Currently, mainstream white light-emitting diodes (WLEDs)
fabricated
by combining a blue LED chip with Y 3 Al 5 O 12 :Ce 3+ yellow phosphor suffer from poor color rendering,
unstable color temperature, and blue light hazards due to the lack
of a red spectral component. In this study, a low-cost, easily synthesizable,
nontoxic, and highly stable red-emitting Cu(I)-based hybrid material, Cu 2 I 2 (2-et-3-mepz) (2-et-3-mepz = 2-ethyl-3-methylpyrazine),
is developed. This red-emitting material exhibits intense red emission
under ultraviolet/blue light excitation, with an emission peak at
628 nm and CIE chromaticity coordinates of (0.628, 0.372), which are
close to the standard red light. Experimental and theoretical analyses
reveal that its luminescence mechanism primarily originates from hybrid
metal/halide-to-ligand charge transfer ( 3 M/HLCT) and cluster-centered
( 3 CC) transitions. A WLED device fabricated by combining Cu 2 I 2 (2-et-3-mepz) with commercial blue and green phosphors
achieves an Ra value as high as 93.0. Furthermore, Cu 2 I 2 (2-et-3-mepz) also exhibits excellent scintillation performance
with a light yield of 12,647 photons MeV –1 . A flexible
scintillation screen prepared using this material achieves a spatial
resolution of 9.45 lp mm –1 , demonstrating its potential
application value in X-ray imaging. This work not only achieves high-performance
WLED devices but also provides a new strategy for developing low-cost,
environmentally friendly red-emitting materials.

Electrochemical CO 2 reduction to highly selective ethylene is particularly desirable yet challenging. The competition between methane and ethylene in the CO 2 RR process underscores the critical necessity of designing the catalyst for ethylene by suppressing the methane pathway. Herein, we introduced controllable Cu(II)/Cu(I) interfaces by regulating the ratio of ionic liquids (ILs) and H 2 O. The Cu x O-IL4 catalyst with main CuO/Cu 2 O structure, exhibits a high Faraday efficiency toward C 2 H 4 (∼54 %) and less CH 4 (∼1 %) under 250 mA·cm −2 current density at −1.18 V (vs RHE). Experimental and theoretical analyses demonstrate that Cu(I) sites favor CO bridge formation, while Cu(II) sites preferentially generate CO atop on Cu x O-IL4 catalysts. The Cu(II)/Cu(I) interface in Cu x O-IL4 facilitates the generation of CO bridge intermediates, thereby lowering the energy barrier for *CO dimerization and also preventing *CO hydrogenation to CH 4 formation, thus enhancing the selectivity of CO 2 -to-ethylene conversion. The study provides a novel approach for precisely controlling CO adsorption configurations to enhance CO 2 RR selectivity toward high-value multicarbon products formation.

Comprehensive Summary Traditional inorganic single-crystal scintillators face several adverse conditions, such as high cost, complex process and poor flexibility. To avoid these shortcomings, in this work, the low-cost benzyl bromide is used to efficiently synthesize the 0D organic-inorganic hybrid copper(I) halide compound Cu 5 I 4 Br 3 (bz-ted) 2 (Compound Y ) by the room-temperature solution method. The yields of the ligand (95%) and the scintillator (97%) are nearly 100%, and a single-batch production of 10.29 g is achieved, while the cost of the ligand raw material is drastically reduced. This material has an “all-in-one” (AIO) structure, with intramolecular π···π stacking interaction and Cu···Cu cuprophilic interactions together enhancing structural rigidity, resulting in its remarkable thermal, air, and light stability. Notably, Compound Y exhibits excellent yellow light-emitting properties with an outstanding photoluminescent quantum yield (PLQY = 90.66%) and an ultrahigh external quantum efficiency (EQY = 81.59%), which is investigated to be the emission from the self-trapped excitons (STEs). Moreover, it has favorable scintillation properties, with a low detection limit of 2.426 μGy air ·s −1 and a satisfactory spatial resolution of 17.7 lp·mm −1 . When blended with styrene-ethylene-butene-styrene (SEBS) to fabricate the flexible scintillation screen, it can withstand 200% stretching and 0°–90° bending, with no halo distortion in imaging. This work not only promotes the commercial application of AIO-structured materials, but also provides a new approach for the large-scale fabrication of low-cost and high-performance flexible X-ray scintillators.

