The Effect of Zinc Oxide Nanoparticles for Enhancing Rice (Oryza sativa L.) Yield and Quality

Haipeng Zhang,Rui Wang,Zhiqing Chen,Peiyuan Cui,Hao Lu,Yanju Yang,Hongcheng Zhang

Zinc oxide nanoparticles (ZnO NPs) have been applied widely in agriculture, and many studies were conducted to evaluate the effect of ZnO NPs on plant growth. So far, few studies have been investigated with regard to the potential effect of ZnO NPs on cereal yield formation or Zn content in grains. Herein, we used a pot experiment, which was conducted involving five dosages of Zn (0.3, 0.6, 1.2, 2.4, and 4.8 g Zn pot−1), to evaluate the impacts which ZnO NPs made in rice yield, dry matter accumulation, rice quality and grain Zn contents. The results demonstrated that ZnO NPs increased the production of grain, dry matter accumulation and particulate Zn content. Compared with control treatment, ZnO NPs application presented higher rice yield with more panicle number (4.83–13.14%), spikelets per panicle (4.81–10.69%), 1000-grain weight (3.82–6.62%) and filled grain rate (0.28–2.36%). Additionally, the dry jointing, heading and mature periods, LAI, SPAD values, and photosynthetic potentials of ZnO NPs were all significantly higher relative to treatment without ZnO NPs. The more photosynthetic substances and higher dry matter accumulated in the whole rice growing stage resulted in higher rice grain yield. Furthermore, ZnO NPs increased brown rice rate, milled rice rate, head rice rate, chalkiness size, chalkiness grain rate, chalkiness degree, amylose content and protein content, improving rice processing and appearance qualities. For the Zn nutrition in rice grain, ZnO NPs application significantly increased the Zn content of edible polished rice and promoted the relocation of Zn from the aleurone layer. This study effectively demonstrated that ZnO NPs could be a potential high-performed fertiliser for enhancing rice yield and quality. Keywords:Zinc oxide nanoparticles;yield formation;rice quality;photosynthetic potential

Biomimetic dandelion-like magnetic nanoparticles for capture and detection of S. aureus and L. monocytogenes

Guanhua Chen,Zhengzheng Wang,Mei Deng,Fangbin Xiao,Xuekun Bai,Hengyi Xu

Foodborne pathogenic microorganism pose a serious threat to public health security; thus, constructing quick and sensitive methods for early screening foodborne pathogenic microorganism is necessary for human health. Teicoplanin (Teic) is a broad-spectrum biometric molecule, which can recognize Gram-positive (G + ) bacteria. In this study, using bovine serum albumin (BSA) and PEG 2k as “carrier” and “linker”, respectively. Teic was wired to magnetic beads (MBs) to form the biomimetic dandelion-like magnetic nanoparticles (MBs-PEG-BSA-Teic) to capture Staphylococcus aureus ( S. aureus ) and Listeria monocytogenes ( L. monocytogenes ). The MBs-PEG-BSA-Teic displayed excellent capture ability of S. aureus (> 93%) and L. monocytogenes (> 86%). After separation, the fluorescence probes were added, forming a “sandwich” structural compounds, namely, MBs-PEG-BSA-Teic~bacteria~fluorescence probes. The fluorescence intensity of the probes was measured by a multimode microplate reader. Consequently, this novel method showed high detection sensitivity to target model bacteria, and the detection limit of S. aureus and L. monocytogenes in PBS buffer solution and juice were achieved 10 1 CFU/mL, spinach and ground beef samples were achieved 10 2 CFU/g. The method has easy operation and high sensitivity, and it is less interfered by food matrix or negative bacteria; thus, this method is suitable for early screening G + pathogen in suspected food samples.

Identification and Characterization of the Seed Storage Proteins and Related Genes of Cannabis sativa L.

Sun Xin,Sun Yao,Li Yao,Wu Qiong,Wang Lei

Hemp (Cannabis sativa L.) seed is emerging as a novel source of plant protein owing to its rich protein content and reasonable nutritional structure. In the current study, the storage proteins of hemp seed were extracted using different methods. The modified Osborne method yielded maximum extraction of the hemp seed storage proteins, while degreasing had little effect on the hemp seed protein (HSP) extraction. Protein identification results revealed that 11S globulin (edestin) was the most abundant protein in hemp seed, and the molecular weights of the two subunits of this protein were ~35 and 20 kDa, respectively. The second most abundant protein was 2S albumin (Cs2S), with a molecular weight of ~14–15 kDa. The least abundant protein was 7S vicilin-like protein (Cs7S), with a molecular weight of ~47 kDa. Subsequently, gene families encoding these three storage protein classes, including three genes for edestin, two for Cs2S, and one for Cs7S, were cloned and then analyzed for amino acid composition and structure. The three edestins were different in their amino acid sequences and calculated molecular weights. The analysis of coding sequences revealed a higher percentage of similarity (62.7%) between Edestin1 and Edestin3, while the similarity decreased significantly to ~57% between Edestin1 and Edestin2, and 58% between Edestin2 and Edestin3. The calculated protein molecular weight was the highest for the protein encoded by Edestin1 and the smallest for the protein encoded by Edestin2. All three edestins were rich in arginine, while Edestin3 had a higher methionine content relative to that in the other two, which proved that Edestin3 had a better nutritional value. Cs2S and Cs7S were different from those reported in previous studies. Therefore, it could be inferred that amino acid composition varies with different hemp cultivars. The current research brought significant theoretical advance in illuminating the understanding of hemp seed storage protein and would have significance for future research on improving the nutritional quality of hemp seed and developing bioactive peptides.

