The sensitive detection of foodborne pathogenic and rapid antibiotic susceptibility testing (AST) is of great significance. This paper reports the enzyme-triggered in situ synthesis of yellow emitting silicon nanoparticles (SiNPs) and the detection of Escherichia coli ( E. coli ) O157:H7 in food samples and the rapid AST. The rapid counting of E. coli O157:H7 has been achieved through direct visual observation, equipment detection, and smartphone digitalization. A simple detection platform based on smartphone senses and cotton swabs has been established. Meanwhile, rapid AST based on enzyme-catalyzed SiNPs can intuitively obtain colorimetric samples. This paper established a system for bacterial enzyme-triggered in situ synthesis of SiNPs, with high responsiveness, luminescence ratio, and specificity. The detection limit for E. coli O157:H7 can reach 100 CFU/mL during 5 h, and the recovery efficiency ranges from 90.14% to 110.16%, which makes it a promising strategy for the rapid detection of E. coli O157:H7 and AST.

Hypertensive heart failure pathogenesis involves angiotensin II (Ang II)-mediated mechanisms through both hemodynamic overload and direct cellular signaling pathways. This study investigates the therapeutic potential of Portulaca oleracea L. (POL) extract and its active constituent Oleracein E (OE) in attenuating Ang II-induced pathological cardiac remodeling and subsequent heart failure progression. An experimental model of hypertensive heart failure was established in C57BL/6 mice through continuous subcutaneous infusion of Ang II for 4 weeks, with Oleracein E (OE) intervention administered during the final 2 weeks of the protocol. To elucidate the molecular mechanisms underlying OE's cardioprotective effects, we employed an integrated approach combining RNA sequencing analysis, molecular docking simulations, and target validation through drug affinity response target stability (DARTS) and cellular thermal shift assay (CETSA) techniques. The experimental results demonstrated that both POL extract and its active component OE exerted significant cardioprotective effects against Ang II-induced cardiac dysfunction in murine models, primarily through attenuation of pathological cardiac hypertrophy and suppression of inflammatory responses. Transcriptomic profiling via RNA sequencing identified the MAPK signaling pathway as a critical mediator of these protective effects. Subsequent transcription factor analysis revealed STAT2 as a key regulatory component in this pathway. Importantly, OE treatment effectively mitigated inflammatory responses in both in vivo cardiac tissues and in vitro cultured cardiomyocytes by specifically inhibiting the Ang II-activated MAPKs/STAT2 signaling cascade. Genetic ablation experiments further confirmed the essential role of this pathway, as the anti-inflammatory efficacy of OE was completely abolished in cardiomyocytes with MAPKs or STAT2 deficiency. Molecular interaction studies employing in silico docking simulations combined with experimental validation through DARTS and CETSA techniques provided direct evidence of physical binding between OE and the STAT2 protein. Our findings demonstrate that Oleracein E (OE) confers cardioprotection against Ang II-induced myocardial injury through a novel mechanism involving STAT2 targeting and subsequent suppression of MAPK signaling-mediated inflammatory cascades. These results not only elucidate a previously unrecognized pharmacological pathway but also suggest that OE and its derivatives hold significant therapeutic promise for the clinical management of hypertensive heart failure, potentially offering a targeted approach to modulate pathological cardiac remodeling.

Conventional extraction techniques for fat-soluble nutrient analysis in foods may present certain limitations, including variable or limited efficiency, high solvent consumption, lengthy procedures, and potential environmental and health concerns. This study evaluates deep eutectic solvents (DESs) as a green and efficient alternative for extracting vitamin E and carotenoids from vegetables and fruits. Conductor-like screening model for real solvents (COSMO-RS) simulation and experimental screening identified the menthol-thymol (1:2) DES as optimal due to the balanced distribution of polar and nonpolar sites within its components. A novel dispersive liquid-liquid microextraction method based on this DES (DES-DLLME) was developed and integrated with HPLC for the simultaneous quantitation of vitamin E and carotenoids. The optimized method demonstrated excellent analytical performance with low detection limits (0.01-0.18 µg/mL), satisfactory recovery rates (80.04-111.95%), and high reproducibility (relative standard deviation, RSD < 16.83%). When applied to a range of Chinese food samples, the obtained nutrient profiles were largely consistent with USDA reference data, with observed variations likely due to factors such as geographic origin, cultivation practices, and post-harvest processing. This DES-based approach offers an efficient, reproducible, and environmentally friendly method for determining fat-soluble vitamins and carotenoids in complex food matrices, offering significant value for food quality assessment and nutritional research.

