The establishment of high-performance anti-fouling nanofiltration membranes via cooperation of annular supramolecular Cucurbit[6]uril and dendritic polyamidoamine

Jing Cai,Xue-Li Cao,Yi Zhao,Fu-Yi Zhou,Zhaoliang Cui,Yong Wang,Shi-Peng Sun

In this work, annular supramolecular cucurbit[6]uril (CB6) and dendritic polyamidoamine (PAMAM) were employed to construct high-performance nanofiltration (NF) membranes via one-step interfacial polymerization (IP) process. The obtained membrane features not only the enlarged polyamide tunnels based on the large steric hindrance of CB6 and PAMAM but also the novel hydrophobic rotaxane tunnels based on the host-guest chemistry between CB6 and piperazine (PIP) or PAMAM. The CB6/PAMAM-modified membrane achieved nearly four times enhancement of pure water permeability (PWP) compared to the unmodified traditional membranes with still high Na 2 SO 4 rejection. This indicates the positive contribution of CB6/PAMAM-modification to overcome the “trade-off” effect limiting NF development. In addition, the modified membrane shows the industry-leading separation performance for monovalent and bivalent anions due to the cooperation between the modified pore-size and surface charge. More valuably, CB6/PAMAM-modified membrane manifests the strong anti-fouling property not only in inorganic salt system but also in organic pollutant system based on the modified surface characteristics, indicating its broader application fields. This provides a feasible method to prepare a high-performance anti-fouling NF membrane.

A novel insight into fluorescent sensor for patulin detection using thiol-terminated liposomes with encapsulated coumarin-6 as signal probe

Dongxia Nie,Dakai Guo,Qingwen Huang,Wenbo Guo,Jianhua Wang,Zhihui Zhao,Zheng Han

Nanocomposites of thiol-terminated liposomes encapsulating with coumarin-6 ( [email protected] ) were successfully prepared via a simple thin-film dispersion method to construct a novel fluorescent sensor for selectively detecting trace amounts of patulin. Thiol group ( SH) was designed not only as a specific recognition element to capture patulin, but also as a convenient linker for the further separation of patulin-derivatized and un-derivatized [email protected] by combination with magnetic NH 2 [email protected] 3 O 4 nanoparticles. Liposomes primarily provided an effective platform with a large surface area for facilitated accommodation of large amounts of recognition element ( SH group) and fluorophore (coumarin-6). In such an assay protocol, a reliable link was established between the changes in fluorescent intensity (ΔF) from the nanocomposites of [email protected] /NH 2 [email protected] 3 O 4 and the concentrations of patulin in the range of 0.05–20 ng mL −1 , with the correlation coefficient of 0.996. This approach also effectively eliminated the background interference from other mycotoxins and metal ions. Under the optimal conditions, the designed sensor displayed excellent performance for patulin analysis, with an extremely low detection limit (0.033 ng mL −1 ), high recovery (96.2–107.6 %) and excellent selectivity. Furthermore, the feasibility of its applications has also been demonstrated in the analysis of real juice samples, providing a novel tactics for rational design of a fluorescence assay, for point-of-care diagnostics of patulin, with great potential to be extended to other hazardous compounds by substitution of the recognition elements.

Potential Mechanisms of 3, 4-Oxo-Isopropylidene-Shikimic Acid in Ameliorating 2, 4, 6-Trinitrobenzenesulfonic Acid-Induced Colitis in Rats

