhen-Yu,Xu Xian-Bing

Vortex-assisted liquid-liquid micro-extraction (VALLME) was applied for eliminating the matrix effect and improving the sensitivity of eugenol determination by headspace solid phase micro-extraction (HS-SPME) coupled with gas chromatography-mass spectrometer (GC-MS) in fish samples. Under the optimum condition of VALLME and HS-SPME, the good linearity of eugenol in fish samples was obtained in the range from 15.0 to 750.0 μg/kg. The limit of detection (LOD) and the limit of quantification (LOQ) for eugenol were 0.5 and 1.5 μg/kg, respectively. The recovery of eugenol was ranging from 95.4 to 119.9% and the relative standard deviation (RSD %) for eugenol was lower than 20%. The results turned out that VALLME followed HS-SPME-GC-MS was a simple, rapid, and accurate method for the determination of eugenol in fish samples. The levels of eugenol in different parts of real samples were tested by the developed method, and the results concluded that the developed method was compatible with GC-MS analysis with long-term application and had excellent repeatability, linearity, and sensitivity.

Rapid in vivo determination of fluoroquinolones in cultured puffer fish (Takifugu obscurus) muscle by solid-phase microextraction coupled with liquid chromatography-tandem mass spectrometry

Yijia Tang,Jianqiao Xu,Le Chen,Junlang Qiu,Yuan Liu,Gangfeng Ouyang

Fluoroquinolones (FQs) are a group of antimicrobial agents that have been widely used for therapeutic purposes in clinical medicine for human and veterinary diseases, as well as in aquaculture production. Their residues, however, may survive in foods of animal origin, thus causing health risks for human. In this study, a rapid and sensitive method based on in vivo solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed to detect the residues of five FQs in cultured puffer fish ( Takifugu obscurus ). In vitro fiber evaluation experiment demonstrated that, compared with the thicker polydimethylsiloxane (PDMS) coating (165   µm), the custom-made biocompatible C18-PAN fibers (45   µm) exhibited much higher extraction efficiencies towards FQs (approximately 9–31 times). The custom-made C18-PAN coating also possessed excellent reproducibility in fish muscle with the intra-fiber relative standard deviations (RSDs) ranging from 11.2% to 14.3% (n = 6) and inter-fiber RSDs ranging from 13.1% to 16.1% (n = 6), which was suitable for in vivo sampling. The custom-made SPME fibers were subsequently applied to determine fluoroquinolones in dorsal-epaxial muscle of living puffer fish. The accuracies were verified through the comparison with traditional liquid extraction (LE) method, and the sensitivities were demonstrated to be satisfying with the limits of detection (LODs) ranging from 0.3   ng   g −1 to 1.5   ng   g −1 . In general, this study presented a convenient and high-efficient method to determine fluoroquinolones in puffer fish by in vivo sampling.

Toxic effects and comparison of common amino antioxidants (AAOs) in the environment on zebrafish: A comprehensive analysis based on cells, embryos, and adult fish

Chenyang Rao,Fuhao Chu,Fang Fang,Dongfang Xiang,Bo Xian,Xiaying Liu,Shaopan Bao,Tao Fang

The ubiquity of amino antioxidants (AAOs) in the environment has attracted increasing attention, given their potential toxicity. This investigation represents a pioneering effort, systematically scrutinizing the toxicological effects of four distinct AAOs across the developmental spectrum of zebrafish, encompassing embryonic, larvae, and adult stages. The results indicate that four types of AAO exhibit varying degrees of cell proliferation toxicity. Although environmentally relevant concentrations of AAOs exhibit a comparatively circumscribed impact on zebrafish embryo development, heightened concentrations (300 μg/L) of N -(1,3-dimethylbutyl)- N′ -phenyl- p -phenylenediamine (6PPD) and N -isopropyl- N′ -phenyl- p -phenylenediamine (IPPD) distinctly evoke developmental toxicity. Behavioral analysis results indicate that at concentrations of 20 and 300 μg/L, the majority of AAOs significantly reduced the swimming speed and activity of larvae. Moreover, each AAO triggers the generation of reactive oxygen species (ROS) in larvae, instigating diverse levels of oxidative stress. The study delineates parallel toxicological patterns in zebrafish exposed to 300 μg/L of 6PPD and IPPD, thereby establishing a comparable toxicity profile. The comprehensive toxicity effects among the four AAOs is as follows: IPPD >6PPD >  N -Phenyl-1-naphthylamine (PANA) > diphenylamine (DPA). These findings not only enrich our comprehension of the potential hazards associated with AAOs but also provide data support for structure-based toxicity prediction models.