This study assessed the efficacy of dietary vitamin E in ameliorating growth and liver function in Jinhu grouper compromised by the inclusion of oxidized fish oil (OFO; peroxide value: 121.68 mmol/kg) in the diet, compared with a fresh fish oil control (FFO; peroxide value: 1.25 mmol/kg). The eight-week feeding trial utilized five experimental diets that were isonitrogenous and isolipidic: FFO, OFO (0 mg/kg VE), or OFO supplemented with 200 (VE1), 400 (VE2), or 600 (VE3) mg/kg vitamin E. Fish receiving oxidized fish oil showed slower growth, indicated by reduced weight gain rate (WGR) and increased feed conversion ratio (FCR). Intake of oxidized lipids altered lipid metabolism, lowering serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein cholesterol (HDL-C) but elevating low-density lipoprotein cholesterol (LDL-C) and malondialdehyde (MDA) levels. Hepatic antioxidant capacity was compromised, as revealed by the concurrent suppression in both the activities and gene expression levels of key antioxidant enzymes (SOD, CAT, and GPx). Concurrently, oxidized fish oil induced liver inflammation, significantly upregulating pro-inflammatory cytokine genes (notably il-1β , il-6 , il-8 , tnf-α ), and caused histopathological damage including hepatocyte nuclear displacement and lipid droplet accumulation. Vitamin E supplementation ameliorated these effects. The VE2 group (400 mg/kg) maximally restored growth performanc, reduced serum MDA and LDL-C levels, enhanced hepatic antioxidant enzyme activities and gene expression, suppressed inflammatory cytokine expression, and attenuated histopathological lesions. In conclusion, dietary vitamin E supplementation at 400 mg/kg effectively counteracts growth impairment and hepatotoxicity induced by oxidized fish oil in Jinhu grouper.

A simple and valid method for rapid screening of cathepsin B inhibitors from traditional Chinese medicines (TCMs) was established by the combination of immobilized enzyme microreactor (IMER) and capillary electrophoresis. Cathepsin B was immobilized on the inner surface of the capillary by glutaraldehyde method. The separation of substrate and product could be finished by baseline within 3 min. The activity of the immobilized cathepsin B remained approximately 90% after 50 runs. The quantification and statistical analysis of the product peak area was used to evaluate the catalytic activity of cathepsin B. The value of Michaelis-Menten constant of cathepsin B was 0.85 mM. The half-maximal inhibitory concentration (IC 50 ) of L -trans-Epoxysuccinyl-leucylamido(4-guanidino)butane (E-64) was measured as 36.08 nM, which indicated that the cathepsin B reactor was successfully developed and was feasible for inhibitorscreening. The raised method was then applied to discover the inhibitory potential of 17 standard compounds from traditional Chinese medicines. Five natural products, including kaempferol, rutaecarpine, evodiamine, theophylline, lycobetaine showed potential inhibition for cathepsin B. Additionally, molecular docking study was investigated for supporting the interaction between enzyme and inhibitors.

It was the first report that the concentrated hydrolyzates from the enzymatic hydrolysis of dilute NaOH (3 wt%)-soaking rice straw at 30 °C was used to form [Bmim]PF 6 -hydrolyzate (50:50, v/v) media for bioconverting ethyl 4-chloro-3-oxobutanoate (COBE) into ethyl ( R )-4-chloro-3-hydroxybutanoate [( R )-CHBE] (>99% e.e. ) with recombinant E. coli CCZU-A13. Compared with pure glucose, the hydrolyzates could promote both initial reaction rate and the intracellular NADH content. Furthermore, emulsifier OP-10 (20 mM) was employed to improve the reductase activity. Moreover, Hp-β-cyclodextrin (0.01 mol Hp-β-cyclodextrin/mol COBE) was also added into this bioreaction system for enhancing the biosynthesis of ( R )-CHBE from COBE by E. coli CCZU-A13 whole-cells. The yield of ( R )-CHBE (>99% e.e. ) from 800 mM COBE was obtained at 100% in the [Bmim]PF 6 -hydrolyzate (50:50, v/v) media by supplementation of OP-10 (20 mM) and Hp-β-CD (8 mM). In conclusion, an effective strategy for the biosynthesis of ( R )-CHBE was successfully demonstrated.

