Recently, stimuli-responsive gene carriers have been widely studied to overcome the extra- and intracellular barriers in cancer treatment. In this study, we modified polydopamine nanoparticles with low-molecular weight polyethylenimine (PEI 1.8k ) and polyethylene glycol-phenylboronic acid (PEG-PBA) to prepare pH-responsive gene carrier PDANP-PEI-rPEG. PBA and polydopamine could form pH-responsive boronate ester bonds. Non-responsive PDANP-PEI-nPEG and non-PEGylated PDANP-PEI were also studied as control. Both PDANP-PEI-rPEG/DNA and PDANP-PEI-nPEG/DNA complexes remained stable in the pH environment of blood circulation or extracellular delivery (pH 7.4) owing to the PEG modification. And after being internalized into endosomes, the boronate ester bonds could be cleaved. The pH responsive ability of PDANP-PEI-rPEG might facilitate complexes dissociation and gene release inside cells. The transfection level of PDANP-PEI-rPEG/DNA complexes was about 100 times higher than that of PDANP-PEI-nPEG/DNA complexes with the same mass ratios. Moreover, after NIR light irradiation at the power density of 2.6 W/cm 2 for 20 min, the good photothermal conversion ability of PDANP resulted in quick endosomal escape. The transfection level of PDANP-PEI-rPEG/DNA complexes doubled, even higher than that of lipofectamine 2000/DNA complexes. This was also confirmed by Bafilomycin A1 inhibition test and CLSM observation. In response to the acidic pH within cancer cells and the NIR light irradiation, the PDANP-PEI-rPEG carrier could overcome multiple obstacles in gene delivery, which was promising for further application in gene therapy.

Amphiphilic polymer carriers (PEG–St– R ) were prepared from cassava starch and their pH response was investigated. First, hydrophobic tapioca starch polymer (St– R ) was prepared with octyl acyl as the hydrophobic group. The hydrophilic group polyethylene glycol (mPEG) was then introduced into the polymer by esterification to produce amphiphilic tapioca starch polymer (PEG–St– R ). Its self-assembly behavior was characterized using fluorescent probes. The morphology of PEG–St– R was investigated by transmission electron microscopy (TEM). Loading of the anti-cancer drug curcumin was used to assess the delivery and slow-release performance of the amphiphilic tapioca starch polymer. Cumulative drug release was explored at various pH conditions, with the greatest release from drug-loaded micelles being observed under acidic conditions and stable in a neutral environment. These results provide a theoretical basis for the preparation of pH-responsive nanomicelle carriers, and a platform for the preparation of novel amphiphilic starch-based polymers.

A fast, simple, green and low-cost synthesis method in aqueous solution was successfully established for the preparation of l -tyrosine protected Cu nanoclusters (Cu NCs). Subsequently, the as-prepared Cu NCs was characterized by UV–vis absorption spectroscopy, fluorescence spectroscopy, transmission electron microscopy (TEM) and X-ray photoelectron spectroscopy (XPS). It exhibited good water solubility, excellent photostability and high stability toward high concentration of sodium chloride. The Cu NCs revealed maximum excitation and emission peaks at 430 and 477 nm, respectively. Importantly, the Cu NCs showed a rapid response, a liner relation over the pH ranges from 5.02 to 11.92 and could be repeated for five consecutive cycles without fluorescence decay. Further, the as-prepared Cu NCs was applied for the determination of pH value in water samples. Simultaneously, all above features showed that l -tyrosine protected Cu NCs had great potential for sensing of pH in biological field.

Stimuli-responsive surfaces have the potentials to solve the trade-off between biocompatibility and antibacterial performance by switching specific function on-demand, which makes them a promising option for developing biocompatible antibacterial surfaces. In this study, a pH-responsive antibacterial surface was created by grafting polycationic brushes and forming an acid-sensitive zwitterionic structure through a spontaneous amino-yne click reaction. Under physiological conditions, the zwitterionic structure provided the surface with good biocompatibility, as confirmed by lower hemolysis and cytotoxicity, as well as low adhesion and activation of platelets. When bacteria begin to colonize on the biomaterial surface, the decrease in local pH can break the acid-sensitive enamine bond and expose the polycations, activating the bactericidal activity. The good and on demand bactericidal activity was verified by the stimulated and authentic bacterial infection conditions. This study highlights the potential of stimuli-responsive zwitterionic surfaces as a valuable option for infection-resistant medical devices .

