New Approach Methodologies (NAMs) reveal triphenyl phosphate (TPP) disrupts lipid metabolism in estuarine fish via PPARγ activation: A concentration-dependent transcriptomic and molecular docking study

Jinzhi Ren,Ying Peng,Chenyang Sun,Yining Zhang,Jianjun Li,Simin Xia,Caixia Gao,Yimeng Wang,Xiangping Nie

There is an urgent need for a more efficient and ethical approach to chemical toxicity assessment. The emergence of New Approach Methodologies (NAMs) based on fish embryos transcriptomics provides a promising alternative. Triphenyl phosphate (TPP) — a pervasive contaminant in estuaries — disrupts lipid homeostasis in mammals via PPARγ activation, yet its ecotoxicological impacts on estuarine fish remain unknown. Leveraging NAMs, this study integrates concentration-dependent transcriptomics (CDT), molecular docking, and phenotypic toxicity assays to unravel effects of TPP on embryonic development and lipid metabolism in the benthic fish Mugilogobius chulae . We found that TPP induced significant lethality (LC₅₀ = 1.86 × 10 6  ng/L) and teratogenicity (EC₅₀ = 5.27 × 10 5  ng/L), suppressing spontaneous movement, heart rate, and hatching. At concentrations of 5270 ng/L and 52,700 ng/L, TPP elevated triglycerides, total cholesterol, and hepatotoxicity markers (alanine aminotransferase / aspartate aminotransferase). CDT analysis identified fatty acid metabolism pathways as the most sensitive targets, with TPP upregulating PPARγ while downregulating CD36 and ACSL5 — indicating blocked fatty acid oxidation and explaining lipid accumulation. Crucially, co-exposure with PPARγ antagonist GW9662 reversed metabolic disorders, validating the mechanistic role of PPARγ. Furthermore, molecular docking confirmed that TPP binds PPARγ with agonist-like affinity. This work establishes a NAM-based framework for deciphering the toxicity of contaminants of emerging concern in estuarine ecosystems, offering novel tools for ecological risk assessment.

Microwave-assisted pyrolysis of fish-scale biochar by magnetic ionic liquid activation for the soild phase extraction of benzophenones ultraviolet filters from sludge-amended soil

Shiqian Gao,Min Pan,Jiarui Zhou,YiZhou Shen,Jianhui Wang,Xin He

The magnetic ionic liquid-modified fish-scale biochar (MIL/FSBC) was prepared via microwave-assisted pyrolysis (MWP) processes. The hydroxyapatite (HAP) were efficiently separated from the interior of collagen fibrils with magnetic ionic liquid [C 4 min]FeCl 4 . The synergistic activation of HAP liberation combined with MIL process significantly increased the MWP efficiency of fish scale biomass. The morphology and texture properties of MIL/FSBC magnetic nanoparticles (MNPs) were analyzed using SEM, TEM, XRD, XPS, VSM and BET/BJH. The MIL/FSBC MNPs were employed to extract and enrich the benzophenone (BP-1, BP-3 and BP-12) ultraviolet filters from sludge-amended soil sample using magnetic dispersive solid phase extraction (MDSPE) coupled with microwave assisted extraction (MAE) technique. The important parameters influencing the extraction efficiency of the target analytes were investigated, including the amount of adsorbent, pH, microwave extraction temperature and time, NaCl concentration and desorption conditions. The average relative recoveries of BPs exceeded 90.5% using 20.0 mg of MIL/FSBC for extraction over 20.0 min at 35 °C. The matrix effect ranges from −11.42% to 6.2%, and it is slight at low concentrations. The limits of detection (LODs) and limits of quantification (LOQs) for the calibration curve were in a range of 4.3–14.3 ng/kg and 14.9–47.1 ng/kg, respectively. The monitored stability (79.7–106.3%) in soil samples over 90 days confirmed the environmental persistence of BPs in the soil. The developed method was applied to determine BPs in sludge-amended soil and sediment samples, with acceptable recoveries (84.2–102.6%). The typical chemisorption with a heterogeneous multilayer adsorption mechanism was confirmed using the quasi-secondary kinetic model and the Freundlich adsorption isotherm model. The results confirmed that the proposed method was practicable for the analysis of ultraviolet filters pollutant in actual soil samples.

