Colonic delivery and controlled release of curcumin encapsulated within plant-based extracellular vesicles loaded into hydrogel beads

Hang Liu,Yikun Liu,Shengfeng Peng,Lei Zhou,David Julian McClements,Suqiong Fang,Wei Liu

In this study, the application of banana-derived extracellular vesicles (EVs) as natural carriers for hydrophobic nutraceuticals was investigated for the first time. Curcumin was solubilized in an amorphous state within the hydrophobic domains of the EVs in banana juice. The bioaccessibility of the curcumin was considerably higher in the curcumin-loaded EVs (CEVs) than for pure curcumin crystals. The retention/release behavior of the curcumin in the CEVs was regulated by incorporating the curcumin-loaded banana juice into hydrogel beads, which were assembled from sodium alginate and banana pectin. The pectin and D-galacturonic acid contents of the banana juice were 0.87 % ± 0.11 % and 0.58 % ± 0.08 %, respectively. With the introduction of sodium alginate, the sphericity, hardness, chewiness, water holding capacity, surface smoothness and other physiochemical properties of the hydrogel beads were improved. Fourier Transform infrared analysis showed that hydrogen bonding played an important role in the formation of the hydrogel beads. The hydrogel beads had a higher swelling ratio under neutral conditions than under acidic conditions. Encapsulation of the curcumin-loaded banana juice within the hydrogel beads retarded curcumin release and enhanced the therapy potential of ulcerative colitis, which indicated that these delivery systems could be designed to regulate the release profile of the CEVs. Encapsulation of the curcumin also improved its resistance to heating, exposure and storage. In summary, we have shown that encapsulation of curcumin within plant-based EVs loaded into hydrogel beads can improve its functional performance.

Antibacterial antioxidant polyurethane/curcumin patch with anti-swelling ability for tissue adhesion and treatment of oral ulcers

Xiaolei Guo,Lin Yang,Nan Sheng,Xirui Long,Zhen Li,Feng Luo,Jiehua Li,Hong Tan

Oral ulcers severely affect the patient’s mood, diet, and language activities and are mainly treated by topical administration of a patch. However, the currently commercially available patches have a limited treatment effect because of the short action time in the oral cavity. In this study, we present a novel adhesive patch made from trifunctional polycaprolactone crosslinked waterborne polyurethane (CDCPU) loaded with curcumin for treating oral ulcers. The long-chain triol cross-linking points in the polyurethane can limit water entry, while stabilizing the curcumin loading in the emulsion. Meanwhile, the interaction between curcumin and polyurethane improved the anti-swelling (swelling rate of 37.7 ± 4.9 %), water resistance (water absorption rate of 66.6 ± 5.6 %), and mechanical properties of the patch. The adhesion strength of the CDCPU patch to porcine oral mucosa and skin was 11.3 ± 1.7 kPa and 14.7 ± 2.5 kPa, respectively. The addition of curcumin provides the CDCPU patch with effective antibacterial and antioxidant functions. Moreover, the CDCPU patch was effective in promoting mucosal epithelialization and ulcer healing in a rat oral ulcer model. The development of this adhesive patch provides a new strategy for the treatment of oral ulcers.

Construction of enzymatic crosslinking beta-lactoglobulin-camellia oil emulsion gels: microstructural characterization, stability, and curcumin delivery behavior

Liwei Zhao,Taijiao Xiang,Jiaoyu Wang,Ziming Xu,Danli Wang,Tinglan Yuan,Ling Li,Haina Yuan,Gongshuai Song,Mengna Zhang,Jinyan Gong

Protein-based emulsion gels with transglutaminase (TG)-catalyzed crosslinking exhibit great potential for delivering hydrophobic bioactive substances. However, effects of TG-catalyzed crosslinking modifications under different reaction times on the structural and textural characterization, stability, and delivery behavior have rarely been reported. This study investigated the effect of TG-catalyzed crosslinking on the physicochemical, structural, and functional properties of β-lactoglobulin (β-LG) - camellia oil (CAO) emulsion gels by cold-gelation under different reaction times. Results revealed that prolonged reaction times could improve the formation of intramolecular and intermolecular covalent bonds between lysine and glutamine residues. Rheological analysis showed an increase in both the storage modulus (G’) and loss modulus (G’’), indicating enhanced the gel elasticity and stability. Furthermore, the oxidative stability, the water-holding capacity, and functional stability (freeze-thaw, UV-light, thermal, and salt stability) of the emulsion gels were enhanced by the TG-catalyzed crosslinking. In the in vitro digestion process, the encapsulation efficiency and bioaccessibility of curcumin (Cur) were enhanced in the TG-catalyzed crosslinking β-LG-CAO emulsion gel system when the reaction time exceeded 3 h. The dense gel structure, improved stability, and great Cur delivery behavior of the emulsion gel could be formed by TG-catalyzed crosslinking at 4 h. This study benefited the encapsulation and release behavior of Cur in oil-in-water emulsions and the application in food and nutraceutical products.

