Grafting of syringic acid onto fucoidan: Enhanced functional properties and therapeutic potential in histamine-induced liver injury

Mengyao Zhang,Jiali Tang,Jiaying Wang,Chenchen Qi,Guohui Chen,Wei Chen,Chengtao Wang

This study explores the grafting of syringic acid onto fucoidan and assesses the resultant graft's (F S) enhanced functional properties and therapeutic potential in a histamine-induced liver injury model. Utilizing a redox system of ascorbic acid and hydrogen peroxide, the grafting process achieved a grafting rate of 290.45 mg CAE/g, confirmed through UV–Vis spectroscopy, FT-IR spectroscopy, scanning electron microscopy, and thermogravimetric analysis. In vivo investigations using histamine-sensitive mice demonstrated that F S significantly mitigated histamine-induced damage, evidenced by improvements in body weight, organ index, and colon length. F S exhibited superior efficacy in restoring liver function indices (ALT, AST, ALP, GGT), enhancing antioxidant defenses (GSH, SOD), and reducing oxidative stress markers (MDA). Anti-inflammatory effects included reduced levels of pro-inflammatory cytokines (TNF- α , IL-6, IL-1 β ) and modulation of apoptosis-related proteins (BCl 2 , BAX). Additionally, F S upregulated key antioxidant genes in the Nrf2/KEAP1 pathway and downregulated inflammatory genes in the NF- κ B pathway. Restoration of colonic tight junction proteins (ZO-1, Occludin, Claudin-1) and normalization of gut microbiota composition further underscored the therapeutic potential of F S. The study highlights the significant enhancement in functional properties of fucoidan through SA grafting, presenting F S as a promising candidate for developing functional foods and nutraceuticals aimed at preventing and mitigating liver damage and related disorders.

Double-network PVA/γ-PGA/fucoidan hydrogels with favourable biocompatibility and anti-inflammatory activity for wound healing

Aoyu Li,Jun Yang,Yingying He,Changfeng Qu,Jinlai Miao

Acute wound healing still faces numerous challenges, such as excessive inflammatory response, prolonged hemostasis time, and insufficient tissue regeneration, which often lead to delayed wound healing and scar formation. To address these issues, this study developed a multifunctional “physical-ionic” dual-network hydrogel (PPF hydrogel). This hydrogel combines an elastic polyvinyl alcohol (PVA) scaffold with a rigid ionic network composed of γ-polyglutamic acid (γ-PGA) and fucoidan, cross-linked via Ca 2+ . The resulting hydrogel possesses a highly interconnected porous structure, exhibiting excellent mechanical strength and moderate tissue adhesion. In vitro and in vivo evaluations showed that the PPF hydrogel has good blood and cell compatibility, rapidly achieving hemostasis and promoting the proliferation and migration of L929 fibroblasts. In a mouse full-thickness skin resection model, the PPF significantly accelerated wound closure while reducing epidermal hyperplasia and promoting orderly collagen deposition. Mechanistically, the PPF effectively modulates the immune microenvironment by downregulating pro-inflammatory cytokines (TNF-α, IL-6) and upregulating anti-inflammatory factors (IL-10) and VEGF. This regulation promotes M2 macrophage polarization and angiogenesis, thereby accelerating the transition from the inflammatory phase to the remodeling phase. This dual-network hydrogel offers a promising therapeutic strategy for high-quality acute wound repair in regenerative medicine.

Preparation of Hydroxypropyl Methylcellulose-Fucoidan-Citral Nanoemulsion Composite Film: Characterization and Preservative Application in Ostrea rivularis Gould

Hu Yaqin,Liu Guangyu,Wang Dongxue,Yuan Chunhong,Wu Tiantian,Huang Jiayin,Xue Changfeng,Hu Zhiheng

Ostrea rivularis Gould ( O. rivularis ) is nutritionally rich oyster species highly perishable due to endogenous enzymatic activity and microbial proliferation. Currently, active packaging incorporating natural antimicrobials offers a promising approach to improve the quality and shelf-life of aquatic products. In this study, an active composite film (HPMC-FCD-CNE) was developed by incorporating citral nanoemulsion (CNE) into a hydroxypropyl methylcellulose (HPMC) and fucoidan (FCD) matrix. The film with 4% (v/v) CNE exhibited optimal mechanical and barrier properties alongside enhanced antioxidant and antibacterial activities. During refrigerated storage at 4 °C, oysters wrapped with the HPMC-FCD-CNE demonstrated a 4-day extension in shelf-life compared to unwrapped controls, with superior sensory acceptability and delayed quality deterioration, as indicated by reduced total volatile basic nitrogen (TVB-N), K value, and microbial counts. These findings highlight the potential of HPMC-FCD-CNE as an effective active packaging strategy to preserve oysters and other aquatic products under refrigeration.

