Dual humidity-pH responsive cellulose-based phosphorescent paper for multimodal anti-counterfeiting

Wang Qing,Yu Mingguang,Chen Guangxue,Chen Zijian,Zhong Yingying,Mo Qijie,Wang Honghu

Herein, a facile strategy for fabricating cellulose-based paper with dual humidity-pH responsive room-temperature phosphorescence (RTP) through molecular excited-state modulation was reported. The phosphorescent system was constructed by immobilizing ethylenediamine-mediated condensation product of 4-bromo-1,8-naphthalic anhydride (forming 1,8-naphthalimide derivatives) onto cellulose matrix via hydrogen-bond rigidification. Post-papermaking and drying processes, the developed material exhibits remarkable excitation-dependent dual emission: orange-red phosphorescence ( τ  = 6.3 ms) under ambient conditions and blue fluorescence ( τ  = 3.3 ns) under saturated humidity. Notably, the humidity-responsive luminescence switching demonstrates excellent reversibility after drying cycles. Furthermore, the incorporated naphthalimide moiety preserves its inherent pH sensitivity, enabling simultaneous pH detection through emission wavelength shifts. Systematic characterization reveals that the humidity response originates from water-molecule regulated intersystem crossing efficiency, while the pH response stems from protonation-induced electronic structure modifications. This work establishes a novel paradigm for designing multi-stimuli responsive smart materials through rational manipulation of molecular excited states, demonstrating potential applications in paper-based anti-counterfeiting technologies.

Injectable and self-healing polysaccharide-based hydrogel for pH-responsive drug release

Chao Qian,Tingbin Zhang,Joel Gravesande,Charles Baysah,Xiaoyan Song,Jinfeng Xing

Injectable hydrogels with self-healing and pH-responsive property are appealing for biomedical applications. Herein, we developed a facile and green method to prepare a multifunctional polysaccharide-based hydrogel as a new carrier of drug. The hydrogels were prepared by forming reversible chemical bond between carboxyethyl-modified chitosan (CEC) and aldehyde modified hyaluronic acid (A-HA). The morphology and rheological property of the hydrogels with different solid content were systematically characterized. Owing to the dynamic equilibrium of the Schiff base bonds between amine groups on CEC and aldehyde groups on A-HA, the rapid self-healing performance of hydrogels was confirmed through qualitative and quantitative methods without any external stimulus. The pH-responsive behaviour was demonstrated by equilibrium swelling and in vitro Doxorubicin (Dox) release in PBS medium with various pH. In acidic condition, Dox can be release more rapidly compared with weak alkaline medium. Furthermore, the kill effect of Dox released from hydrogels for cancer cells was investigated. In vitro degradation and cytotoxicity examinations showed that the hydrogel is biodegradable and biocompatible. Therefore, such polysaccharide-based injectable self-healing and pH-responsive hydrogel is a promising candidate as drug delivery carrier.

Qinying Shi,Ying Lu,Guannan Zhang,Xin Yang,Rong Li,Guoliang Zhang,Xiudong Guo,Jianbo Song,Qin Ding

The recurrence and bone defect of malignant osteosarcoma postsurgical treatment have gained remarkable attention. Therefore, the development of multifunctional treatment platform is urgently desirable to achieve efficient tumor treatment and bone regeneration. In this paper, a multifunctional nanomaterial using mesoporous silica (MSN) as platform modified with quercetin (Qr), collagen (Col) and dopamine (PDA) was developed. Our findings demonstrated that the nanoparticles designed in this work had excellent photothermal properties and pH responsiveness. In addition, the nanoparticles had outstanding anti-tumor ability and could killed Saos-2 cells within 10 min under 808 nm laser irradiation owing to the synergistic effect of hyperthermia and Qr. Besides, the modification of PDA and Col endows the nanoparticles with excellent osteogenic activity.