Yan Yan,Pengchong Wang,Ying Sun,Yalin Dong,Jianfeng Xing

Previously, we reported that 3, 4-oxo-isopropylidene-shikimic acid (ISA) has therapeutic potential in experimental colitis in rats. This study aimed to elucidate the potential mechanisms of ISA on the inflammatory response in rats with 2, 4, 6-trinitrobenzenesulfonic acid-induced colitis. After the induction of colitis, rats were orally administered ISA for 12 days. Then, the expression levels of inflammatory cytokines, cell adhesion molecules, and matrix metalloproteinase (MMP) in the blood and colon tissues, and the protein level of nuclear factor kappa B (NF-κB) p65 in cytoplasm and nucleus of colon tissues were evaluated. As a result, an enhanced inflammatory response was observed in rats with experimental colitis. However, the treatment with ISA significantly ameliorated the inflammatory response, which was manifested as a significant decrease in the levels of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, interferon (IFN)-γ, IL-8, TNF-α mRNA, P-selectin, E-selectin, intercellular cell adhesion molecule-1, MMP9 and MMP9 mRNA in rat blood and colon tissues, respectively, and a significant decrease in the levels of IFN-γ/IL-4, and the NF-κBp65 activity coefficient. Therefore, the therapeutic effect of ISA on experimental colitis may be related to its inhibitory effect on the expression of cytokines, adhesion molecules, and MMP9, which may be involved in the inhibition of the activation and nuclear translocation of NF-κBp65.

Repetitive transcranial magnetic stimulation protects mice against 6-OHDA-induced Parkinson’s disease symptoms by regulating brain amyloid β1–42 level

Ba Fang,Zhou Yuxin,Zhou Jie,Chen Xueyun

Repetitive transcranial magnetic stimulation (rTMS) is a technique protecting neurons against diverse neurodegenerative disorders by delivering magnetic stimuli into the brain through the intact scalp. In the current study, the protection effect of rTMS on Parkinson’s disease (PD) and the associated mechanism driving the treatment were explored. The PD symptoms were induced using 6-OHDA in mice, and the effect of rTMS of two frequencies (1 Hz and 10 Hz) on the cognitive behaviors and neuron viability was detected. Afterwards, the level of Aβ 1–42 and activity of MKK7-ERK-Fos-APP axis under the administration of rTMS were recorded as well. The intracranial injection of 6-OHDA impaired the cognitive behaviors of the mice in the test of Morris water maze as well as reducing the viability and number of neurons in PD mice. After the treatment of rTMS of both frequencies, the cognitive function of mice was improved and the neuron viability and number were restored in mice brain tissues. The administration of rTMS also increased the cerebrospinal fluid (CSF) level of Aβ 1–42 in PD mice, which was accompanied by the suppressed levels of p-MKK7, p-ERK1/2, p–c-Fos, and APP. Moreover, the effect of rTMS on mice nerve system was all exerted in a frequency-dependent manner. In conclusion, the findings outlined in the current study affirmed the protection effect of rTMS against PD. The anti-PD function of rTMS was associated with the suppression of MKK7-ERK-Fos-APP axis, which subsequently resulted in the increased CSF Aβ 1–42 level and decreased brain Aβ 1–42 level.

Curcumin Ameliorates Chronic Renal Failure in 5/6 Nephrectomized Rats by Regulation of the mTOR/HIF-1α/VEGF Signaling Pathway

Yangbiao He,Xujun Lang,Dong Cheng,Zhihao Yang

Previous studies implicated the mammalian target of rapamycin (mTOR)/hypoxia-inducible factor-1α (HIF-1α)/vascular endothelial growth factor (VEGF) pathway in renal fibrosis and found that curcumin could suppress the expression of mTOR. Therefore, the aim of the present study was to investigate the therapeutic effects of curcumin against chronic renal failure (CRF) in a rat model induced by 5/6 nephrectomy through inhibition of mTOR/HIF-1α/VEGF signaling. A total of 70 male Sprague–Dawley rats were divided into seven groups: a sham group, a CRF group, and five treatment groups. Except for the sham rats, all rats underwent 5/6 nephrectomy to induce CRF. The 5/6 nephrectomized rats received treatment with curcumin vehicle, everolimus vehicle, curcumin, everolimus, or the combination of curcumin and everolimus. Everolimus, a specific inhibitor of mTOR, was used as a positive control. At the end of treatment, blood biochemical indexes, proteinuria and the kidney index were detected. Moreover, histological change was examined by hematoxylin and eosin staining, and protein expression levels were detected by Western blotting. The blood biochemical indexes, proteinuria, and kidney index were increased in the CRF group as compared to the sham group, which was accompanied by marked activation of the mTOR/HIF-1α/VEGF pathway. However, curcumin, as well as everolimus, restored or ameliorated these changes. These results indicate that activation of the mTOR/HIF-1α/VEGF signaling pathway plays an important role in the occurrence and development of CRF, and that curcumin has renoprotective effects by blocking activation of this pathway.