Bioorthogonal information storage in l-DNA with a high-fidelity mirror-image Pfu DNA polymerase

Fan Chuyao,Deng Qiang,Zhu Ting F.

Natural DNA is exquisitely evolved to store genetic information. The chirally inverted l -DNA, possessing the same informational capacity but resistant to biodegradation, may serve as a robust, bioorthogonal information repository. Here we chemically synthesize a 90-kDa high-fidelity mirror-image Pfu DNA polymerase that enables accurate assembly of a kilobase-sized mirror-image gene. We use the polymerase to encode in l -DNA an 1860 paragraph by Louis Pasteur that first proposed a mirror-image world of biology. We realize chiral steganography by embedding a chimeric d -DNA/ l -DNA key molecule in a d -DNA storage library, which conveys a false or secret message depending on the chirality of reading. Furthermore, we show that a trace amount of an l -DNA barcode preserved in water from a local pond remains amplifiable and sequenceable for 1 year, whereas a d -DNA barcode under the same conditions could not be amplified after 1 day. These next-generation mirror-image molecular tools may transform the development of advanced mirror-image biology systems and pave the way for the realization of the mirror-image central dogma and exploration of their applications.

Determination of Acetamiprid by Fluorescence Monitoring of a Glycine-L-Histidine Copper-Organic Framework Aptasensor

Zhipeng Su,Fanyan Ye,Kaiyu He,Tao Yang,Wang Li,Jiali Ren

The sensitive determination of pesticides constitutes an important aspect in ensuring food safety. Due to the disadvantages of traditional methods, such as low sensitivity, complex operation, and high cost, the development of an efficient and economical biosensor to detect pesticide residues is required. In this work, the dipeptide glycyl-L-histidine (GH) and divalent copper ions (Cu2+) were employed as bioorganic linkers and metal ions in the synthesis of the peptide-based metal organic framework (MOF) material GH-Cu BioMOF. Because of its properties of fluorescence quenching and adsorption of nucleic acids, GH-Cu BioMOF was used to develop a distinctive aptamer based method for the determination of pesticide residues in food. The GH-Cu BioMOF-based aptasensor was used for acetamiprid and demonstrated a linear response in the range from 0.15 to 3.00 µM with a detection limit of 137 nM, verifying the feasibility of this method for pesticide determination in food.

Nonionic and Water-Soluble Poly(d/l-serine) as a Promising Biomedical Polymer for Cryopreservation

Yuling Sun,Jie Liu,Zhibo Li,Jianjun Wang,Yanbin Huang

Water-soluble, biodegradable, nonionic, and biocompatible polymers with multiple functional groups are highly desired for biomedical applications. Here, we report that water-soluble nonionic poly(d/l-serine) is chirality-controllable, biodegradation-controllable, and non-cytotoxic. Hence, it can be a highly sought-after alternative to the widely used poly(ethylene glycol), with an additional advantage of having multiple hydroxyl groups for further functionalization. As one example of its biomedical applications, poly(d/l-serine) demonstrated an obvious cryoprotective effect on the red blood cells. The usage of poly(d/l-serine) in the cryopreservation field would be of great promise to resolve the difficulties in separating cryoprotectants due to toxicity.

The Great Capacity on Promoting Melanogenesis of Three Compatible Components in Vernonia anthelmintica (L.) Willd.