A peptide-decorated and curcumin-loaded mesoporous silica nanomedicine for effectively overcoming multidrug resistance in cancer cells

Xian Sun,Yingping Luo,Liwei Huang,Bo-Yang Yu,Jiangwei Tian

Multidrug resistance (MDR) is a major obstacle to the success of cancer chemotherapy, which is closely associated with the overexpression of P-glycoprotein (P-gp) and the endolysosomal capture of drug. Therefore, to explore a strategy that could down-regulate P-gp expression and promote drugs escaping from the endolysosome is highly desired for overcoming MDR in chemotherapeutic treatment. This work reports a peptide-functionalized mesoporous silica nanomedicine (MSN) for encapsulation of curcumin (CUR) and doxorubicin (DOX) to significantly reverse the MDR in consequence of rapid endolysosomal escape and the inhibition of P-gp function. The nanomedicine is modified with the peptide of GFLGHHHRRGDS to encapsulate CUR and DOX with high loading efficiency and content. It can be selectively uptaken by the DOX resistant MCF-7/ADR cells via the specific recognition between RGDS and αvβ3 integrin to enter into lysosomes. Afterwards, the GFLG is cleaved by the lysosomal cathepsin B to release CUR and DOX from MSN. Furthermore, the imidazole-rich HHHR peptide can induce effective lysosomal membrane permeability which is beneficial for drug escape from lysosome, CUR-mediated inhibition of P-gp, and nuclear import of DOX in MCF-7/ADR cells. Confocal fluorescence imaging, flow cytometry and MTT assays demonstrate that the combination of CUR and the peptide-functionalized MSN markedly enhances the intracellular DOX concentration and achieves a successful chemotherapeutic treatment to MCF-7 or MCF-7/ADR cells, thus this work offers an effective strategy to overcome MDR.

Nanoparticles based on chitosan hydrochloride/hyaluronic acid/PEG containing curcumin: In vitro evaluation and pharmacokinetics in rats

Yurui Xu,Sajid Asghar,Liu Yang,Zhipeng Chen,Hongying Li,Wenwen Shi,Yibo Li,Qinqing Shi,Qineng Ping,Yanyu Xiao

Nanoparticles based on chitosan hydrochloride (CSH)-hyaluronic acid (HA)-PEG were prepared for delivering curcumin (CUR) (CUR-PNPs) to brain tumor. CUR-PNPs of 245.9   nm and spherical morphology were obtained at optimized CSH/HA/PEG20000/CUR ratios with negative charge of about −27.2   mV and EE of approximately 93.3%. Cytotoxicity studies showed that CUR-PNPs improved drug’s anticancer activity in rat glioma cells (C6). The cellular uptake mechanism study showed active targeting of CUR-PNPs into C6 cells by HA mediated endocytosis . Clathrin-coated pit mediated endocytosis, clathrin-mediated endocytosis and macropincytosis were also identified as the entry pathways of PNPs into C6 cells. Pharmacokinetics of preparations in rats after i.v. administration further proved the superiority of CUR-PNPs (3.98 times greater than the area under the curve of CUR solution). In conclusion, CUR-PNPs might be a promising carrier for the therapy of brain tumors.

Delivery of curcumin by directed self-assembled micelles enhances therapeutic treatment of non-small-cell lung cancer