Flavonoid compounds in Tetrastigma hemsleyanum Diels et Gilg ( T. hemsleyanum ) are essential for plant stress responses, yet their biosynthetic pathways and regulatory mechanisms remain poorly understood. This study identified and characterized two flavanone-3-hydroxylase ( F3H ) genes, Th F3H1 and Th F3H2, from T. hemsleyanum , which belong to the 2ODDs-DOXC28 subfamily. Tissue-specific expression analysis revealed that both genes are highly expressed in tuberous roots, with distinct stress response patterns observed. Sequence and phylogenetic analyses confirmed that these genes contain typical structural domains of the 2ODD family and are closely related to F3H proteins from various plant species. Subcellular localization studies indicated that Th F3H1 is predominantly cytoplasmic, while Th F3H2 is found both in the cytoplasm and the nucleus. In vitro enzymatic assays demonstrated that both enzymes catalyze the conversion of naringenin to dihydrokaempferol and small amounts of apigenin, with Th F3H2 exhibiting higher catalytic efficiency ( k cat / K m = 0.0093 s −1 ·μM −1 ). AlphaFold2 predictions and molecular docking analysis revealed the binding modes of Th F3H1 with itssubstrates. Site-directed mutagenesis further identified four key catalytic residues (Glu122, Lys196, Lys326, and Asp330). Furthermore, both genes were significantly upregulated under salt stress, and their heterologous expression enhanced the salt tolerance of Escherichia coli , underscoring their critical roles in plant stress responses. These findings enhance the understanding of plant F3H function and provide a molecular foundation for improving the quality and stress resistance of T. hemsleyanum .

Cardiovascular diseases remain a leading cause of death worldwide, with acute myocardial infarction (AMI) accounting for a significant proportion of fatalities. The rapid and accurate detection of cardiac troponin I (cTnI) is critical for early diagnosis of AMI. We report an innovative, highly sensitive, solidly mounted resonator (SMR) biosensor based on a c-axis-tilted scandium-doped aluminum nitride (Sc0.1Al0.9N) film for label-free cTnI detection. The SMR device, fabricated by magnetron sputtering and incorporating an acoustic Bragg reflector, is designed to operate in the shear acoustic mode. The sensor demonstrates a quality factor of 2030 and an effective electromechanical coupling coefficient of 2.6%. Functionalized with a self-assembled monolayer and monoclonal antibodies, the sensor achieves a detection limit of 3 pg/mL for cTnI in serum, with a rapid response time of 100 s and high selectivity against common interferents. The device also exhibits significant potential for integration with CMOS electronics, supporting the development of miniaturized, point-of-care (POC) diagnostic systems for cardiovascular diseases.

Electrochemical sensing devices often suffer from biofouling due to nonspecific adsorption of biomolecules and microorganisms in complex biological matrices. To address this challenge, we developed an efficient antifouling and self-sterilizing sensing interface by grafting polyethylene glycol (PEG) onto two-dimensional Ti 3 C 2 T x MXene nanosheets for the accurate detection of cardiac troponin I (cTnI), a gold-standard biomarker for acute myocardial infarction (AMI). The Mn 2+ -intercalated Ti 3 C 2 T x (Mn-Ti 3 C 2 T x ) exhibited enhanced electrochemical activity in a three-electron oxygen reduction reaction (3e - ORR), enabling in situ self-sterilization by activating molecular oxygen to generate hydroxyl radicals (•OH), which demonstrated potent antibacterial effects against both Gram-negative and Gram-positive bacteria. Density functional theory calculations revealed that Mn doping in MXene significantly reduced the free energy barrier of *OOH intermediate, promoting H 2 O 2 formation and subsequent reduction to •OH. The PEGylated Mn-Ti 3 C 2 (PEG@Mn-Ti 3 C 2 T x ) composite combined the superior hydrophilicity of PEG for effective protein repulsion with the enhanced conductivity of Mn-Ti 3 C 2 T x , enabling sensitive biosensing. Critically, the fabricated biosensor demonstrated high specificity for cTnI against common interferents and achieved reliable detection in spiked serum samples. It exhibited a broad detection range from 0.1 pg mL -1 to 500 ng mL -1 with an ultralow limit of 0.03 pg mL -1 . This work presents a novel strategy for combating biofouling and paves the way for designing robust biosensors capable of accurate and long-term operation in clinically relevant environments, holding great promise for point-of-care diagnostics.