Effects of hydroxyproline supplementation in low fish meal diets on collagen synthesis, myofiber development and muscular texture of juvenile Pacific white shrimp (Litopenaeus vannamei)

Menglin Shi,Haomin Li,Tianyu Chen,Bocheng Huang,Xiaoyue Li,Xiaohui Dong,Shuyan Chi,Qihui Yang,Hongyu Liu,Junming Deng,Beiping Tan,Shuang Zhang,Shiwei Xie

This experiment aimed to evaluate the impact of dietary hydroxyproline (Hyp) supplementation on the flesh quality of juvenile Pacific white shrimp ( Litopenaeus vannamei ) fed a low fishmeal diet. Six formulated diets included one high fishmeal (HF; 25% fishmeal content) and five low fishmeal diets (10% fishmeal content) with 0%, 0.2%, 0.4%, 0.6% and 0.8% Hyp (LF0, LF2, LF4, LF6 and LF8, respectively). Each diet was assigned to four replicates and 40 shrimp (0.32 ± 0.00 g) per replicate were fed four times a day for 8 weeks. Dietary Hyp supplementation had little effects on growth performance, but significantly increased the contents of Hyp, prolyl 4-hydroxylases (P4Hs), and collagen ( P < 0.05). The meat yield, springiness, hardness, chewiness, and cohesiveness of muscle were the highest in the LF2 group. Cooking loss and freezing loss of muscle were the lowest in the LF2 group ( P < 0.05). Dietary supplementation with 0.4% Hyp increased the myofiber density and decreased the myofiber diameter of muscle ( P < 0.05). Supplementation of Hyp in the diet up-regulated the mRNA expression of smyhc5 , smyhc15 , col1a1 , col1a2 , igf-1f , tgf-β and tor , and down-regulated the mRNA expression of smyhc 1 , smyhc 2 , smyhc 6a ( P < 0.05). Supplementation of Hyp in the diet up-regulated the protein expression of P-4E-BP1, P-AKT, AKT and P-AKT/AKT ( P < 0.05). These results suggested that the addition of 0.4% Hyp to low fishmeal diets improved the flesh quality of L. vannamei .

Determination of a major allergen in fish samples by simple and effective label-free capillary electrophoretic analysis after background suppression in ion-exchange chromatography

Linglin Fu,Jinru Zhou,Chong Wang,Yan Zhang,Aijin Ma,Yanbo Wang

A simple analytical method using ion-exchange chromatography (IEC) coupled with capillary zone electrophoresis (CZE) was newly developed for detection and quantification of parvalbumin (PV), a major allergen, in fish. The procedure of IEC for simple enrichment of PV crude extract was optimized. By using 25 mmol/L borate-borax buffer with 15 kV separation voltage at pH 9.2 for CZE separation, the migration time, separation efficiency and electrophoretic resolution greatly improved. Under the optimal conditions, PV was determined in 2.8 min, with the limit of detection (LOD) at 0.71 μg/mL (S/N = 3) and the recoveries at 89.6%–104.7%. We also found that only when the concentration of PV was above LOD reported here was the protein capable of stimulating human mast cell degranulation, indicating the biological significance of the LOD. Finally, the use of this method to analyze fish samples with simple sample preparation highlights the applicability for detection of allergens in seafood matrices.

Rapid detection of malachite green residues in fish using a surface-enhanced Raman scattering-active glass fiber paper prepared by in situ reduction method

Di Deng,Qiuyuan Lin,Hua Li,Zhipeng Huang,Yunyan Kuang,Hui Chen,Jilie Kong

Food safety issues, especially those related to drug residues , have caused concern in recent years. Malachite green (MG), which is frequently used as ectoparasiticide, fungicide , and antiseptic in fish farming, is poisonous and carcinogenic . Several technologies and methods have been developed to accurately detect MG, but they can be time-consuming and expensive. Herein, we prepared a glass fiber paper as a surface-enhanced Raman scattering substrate for rapid and convenient detection of MG residues in fish. Uniform silver nanoparticles were deposited on the glass fiber paper by two in situ reduction steps of silver nitrate. The Raman signal of MG was detected and recorded using a portable Raman spectrometer in 10 s. There was a good linear relationship between the surface-enhanced Raman scattering signal intensity and the concentration of MG between 1 × 10 −7 and 1 × 10 −5 mol/L and the limit of detection was 5 × 10 −10 mol/L. Residual MG in fish samples was extracted and detected with a recovery rate range of 98.9–119.4%. This method provides a novel way to detect MG in fish.

