Lateral flow assays (LFAs) hold great promise for point-of-care testing, especially in resource-poor settings. However, the poor sensitivity of LFAs limits their widespread applications. To address this, we developed a novel device by integrating dialysis-based concentration method into LFAs. The device successfully achieved 10-fold signal enhancement in Human Immunodeficiency Virus (HIV) nucleic acid detection with a detection limit of 0.1 nM and 4-fold signal enhancement in myoglobin (MYO) detection with a detection limit of 1.56 ng/mL in less than 25 min. This simple, low-cost and portable integrated device holds great potential for highly sensitive detection of various target analytes for medical diagnostics, food safety analysis and environmental monitoring.

Lateral flow assays (LFAs) as rapid analytical techniques promise to be widely used in point-of-care (POC) diagnostics because of their affordability and simplicity. However, LFAs still suffer from low sensitivity in detection of various biomarkers, e.g., nucleic acids. In this study, we developed a simple and general one-step signal amplification strategy, which employed oligonucleotide-linked gold nanoparticle (AuNP) aggregates to enhance the sensitivity in nucleic acid lateral flow (NALF) assays. Using a nucleic acid sequence of human immunodeficiency virus type 1 (HIV-1) as a model analyte, we observed that the detection limit of the developed NALF assay was 0.1 nM, which was improved by 2.5-fold compared with that of a non-signal amplification approach. The methodology described here could be used to detect a broad range of nucleic acids, and the general signal amplification approach could be potentially adopted in other types of LFAs.

Natural killer (NK) cells, natural killer T (NKT) cells, and T lymphocytes were analyzed by using a flow cytometer in 225 human immunodeficiency virus (HIV)-positive individuals infected through the past sale of blood and plasma without receiving antiretroviral therapy in the People’s Republic of China. According to CD4 T-cell counts these HIV-infected adults were stratified into three groups: long-term slow progressors, HIV-infected subjects, and AIDS patients. NK cell counts in long-term slow progressors were higher compared to HIV infection and AIDS patients (P < 0.05) and lower compared to normal controls (P < 0.05), whereas NKT cell counts in slow progressors and the HIV infection group were not different from those of normal controls. NK cell counts in HIV-seropositive subjects were positively correlated with CD4 T-cell counts (P < 0.05), and NKT cell counts were positively correlated with CD4 T-cell and CD8 T-cell counts (P < 0.05). The CD8 T-cell counts were higher in slow progressors compared to those with HIV infection, AIDS patients, and normal controls. These results indicated that HIV infection causes alterations of NK cells and T cells in slow progressors, HIV-infected subjects, and AIDS patient groups, but no difference was found in NKT cell counts and percentages in slow progressors and the HIV-infected group compared to normal controls.

Human Immunodeficiency Virus (HIV) infection is characterized by the depletion of human CD4 + T cells, in which ferroptosis has been identified as a contributing mechanism. Dysregulation of the canonical Wnt/β-catenin signaling pathway is strongly associated with HIV infection. Our findings demonstrate that Wnt7a activates this pathway. Subsequently, β-catenin translocates to the nucleus, where it interacts with the transcription factor 4 (TCF4) to regulate ferroptosis in T lymphocytes from HIV-infected individuals. Analysis of peripheral blood mononuclear cells (PBMCs) from HIV-infected individuals revealed significantly decreased expression levels of Wnt7a, along with key Wnt/β-catenin pathway components (β-catenin, TCF4) and the ferroptosis regulator GPX4, compared to healthy controls. Concurrently, PBMCs from HIV-infected individuals exhibited elevated levels of ferroptosis. We established an in vitro HIV-infected T lymphocyte model by expanding patient-derived PBMCs using a CD3/CD28/CD2 T cell activator and IL-2. Lentiviral transduction was used to stably overexpress Wnt7a in this model. Wnt7a overexpression successfully activated the Wnt/β-catenin pathway and significantly reduced ferroptosis levels. Furthermore, pharmacological manipulation of ferroptosis modulated Wnt7a expression: treatment with the ferroptosis inhibitor Liproxstatin-1 and Ferrostatin-1 increased Wnt7a expression, while exposure to the ferroptosis inducer RSL-3 decreased it. Flow cytometry analysis confirmed that overexpressing Wnt7a significantly increased the number of CD4 + T cells compared to the control. In summary, Wnt7a activates the Wnt/β-catenin signaling pathway to reduce ferroptosis in T lymphocytes from HIV-infected individuals, likely by targeting GPX4, suggesting a potential new strategy for treating HIV infection by restoring the T lymphocyte population.

Proflavine could recognise HIV RRE (Rev response element) RNA, thus inhibiting its interaction with Rev peptide. Owing to the non-significant difference in fluorescence polarization (FP) signal between the proflavine and proflavine-RRE complex, it is not practicable for the identification of antagonists of HIV Rev peptide using proflavine as a FP probe. In the present study, graphene oxide (GO) was introduced to amplify the FP signal to enhance the sensitivity for the identification of Rev antagonists, thereby enabling the development of a primary GO-enhanced FP biosensor for the discovery of Rev antagonists. The effect of the amount of GO on the FP changes was investigated via displacement assay using a known Rev antagonist, neomycin B. The mechanism underlying the changes in FP caused by the addition of neomycin B was explored in the presence of GO. Finally, the performance of this biosensor was further evaluated using other 4 test drugs. The introduction of GO to this interaction system increased the practicability for the discovery of Rev antagonists using proflavine as a probe, and this label-free method does not suffer from limitations caused by fluorescent covalent labeling, thus providing a novel strategy for the characterization of ligand-RNA binding interactions.