Ovarian cancer is a deadly gynecological malignancy often diagnosed at an advanced stage, with few treatment options available. Polyphyllin I (PPI), the primary active ingredient of Paris polyphylla , has shown excellent anti-cancer activity against various cancers. However, studies on PPI use and ovarian cancer are limited, and the potential mechanisms remain unclear. This study explored the effects of anti-ovarian cancer and the potential mechanisms of action of PPI. Methods such as MTT, colony formation, three-dimensional spheroid formation, flow cytometry, and western blotting were employed to assess the anti-proliferative effects of PPI on different ovarian cancer cell lines. RNA sequencing was used to discover potential mechanisms by evaluating the effects of PPI on transcriptional signatures and identifying differences in gene expression patterns. The results indicate that PPI demonstrated dose- and time-dependent anti-proliferative effects on ovarian cancer cell lines. PPI also up-regulated 1518 genes and down-regulated 800 genes. The genes showed enrichment in the “apoptosis” gene sets, while exhibiting negative enrichment in the “cell cycle” and “Myc signaling pathway” gene sets. PPI caused cell cycle arrest at the G0/G1 phase by increasing the expression of p21 and p27 and inhibiting the activity of the CDK2/cyclin E complex. PPI triggered cell apoptosis, which was associated with elevated cellular reactive oxygen species and mitochondrial membrane potential depolarization. Finally, PPI reduced c-Myc protein levels and affected mRNA levels of c-Myc pathway-related genes. PPI effectively suppressed ovarian cancer cell proliferation, indicating its potential as a treatment option for ovarian cancer.

The stimulator of interferon genes (STING) has emerged as a promising target for cancer immunotherapy. However, STING agonists alone have demonstrated limited clinical success. Indoleamine 2,3-dioxygenase 1 (IDO1) has been implicated as a key resistance mechanism that undermines the antitumor effects of STING activation, yet the cell-type-specific roles and the combined impact of STING and IDO1 on tumor immune orchestration remain poorly understood. In this study, we report that a co-delivery nanosystem of STING agonists and IDO1 inhibitors, rather than their simple combination, yields synergistic antitumor responses. This nanomedicine exhibits tropism toward myeloid cell populations in vivo , particularly dendritic cells (DCs), and facilitates pH-responsive cytosolic delivery of both agents. As a result, it enhances antigen cross-presentation, DC maturation, and cytokine secretion, driving effective priming of cytotoxic T lymphocytes. Compared to monotherapies or physical nanoparticle combinations of STING agonists and IDO1 inhibitors, this two-in-one nanomedicine significantly suppresses tumor growth and confers a survival benefit. Depletion of STING in the host ( Tmem173 −/− mice) or deficiency in type I conventional DCs (cDC1s, Batf3 −/− mice) abolishes therapeutic efficacy, whereas depletion of STING in tumor cells exerts minimal impact. A bone marrow chimera experiment further supports the STING-cDC1 dependence. Its capability to overcome tumor resistance and the underlying mechanism were further investigated in a well-established tumor model. Our findings highlight the cooperative role of STING activation and IDO1 blockade within cDC1s and introduce an integrated drug delivery system to combat immune tolerance in cancer therapy.

Upconversion nanoparticles (UCNPs)-based fluorescence lateral flow immunoassay (FLFIA) has significant application prospects in clinical diagnosis, but it still suffers from relatively low sensitivity in detecting trace amounts of biomarkers. Here, we report for the first time a strong fluorescence-emitting core-shell-shell NaLuF 4 @NaLuF 4 :Yb,Er@NaLuF 4 UCNP as the FLFIA probe. The UCNPs-based FLFIA platform enables point-of-care detection of cardiac troponin I (cTnI) with high sensitivity, accuracy, and resistance to autofluorescence interference. NaLuF 4 :Yb,Er encapsulated in the intermediate shell layer can effectively reduce the cross-relaxation and surface quenching of the luminescence center, resulting in a higher quantum yield. Compared with the conventional core-shell NaLuF 4 :Yb,Er@NaLuF 4 UCNPs, the fluorescence intensity increases by 2.4 times. The UCNPs-based FLFIA was further integrated with a portable smartphone platform for rapid, on-site detection. The UCNPs probe enabled sensitive cTnI detection, with a detection limit of 0.035 ng/mL in buffer and 0.059 ng/mL in serum, and a detection range of 0.05–100 ng/mL. In testing clinical samples, the results from the platform showed excellent correlation with hospital results. We anticipate that the NaLuF 4 @NaLuF 4 :Yb,Er@NaLuF 4 UCNPs can serve as a promising immunolabeling nanoprobe for highly sensitive and accurate FLFIA detection. Graphical