Wen-Ting Zhu,Sheng-Yao Liu,Lei Wu,Hua-Li Xu,Jun Wang,Guo-Xin Ni,Qing-Bing Zeng

Background It has been widely reported that curcumin (CUR) exhibits anticancer activity and triggers the apoptosis of human A549 non-small-cell lung cancer (NSCLC) cells. However, its application is limited owing to its poor solubility and bioavailability. Therefore, there is an urgent need to develop a new CUR formulation with higher water solubility and better biocompatibility for clinical application in the future. Materials and methods In this study, CUR-loaded methoxy polyethylene glycol–polylactide (CUR/mPEG–PLA) polymeric micelles were prepared by a thin-film hydration method. Their characteristics and antitumor effects were evaluated subsequently. Results The average size of CUR/mPEG–PLA micelles was 34.9±2.1 nm with its polydispersity index (PDI) in the range of 0.067–0.168. The encapsulation efficiency and drug loading were 90.2%±0.78% and 9.1%±0.07%, respectively. CUR was constantly released from the CUR/mPEG–PLA micelles, and its cellular uptake in A549 cells was significantly increased. It was also found that CUR/mPEG–PLA micelles inhibited A549 cell proliferation, increased the cell cytotoxicity, induced G2/M stage arrest and promoted cell apoptosis. Moreover, the CUR/mPEG–PLA micelles suppressed the migration and invasion of A549 cells more obviously than free CUR. Additionally, CUR/mPEG–PLA micelles inhibited human umbilical vein endothelial cells migration, invasion and corresponding tube formation, implying the antiangiogenesis ability. Its enhanced antitumor mechanism may be related to the reduced expression of vascular endothelial growth factor, matrix metalloproteinase (MMP)-2, MMP-9 and Bcl-2 as well as the increased expression of Bax. Conclusion The mPEG–PLA copolymer micelles can serve as an efficient carrier for CUR. The CUR/mPEG–PLA micelles have promising clinical potential in treating NSCLC.

Enhanced effect in combination of curcumin- and ketoconazole-loaded methoxy poly (ethylene glycol)-poly (ε-caprolactone) micelles

Fangfang Teng,Peizong Deng,Zhimei Song,Feilong Zhou,Runliang Feng

In order to enhance water-solubility and realize controlled release while keeping synergistic effects of ketoconazole and curcumin , drug-loaded methoxy poly (ethylene glycol)-b-poly (ε-caprolactone) micelles were prepared through thin membrane hydration method. Transmission electric microscopy and dynamitic light scattering characterization revealed the formation of ketoconazole- and curcumin-loaded micelles with an average size of 44.70   nm and 39.56   nm, respectively. The drug-loaded micelles endowed the two drugs’ slow controlled release with water-solubility enhanced to 85 and 82000 folds higher than the corresponding raw drugs, respectively. In vitro antifungal activity test, chequerboard test and inhibition zone test indicated that efficacy of ketoconazole-loaded micelles was improved by introduction of curcumin-loaded micelles with a low fractional inhibitory concentration index (0.073). Biofilm formation inhibition assay also demonstrated that participation of curcumin-loaded micelles obviously strengthened the inhibition of fungal biofilms formation induced by ketoconazole-loaded micelles. The high synergistic activity of combinations is encouraging and the MPEG-PCL micelle is a potential drug delivery system for the combination of ketoconazole and curcumin.

Boron and nitrogen co-doped carbon dots as a sensitive fluorescent probe for the detection of curcumin

Wei Bian,Xuan Wang,Yakun Wang,Haifen Yang,Jialin Huang,Zongwei Cai,Martin M. F. Choi

In this present study, a fluorescent probe was developed to detect curcumin, which is derived from the rhizomes of the turmeric. We used a simple and economical way to synthesize boron and nitrogen co-doped carbon dots (BNCDs) by microwave heating. The maximum emission wavelength of the BNCDs was 450 nm at an excitation wavelength of 360 nm. The as-prepared BNCDs were characterized by multiple analytical techniques such as transmission electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, and infrared spectroscopy. The synthesized carbon nanoparticles had an average particle diameter of 4.23 nm. The BNCDs exhibited high sensitivity to the detection of curcumin at ambient conditions. The changes of BNCDs fluorescent intensity show a good linear relationship with the curcumin concentrations in the range 0.2–12.5 μM. This proposed method has been successfully applied to detect the curcumin in urine samples with the recoveries of 96.5–105.5%.

Amperometric determination of NADH based on a poly-Ni(II)–curcumin composite film modified glassy carbon electrode

Xiaoqian Ouyang,Yaping Ding,Xi Ba,Shuqing Gu

A poly-Ni(II)–curcumin (curcumin: 1,7-bis[4-hydroxy-3-methoxyphenyl]-1,6-heptadiene-3,5-dione) composite film modified electrode was prepared for amperometric determination of β-nicotinamide adenine dinucleotide (NADH). Scanning electron microscopy and Energy Dispersive X-ray spectroscopy were used to examine the morphology and interface property of the Ni(II)–curcumin film. Under optimum conditions, the amperometric detection of NADH provided a wide linear detection range (0.3–300 μM), high sensitivity (230.02 μA mM−1 cm−2) and low limit of detection (LOD = 0.18 μM, S/N = 3). Moreover, the modified electrode showed good stability and reproducibility, indicating that it can improve the analysis signal of NADH in a very convenient way.