Insights into the balance of safety, flavor, and texture in fermented fish: A study based on full-length sequencing of fermented largemouth bass (Micropterus salmoides)

Xianhao Zhang,Bingrui Guo,Bing Dai,Shimiao Wang,Siqi Zhang,Sufang Zhang,Chaofan Ji,Beiwei Zhu,Xinping Lin

In this study, the effect of fermentation on the safety, flavor, and texture of largemouth bass (Micropterus salmoides ) was investigated. Compared to fresh fish, the proteins formed a network structure that enhanced bound water proportion, texture hardness, and water holding capacity, while it reduced cooking losses. TVB-N and TBARS levels increased gradually but stayed within safe limits. However, histamine accumulation surpassed the recommended limit at 56.84 ± 0.17 mg/100 g in late stages. A total of 67 volatile compounds were detected, with 13 identified as key flavor compounds (OAV > 1). 3rd-generation full-length 16S rRNA gene sequencing revealed a microbial community shift from high diversity to dominance by strains like Vibrio anguillarum , Vagococcus fessus , and Vagococcus coleopterorum . Among these, 10 species were highly correlated with flavor formation. This study reveals quality patterns in fermented largemouth bass, providing insights into achieving a balance among safety, flavor, and texture in fermented fish.

Inspired by human olfactory system: Deep-learning-assisted portable chemo-responsive dye-based odor sensor array for the rapid sensing of shrimp and fish freshness

Chengbin Jiang,Alan J.X. Guo,Yuwen Li,Yufan Wang,Jianfeng Sun,Zhizhou Chen,Xin Chen,Qianyun Ma

Colorimetric sensor arrays (CSAs) can detect the volatile organic compounds (VOCs) produced during food storage, enabling odor visualization for freshness monitoring. Herein, sixteen chemo-responsive dyes that undergo observable color changes to VOCs are absorbed on ammonium quaternized cellulose nanofibres (C-CNFs) to design a paper-based CSA. The strong ionic interactions between the dyes and C-CNFs immobilize the dyes onto the sensor without leakage and provide stable and consistent colorimetric change even at a high relative humidity (RH = 60 %). Owing to the three-dimensional porous structure of C-CNFs, the CSA demonstrates high sensitivity for detecting VOCs produced during shrimp and fish spoilage. The limits of detection for ammonia, dimethylamine, trimethylamine, cadaverine, and putrescine are 2, 21, 3, 1, and 1 ppm, respectively. By selecting dyes covering a wide range of VOCs, the color changes in the CSA can accurately differentiate fresh, less fresh, slightly spoilt, and spoilt shrimp and fish. These color changes are further digitized and used to train a convolutional neural network (CNN). The CNN method has high accuracy (99 %) for determining abovementioned freshness levels. The combination of CSA and CNN is a promising approach for fabricating a portable sensing platform for the advanced non-destructive determination of food freshness levels.

Effects of grilling process on the formation pattern of heterocyclic amines and advanced glycation end products and physicochemical quality of premade grilled grass carp fish blocks

Zheyuan Xu,Haixin Yin,Yanshun Xu,Wenshui Xia,Dawei Yu

Grilled grass carp skin contained only 23–39% of total HAs and 36–65% of AGEs found in flesh. • HAs correlated with Trp/GO, while AGEs were linked to Lys/MGO under grilling conditions. • Temperature-dependent differentiation of HAs and AGEs formation was found in grilled grass carp. This study found a temperature-dependent pattern in heterocyclic amines (HAs) and advanced glycation end-products (AGEs) formation, as well as the physicochemical quality attributes of pre-made grilled grass carp. Raising the center temperature from 50 °C to 80 °C increased total HAs by 20% and total AGEs (CML + CEL) by 107%. At grilling temperatures of 220–280 °C, HAs formation dominated, with tryptophan, glyoxal, and creatine/creatinine as key precursors. In contrast, AGE formation prevailed at 310 °C, primarily involving lysine and methylglyoxal. Notably, grilled fish skin contained only 23–39% of total HAs and 36–65% of AGEs found in flesh. Prolonged grilling increased hardness, chewiness, gumminess, and yellowness (b*) but reduced lightness (L*) and moisture content, the latter showing strong negative correlation with HAs ( r  = −0.89) and moderate correlation with AGEs ( r  = −0.51). These findings provide science-based guidance for optimizing grilling conditions to balance safety and quality.