Signal amplification techniques are highly desirable for the analysis of low-level targets that are closely related with diseases and the monitoring of important biological processes. However, it is still challenging to achieve this goal in a facile and economical way. Herein, we developed a novel dual signal amplification strategy by combining urease catalysis with the release of Ag+ from silver nanoparticles (AgNPs). This strategy was used for quantifying a DNA sequence (HIV-1) related with human immunodeficiency virus (HIV). DNA target HIV-1 hybridizes with the capture DNA probe on magnetic beads and the reporter DNA probe on AgNPs, forming a sandwich complex. The captured AgNPs are then transformed into numerous Ag+ ions that inactivate numerous ureases. Without catalytic production of ammonia from urea, the substrate solution shows a low pH 5.8 that will increase otherwise. The pH change is monitored by a pH indicator (phenol red), which allows for colorimetric detection. The proposed approach is sensitive, easy to use, economic, and universal, exhibiting a low detection limit of 9.7 fM (i.e., 1.94 attomoles) and a dynamic linear range of 4 orders for HIV-1 sequence detection.

Human immunodeficiency virus (HIV) remains a significant health issue with no vaccine available. Targeting the HIV-1 protease (HIV-1 PR) is essential in treatment, with inhibitors like lopinavir being historically an important component. However, lopinavir alone has low bioavailability and causes side effects when combined with ritonavir, leading to efforts to develop improved analogs. In this study, we designed and synthesized a phosphinic analog of lopinavir, PL1, by replacing its hydroxyethylene moiety with a phosphinic group. The inhibitory evaluation demonstrated a promising inhibition potency of PL1 against HIV-1 PR, supported by molecular dynamics simulations predicting its favorable interactions. Integrating AI-based docking tools confirmed PL1’s strong potential, with an IC 50 /EC 50 of 1.32 × 10 −8 mol/L and a binding affinity of 6.142, surpassing lopinavir’s 5.928. PL1’s binding efficacy index of 0.0119, comparable to lopinavir’s 0.0132, indicates efficient inhibition. Importantly, PL1 is less toxic than lopinavir, making it a promising lead molecule for HIV-1 PR targeting.

The regulator of expression of virion (Rev) protein binds specifically to the Rev-responsive element (RRE) RNA in order to regulate the expression of the human immunodeficiency virus (HIV)-1 genes. Fluorescence indicator displacement assays have been used to identify ligands that can inhibit the Rev–RRE interaction; however, the small fluorescence indicators cannot fully replace the Rev peptide or protein. As a result, a single rhodamine B labeled Rev (RB-Rev) model peptide was utilized in this study to develop a direct and efficient Rev–RRE inhibitor screening model. Due to photon-induced electron transfer quenching of the tryptophan residue on the RB fluorophore, the fluorescence of RB in Rev was weakened and could be dramatically reactivated by interaction with RRE RNA in ammonium acetate buffer (approximately six times). The interaction could reduce the electron transfer between tryptophan and RB, and RRE could also increase RB fluorescence. The inhibitor screening model was evaluated using three known positive Rev–RRE inhibitors, namely, proflavin, 6-chloro-9-[3-(2-chloroethylamino)propylamino]-2-methoxyacridine (ICR 191), and neomycin, as well as a negative drug, arginine. With the addition of the positive drugs, the fluorescence of the Rev–RRE decreased, indicating the displacement of RB-Rev. This was confirmed using atomic force microscopy (AFM) and the fluorescence was essentially unaffected by the addition of arginine. The results demonstrated that RB-Rev can be used as a fluorescent probe for recognizing small ligands that target RRE RNA. The Rev–RRE inhibitor screening model offers a novel approach to evaluating and identifying long-acting Rev inhibitors.

Nevirapine, as one of the antiretroviral drugs, is widely used for the treatment of HIV/AIDS. With the rapid growth in demand for precision medicine and point-of-care rapid diagnostics, electrochemical detection technology has emerged as a key driver of point-of-care testing due to its high sensitivity, potential for miniaturization, low power consumption, and cost-effectiveness. In this experiment, a bimetallic MOF nanocomposite, NiCu-MOF, was synthesized via the solvothermal method and combined with MWCNTs and Ti 3 C 2 via ultrasonication to construct a nevirapine electrochemical sensor for detecting nevirapine. Under optimized experimental conditions, the linear range of nevirapine is 0.01–10.0 μM and 10.0–50.0 μM, with a detection limit of 1.2 nM(0.32 ng/mL). When applied to actual sample testing, the sensor was found to be suitable for the detection of nevirapine in actual samples, yielding recovery rates of 98.71–106.3 % and demonstrating good practical application value. Additionally, the sensor exhibits high selectivity and resistance to interference, and employs a simple synthesis process for the nevirapine detection in serum and urine.