Yifan Lai,Qingyuan Feng,Rui Zhang,Jing Shang,Hui Zhong

To investigate a possible methodology of exploiting herbal medicine and design polytherapy for the treatment of skin depigmentation disorder, we have made use ofVernonia anthelmintica (L.) Willd., a traditional Chinese herbal medicine that has been proven to be effective in treating vitiligo. Here, we report that the extract ofVernonia anthelmintica (L.) Willd. effectively enhances melanogenesis responses in B16F10. In its compound library, we found three ingredients (butin, caffeic acid and luteolin) also have the activity of promoting melanogenesis in vivo and in vitro. They can reduce the accumulation of ROS induced by hydrogen peroxide and inflammatory response induced by sublethal concentrations of copper sulfate in wild type and green fluorescent protein (GFP)-labeled leukocytes zebrafish larvae. The overall objective of the present study aims to identify which compatibility proportions of the medicines may be more effective in promoting pigmentation. We utilized the D-optimal response surface methodology to optimize the ratio among three molecules. Combining three indicators of promoting melanogenesis, anti-inflammatory and antioxidant capacities, we get the best effect of butin, caffeic acid and luteolin at the ratio (butin:caffeic acid:luteolin = 7.38:28.30:64.32) on zebrafish. Moreover, the effect of melanin content recovery in the best combination is stronger than that of the monomer, which suggests that the three compounds have a synergistic effect on inducing melanogenesis. After simply verifying the result, we performed in situ hybridization on whole-mount zebrafish embryos to further explore the effects of multi-drugs combination on the proliferation and differentiation of melanocytes and the expression of genes (tyr, mitfa, dct, kit) related to melanin synthesis. In conclusion, the above three compatible compounds can significantly enhance melanogenesis and improve depigmentation in vivo. Keywords:melanogenesis;ROS;inflammatory;zebrafish;butin;caffeic acid;luteolin;synergistic effect

Electrospun polylactic acid nanofiber membranes containing Capparis spinosa L. extracts for potential wound dressing applications

Peng Zhu,Xingqun Zhang,Yunlong Wang,Changen Li,Xianzhu Wang,Jiancheng Tie,Ying Wang

Capparis spinosa L. (CSL) is a medicinal plant with high antibacterial activity against a variety of pathogens and antioxidation properties. In this paper, for the first time, nanofiber membranes of polylactic acid (PLA) containing 0, 4, 7,and 10 wt% CSL ethyl acetate extract were fabricated by electrospinning. Scanning electron microscopy showed that the fiber diameter decreased after adding CSL to the PLA nanofibers. Fourier transform infrared spectroscopy confirmed that CSL was successfully incorporated in the matrix. The water contact angle test proved that the addition of CSL improved the hydrophilic properties of the material. Moreover, the addition of CSL improved the oxidation resistance of the composite fiber membrane. A burst drug release from the composite nanofibers occurred within the first 12 hr, followed by slow release over a prolonged period of time. As the concentration of CSL increased, the inhibition ability of nanofibers against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) gradually increased. In summary, due to their good mechanical, antioxidant, and antibacterial properties, CSL/PLA nanofiber membranes may possess potential applications as wound dressing materials.

Micro-distribution of arsenic and polycyclic aromatic hydrocarbons and their interaction in Pteris vittata L.

Xiao-Yong Liao,Xue-Gang Gong,Li-Li Zhang,Daniel P. Cassidy

Interactive effects of inorganic arsenic (As) species and polycyclic aromatic hydrocarbons (PAHs) on their uptake, accumulation and translocation in the hyperaccumulator Pteris vittata L. ( P. vittata ) were studied hydroponically. The presence of PAHs hindered As uptake and acropetal translocation by P. vittata , decreasing As concentrations by 29.8%–54.5% in pinnae, regardless of the initial As speciation. The inhibitive effect of PAHs was 1.6–8.7 times greater for arsenite [As(III)] than for arsenate [As(V)]. Similarly, inorganic As inhibited the uptake of fluorene (FLU) and benzo[a]pyrene (BaP) by P. vittata roots by 0.4%–21.7% and by 33.1%–69.7%, respectively. Interestingly, coexposure to As and PAHs slightly enhanced the translocation of PAHs by P. vittata with their concentrations increased 0.3 to 0.8 times in shoots, except for the As(III)+BaP treatment. The antagonistic interaction between As and PAHs uptake is likely caused by competitive inhibition or oxidative stress injury. By using synchrotron radiation micro X-ray fluorescence imaging, high concentrations of As were found distributed throughout the microstructures far from main vein of the pinnae when coexposed with PAHs, the opposite of what was observed with exposure to As only. PAHs could also significantly inhibit the accumulation and distribution of As in vascular bundles in rachis treated with As(III). The results of two-photon laser scanning confocal microscopy revealed that PAHs were mainly distributed in the vascular cylinder, epidermal cells, vascular bundles, epidermis and vein tissues, and this was independent of As speciation and treatment. This work offers new positive evidence for the interaction between As and PAHs in P. vittata , presents new information on the underlying mechanisms for interactions of As and PAHs affecting their uptake and translocation within P. vittata L., and provides direction for future research on the mechanisms of PAHs uptake by plants.