Fabrication and characterization of waste fish scale-derived gelatin/sodium alginate/carvacrol loaded ZIF-8 nanoparticles composite films with sustained antibacterial activity for active food packaging

Yongshi Li,Peng Shan,Fuyou Yu,Hui Li,Lincai Peng

An environmental-friendly composite films containing waste fish scale-derived gelatin (FSG), sodium alginate (SA) and carvacrol loaded ZIF-8 (CV@ZIF-8) nanoparticles were designed and fabricated to develop active food packaging materials capable of sustained antibacterial activity. The microstructure and physicochemical properties of the FSG/SA/CV@ZIF-8 composite films were investigated. The incorporation of CV@ZIF-8 into FSG/SA matrix significantly enhanced the UV-light blocking and the elongation at break, improved water resistance and reduced water vapor permeability, and improved the thermal stability of composite film. The FSG/SA/CV@ZIF-8 film not only exhibited strong antioxidant activity with DPPH radical scavenging rate of 92.35 %, but also showed the satisfactory and long-acting antibacterial ability against E. coli and S. aureus due to slow release of CV from composite film. Strawberry preservation experiment revealed that FSG/SA/CV@ZIF-8 film decelerated the texture deterioration and retarded the growth of spoilage microorganism, resulting in the prolonged shelf-life of 8 days under ambient condition, indicating its promising application prospect in food preservation packaging.

Integrated one–dimensional CuO–nanowire arrays/Cu–foam nanoarchitecture for ultrahigh sensitive and non–enzymatic electrochemical determination of histamine levels in different–bacteria fermented mandarin fish

Xun Zhou,Yongxiang Wu,Yao Jiang,Chen Li,Longping Xu,Peng Cui,Xinsong She

Integrated one–dimensional CuO–nanowire arrays/Cu–foam (CuO–NWAs/Cu–foam) nanostructure, which was fabricated by oxidation and calcination, has been newly utilized as non–enzymatic electrocatalytic electrode for exploring histamine level. Under optimal condition of pH at 13 and potential at 0.55 V ( vs Ag/Ag/Cl), the sensitivity of CuO–NWAs/Cu–foam electrode towards non–enzymatic electrochemical histamine determination presented as high as 12.94 mA mM −1 cm −2 , linear range spanned between 0.5 and 1046 μM, a detection limit (S/N = 3) was about 44 nM. The unprecedented causes of ultrahigh sensitivity were physically contributed to enhancing active surface area and declining charge transfer resistance. More crucially, the outstanding selectivity, stability and reproducibility facilitated its practical capacity on evaluating histamine levels in different–bacteria fermented mandarin fish, which triggered the potential feasibility for commercializing non–enzymatic electrochemical determination of histamine with high sensitivity, reliable precision and low expenditure.

A multiple signal amplification photoelectrochemical biosensor based on biotin-avidin system for kanamycin sensing in fish and milk via synergism of g-C3N4 and Ru@SiO2