Generation of functional single-chain fragment variable from hybridoma and development of chemiluminescence enzyme immunoassay for determination of total malachite green in tilapia fish

Jiexian Dong,Zhenfeng Li,Yu Wang,Maojun Jin,Yudong Shen,Zhenlin Xu,A.M. Abd El-Aty,Shirley J. Gee,Bruce D. Hammock,Yuanming Sun,Hong Wang

To determine malachite green (MG) and its major metabolite, leucomalachite green (LMG) residual levels in tilapia fish, chemiluminescent enzyme immunoassay (CLEIA) was developed based on a single-chain variable fragment (scFv)-alkaline phosphatase (AP) fusion protein. At first, V H and V L gene sequences were cloned from hybridoma cell lines secreting monoclonal antibody against LMG, and then thoroughly by database-assisted sequence analysis. Finally, the productive V H and V L were assembled to an intact scFv sequence and engineered to produce scFv-AP fusion protein. The fusion protein was further identified as a bifunctional reagent for immunoassay, then a sensitive one-step CLEIA against LMG was developed with a half-maximal inhibitory concentration (IC 50 ) and limit of detection (LOD) of 1.3 and 0.04 ng/mL, respectively. The validation results of this novel competitive CLEIA was in line with those obtained by classical HPLC method for determination of total MG in spiked and field incurred samples.

Dietary supplementation of chenodeoxycholic acid improved the growth performance, immune response and intestinal health of juvenile Penaeus monodon fed a low fish-meal diet

Shiwei Xie,Dan Wei,Lixia Tian,Yongjian Liu

Bile acid metabolism in aquatic animals usually is suppressed when fish meal (FM) is replaced by plant proteins. An eight weeks feeding experiment was conducted to evaluate the effects of dietary chenodeoxycholic acid (CDCA) supplementation on growth, immune response and intestinal health of Penaeus monodon fed a low FM diet. Three diets were formulated: high FM (25 %) diet (HF), low FM (15 %) diet (LF), and low FM diet supplemented with 0.1 % CDCA (LFC). Shrimp fed Diet LF showed lower weight gain and feed efficiency than those fed Diet HF. Low dietary FM up-regulated the mRNA level of xbp1 , and down-regulated the mRNA levels of sod , iap and NF-κB signaling pathway-related genes ( traf6 , tube ). Dietary supplementation of CDCA up-regulated the expression of relish and anti-oxidative-related genes ( sod and cat ) in the hepatopancreas and midgut. Low dietary FM impaired the intestinal fold and induced the endoplasmic reticulum swollen of intestinal epithelial cell, dietary supplementation of CDCA alleviated this problem. The intestinal microbiota diversity was lower in shrimp fed Diet LF than those fed HF. Dietary supplementation of CDCA in a low FM diet increased the intestinal microbiota diversity by decreasing the relative abundance of dominant phylum Proteobacteria. Low dietary FM increased the relative abundance of a dominant pathogenic bacterium ( Vibrio ), which was decreased by the supplementation of CDCA. These results indicated that dietary supplementation of CDCA in a low FM diet improved the growth performance, immune response and intestinal health of shrimp.

Dian-Bing Zhou,Ya-Bing Xiao,Fang Han,Ya-Ning Lv,Lei Ding,Wei Song,Yu-Xin Liu,Ping Zheng,Da Chen

A novel magnetic metal-organic frameworks (Fe 3 O 4 @UiO-66-SH) was successfully prepared by coating Fe 3 O 4 nanospheres with sulfur-functionalized UiO-66. The Fe 3 O 4 @UiO-66-SH possesses both the magnetic properties of Fe 3 O 4 and the diverse properties of metal-organic framework (MOF) in one material, which has the superiority of high surface area, easy-operation and strong adsorb ability with mercury, is used for the magnetic solid-phase extraction of methylmercury (MeHg + ) and inorganic mercury (Hg 2+ ) in water and fish samples. The analyzes were conducted by high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS). The different pretreatment conditions influencing the extraction recoveries of Hg 2+ and MeHg + , including adsorbent amount, pH, extraction time, elution solvent, elution volume, desorption time, co-existing ions and dissolved organic materials were investigated. Under the optimized conditions, the limits of detection (LODs) of Hg 2+ and MeHg + for water samples were 1.4 and 2.6 ng L −1 , and the limits of quantification (LOQs) of Hg 2+ and MeHg + for water samples were 4.7 and 8.7 ng L −1 . The enrichment factors (EFs) were 45.7 and 47.6 fold for Hg 2+ and MeHg + , respectively. The accuracy of the proposed method was demonstrated by analyzing the certified reference material of fish tissue (GBW10029) and by determining the analyte content in spiked water and fish samples. The determined values were in good agreement with the certified values and the recoveries for the spiked samples were in the range of 84.5–96.8%.