Preparation of a fucoidan-KCl-based sodium salt substitute and its application in improving chicken mince quality

Ziyan Li,Weijie Wu,Ruiling Liu,Huizhi Chen,Xiangjun Fang,Hangjun Chen,Ben Niu,Haiyan Gao

Potassium chloride (KCl), a sodium substitute, faces challenges due to its metallic bitterness. This study investigated the bitterness-masking effect of fucoidan (Fuc), a sulfated polysaccharide, on KCl and its impact on chicken mince quality. Sensory evaluation, rheological analysis, and simulated K + release tests revealed that 1.5 % Fuc reduced KCl bitterness by 73 % while enhancing perceived saltiness by 19 %. Fuc binds to K + via sulfate groups, increasing solution viscosity and slowing oral K + release. Applied to chicken mince, Fuc-KCl solutions improved gel water-holding capacity (WHC) by forming a dense protein-polysaccharide network, reducing cooking loss, and enhancing textural properties (e.g., chewiness and elasticity). LF-NMR and SEM confirmed that Fuc restructured water distribution and refined gel porosity. Molecular docking identified key interactions between Fuc and myosin residues (Thr277, Lys432), stabilizing the gel matrix. This polysaccharide-based strategy effectively masks KCl bitterness, reduces sodium content, and enhances meat quality, offering a novel approach for low-sodium food innovation.

Induction of degradation of fucoidan oligosaccharides via low-temperature plasma treatment for enhanced structural characteristics and bioactivities

Meiyu Yuan,Ruijuan Fan,Kaitao Peng,Xian Cui,Xiaoxiao Song

Low-temperature plasma (LP) is an emerging advanced oxidation technology that can rapidly and efficiently degrade refractory organic compounds without introducing secondary pollutants. Further investigation is required into the mechanisms of polysaccharide degradation via plasma technology and the enhancement of the bioactivity of the resulting degradation products. In this study, we systematically investigated the degradation mechanisms, structural transformations, and functional potential of fucoidan (Fuc) using LP technology. The critical parameters of the LP treatment were optimized based on the Box–Behnken design, leading to the successful preparation of Fuo-30 T. Kinetic analysis revealed that the degradation process of Fuc followed second-order reaction kinetics. Structural characterization demonstrated that the active radicals generated by LP preferentially cleave glycosidic bonds in glucuronic acid (Glc p A) residues and β-1,6-linked side chains. Fuo-30 T significantly improves DSS-induced colitis. Mechanistically, it inhibits the activation of the NF-κB and MAPK pathways, enhances the stability of Nrf2 and PPARγ, and effectively reduces oxidative stress and inflammatory responses. Another possible mechanism involves lipid metabolism, particularly that of arachidonic acid. Fuo-30 T intervention significantly decreased the production of proinflammatory factors (PGE2, TXB2, and Hete) by inhibiting the cyclooxygenase and lipoxygenase pathways, thereby improving the TCA cycle. Finally, Fuo-30 T significantly upregulated the expression of tight junction proteins in the intestinal epithelium by reshaping the gut microbiota structure, which synergistically improved intestinal microecology and mucosal immune homeostasis.

Protective Effects of Fucoidan on Iodoacetamide-Induced Functional Dyspepsia via Modulation of 5-HT Metabolism and Microbiota

Tianxu Liu,Muyuan Ma,Yonglin Wu,Ismail Muhammad Asif,Daosen Chen,Lichong Liu,Minghui Zhang,Yijie Chen,Bin Li,Ling Wang