Jun Yao

Background The residues of kanamycin can accumulate in the human body for a long time and pose serious health risks, including hearing loss , kidney poisoning, and drug allergic reactions. Therefore, it is crucial to develop a rapid, highly sensitive, and low-cost method for detecting kanamycin residues in foods. However, the current methods have limitations such as low sensitivity, expensive instruments, and multiple steps, which make them impractical for use in resource-limited environments and emergencies. In this study, the creation of a multiple-signal amplification photoelectrochemical biosensor to address these aforementioned issues is discussed. Results Herein, we proposed a multiple signal amplification photoelectrochemical (PEC) biosensor based on carboxylated g-C 3 N 4 and avidin functionalized Ru@SiO 2 for the ultrasensitive detection of kanamycin. The carboxylated g-C 3 N 4 was a highly efficient photoactive substance for amplifying photoelectric signals and a substrate for aptamer immobilization. The DOS and PDOS of g-C 3 N 4 were studied by simulation, and the sensing mechanism of the probe at the molecular level was revealed. Meanwhile, using Ru@SiO 2 as a signal amplifying unit, through the cooperative work between Ru@SiO 2 and g-C 3 N 4 , the photoelectric signal could be double amplified to produce an excellent photocurrent response. Under optimized conditions, the photocurrent response of the PEC biosensor to kanamycin was obtained at concentrations from 0.1 nM to 1000 nM with a lower detection limit of 4.1052 × 10 −11  mol L −1 . This protocol demonstrates high sensitivity, brilliant specific recognition ability, excellent reproducibility, and acceptable stability. Significance The first combination of g-C 3 N 4 and avidin-Ru@SiO 2 as photocurrent materials greatly enhanced the sensitivity of the PEC biosensors. Moreover, the specificity and sensitivity of the PEC biosensor were further improved through the specific interaction between kanamycin and aptamer. The photoelectric conversion mechanism based on g-C 3 N 4 and two pathways for enhancing the photocurrent by Ru(byp) 3 2+ were proposed. Through simulations of the DOS and PDOS of g-C 3 N 4 , the sensing mechanism of the probe at the molecular level was revealed. Under the optimum conditions, the PEC biosensor exhibited a wide linear concentration range and a low detection limit.

Preparation and properties of ferulic acid-sugar beet pulp pectin ester and its application as a physical and antioxidative stabilizer in a fish oil-water emulsion

Haiming Chen,Hui Niu,Hailing Zhang,Yonghuan Yun,Wenxue Chen,Qiuping Zhong,Weijun Chen,Xiong Fu

A ferulic acid-sugar beet pulp pectin complex (FA-SBPP) was prepared using an immobilized enzyme (Novozym 435®) as the catalyst at 60 °C in a vacuum-controlled system. The structure of FA-SBPP was characterized by FT-IR and NMR ( 1 H and 13 C). In addition, the antioxidant activity of FA-SBPP was evaluated according to the DPPH free radical scavenging ability and ORAC values. Moreover, the physical and oxidation stability of the emulsion was evaluated by particle size distribution, cream index (CI), peroxide value (POV), and 2-thiobarbituric acid reactive substance (TBARS) formation. The results showed that esterification between FA and SBPP was confirmed, and the reaction mainly occurred at the C-2, C-3, and C-6 positions. When the concentration was 1.0 mg/mL, the DPPH scavenging activity and total antioxidant activity of FA-SBPP-3 were 80.03% and 355.72 μmol TE/g, respectively. Compared with SBPP, FA-SBPP-stabilized emulsions exhibited significant smaller droplet sizes and lower CIs, POVs and TBARS amounts. Thus, the introduction of FA changed not only the chemical reactivity but also the polarity of SBPP, thereby improving its antioxidant ability and affinity for the oil-water interface. Thus, we provide a multifunctional stabilizer that can reduce the dose of antioxidants or even replace them in oil-in-water emulsions.

A novel reductive graphene oxide-based magnetic molecularly imprinted poly(ethylene-co-vinyl alcohol) polymers for the enrichment and determination of polychlorinated biphenyls in fish samples

Saichai Lin,Ning Gan,Jiabin Zhang,Xidong Chen,Yuting Cao,Tianhua Li

The novel reductive graphene oxide-based magnetic molecularly imprinted poly(ethylene- co -vinyl alcohol) polymers (rGO@m-MIPs) were successfully synthesized as adsorbents for six kinds of polychlorinated biphenyls (PCBs) in fish samples. rGO@m-MIPs was prepared by surface molecular imprinting technique. Besides, Fe 3 O 4 nanoparticles (NPs) were employed as magnetic supporters, and rGO@Fe 3 O 4 was in situ synthesis. Different from functional monomer and cross-linker in traditional molecularly imprinted polymer, here, 3,4-dichlorobenzidine was employed as dummy molecular and poly(ethylene- co -vinyl alcohol) was adopted as the imprinted polymers. After morphology and inner structure of the magnetic adsorbent were characterized, the adsorbent was employed for disperse solid phase extraction toward PCBs and exhibited great selectivity and high adsorption efficiency. This material was verified by determination of PCBs in fish samples combined with gas chromatography-mass spectrometry (GC-MS) method. According to the detection, the low detection limits (LODs) of PCBs were 0.0035–0.0070 µg l −1 and spiked recoveries ranged between 79.90 and 94.23%. The prepared adsorbent can be renewable for at least 16 times and expected to be a new material for the enrichment and determination of PCBs from contaminated fish samples. Effects of Dietary Fish Oil Levels on Growth Performance, Lipid Metabolism, Hepatic Health, Nonspecific Immune Response, and Intestinal Microbial Community of Juvenile Amur Grayling (Thymallus grubii)