Fate of 6:2 fluorotelomer sulfonic acid in pumpkin (Cucurbita maxima L.) based on hydroponic culture: Uptake, translocation and biotransformation

Shuyan Zhao,Tiankun Liang,Lingyan Zhu,Liping Yang,Tianqi Liu,Jia Fu,Bohui Wang,Jingjing Zhan,Lifen Liu

6:2 fluorotelomer sulfonic acid (6:2 FTSA) is currently used as an alternative to perfluorooctanesulfonate (PFOS) and is widely detected in the environment. The uptake, translocation and biotransformation of 6:2 FTSA in pumpkin ( Cucurbita maxima L.) were investigated by hydroponic exposure for the first time. The root concentration factor (RCF) of 6:2 FTSA was 2.6–24.2 times as high as those of perfluoroalkyl acids (PFAAs) of the same or much shorter carbon chain length, demonstrating much higher bioaccumulative ability of 6:2 FTSA in pumpkin roots. The translocation capability of 6:2 FTSA from root to shoot depended on its hydrophobicity . Six terminal perfluorocarboxylic acid (PFCA) metabolites, including perfluoroheptanoic acid (PFHpA), perfluorohexanoic acid (PFHxA), perfluoropentanoic acid (PFPeA), perfluorobutanoic acid (PFBA), perfluoropropionic acid (PFPrA) and trifluoroacetic acid (TFA) were found in pumpkin roots and shoots. PFHpA was the primary metabolite in roots, while PFBA was the major product in shoots. 1-aminobenzotriazole (ABT), a cytochromes P450 (CYPs) suicide inhibitor, could decrease the concentrations of PFCA products with dose-dependent relationships in pumpkin tissues, implying the role of CYP enzymes involved in plant biotransformation of 6:2 FTSA. This study indicated that the application of 6:2 FTSA can lead to the occurrence of PFCAs (C2–C7) in plants.

Amino-terminated supramolecular cucurbit [6] uril pseudorotaxane complexes immobilized on magnetite@silica nanoparticles: A highly efficient sorbent for salvianolic acids

Qian Zhang,Dong-Dong Zhou,Jia-Wei Zhang,Die Gao,Feng-Qing Yang,Hua Chen,Zhi-Ning Xia

In this study, a novel amino-terminated supramolecular cucurbit [6] uril (CB [6]) pseudorotaxane motif coated magnetic nanoparticles (MNPs) was successfully fabricated through cucurbit [6] uril promoted azide-alkyne cycloaddition. The physicochemical properties of the obtained functional MNPs were investigated by scanning electron microscope, transmission electron microscope, energy dispersive X-ray spectroscopy, Fourier transform infrared spectroscopy, X-ray diffractometer, vibrating sample magnetometer, thermal gravimetric-derivative thermogravimetric analysis , and Zeta potential analyzer. Due to the abundant carboxyl groups in the structure of CB [6] and amino residue on the surface of the pseudorotaxane motif MNPs, the prepared materials were employed to extract salvianolic acids (SAs) for the first time. A series of significant parameters in the extraction and elution process, including sample pH, acetonitrile percentage, ionic strength and extraction temperature, as well as eluent type and desorption time, were examined systematically. Based on the results, the electrostatic interaction , van der Waals interaction , hydrophilic interaction and hydrogen bonding formation between the analytes and sorbent were speculated. The equilibrium state of static adsorption study indicated the outstanding adsorption capacity of pseudorotaxane motif MNPs toward six SAs to be 15–80 mg/g, providing much better adsorption affinity for SAs compared with MNPs only modified with amino, and their adsorption behaviors were all fitted with Freundlich adsorption isotherm model. In addition, the developed pseudorotaxane motif MNPs offered good recyclability and stability. Taking advantage of these merits, four SAs from Salvia miltiorrhiza Bunge were successfully extracted using the prepared pseudorotaxane motif MNPs with relative recovery in the range from 95.1% to 106.5% and RSDs below 9.7%. These results demonstrated that the synthesized pseudorotaxane motif MNPs showed potential as a candidate in the extraction of SAs.

