Cuproptosis, a potential target for the therapy of Diabetic Critical Limb Ischemia

Tao Bai,Luhao Wang,Zhentao Qiao,Zhiwei Wang

Diabetic patients are considered as the high risk population to develop critical limb ischemia (CLI), a peripheral vascular disease (PVD) resulted from atherosclerosis. Cuproptosis is a novel copper-dependent cell death that has shown the regulatory role in diabetes, while its effect on diabetic CLI has not been explored yet. In this study, Diabetic CLI mice was induced by femoral artery ligation (FAL) on diabetic mice. Endothelial injury in diabetic CLI was mimicked in human microvascular endothelial cells (HMEC-1) via the induction with high glucose (HG) and nutrient deprivation (ND). Besides, copper chelator Ammonium Tetrathiomolybdate (TM), which has shown the anti-cuproptosis property, was administrated to explore its potential effects on diabetic CLI mice and HG/ND-induced HMEC-1 cells. Strikingly, obvious cuproptosis was found in the gastrocnemius muscles of diabetic CLI mice and HG/ND-induced HMEC-1 cells, as evidenced by the copper overload and dysregulated cuproptosis-related proteins (such as Fe-S cluster proteins, copper exporter ATP7A, and copper importer SLC31A1). More importantly, TM protected against the hindlimb ischemic damages in diabetic CLI mice and alleviated cuproptosis-associated cell deaths in HG/ND-induced HMEC-1 cells. In summary, this study indicates the involvements of cuproptosis in diabetic CLI, and provides novel insights into copper chelator TM on diabetic CLI therapy.

Spectroelectrochemistry study on the surface modification of chalcopyrite by Cu2+ and its depressive response to flotation collector adsorption by negative potential

Kecan Zhang,Shan Lei,Yu Miao,Bao Guo

In the flotation beneficiation of copper and molybdenum co-occurrence ores, the efficient depression of chalcopyrite is crucial for successful copper-molybdenum separation. However, the consumption of conventional depressant NaHS is huge. There is greater susceptibility of chalcocite and other secondary copper sulfide minerals to electrochemical reaction in comparison to chalcopyrite. Given the irreversible desorption behavior of collector molecules on chalcocite upon negative potential application, electrochemically regulated flotation can be effectively utilized to diminish the utilization of conventional depressants. In this work, electrochemistry and shell-separated nanoparticle-enhanced Raman spectroscopy (SHINERS) were used to explore the transformation of chalcopyrite surface to chalcocite-like species under the combined action of Cu 2+ and negative potential. The results show that the similarity of cyclic voltammetry (CV) between the modified chalcopyrite and chalcocite, increases with the pretreatment potential becoming increasingly negative. Electrochemical impedance spectroscopy (EIS) also reflects their similar adsorption and desorption behavior in response to ammonium dibutyl dithiophosphate (ADD), a typical flotation collector molecule. In addition, the spectroelectrochemical directly reflects the irreversible desorption of ADD molecules from the modified chalcopyrite surface at − 0.7 V. The research results lay a theoretical foundation for the efficient Cu-Mo separative flotation by depressing chalcopyrite under the combination of Cu 2+ and negative potential.

Unveiling the Role of Copper Valence States in Enhancing the Catalytic Performance of Copper-Modified ZSM-5 for Direct Methane Conversion

Yunhan Pu,Huajie Zhang,Yanjun Li,Chuan Yu,Xiaofei Song,Chen Yang,Mingli Fu

The conversion of methane (CH4) to methanol (CH3OH) under mild conditions remains a significant challenge in catalysis. In this study, we introduce a method to adjust the surface valence states of copper species in Cu-ZSM-5 catalysts by annealing under different atmospheres (N2, air, and H2). Among these, the 10% Cu-ZSM-5 catalyst calcined in H2showed outstanding performance, achieving a methanol productivity of 8.08 mmol/(gcat·h) and 91% selectivity at 70 °C and 3 MPa using H2O2as the oxidant. Comprehensive characterization revealed that H2annealing optimized the Cu surface to a lower valence state (predominantly Cu+), enhancing CH4adsorption and promoting H2O2activation to generate ·OH and ·CH3radicals, which drive selective CH3OH formation. In situ DRIFTS and radical trapping experiments further confirmed the critical role of Cu+in facilitating C-H bond cleavage and suppressing overoxidation.