Effective extraction and recovery of linarin from Chrysanthemum indicum L. flower using deep eutectic solvents

Na Guo,Yong-Peng Zou,Hong-Kun Li,Ping Kou,Zhi-Ming Liu,Yu-Jie Fu

Conventional approaches for extraction and enrichment of phytochemicals from plant matrix have many drawbacks such as high organic solvent consumption, low extraction yields, time-consuming and large labor for the enrichment process. In the present study, ultrasound-assisted deep eutectic solvents extraction (UAE-DESs) was developed for extraction and determination of the linarin from Chrysanthemum indicum L. flower (CIF). And a feasible strategy was performed to separate and recover the linarin from denatured DESs using AB-8 macroporous resins as adsorbents. Under the optimal extraction conditions, the maximum linarin extraction yield reached 14.23 mg/g with liquid–solid ratio 32 mL/g, extraction power 340 w, extraction time 32 min and deep eutectic solvent of choline chloride-ethylene glycol with the molar ratio 1:2 and water content 30% (v/v) as a extraction system, which was 1.21 times higher than 80% ethanol extraction. After macroporous resins enrichment, the recovery efficiency of linarin reached 81.55%. In addition, the DESs could be regenerated for extraction process, and macroporous resins could be further used for enrichment process with desired performance. This study provided an effective and convenient extraction and recovery method for linarin determination from CIF which was also applicable to other natural plants.

Highly Efficient Enzymatic Conversion of Rutin to Isoquercitrin and l-Rhamnose Using Deep Eutectic Solvents

Deqing Wang,Pu Zheng,Pengcheng Chen,Dan Wu

Deep eutectic solvents (DESs), considered to be “green media”, have recently been employed in multiple applications including biotransformations. Here, a DES was used in the production of isoquercitrin and l-rhamnose from rutin by enzymatic transformation with recombinant α-l-rhamnosidase from Aspergillus niger. A solution synthesized from choline chloride and urea (ChCl-U) performed best in the conversion of rutin among 24 kinds of DESs. When 130 g/L rutin was used as the substrate, the conversion rate was 100% and the productivity of l-rhamnose and isoquercitrin was 208.68 mM/h under optimized conditions in ChCl-U solution, which was 1.12 times higher than that in buffer (186.92 mM/h). The recycling of ChCl-U solution was also investigated by aqueous two-phase system extraction of l-rhamnose, and ChCl-U/K2HPO4 was screened as the suitable extraction solvent. When the ChCl-U content was 34%, the extraction efficiency of l-rhamnose was 79.06% with a purity and primary crystallization yield of 96.28 and 66.49%, respectively. After five cycles of ChCl-U solution, the average enzymatic conversion rate of recycling was 95.63% with an average purity of 94.22% of the isoquercitrin prepared. After 10 cycles of reuse, the residual conversion rate was 61.14%. The results indicate that the DES has obvious advantages in biocatalytic processes involving flavonoid conversion.

Melatonin mitigates cadmium and aluminium toxicity through modulation of antioxidant potential in Brassica napus L

A. Sami,F. A. Shah,M. Abdullah,X. Zhou,Y. Yan,Z. Zhu,K. Zhou

Melatonin has emerged as an essential molecule in plants, due to its role in defence against metal toxicity. Aluminium (Al) and cadmium (Cd) toxicity inhibit rapeseed seedling growth. In this study, we applied different doses of melatonin (50 and 100 µ m ) to alleviate Al (25 µ m ) and Cd (25 µ m ) stress in rapeseed seedlings. Results show that Al and Cd caused toxicity in rapeseed seedling, as evidenced by a decrease in height, biomass and antioxidant enzyme activity. Melatonin increased the expression of melatonin biosynthesis-related Brassica napus genes for caffeic acid O-methyl transferase ( BnCOMT ) under Al and Cd stress. The genes BnCOMT-1 , BnCOMT-5 and BnCOMT-8 showed up-regulated expression, while BnCOMT-4 and BnCOMT-6 were down-regulated during incubation in water. Melatonin application increased the germination rate, shoot length, root length, fresh and dry weight of seedlings. Melatonin supplementation under Al and Cd stress increased superoxide dismutase, catalase, peroxidase, ascorbate peroxidase, proline, chlorophyll and anthocyanin content, as well as photosynthesis rate. Both Cd and Al treatments significantly increased hydrogen peroxide and malondialdehyde levels in rapeseed seedlings, which were strictly counterbalanced by melatonin. Analysis of Cd and Al in different subcellular compartments showed that melatonin enhanced cell wall and soluble fractions, but reduced the vacuolar and organelle fractions in Al- and Cd-treated seedlings. These results suggest that melatonin-induced improvements in antioxidant potential, biomass, photosynthesis rate and successive Cd and Al sequestration play a pivotal role in plant tolerance to Al and Cd stress. This mechanism may have potential implications in safe food production.