The influence of montmorillonite on the processing and properties of anionic polyamide 6/montmorillonite composites via vacuum assisted resin infusion

Wenqi Wang,Jie Ying,Jikui Wang,Qiang Wang

In this article, the vacuum assisted resin infusion (VARI) process was employed to prepare anionic polyamide 6/montmorillonite (APA6/MMT) nanocomposites. The ɛ-caprolactam and MMT mixture are infused into the mold by vacuum. At the same time, the anionic polymerization of ɛ-caprolactam proceeds all the way which results in fast viscosity rise. To make the VARI process has more practical application value, enough infusion time is necessary. Three types of MMT were chosen in this paper: Na + -MMT, OMMT-CTAB, and OMMT-AUA. In order to study the effects of MMT on infusion process and material's performance, conversion, and temperature test, X-ray diffraction analysis, transmission electronic microscopy, thermogravimetric analysis, Molau test, and mechanical test were performed. The results indicated that the OMMT-AUA can both further improve the material's performance and has longer infusion time for VARI process. POLYM. Determination of nitenpyram and 6-chloronicotinic acid in environmental samples by ion chromatography coupled with online photochemically induced fluorescence detector

Nadeem Muhammad,Yun Zhang,Weixia Li,Yong-Gang Zhao,Amjad Ali,Qamar Subhani,Tariq Mahmud,Junwei Liu,Hairong Cui,Yan Zhu

A simple, cost-effective, sensitive, and quick method for the determination of nitenpyram and its metabolite 6-chloronicotinic acid in environmental samples was developed by coupling an ion chromatograph with a fluorescence detector and a post-column photochemical reactor. This developed analytical method involved a rapid sample extraction by modified and miniaturized quick, easy, cheap, effective, rugged, and safe method followed by isocratic ion chromatographic separation of nitenpyram and 6-chloronicotinic acid into an IonPac ™ AS11-HC column protected by IonPac ™ AG11A guard column by running 30 mM NaOH + 10% acetonitrile mobile phase. A homemade post-column photochemical reactor was also integrated with the ion chromatographic system for online transformation of both analytes into their respective highly fluorescent photoproduct in basic media without using an extra pump. The developed method was validated by following SANTE/11945/2015 guidelines on analytical quality control and validation procedures. The method showed a good linear response (r > 0.999), improved limit of detection (0.101–0.132 μg/L), minimum or no matrix effect, excellent recoveries (90.2–100.10%) and relative standard deviations were found to be ≤6.50%.

New strategy towards flame retardancy through design, synthesis, characterization, and fire performance of a chain extender in polyamide 6 composites

Miao-Jun Xu,Hai-Chao Liu,Kun Ma,Bin Li,Zhi-Yong Zhang

A novel flame-retardant chain extension agent (9,10-dihydro-9-oxa-10-phosphaphenanthrene-10-oxide)-3,5-triglycidyl isocyanurate defined as DOPO-TGIC was synthesized, and its chemical structure was well characterized and confirmed. The effect of DOPO-TGIC as a synergistic agent on the flame retardancy, rheology, thermal and mechanical properties of polyamide 6/aluminum diethylphosphinate (PA6/AlPi) composites were investigated in details. The results demonstrated that PA6/AlPi/DOPO-TGIC composites (1.6 mm) successfully passed UL-94 V-0 rating with the limiting oxygen index value of 30.5% when the total loading amount of AlPi/DOPO-TGIC was 10 wt%. In order to achieve the equal flame-retardant level, the individual AlPi was required 14 wt%. The incorporated DOPO-TGIC improved the complex viscosity of PA6/AlPi/DOPO-TGIC composites due to the chain extending reaction between epoxide groups in DOPO-TGIC and the terminal groups of PA6 matrix. The mechanical performance of PA6/AlPi/DOPO-TGIC composites was also improved compared with that of PA6/AlPi composites. DOPO-TGIC stimulated to the formation of more sufficient and compact char layer during combustion. The higher melt viscosity and compact char layer of PA6 composites effectively constrained the volatilization of flammable gases, thus the heat release was reduced. Consequently, the introduction of DOPO-TGIC simultaneously enhanced the flame retardant and mechanical properties of PA6/AlPi/DOPO-TGIC composites compared with that of PA6/AlPi composites. POLYM. Strontium fructose 1, 6-diphosphate alleviate cyclophosphamide-induced oligozoospermia by improving antioxidant and inhibiting testicular apoptosis via FAS/FASL pathway