Magnesium hydroxide nanoplates: a pH-responsive platform for hydrophobic anticancer drug delivery

Mingyi Guo,Faheem Muhammad,Aifei Wang,Wenxiu Qi,Nan Wang,Yingjie Guo,Yen Wei,Guangshan Zhu

The cost of conventional chemotherapeutic drugs is substantially high, and biomedical researchers are constantly hunting for cheap and effective chemotherapeutic alternatives. Recently, curcumin has emerged as a cost effective anticancer remedy, however, the low bioavailability of curcumin has been a major impediment to its successful utilization for disease management. In this work, we developed a highly biocompatible magnesium hydroxide as an intelligent nanocarrier for delivering curcumin into cancer cells. Curcumin was loaded onto magnesium hydroxide nanoplates via a complexation strategy. Furthermore, these drug conjugated nanoparticles not only achieve efficient loading of a highly hydrophobic drug, but also exhibit pH responsive release in extracellular or intracellular acid environments, validated by in vitro drug release, confocal microscopy and MTT assay. These biocompatible nanoplates can be promising candidates for the further development of smart drug delivery nanodevices.

pH-responsive pseudorotaxane between comblike PEO-grafted triblock polymer and α-cyclodextrin

Li Xiubing,Xiao Junjie,Wang Xi,Yi Chunwang,Su Shengpei

The pH-responsive inclusion complexation of comblike triblock polymer, P2VP- b -PPEOMA- b -P2VP, with α-cyclodextrin (α-CD) was studied. The triblock polymer was prepared by reversible addition–fragmentation chain transfer polymerization (RAFT) and formed inclusion complexes (ICs) after selective threading of the PEO segment of the triblock polymer through the cavities of α-CD units. For comparison, PPEOMA homopolymer was prepared, and the inclusion complexation with α-CD was also studied. The formed ICs were characterized by XRD and 1 H NMR. The results revealed that P2VP- b -PPEOMA- b -P2VP can form ICs with α-CD even when forming micelles, and the introduction of P2VP had a great influence on the solution property and the stoichiometry of EO to CD of the inclusion complexes depending on the concentration and the pH of the solution.

Hyaluronidase and pH Dual-Responsive Nanoparticles for Targeted Breast Cancer Stem Cells

Li Weinan,Zhang Xiaoyu,Nan Yang,Jia Li,Sun Jialin,Zhang Lina,Wang Yanhong

pH-responsive and CD44 receptor-mediated targeted nanoparticles for eliminating cancer stem cells (CSCs) were developed based on complexes of PEG-poly(β-amino ester) (PEG-PBAE) micelles (PPM) coated with hyaluronic acid (HA) (HA-coated PPM complex, or HPPMc). Thioridazine (Thz) was loaded into HPPMc with a decent drug loading content. The release results of the drug in vitro showed that Thz was released from the HPPMc, which was stimulated by both the acidic pH and specific enzymes. Cytotoxicity studies on mammospheres (MS) revealed that the toxicity potential of Thz-loaded HPPMc (Thz–HPPMc) at pH 5.5 was better than drug solutions. Compared with that at pH 7.4, a higher cellular uptake of a coumarin-6 (C6)-labeled complex at pH 5.5 was observed, which demonstrated that complexes were efficiently taken up in MS. Meanwhile, free HA competitively inhibited the cellular uptake of HPPMc, which revealed that the uptake mechanism was CD44 receptor-mediated endocytosis. Within the acidic endolysosomal environment, the protonation of PBAE facilitated the escape of the complex from the lysosome and releases the drug. The results of in vivo distribution studies and tumor suppression experiments showed that HPMMc could stay in the tumor site of BALB/c nude mice for a longer period of time, and Thz–HPPMc could significantly improve the tumor-suppressing effect. All these results demonstrated the great potential of the multifunctional nanoparticle system for eliminating CSCs.

Injectable pH-responsive hydrogel for combinatorial chemoimmunotherapy tailored to the tumor microenvironment

Gu Jun,Zhao Gang,Yu Jiangkun,Xu Pei,Yan Jiabin,Jin Zhengshuai,Chen Sheng,Wang Yong,Zhang Leshuai W.,Wang Yangyun