Magnetic harvesting of microalgae depends on the interface interactions between microalgal cells and flocculants, and thus it is important to find a reasonable evaluation method understand these interface interactions. In this effort, we discussed the formation mechanism and pH regulation of microalgae magnetic harvesting via magnetic chitosan (Fe 3 O 4 @SiO 2 -CS) synthesized by a micro-emulsion method. The flocculation performances and mechanisms during the harvesting of microalgae by the novel flocculant were evaluated. The synthesis process and structure were determined by XRD, FT-IR, SEM and XPS, and the results indicated that Fe 3 O 4 @SiO 2 -CS was successfully prepared. The collection efficiency of 96.52 % was achieved for Chlorella vulgaris within 3 min using Fe 3 O 4 @SiO 2 -CS with the Fe 3 O 4 @SiO 2 and CS mass ratio of 1:3 under pH 1.0. Moreover, this study introduced the two-dimension fractal dimension and extended Derjaguin-Landau-Verwey-Overbeek (E-DLVO) model to evaluate the harvesting performances and mechanisms of Fe 3 O 4 @SiO 2 -CS. Experimental results showed that a strong electrostatic interaction was formed under acidic conditions between Fe 3 O 4 @SiO 2 -CS and microalgae cells with D 2 = 1.82 (fractal dimension value). The hydrophobic tryptophan-like amino acid was observed in the slime-extracellular polymeric substances (S-EPS) secreted by microalgae. S-EPS was found to be a significant substance to promote harvesting performance when the system was neutral or alkaline. This work provides deeper insights into microalgae harvesting by Fe 3 O 4 @SiO 2 -CS as well as reasonable mechanisms to illustrate the interaction of Fe 3 O 4 @SiO 2 -CS and microalgae under different pH values.

Currently, very limited therapeutic approaches are available for the drug treatment of atherosclerosis(AS). H 2 S-donor is becoming a common trend in much life-threatening research. Several studies have documented that H 2 S-lyase is predominantly present in endothelial cells. N-Acetylneuraminic acid (SA), natural carbohydrate, binds specifically to the E -selectin receptor of endothelial cells. Meanwhile, recent studies related to Chondroitin sulfate have excellent target binding ability with CD44 receptor. We conjecture that the N-Acetylneuraminic acid and Chondroitin sulfate modified nanomicelles not only enhances the accumulation of the drug but also cleaves the H 2 S donor in the lesion, thus one stone two birds. Given these findings, we synthesized two kinds of nanoparticles, Carrier I (SCCF) and Carrier II (SCTM), for atherosclerosis to validate our guesses. Initially, S-allyl-L-cysteine and 4-methoxyphenylthiourea were used as H 2 S donors for SCCF and SCTM, respectively. After the introduction of ROS-sensitive groups. Then, micelles with N-Acetylneuraminic acid and Chondroitin sulfate were prepared to load rapamycin(RAP). Further, in atherosclerosis Oil Red O staining (ORO) results confirmed remarkable treatment effect with [email protected] and [email protected] Thus, we conclude that the effect of dual-targeting nanomicelles with ROS-sensitive H 2 S donor based on N-Acetylneuraminic acid and Chondroitin sulfate will have a better role in atherosclerosis.