J. Li,Y. Hu,Q. Zhang,B. Ma,Z. Wu,Y. Wang,J. Sun,J. Zhu,H. Ying,P. Ouyang

Summary This study investigated the treatment effects of a new compound, strontium fructose 1, 6-diphosphate (FDP-Sr), in cyclophosphamide (CP)-induced oligozoospermia. FDP-Sr, with extra high-energy supply, could reverse male hypogonadism in the testis. Male Wistar rats were randomly divided into three groups: control group (vehicle treated), CP group and CP + FDP-Sr group. Both CP group and CP + FDP-Sr groups were orally administered CP (20 mg kg −1 ) consecutively for the first 7 days to establish CP-induced testicular toxic models. Subsequently, CP group was given orally distilled water per day, whereas CP + FDP-Sr group was received FDP-Sr (200 mg kg −1 ) for 49 days. Compared to the CP group, the FDP-Sr group showed significantly increased levels of serum testosterone, testis relative weights and epididymal sperm counts in rats. In addition, rats treated by FDP-Sr showed the recuperative activities of testicular marker enzymes and normalised levels of antioxidants in tissue. Testicular protection of FDP-Sr was further demonstrated by enhancing expression of P450scc, reducing ability of FAS/FASL and generating cytoprotection in the histopathological study. FDP-Sr appeared to possess an ability to attenuate CP-induced reproduction toxicity via the activation of antioxidants and steroidogenesis enzymes, and alleviate oligozoospermia via inhibition of testicular apoptosis by FAS/FASL pathway.

Experimental and Theoretical Investigations on the Supermolecular Structure of Isoliquiritigenin and 6-O-α-D-Maltosyl-β-cyclodextrin Inclusion Complex

Bin Li,Benguo Liu,Jiaqi Li,Huizhi Xiao,Junyi Wang,Guizhao Liang

Isoliquiritigenin (ILTG) possesses many pharmacological properties. However, its poor solubility and stability in water hinders its wide applications. The solubility of bioactive compounds can often be enhanced through preparation and delivery of various cyclodextrin (CD) inclusion complexes. The 6-O-α-d-maltosyl-β-CD (G2-β-CD), as one of the newest developments of CDs, has high aqueous solubility and low toxicity, especially stable inclusion characteristics with bioactive compounds. In this work, we for the first time construct and characterize the supermolecular structure of ILTG/G2-β-CD by scanning electron microscopy (SEM), ultraviolet-visible spectroscopy (UV), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffractometry (XRD). The solubility of ILTG in water at 25 °C rises from 0.003 to 0.717 mg/mL by the encapsulation with G2-β-CD. Our experimental observations on the presence of the ILTG/G2-β-CD inclusion complex are further supported by the ONIOM(our Own N-layer Integrated Orbital molecular Mechanics)-based QM/MM (Quantum Mechanics/Molecular Mechanics) calculations, typically substantiating these supermolecular characteristics, such as detailed structural assignments, preferred binding orientations, selectivity, solvent effects, interaction energies and forces of the ILTG/G2-β-CD inclusion complex. Our results have elucidated how ILTG interacts with G2-β-CD, demonstrating the primary host-guest interactions between ILTG and G2-β-CD, characterized by hydrogen bonds, hydrophobic interactions, electrostatic forces, and conformational effects, are favored for the formation of the ILTG/G2-β-CD inclusion.Keywords:isoliquiritigenin;6-O-α-d-Maltosyl-β-CD;inclusion complex;ONIOM calculation