Multi-dimensional molybdenum-doped CuCoSe nanoflowers/CdSe nanodots/CuO nanowires for asymmetric supercapacitors

Xingyue Wu,Baozhu Yang,Yiping Qiu,Junyao Li,Yong Qin,Yong Kong

Three-dimensional (3D) Mo-doped CuCoSe (Mo-CuCoSe) nanoflowers and zero-dimensional (0D) CdSe nanodots are successively grown on the surface of one-dimensional (1D) CuO nanowires through a two-step electrodeposition strategy. As the supporting matrix, the 1D CuO nanowires can greatly alleviate the aggregation of the Mo-CuCoSe. The 3D Mo-CuCoSe nanoflowers show unique hydrangea-like structure, which can not only enhance the diffusion of electrolyte ions/electrons, but also facilitate the Faraday redox reactions. The 0D CdSe nanodots play a pivotal role in the enhancement of capacitance retentions. Due to the synergistic effect of CuO nanowires, Mo-CuCoSe nanoflowers and CdSe nanodots, the multi-dimensional CuO/Mo-CuCoSe/CdSe shows a high specific capacitance of 4600 mF cm −2 (at 1.0 mA cm −2 ) and excellent cyclic stability. Further, activated carbon (AC) and the multi-dimensional CuO/Mo-CuCoSe/CdSe are assembled to construct an asymmetric supercapacitor (ASC), which exhibits a maximum energy density of 30.3 Wh kg −1 at 214.2 W kg −1 .

Cu-Deficient CuInSe Quantum Dots for “Turn-On” Detection of Adenosine Triphosphate in Living Cells

Jilei Chen,Lu Zhang,Shulan Li,Feng-Lei Jiang,Peng Jiang,Yi Liu

Copper indium selenide (CISe) quantum dots (QDs) are a class of extremely appealing candidates for various biological applications due to their composition- and size-dependent photoluminescence and low toxicity. However, the synthesis of high-quality water-soluble CISe QDs with controlled composition in the aqueous phase is still a great challenge, which limits their biological applications. Moreover, the effects of the copper content in CISe QDs on the fluorescence and the corresponding mechanism are still subject to debate. Herein, a composition-tunable aqueous synthesis of high-quality Cu-deficient CISe QDs was presented, and the effects of the copper content on the fluorescence of CISe QDs was studied by the time-resolved fluorescence. The composition of CISe QDs could be flexibly controlled by manipulating the reaction temperature, the reaction time, and the Cu/In precursor ratio. The time-resolved fluorescence of CISe QDs with different Cu/In ratios suggested that the donor–acceptor pair (DAP) recombination was enhanced gradually with the increase of copper vacancies, resulting in the enhancement of fluorescence. However, if copper vacancies achieved an excessive concentration, the fluorescence would decrease possibly due to Auger recombination. The optimized CISe QDs with the maximum quantum yield of 10.3% were obtained with the Cu/In ratio, reaction temperature, and reaction time of 1/5, 170 °C, and 1 h, respectively. Based on the intense emission of the Cu-deficient CISe QDs, a highly sensitive “turn-on” fluorescent probe was developed for the detection of adenosine triphosphate (ATP) in living cells with a limit of detection of 56 nM. This work provides a paradigm for the aqueous-phase synthesis and applications of ternary I–III–VI QDs.