Molecular interaction of darunavir (DRV), a HIV protease inhibitor with calf thymus deoxyribonucleic acid (ct-DNA) was studied in physiological buffer (pH 7.4) by multi-spectroscopic approaches hand in hand with viscosity measurements and molecular docking technique. The UV absorption and fluorescence results together revealed the formation of a DRV–ct-DNA complex having binding affinities of the order of 10 3 M − 1 , which was more in keeping with the groove binding. The results that DRV bound to ct-DNA via groove binding mode was further evidenced by KI quenching studies, viscosity measurements, competitive binding investigations with EB and Rhodamine B and CD spectral analysis. The effect of ionic strength indicated the negligible involvement of electrostatic interaction between DRV and ct-DNA. The thermodynamic parameters regarding the binding interaction of DRV with ct-DNA in terms of enthalpy change (Δ H 0 ) and entropy change (Δ S 0 ) were − 63.19 kJ mol − 1 and − 141.92 J mol − 1 K − 1 , indicating that hydrogen bonds and van der Waals forces played a predominant role in the binding process. Furthermore, molecular simulation studies suggested that DRV molecule was prone to bind in the A-T rich region of the minor groove of DNA.

This research aimed to analyze the biocompatibility and growth behavior of human immunodeficiency virus (HIV)-associated colorectal cancer (CRC) cells on a polylactic acid/polyglycolic acid (PLA-PCL) composite nanofiber scaffold (CNS) using proteomic analysis. The PLA-PCL CNS was prepared
using a gel extraction phase separation (GEPS) method in a dioxane/ethanol solvent system, and the effects of gel temperature, PLA-PCL mass ratio (MR), solvent-to-polymer MR on the structure and properties of the PLA-PCL CNS were analyzed. Subsequently, Lappaol F (LAF) was incorporated into
the PLA-PCL CNS using electrospinning (ES) technology, and tissue specimens were obtained from HIV-associated CRC patients to investigate the impacts of LAF-PLA-PCL CNS on the growth behavior of HIV-associated CRC cells through mass spectrometry and bioinformatics analysis. Field emission
scanning electron microscopy (FE-SEM) revealed that the CNS could be obtained at gel temperatures between −20 °C and −10 °C, PLA-to-PCL MRs of 1:1, 3:2, 7:3, and ethanol content of 5%–15%. X-ray diffraction (XRD) and differential scanning calorimetry (DSC) showed
that the PLA-to-PCL MR of 70:30 exhibited compatibility and crystallinity of the PLA-PCL CNS, while the porosity increased with an increase in PCL content. Fourier-transform infrared spectroscopy (FTIR) analysis indicated a good biological activity of the PLA-PCL CNS. The relative cell proliferation
(RCP) and cytotoxicity grades exhibited no marked differences (P >0.05) between the cancer cells cultured with DMEM and those with PLA-PCL scaffold extract over time. Proteomic analysis identified 127 differentially expressed proteins (DEPs) in HIV-associated CRC cells co-cultured
with LAF-PLA-PCL CNS, and Gene Ontology (GO) protein and KEGG pathway enrichment analysis (KEGG analysis) revealed that the LAF-PLA-PCL NCS could affect the cell cycle (CC) of HIV-associated CRC cells. In conclusion, the PLA-PCL NCS exhibited good compatibility, crystallinity, and biological
activity, while the LAF-PLA-PCL NCS showed the potential to inhibit cell proliferation by affecting the CC.

Darunavir (DRV), a second-generation HIV protease inhibitor, is widely used across the world as an important component of HIV therapy. The interaction of DRV with bovine serum albumin (BSA), a major carrier protein, has been studied under simulated physiological conditions (pH 7.4) by multi-spectroscopic techniques in combination with molecular modeling. Fluorescence data revealed that the intrinsic fluorescence of BSA was quenched by DRV in terms of a static quenching procedure due to the formation of the DRV-BSA complex. The results indicated the presence of single weak affinity binding site (~ 10 3 M − 1 , 310 K) on protein. The thermodynamic parameters, namely enthalpy change (Δ H 0 ), entropy change (Δ S 0 ) and Gibbs free energy change (Δ G 0 ) were calculated, which signified that the binding reaction was spontaneous, the main binding forces were hydrogen bonding and van der Waals forces . Importantly, competitive binding experiments with three site probes, phenylbutazone (in sub-domain IIA, site I), ibuprofen (in sub-domain IIIA, site II) and artemether (in the interface between sub-domain IIA and IIB, site II’), suggested that DRV was preferentially bound to the hydrophobic cavity in site II’ of BSA, and this finding was validated by the docking results. Additionally, synchronous fluorescence, three-dimensional fluorescence and Resonance Rayleigh Scattering (RRS) spectroscopy gave qualitative information on the conformational changes of BSA upon adding DRV, while quantitative data were obtained with Fourier transform infrared spectroscopy (FT-IR).