Hydrogels have attracted widespread attention in anticounterfeiting due to their unique physical/chemical properties and designability. However, hydrogels’ poor mechanical properties and sluggish response to chemical stimuli pose challenges for their wide application. A fluorescent tough organohydrogel capable of freeform writing of information is reported in this work. By incorporation of the fluorescent monomer 7-methylacryloxy-4-methylcoumarin into the polyacrylamide network in a covalently cross-linked manner while intertwining with the carboxymethyl cellulose sodium network, a fluorescent tough organohydrogel with a dual-network structure is prepared. The organohydrogel shows acid–base-mediated adjustable fluorescence through the transformation of fluorescent monomers. Ion printing and electrical stimulation design achieved free information storage and encryption. In addition, the prepared organohydrogel has good antifreezing properties and can be encrypted and decrypted at subzero temperatures. The encrypted information in the organohydrogel can be read only after UV-light irradiation. These patterned fluorescent organohydrogels should find applications in protected message displays for improved information security.

Chemodynamic therapy (CDT) and photothermal therapy (PTT) have been widely studied as treatment alternatives to anbiotics to avoid the occurrence of bacterial resistance because of their negligible bacterial resistance and high antibacterial efficiencies. Herein, we developed an infection microenvironment-responsive nanogel (STF) to successfully achieve a PTT synergized CDT for bacterial infection. Through a one-step/pot method, STF nanotherapeutics consisting of Fe3+/sodium alginate (SA) based nanogels and Fe3+/tannic acid (TA) based metal–phenolic networks (MPNs) were conveniently prepared by the coordination-triggered simultaneous self-assembly of SA, TA, and Fe3+ ions. Afterward, their photothermal and catalytic performances were intensively investigated. Moreover, the sterilization effect of the STF nanogel was evaluated through in vitro and in vivo bactericidal assays. The STF nanogel exhibited a photothermal enhanced peroxidase-like activity because of the good photothermal effect of MPNs. In addition, its Fenton activity was further accelerated by an extra amount of Fe2+ derived from the degradation of Fe3+-chelating agents (SA) and reduction of TA. Together, the STF nanogel possessed a photothermal/Fe2+ dual-augmented catalytic property. In vitro and in vivo antibacterial assays proved its satisfactory bactericidal efficiency owing to PTT/Fe2+ dual-enhanced CDT. Our research findings suggested that the developed intelligent bactericidal nanogel has great potential to be used as an antibiotic alternative to avoid bacterial resistance in future clinical treatments.

Keratin is a good candidate for drug carrier due to its good biocompatibility, low immunogenicity, redox responsiveness, and abundant renewable sources. Herein, doxorubicin (DOX) was first conjugated with keratin through a pH-sensitive hydrazone linkage, and then prepared into particulate drug carrier via desolvation method. The size, morphology, and surface potential of keratin-DOX nanoparticles (KDNPs) were characterized by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The drug release results showed that KDNPs performed an excellent pH-sensitive behavior under acidic tumor microenvironment. Cytotoxicity assay by MTT confirmed that KDNPs exhibited the enhanced cytotoxicity against A549 cells. Furthermore, KDNPs had higher therapeutic efficiency in vivo than free DOX . Hemolysis assay indicated that KDNPs was blood compatible. All the results identified that KDNPs are well suited as an ideal drug carrier.