A highly phosphorescent molecular Au(I) cluster capable of rapid, sensitive, and selective detection of cyanide has been successfully fabricated. The origin of the outstanding sensing performance of the molecular Au(I) cluster toward cyanide is justified by X-ray absorption near edge structure (XANES) and extended X-ray absorption fine structure (EXAFS) analyses. The response mechanism employed with the molecular Au(I) cluster and the cost-effectiveness in cyanide detection affords several key sensor features, making this molecular Au(I) cluster-based sensor unique compared to other cyanide sensing schemes. Importantly, by exploring the phosphorescent properties of the molecular Au(I) cluster in solid state, we demonstrate the first example of the molecular gold(I) cluster-based macroporous sensing film for colorimetric detection of cyanide in complex samples, including red wine, coffee, juice, and soil. Remarkably, the as-prepared sensing film inherits the sensing ability of the molecular Au(I) cluster, and offers a high mechanical flexibility and novel opportunities for real-time monitoring cyanide release in cassava manufacturing.

Ethylene is a crucial gaseous phytohormone that regulates diverse aspects of plant growth and development. In Arabidopsis thaliana , perception of ethylene is mediated primarily by ETHYLENE RESPONSE 1 (ETR1), a prototypic membrane protein that couples ligand sensing to signal transduction through a transmembrane copper cofactor-dependent ethylene-binding site and a cytoplasmic histidine kinase domain. Previous work established that ETR1 binds copper and forms covalent homodimers, but the functional significance of its dimeric architecture in signal transduction has remained elusive. In this study, we expressed and purified full-length ETR1 from Pichia pastoris, enabling the systematic dissection of its oligomeric state, copper redox chemistry, and ligand binding effects. Using an in vitro luminescent kinase assay, we demonstrate that the Cu(I)-ethylene complex strongly suppresses ETR1 autophosphorylation, but only when the receptor is in its disulfide-linked dimer form. Monomeric ETR1 generated by reductive cleavage retained basal kinase activity yet was insensitive to ethylene. Furthermore, the ethylene antagonists Ag + and 1-MCP reversed the ligand-induced suppression. Mutational analysis revealed that the H353Q variant lacked kinase activity, while the C65S/H69A double mutant retained basal activity but was resistant to ethylene-induced inhibition, implicating both the catalytic histidine residue and the copper-binding site in the regulatory mechanism. Together, these findings establish that ETR1 functions as a dimer-dependent ethylene sensor, in which covalent dimerization is essential for allosteric coupling between the ligand-binding and kinase domains. This work thus provides a definitive biochemical model for ethylene perception and offers a robust platform for future structural and mechanistic studies of plant membrane-receptor signaling.

Metal halide hybrid luminescent materials have attracted significant attention due to their tunable emission properties and excellent photoelectric performance. However, achieving metal halides that exhibit near-infrared (NIR) emission under blue light excitation remains a major challenge. This paper reports a novel 2D copper(I) iodide hybrid material, Cu 2 I 2 (THQx) (THQx = 5,6,7,8-Tetrahydroquinoxaline), which is constructed by linking -Cu 2 I 2 - chains with the THQx ligand and achieves broadband NIR luminescence. It can be effectively excited by blue LEDs and has a peak emission at 730 nm with a full width at half maximum of 159 nm. Theoretical calculations indicate that its luminescence originates from the triplet metal/halide-to-ligand charge transfer ( 3 MLCT/HLCT). This material also exhibits excellent thermal stability, air stability, and photostability, demonstrating potential applications in night vision and biological imaging in NIR phosphor-converted LED (pc-LED) devices. This research not only expands the application boundaries of copper(I) halide hybrid materials, but also provides a new approach for the development of low-cost and environmentally friendly NIR luminescent materials.