As the major polysaccharide in brown algae, fucoidan possesses broad biological abilities and has been reported to improve gastrointestinal health. Functional dyspepsia, a common non-organic disease, is a complex of symptoms mainly characterized by pathogenesis, such as visceral hypersensitivity, gastric dysmotility, and inflammation. To date, the effects of fucoidan in regulating functional dyspepsia with visceral sensitivity remains unclear. In the current study, iodoacetamide was employed to establish a mouse model of visceral hypersensitivity. Meanwhile, fucoidan was orally administrated for fourteen days. Indicators were conducted to evaluate the potential of fucoidan as the ingredient of complementary and alternative medicine for functional dyspepsia, such as levels of serum hormones, expression of receptors, and gut microbial profile. The results show that oral administration of fucoidan led to significant reductions in the secretion of 5-hydroxytryptamine, cortisol, and corticosterone. Additionally, it decreased the expression of 5-hydroxytryptamine-3receptors, with regulation of 5-hydroxytryptamine metabolism and improvement of gut microbial imbalance. The above results suggest fucoidan could ameliorate visceral hypersensitivity by modulating 5-HT metabolism and microbiota. The current findings indicate that fucoidan has potential as a biological component in the adjuvant treatment of functional dyspepsia and for its expanded utilization in the food and medical fields.

Fucoidan from Fucus vesiculosus prevents the loss of dopaminergic neurons by alleviating mitochondrial dysfunction through targeting ATP5F1a

Meimei Xing,Guoyun Li,Yang Liu,Luyao Yang,Youjiao Zhang,Yuruo Zhang,Jianhua Ding,Ming Lu,Guangli Yu,Gang Hu

Parkinson's disease is a neurodegenerative disease that is characterized by the loss of dopaminergic neurons. Fucoidan, which has emerged as a neuroprotective agent, is a marine-origin sulfated polysaccharide enriched in brown algae and sea cucumbers. However, variations in structural characteristics exist among fucoidans derived from different sources, resulting in a wide spectrum of biological effects. It is urgent to find the fucoidan with the strongest neuroprotective effect, and the mechanism needs to be further explored. We isolated and purified four different fucoidan species with different chemical structures and found that Type II fucoidan from Fucus vesiculosus (FvF) significantly improved mitochondrial dysfunction, prevented neuronal apoptosis, reduced dopaminergic neuron loss, and improved motor deficits in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mouse model. Further mechanistic investigation revealed that the ATP5F1a protein is a key target responsible for alleviating mitochondrial dysfunction of FvF to exert neuroprotective effects. This study highlights the favorable properties of FvF for neuroprotection, making FvF a promising candidate for the treatment of PD.

Effect of in vitro simulated digestion on the physicochemical properties and pancreatic lipase inhibitory activity of fucoidan

Chunqi Lu,Qiuya Gu,Xiaobin Yu

BACKGROUND Fucoidan has an anti-obesity effect. However, there are few studies on its mechanism. In this study, we investigated the in vitro and in silico inhibitory properties of fucoidan against pancreatic lipase for the first time. We examined the changes in composition, structure, and pancreatic lipase inhibition of fucoidan during in vitro digestion. RESULTS Simulated saliva-gastrointestinal digestion resulted in a slight decrease in the molecular weight of fucoidan but no significant changes in the monosaccharide composition, sulfate content, and functional groups. Moreover, the digestion process significantly increased the inhibition of pancreatic lipase by fucoidan. The study on the type of inhibition showed that the inhibition of pancreatic lipase by fucoidan belonged to mixed inhibition with competitive inhibition. Molecular docking analysis showed that fucoidan could bind to the active site of pancreatic lipase. CONCLUSION This study indicates that fucoidan can be a potential functional food for anti-obesity. © 2024 Society of Chemical Industry.

Physicochemical characterization of Sargassum fusiforme fucoidan fractions and their antagonistic effect against P-selectin-mediated cell adhesion

Siya Wu,Xu Zhang,Jian Liu,Jianxi Song,Ping Yu,Peichao Chen,Zhiyong Liao,Mingjiang Wu,Haibin Tong

P-selectin, mediated adhesion between endothelium and neutrophils, is a promising target for the therapeutics of acute inflammatory-related diseases. It is reported that brown algal fucoidans can antagonize P-selectin function. However, the fractionation and physicochemical characterization of Sargassum fusiforme fucoidan, and the screening of fucoidan fractions with P-selectin antagonistic capability have not been investigated. In this study, we isolated and fractionated systematically the S. fusiforme fucoidan by ion-exchange chromatography and size exclusion chromatography to obtain eight fucoidan fractions. Their physicochemical characterization was determined by chemical methods, HPLC and FT-IR. The inhibitory capacity of the fucoidan fractions in P-selectin-mediated leukocyte adhesion was evaluated by static adhesion assay and parallel-plate flow chamber. Results showed that fucoidan fractions possessed distinct physicochemical properties, including total carbohydrate, uronic acid and sulfate contents, molecular weight, and monosaccharide compositions. Among all the fucoidan fractions, SFF-32 and SFF-42 showed better blocking ability against P-selectin-mediated cell adhesion.