In cell culture research and biotechnology, precise pH monitoring is crucial for maintaining cellular health and ensuring reliable experimental outcomes. Traditional pH measurement methods, such as glass electrodes and chemical indicators, are often limited by issues such as fragility, calibration requirements, and potential cytotoxicity. This study presents a novel pH sensor based on bacteriorhodopsin (bR), a light-sensitive protein that undergoes conformational changes in response to pH fluctuations, generating a measurable photoelectric signal. The integrated bR-based electrochemical electrode in a flexible pH biosensor is demonstrated, with measurements spanning the physiological pH range of 6.0–8.5. The sensor shows a high correlation (R2= 0.977) between photo-generated current signals and pH, indicating robust performance for real-time, non-invasive pH monitoring. The biocompatibility and non-invasive nature of this sensor make it particularly suitable for continuous monitoring in cell culture environments. The sensor’s practical application is validated by its integration into cell well plates for tracking the pH changes during cell growth, providing valuable insights into metabolic processes and growth conditions. In the future, efforts will focus on enhancing sensor sensitivity, stability, and integration with multi-parameter monitoring systems for more comprehensive cell culture analysis.

pH sensor is an important and practical device with a wide application in environmental protection field and biomedical industries. An efficient way to enhance the practicability of intelligent polymer composed pH sensor is to subtilize the three-dimensional microstructure of the materials, adding measurable features to visualize the output signal. In this work, C. rubi wing scales were combined with pH-responsive smart polymer polymethylacrylic acid (PMAA) through polymerization to achieve a colour-tunable pH sensor with nature gyroid structure. Morphology and reflection characteristics of the novel composites, named G-PMAA, are carefully investigated and compared with the original biotemplate, C. rubi wing scales. The most remarkable property of G-PMAA is a single-value corresponding relationship between pH value and the reflection peak wavelength (λ max ), with a colour distinction degree of 18 nm/pH, ensuring the accuracy and authenticity of the output. The pH sensor reported here is totally reversible, which is able to show the same results after several detection circles. Besides, G-PMAA is proved to be not influenced by the detection angle, which makes it a promising pH sensor with superb sensitivity, stability, and angle-independence.

A novel amphiphilic copolymer consisting of tetraphenylethylene-oxazolidine (TPE-OX) as a pH-sensitive chromophore and poly(ethylene glycol) methyl ether methacrylate (PEGMA) as a water-soluble chain has been well designed and synthesized. A self-assembled polymeric micelle based on the copolymer exhibited clear dual-emission switching between cyan and red with a decrease of pH value in Tris-HCl buffer solution due to the extended conjugation and emerging intramolecular charge transfer effect when opening the spiro-ring of OX moiety. Fluorescence imaging of HepG2 cells stained by the polymeric micelle shows switched luminescence from cyan to red with high selectivity and contrast, indicating that the polymeric micelle was an effective probe for intracellular pH detection. Additionally, the red emission of the polymeric micelle in lysosomes can reversibly switch back to the original cyan emission in response to the two lysosomal activity inhibitors chloroquine and bafilomycin, which demonstrates that the polymeric micelle has potential applications in detecting the activity of lysosomal and further autophagy in cancer cells.

Smart protein-mimetic materials have received increasing interest for many applications. Here, we reported a facile and efficient approach to prepare a class of dual-responsive copoly( l -glutamate)s by ring-opening polymerization (ROP) and post-modification strategy. Both the oligo(ethylene glycol) (OEG) and glutamic acid residues were incorporated to offer thermal- and pH-responsive property simultaneously. We present a systematic study of the influence of both temperature and pH on the solution properties of the copolypeptides. The copolypeptides show entirely reversible pH-dependent thermal-responsive behaviors with tunable phase transition temperatures. It is demonstrated that the sequence of the polymer units can also influence the solution behavior. The α-helix conformation appears to be the dominant secondary conformation, which shows the pH dependence, but generally irrespective of the temperature. We further verify that the reduced circular dichroism (CD) signal upon heating above the clouding point (CP) is entirely due to the aggregation of the polymer chains. All these features make the obtained polypeptides potential candidates for next generation of smart protein-mimetic materials.

In order to increase the loading efficiency of drug carriers, here we demonstrated a microfluidic method to fabricate an asymmetric vesicle, which contains trichloro(1H,1H,2H,2H-perfluoroocty-l)silane (TPS) inner leaflet and lipid outer leaflet. The asymmetric vesicle was characterised by fluorescence microscopy and Fourier transform infra-red spectrum. In vitro cytotoxicity of the vesicles carrying 5-fluorouacil (5-FU) has also been studied.