Although combination chemoimmunotherapy shows promising clinical results for cancer treatment, this approach is largely restricted by variable objective response rate and severe systemic adverse effects of immunotherapeutic antibody and chemotherapeutic drugs. Therefore, an in situ-formed therapeutic silk-chitosan composite scaffold is fabricated in this study to allow local release of the chemotherapeutic drug doxorubicin (DOX) and JQ1 (small molecular inhibitor used for the extraterminal protein BRD4 and bromodomain) with control release kinetics. DOX-JQ1@Gel contains a pH-degradable group that releases therapeutics in a weak acidic tumor microenvironment. The released DOX could directly kill tumor cells or lead to immunogenic cell death, thereby triggering the response of antitumor immunity. Meanwhile, chemotherapy-triggered antigen release and JQ1-mediated PD-L1 checkpoint blockade cumulatively contribute to trigger the response of antitumor immunity. Finally, the DOX-JQ1@Gel is locally injected to evaluate its synergistic cancer therapeutic effect, which is expected to improve objective response rate of immunotherapy and minimize systemic side effects.Graphical Abstract

Non-dye cell viability monitoring by using pH-responsive inverse opal hydrogels

Tengfei Wang,Jingjing Liu,Fuqiang Nie

Recent advances in the field of drug screening focus on accurate, rapid and high-throughput screening methods. In our work, hydrogel inverse opal photonic crystal microspheres (HPCMs) were fabricated through a templating method and exhibited a robust and reversible response to temperature and pH. The response performance was tested under various temperature (25–55 °C) and pH (1.5–7.5) conditions and the reflective peak shifted noticeably within the visible wavelength range. Furthermore, HPCMs were used as drug delivery carriers and not only displayed high doxorubicin (DOX) drug loading but also presented thermo/pH-induced drug release properties. More importantly, these carriers were shown to be good reporters for monitoring cell viability due to their tunable colour variation. This capability was applied to H460 cell cultures with or without DOX. The structure colour of HPCMs varied in different cell culture microenvironments, and cell apoptosis was able to be distinguished. In this way, this fast, non-dyeing method for reporting cell viability in tumour cytotoxicity assays has potential in the field of drug screening and may give new insights into the use of structural colour to report results in drug screening systems.

pH-responsive dithiomaleimide-amphiphilic block copolymer for drug delivery and cellular imaging

Ting Bai,Dongyan Shao,Jianxin Chen,Yifan Li,Ben Bin Xu,Jie Kong

A drug delivery system that is integrated with fluorescent imaging is an emerging platform for tumor diagnostic and therapy. A pH-responsive fluorescent polymer that can respond to the surrounding medium is a desired component with which to construct an advanced drug delivery system with bioimaging characteristics and controllable drug releasing. In this work, we synthesized novel amphiphilic block copolymers of poly(ethylene glycol)- b -poly(2-(diisopropylamino) ethyl methacrylate- co -dithiomaleimide) (PEG- b -poly(DPA- co -DTM)) and poly(ethylene glycol)- b -poly(2-(dibutylamino) ethyl methacrylate- co -dithiomaleimide) (PEG- b -poly(DBA- co -DTM)) with pH-responsiveness and fluorescence. The block copolymers exhibited relatively stable fluorescence properties in different solvent and excitation-independent fluorescence behaviours. By copolymerizing the responsive segments in the molecule chain, the doxorubicin (DOX)-loaded micelles could be triggered to disassemble, thus releasing DOX at the corresponding pH values and yielding a pH-responsive drug release. Targeted deliveries of the drug within the cell were demonstrated by using the carrier responding to different pH values. The best antitumor effect was obtained by PEG- b -poly(DPA- co -DTM), which immediately released DOX as soon as it entered the tumor cells, as a result of responding to the regional pH level (pH = 6.3). The pH-responsive copolymers showed excellent biocompatibilities, as nearly 85% of cells with these fluorescent micelles survive when the testing concentration goes up to 200 μg mL −1 . In all, these pH-responsive and dithiomaleimide-based fluorescent block copolymers hold great potential in future cancer diagnostic and therapeutic techniques.