Microplasma direct writing of a copper thin film in the atmospheric condition with a novel copper powder electrode

Tao Wang,Li Lv,Liping Shi,Baohong Tong,Xingquan Zhang,Guotao Zhang,Jingquan Liu

This study presents the microplasma direct writing of copper film in the open air using a novel copper powder electrode. The powder electrode acted as a high-voltage electrode and the precursor source. The deposition process is realized through ion sputtering of the powder in the microplasma. The bombarded copper nanoparticles were deposited and accumulated on a silicon substrate. The deposited film is composed of copper nanoparticles with sizes in the range from tens to hundreds of nanometers, and its electrical conductivity could reach 3.96 × 10 5 S/m. The morphologies, chemical compositions and electrical property of the thin film can be adjusted by changing the input current, and maskless film deposition with different micropatterns can be achieved by direct microplasma scanning.

A Defective Disc-Like Cu1.96S Anode Material with the Efficient Cu Vacancies for High-Performance Sodium-Ion Storage

Zhihao Wang,Yongyi Lu,Guangdi Zhang,Lingfeng Quan,Mingzu Liu,Haimei Liu,Yonggang Wang

Due to their significant capacity and reliable reversibility, transition metal sulphides (TMSs) have received attention as potential anode materials for sodium-ion batteries (SIBs). Nonetheless, a prevalent challenge with TMSs lies in their significant volume expansion and sluggish kinetics, impeding their capacity for rapid and enduring Na + storage. Herein, a Cu 1.96 S@NC nanodisc material enriched with copper vacancies is synthesised via a hydrothermal and annealing procedure. Density functional theory (DFT) calculations reveal that the incorporation of copper vacancies significantly boosts electrical conductivity by reducing the energy barrier for ion diffusion, thereby promoting efficient electron/ion transport. Moreover, the presence of copper vacancies creates ample active sites for the integration of sodium ions, streamlines charge transfer, boosts electronic conductivity, and, ultimately, significantly enhances the overall performance of SIBs. This novel anode material, Cu 1.96 S@NC, demonstrates a reversible capacity of 339 mAh g −1 after 2000 cycles at a rate of 5 A g −1 . In addition, it maintains a noteworthy reversible capacity of 314 mAh g −1 with an exceptional capacity retention of 96% even after 2000 cycles at 20 A g −1 . The results demonstrate that creating cationic vacancies is a highly effective strategy for engineering anode materials with high capacity and rapid reactivity.

Synergetic Effect of Blended Alkylamines for Copper Complex Ink To Form Conductive Copper Films

Wen Xu,Tao Wang

Cu(II) complex ink consisting of copper formate (Cuf) and a primary alkylamine could yield highly conductive copper films at low heating temperatures without a reducing atmosphere. A synergetic effect of the blended alkylamines on the formation of conductive films was observed. It was found that blending two types of amines with different alkyl chain lengths as ligands could improve the conductivity of copper films, compared with using one of these amines alone. The decomposition mechanism of the Cuf–amine complex and the role of amines with different alkyl chain lengths were investigated. It was found that the decrease in the decomposition temperature and the formation of copper films were attributed to the activating effect and capping effect of the amine, and these two effects were dependent on the alkyl chain length. The relative intensity of the dual effects determined the decomposition rate of the complex and the nucleation and growth of particles. The use of blended amines with different alkyl chain lengths as ligands could balance the two effects and lead to appropriate nucleation and growth rates, so that densely packed copper films with low resistivity could be obtained at low heating temperature in a short time. The Cuf–butylamine–octylamine (Cuf–butyl–octyl) ink with 1:1 molar ratio of the amines showed the best performance. The understanding of the synergetic effect could provide guidance to the design of copper complex inks to control the morphology of the films.

Reversal of cisplatin chemotherapy resistance by glutathione-resistant copper-based nanomedicine via cuproptosis