This study focused on characterizing the volatile profiles and contributing compounds in pan-fried steaks from different Chinese yellow cattle breeds. The volatile organic compounds (VOCs) of six Chinese yellow cattle breeds (bohai, jiaxian, yiling, wenshan, xinjiang, and pingliang) were analyzed by GC-Q-Orbitrap spectrometry and electronic nose (E-nose). Multivariate statistical analysis was performed to identify the differences in VOCs profiles among breeds. The relationship between odor-active volatiles and sensory evaluation was analyzed by partial least square regression (PLSR) to identify contributing volatiles in pan-fried steaks of Chinese yellow cattle. The results showed that samples were divided into two groups, and 18 VOCs were selected as potential markers for the differentiation of the two groups by GC-Q-Orbitrap combined multivariate statistical analysis. YL and WS were in one group comprising mainly aliphatic compounds, while the rest were in the other group with more cyclic compounds. Steaks from different breeds were better differentiated by GC-Q-Orbitrap in combination with chemometrics than by E-nose. Six highly predictive compounds were selected, including 3-methyl-butanal, benzeneacetaldehyde, 2-ethyl-6-methyl-pyrazine, 2-acetylpyrrole, 2-acetylthiazole, and 2-acetyl-2-thiazoline. Sensory recombination difference and preference testing revealed that the addition of highly predictive compounds induced a perceptible difference to panelists. This study provides valuable data to characterize and discriminate the flavor profiles in pan-fried steaks of Chinese yellow cattle. Keywords:Chinese yellow cattle;volatile organic compounds;GC-Q-Orbitrap;E-nose;sensory evaluation;multivariate statistical analysis

We report on a method for ultrasensitive detection and quantification of the pathogen Escherichia coli ( E. coli ), type O157:H7. It is using a tortuous-shaped giant magnetoimpedance (GMI) sensor in combination with an open-surface microfluidic system coated with a gold film for performing the sandwich immunobinding on its surface. Streptavidin-coated super magnetic Dynabeads were loaded with biotinylated polyclonal antibody to capture E. coli O157:H7. The E. coli -loaded Dynabeads are then injected into the microfluidics system where it comes into contact with the surface of gold nanofilm carrying the monoclonal antibody to form the immunocomplex. As a result, the GMI ratio is strongly reduced at high frequencies if E. coli O157:H7 is present. The sensor has a linear response in the 50 to 500 cfu·mL −1 concentration range, and the detection limit is 50 cfu·mL −1 at a working frequency of 2.2 MHz. In our perception, this method provides a valuable tool for developing GMI-based microfluidic sensors systems for ultrasensitive and quantitative analysis of pathogenic bacteria. The method may also be extended to other sensing applications by employing respective immunoreagents.

Insect sex pheromones (SPs) are central to mate-finding behaviour, and play an essential role in the survival and reproduction of organisms. Understanding the roles, biosynthetic pathways and evolution of insect chemical communication systems has been an exciting challenge for biologists. Compared with Lepidoptera, little is known about the mechanisms underlying pheromone biosynthesis in Hemiptera. In this study, we isolated and characterized two new desaturase-like genes, termed Asutdes1 and Asutdes2 , from Adelphocoris suturalis , an important agricultural pest in China. Although the two genes encode an identical protein, Southern blot analysis revealed that they are duplicated genes. The Asutdes2 transcript is more abundant than Asutdes1 in the tissues tested, in particular the metathoracic scent gland and fat body. Silencing Asutdes expression in females by injecting double-stranded RNA ( dsAsutdes ) against a portion of the coding sequence shared by the two genes enhanced the production of (E)-4-oxo-2-hexenal, a component of the A. suturalis SP blend, and dramatically suppressed the sexual attractiveness of A. suturalis females. We conclude that dsAsutdes is associated with the SP biosynthetic pathway in A. suturalis .

An NADH-dependent reductase (SsCR) was discovered by genome data mining. After SsCR was overexpressed in E. coli BL21, recombinant E. coli CCZU-A13 with high reductase activity and excellent stereoselectivity for the reduction of ethyl 4-chloro-3-oxobutanoate (COBE) into ethyl ( R )-4-chloro-3-hydroxybutanoate (( R )-CHBE) was screened using one high-throughput colorimetric screening strategy. After the reaction optimization, a highly stereoselective bioreduction of COBE into ( R )-CHBE (>99% ee ) with the resting cells of E. coli CCZU-A13 was successfully demonstrated in n -butyl acetate–water (10:90, v/v) biphasic system. Biotransformation of 600 mM COBE for 8 h in the biphasic system, ( R )-CHBE (>99% ee ) could be obtained in the high yield of 100%. Moreover, the broad substrate specificity in the reduction of aliphatic and aromatic carbonyl compounds was also found. Significantly, E. coli CCZU-A13 shows high potential in the industrial production of ( R )-CHBE (>99% ee ) and its derivatives.