Preparation of triamcinolone acetonide-loaded chitosan/fucoidan hydrogel and its potential application as an oral mucosa patch

Weiping Zheng,Yuanping Hao,Danyang Wang,Hailin Huang,Fangze Guo,Zhanyi Sun,Peili Shen,Kunyan Sui,Changqing Yuan,Qihui Zhou

Oral inflammatory diseases (OIDs) are among the most common lesions in the oral cavity, affecting the quality of human life and even causing oral cancer. However, most of the current oral mucosa patches still have some limitations, particularly instant, poor mechanical strength and conformability, low adhesion to tissue, and foreign body sensation. Herein, triamcinolone acetonide (TA)-loaded chitosan/fucoidan (CF) composite hydrogels were prepared via chemical crosslinking. The macro/microscopic morphologies and (bio)physicochemical properties of composite hydrogels were investigated. Incorporating fucoidan in chitosan hydrogels greatly enhanced their swelling behavior, mechanical strength, and adhesion properties. Further, the addition of TA in CF hydrogels improved their elastic feature, inhibited inflammatory response, and promoted the formation of mature and well-organized collagen fibers. The developed composite hydrogels displayed not only good antibacterial properties but also good cytocompatibility and histocompatibility . Thus, the designed hydrogels allow the development of oral mucosa patches as a potential treatment for OIDs.

Chitosan–fucoidan encapsulating cinnamaldehyde composite coating films: Preparation, pH-responsive release, antibacterial activity and preservation for litchi

Fengyan Liang,Chusi Liu,Jinwen Geng,Nachuan Chen,Weida Lai,Haitong Mo,Kaidong Liu

In this study, an edible composite film with pH-responsive release was prepared by the formation of Schiff-base imine bonds between chitosan (CS) and oxidized fucoidan (CS–FU) and encapsulating cinnamaldehyde (CA). Fourier-transform infrared, 1 H nuclear magnetic resonance, X-ray photoelectron spectroscopy and gel permeation chromatography confirmed the formation of CS–FU. The result showed that, oxidation degree of FU, degrees of substitution, average molecular weight and yield of CS–FU were 25.57 %, 10.48 %, 23.3094 kDa and 45.63 ± 0.64 %, respectively. Scanning electron microscopy revealed that CA was encapsulated within the CS–FU matrix. Increasing the CA content could improve the mechanical properties and ultraviolet and visible-light resistances of the CS–FU coating films but enhance their water vapor permeabilities. The release of CA increased as the pH decreased, and the antibacterial rate at pH 5 was 2.3-fold higher than that at pH 7, indicating good pH-responsive release and antibacterial properties in mildly acidic environments. Owing to their excellent properties, the CA/CS–FU-0.1 coating films maintained the appearance and quality indices of litchis for at least eight days. Hence, multifunctional composite coating films are prospective eco-friendly and intelligently responsive controlled-release packaging materials for fruit preservation.

Inhibition of the Occurrence and Development of Inflammation-Related Colorectal Cancer by Fucoidan Extracted from Sargassum fusiforme

Xiang Li,Shijun Xin,Xiaoqun Zheng,Liqin Lou,Shiqing Ye,Shengkai Li,Qilong Wu,Qingyong Ding,Ling Ji,Chunrong Nan,Yongliang Lou

Fucoidan has many biological activities, including the inhibitory effect on the development of various cancer types. This study showed that lipopolysaccharide-induced inflammation in FHC cells (normal human colonic epithelial cells) could be reversed using fucoidan at different concentrations. The fucoidan-induced anti-inflammatory effect was also confirmed through in vivo experiments in mice. Compared to the mice of the model group, the ratio of Firmicutes/Bacteroidetes in feces increased and the diversity of gut microbial composition was restored in mice after fucoidan intervention. In colorectal cancer (CRC) cells DLD-1 and SW480, fucoidan inhibited cell proliferation and promoted cell apoptosis. It also blocked the cell cycle of DLD-1 and SW480 at the G0/G1 phase. The animal model of inflammation-related CRC showed that the incidence of tumors in mice was significantly reduced by fucoidan intervention. Furthermore, the administration of fucoidan decreased the expression levels of inflammatory factors such as TNF-α IL-6 and IL-1β in the colonic tissues. Therefore, fucoidan can effectively prevent the development of colitis-associated CRC.