Fluorometric sensing of pH values using green-emitting black phosphorus quantum dots

Yue Qiaoli,Hu Yingying,Tao Lixia,Zhang Baoqian,Liu Chen,Wang Yongping,Chen Chunying,Zhao Jinsheng,Li Chen-Zhong

A fluorometric method is described for “turn-on” sensing of pH values via black phosphorus quantum dots (BPQD). Water-stable BPQD were synthesized by a liquid exfoliation method and characterized by TEM, FT-IR, XPS, and absorption and fluorescence spectra. The nanoparticles of BPQD have a uniform distribution with an average size of 5.2 nm. They exhibit bright green fluorescence, with excitation/emission maxima at 420/515 nm. The fluorescence of the BPQD is likely to arise from the quasi-molecular fluorophores of polycyclic aromatic compounds carrying P-P, P-O-P, and P x O y functions on its surface. The protonation and deprotonation of hydroxyl groups of BPQD causes a different degree of quenching of the BPQD. At pH values below 4.0, protons bind to BPQD to form non-fluorescent ground state complexes. At pH values above 4.0, the hydroxyl groups become deprotonated, and this induces the recovery of fluorescence. The sensor has a linear response in the pH range of 1.0–9.0. It was successfully applied to the determination of the pH values in human urine and serum samples. Schematic representation of the preparation of black phosphorus quantum dots (BPQDs) from powdered BP crystals using liquid-phase exfoliation in N-methyl-2-pyrrolidone solution. The BPQDs display green fluorescence at high pH values but no fluorescence at very low pH values.

Novel Peptide–Polymer Conjugate with pH-Responsive Targeting/Disrupting Effects on Biomembranes

Sijia Wang,Yue Sun,Shouhong Xu,Honglai Liu

Conjugating polymers to peptides has become a new strategy of designing functional antitumor agents for their improved stability and enhanced activity. In this paper, a novel peptide–polymer conjugate PEPc–PMAA with pH responsiveness was designed and synthesized. The isoelectric point of PEPc was studied by dynamic light scattering for the targeting effect. Also, the transmittances of PMAA at different pHs were measured using an ultraviolet–visible spectrophotometer for determining the triggering pH of the disrupting effect. The results showed that PEPc–PMAA was hydrophilic under neutral conditions and changed to be amphiphilic composed of positively charged PEPc and hydrophobic PMAA under acidic conditions. The interactions between PEPc–PMAA and mimic cells were investigated by the measurements of membrane fluidity and cargo leakage from 1,2-dipalmitoyl-sn-glycerol-3-phosphocholine and 1,2-dipalmitoyl-sn-glycerol-3-phospho-(1-rac-glycerol) (DPPG) liposomes. It proved that PEPc–PMAA caused a distinct membrane disturbance of the DPPG liposome at pH 5.5, resulting in more serious cargo leakage. Because of its targeting and disrupting effects on negatively charged biomembranes under acidic conditions, PEPc–PMAA showed its good potential as an antitumor agent.

Removal of ciprofloxacin by persulfate activation with CuO: A pH-dependent mechanism

Shengtao Xing,Wenqing Li,Bo Liu,Yinsu Wu,Yuanzhe Gao

CuO synthesized by a hydrothermal method was used to activate persulfate (PS) for removing ciprofloxacin (CIP) in water. The influences of reaction parameters and coexisting substances on this process were studied. Under the optimal conditions ([CuO] 0  = 6.3 mmol L −1 , [PS] 0  = 1 mmol L −1 , pH 8), CIP was completely degraded and the total organic carbon removal efficiency was 84.2%. Ca 2+ , Mg 2+ and Cl − had little effect on CIP removal, while fulvic acid showed a slight inhibitory effect. By contrast, bicarbonate enhanced CIP degradation and significantly suppressed Cu leaching. Subsequently, the reactive species for CIP degradation were identified by electron paramagnetic resonance and radical scavenging experiments. Although the decomposition efficiency of PS was significantly accelerated by increasing pH, similar amounts of O 2 − , OH, SO 4 − and 1 O 2 were detected at different pH, suggesting the formation of more surface activated PS at higher pH. The reactive species for CIP degradation were found to be dependent on solution pH. CIP was mainly degraded by SO 4 − and 1 O 2 at acidic conditions. The activated PS might have high selectivity towards CIP and its amount might be higher than that of radicals at pH 8. In addition, the uncharged surface of CuO at pH 8 facilitated the adsorption of CIP, which is beneficial to the surface reaction between CIP and activated PS. Hence, the activated PS was the dominated species for CIP degradation at this pH.