Improved bioavailability of curcumin in liposomes prepared using a pH-driven, organic solvent-free, easily scalable process

Ce Cheng,Shengfeng Peng,Ziling Li,Liqiang Zou,Wei Liu,Chengmei Liu

The poor water solubility and bioavailability of curcumin can be improved by encapsulating it into liposomes. However, the existing encapsulation technologies, such as the thin film method and the ethanol injection method, are complex and require the use of organic solvents. In this study, an organic solvent-free and easily scalable encapsulation technique was studied by utilizing the pH-dependent solubility properties of curcumin. Phospholipid was dissolved in water to form liposomes. Curcumin was deprotonated and dissolved under alkaline conditions and then encapsulated into the liposomes after acidification. Morphological observation and X-ray diffraction analysis confirmed that curcumin liposomes had been successfully prepared. Curcumin liposomes prepared by the pH-driven method were stable during storage. During in vitro digestion, curcumin liposomes prepared by the pH-driven method showed similar bioaccessibility to those prepared by the thin film method and higher bioaccessibility than those prepared by the ethanol injection method. The pH-driven method, which is organic solvent-free and easily scalable for industrial production, is thus a promising method for the preparation of curcumin liposomes.

Enhanced Antioxidant and Antibacterial Properties of Polybutylene Adipate-Terephthalate/Curcumin Composite Films Using Surface-Modified Cellulose Nanocrystals

Hashimu Juma,Cunshi Zhao,Qingbo Wang,Yunfeng Guo,Xinyan Fan,Wuming Fan,Linlin Zhao,Jiayi Sun,Dong Wang,Yonggui Wang

Polybutylene adipate-terephthalate (PBAT) offers a convincing ecological alternative to the traditional fossil-based plastics due to its biodegradability and robust mechanical properties. The objective of this study is to develop PBAT-based bio-composite films through incorporating functionalized cellulose nanocrystals (CNC) and curcumin (CUR). In order to improve the interfacial compatibility with the PBAT matrix and co-doping with CUR, CNC was modified using dodecyl succinic anhydride (DxCNC). In this ternary bio-composite system, CUR functioned as a bio-based antioxidant and antimicrobial agent. The presence of CUR also provides excellent UV-shielding properties, whereas the DxCNC effectively enhances the controlled release of CUR. The synergistic effect between DxCNC and CUR in boosting antimicrobial properties, with the inhibition values forE. coliandS. aureusreached 1.82 log CFU/cm2and 2.12 log CFU/cm2, respectively. These findings indicate DxCNC/CUR/PBAT ternary composite films as a promising material for eco-friendly packaging products.

Efficient Delivery of Curcumin by Functional Solid Lipid Nanoparticles with Promoting Endosomal Escape and liver targeting properties

Mengyi Zhang,Ju Liang,Ying Liang,Xuening Li,Wenlan Wu

In the realm of intracellular drug delivery, overcoming the barrier of endosomal entrapment stands as a critical factor influencing the effectiveness of nanodrug delivery systems. This study focuses on the synthesis of an acid-sensitive fatty acid derivative called imidazole-stearic acid (IM-SA). Leveraging the proton sponge effect attributed to imidazole groups, IM-SA was anticipated to play a pivotal role in facilitating endosomal escape. Integrated into the lipid core of solid lipid nanoparticles (SLNs), IM-SA was paired with hyaluronic acid (HA) coating on the surface of SLNs loading with curcumin (CUR). The presence of IM-SA and HA endowed HA-IM-SLNs@CUR with dual functionalities, enabling the promotion of endosomal escape, and specifical targeting of liver cancer. HA-IM-SLNs@CUR exhibited a particle size of ~228   nm, with impressive encapsulation efficiencies (EE) of 87.5% ± 2.3% for CUR. Drugs exhibit significant pH sensitive release behavior. Cellular experiments showed that HA-IM-SLN@CUR exhibits enhanced drug delivery capability. The incorporation of IM-SA significantly improved the endosomal escape of HA-IM-SLN@CUR, facilitating accelerated intracellular drug release and increasing intracellular drug concentration, exhibiting excellent growth inhibitory effects on HepG2 cells. Animal experiments revealed a 3.4-fold increase in CUR uptake at the tumor site with HA-IM-SLNs@CUR over the free CUR, demonstrating remarkable tumor homing potential with the tumor growth inhibition rate of 97.2%. These findings indicated the significant promise of HA-IM-SLNs@CUR in the realm of cancer drug delivery.