Shaoxia Lu,Chang’an Wang,Yang Liu,Bing Liu,Ying Zhang,Honghe Shi,Gefeng Xu,Shicheng Han,Hongbai Liu

This trial was conducted to assess the effects of different levels of dietary fish oil on growth performance, hepatic health, nonspecific immune responses, and intestinal microbial community of Amur grayling ( Thymallus grubii ). Five isonitrogenous diets containing 60 (6FO), 90 (9FO), 120 (12FO), 150 (15FO), and 180 g/kg (18FO) fish oil were fed to triplicate groups of 60 fish per tank for 8 weeks, respectively. The results revealed that specific growth rate (SGR) and weight gain (WG) of fish in the 15FO group were significantly greater than those in the 6FO group ( p < 0.05). Somatic indices and whole-body lipid levels were positively correlated with increases in dietary fish oil levels. Trypsin and lipase activities in 15FO and 18FO groups were significantly higher than those in the 6FO and 9FO groups ( p < 0.05). The activities of intestinal catalase (CAT) and liver superoxide dismutase (SOD), CAT, lysozyme (LZM), alkaline phosphatase (AKP), and acid phosphatase (ACP) improved significantly as the dietary lipid content increased to 185.3 g/kg and decreased thereafter ( p < 0.05). The lipid metabolism-related genes peroxisome proliferator-activated receptor gamma (PPAR γ ) and carnitine palmitoyltransferase 1A (CPT1A) were significantly downregulated and upregulated ( p < 0.05), respectively, in the 15FO group. Immune-related genes in the liver and intestine, such as interleukin (IL-8), were significantly upregulated in the 15FO group ( p < 0.05). The liver sections from 18FO group presented more numerous and larger lipid vacuoles. Both low- (6FO) and high-lipid (18FO) diets reduced the relative abundance of intestinal Lactococcus . The relative abundances of intestinal Staphylococcus and Bacillus (mainly Bacillus anthracis ) increased in the low-lipid diet group and that of Pedobacter increased in the high-lipid diet group. Second-order polynomial analysis of WG and the feed conversion ratio (FCR) for varying levels of dietary lipid revealed that a range of 194.76–198.90 g/kg dietary lipid was optimal for the growth and health of Amur grayling.

Developing a novel pyridine-based ionic porous organic polymer with functional group for highly efficient extraction of phenolic endocrine disruptors in natural water and tilapia fish

Meng Li,Zhi Li,Qianqian Wang,Yihang He,Yixuanfei Gao,Chun Wang,Zhi Wang,Qiuhua Wu

A pyridine-based ionic porous organic polymer bearing hydroxyl groups (OH-iPOP) was developed for the first time by quaternization and crosslinking of 4-pyridinemethanol with 4,4′-bis(chloromethyl)biphenyl. OH-iPOP exhibited strong adsorption capacity for endocrine disruptors (EDs) including bisphenol A, bisphenol F, p-tert-butylphenol and bisphenol B (82.59–183.9 mg g −1 ), much higher than that of commercial adsorbents. The adsorption mechanism of OH-iPOP to EDs involved π-π ⁎ interaction, hydrogen bond and electrostatic interaction. Using OH-iPOP as solid phase extraction adsorbent combined with high-performance liquid chromatography, a reliable method for the sensitive detection of trace EDs in natural water and tilapia fish was established. The detection limits (S/N = 3) were 0.01–0.04 ng mL −1 and 1.00–3.80 ng g −1 for natural water and tilapia fish, respectively. The method recoveries ranged from 81.5 % to 116 %. This work not only develops an efficient adsorbent for the enrichment of EDs, but also provides a new strategy for the synthesis of novel cationic POPs.