Yao Lu,Qingqing Pan,Wenxia Gao,Yuji Pu,Bin He

Platinum-based chemotherapy is widely used to treat various cancers. However, exogenous platinum is likely to cause severe side effects and drug resistance induced by upregulated glutathione (GSH) in cancer cells poses a threat to the management of cancer progression and recurrence. Anticancer copper–organic complexes are excellent candidates to substitute platinum-based chemotherapeutics, exhibiting lower systemic toxicity and even overcoming platinum-based chemotherapy resistance. Here, we report the GSH-resistance of copper(II) bis(diethyldithiocarbamate) (CuET) and its reversal of cisplatin resistance in non-small-cell lung cancer via cuproptosis. Electrochemistry and UV-vis spectroscopy studies demonstrate that CuET possesses a lower reduction potential and the reaction inertness with GSH. Importantly, CuET overcomes the drug resistance of A549/DDP cells and the anticancer effect is hardly affected by intracellular GSH levels. To improve the solubility and bioavailability, bovine serum albumin-stabilized CuET nanoparticles (NPs) are prepared and they have a high drug loading content of 27.5% and excellent physiological stability. In vitro studies manifest that CuET NPs augment the distributions in the cytosol and cytoskeleton, inducing cell death via cuproptosis in A549/DDP cells, which is distinctly different from the apoptosis pattern induced by cisplatin. In vivo antitumor evaluation shows that the nanomedicine has superior biosafety and potent antitumor activity in a cisplatin-resistant tumor model. Our study suggests that copper–organic complex-based nanosystems could be a powerful toolbox to tackle the platinum-based drug resistance and systemic toxicity concerns.

Co-treatment of copper smelting slag and gypsum residue for valuable metals and sulfur recovery

Longgong Xia,Shuheng Cao,Qihou Li,Xingwu Lu,Zhihong Liu

Copper smelting slag and gypsum residue are by-products in copper making processes, and both contain valuable elements, such as copper, sulfur, etc. However, there is more than 0.25 wt% Cu in the smelting slag, which cannot be recovered with present technologies. A sulfidation process, which used the gypsum residue as the sulfur resource, was proposed and studied in treating copper smelting slag. Obtained results showed: 1) CaSO 4 transformed to CaS under reductive decomposing conditions, and reactions between CaS and slag components occurred spontaneously during the slag cooling process; 2) the Fe/SiO 2 mass ratio of the slag, the CaS excess coefficient (α), and the cooling rate (v) had an influence on the terminal phase composition, sulfide particle size, etc.; 3) under optimized conditions (Fe/SiO 2 =1.0: α=5, v  = 1.00°C/min; Fe/SiO 2 =1.3: α=4, v  = 0.25°C/min; Fe/SiO 2 =1.8: α=5, v  = 0.25°C/min), the Cu contents in non-sulfide phases were lower than 0.08 wt%, and the mean particle size of sulfides were 37- 40 μm, which were feasible for flotation recoveries. TCLP (Toxicity Characteristic Leaching Procedure) tests indicated that the co-treatment process improved the stability of these starting materials. Based on samples characterization and thermodynamic analysis, a corresponding reaction mechanism was proposed.

MiR-133a-5p Facilitates Cuproptosis in Hepatocellular Carcinoma Through Targeting of ATP7B

Qiaohui Ren,Xinyue Zhu,Nannan Wang,Kang Yu,Wei Lv,Lianzi Wang,Yan Zang,Dongyue Ma,Xinyi Zhou,Junxiao Yao,Mengjiao Shen,Li Yu,Tao Li

Purpose We explored the effects of miR-133a-5p and ATP7B on cuproptosis in hepatocellular carcinoma. Methods Initially, we assessed the impact of miR-133a-5p on hepatocellular carcinoma (HCC) using CCK-8 assays, cell scratch assays, and flow cytometry. Subsequently, we utilized elesclomol in combination with copper ions to induce cuproptosis in the HCC cell lines PLC/PRF/5 and Huh-7. We evaluated the influence of miR-133a-5p on cuproptosis using CCK-8 assays, cell scratch assays, flow cytometry, and Western blotting. To elucidate the underlying mechanisms, we employed bioinformatics to identify potential downstream target genes of miR-133a-5p and conducted dual-luciferase reporter assays to confirm the binding sites. Finally, we validated the regulatory effect of miR-133a-5p on ATP7B by modulating miR-133a-5p expression through cell transfection experiments. Results The results from the CCK-8 assay, cell scratch assay, and flow cytometry demonstrated that miR-133a-5p significantly inhibits the proliferation and migration of HCC cells while promoting their apoptosis. Furthermore, Elesclomol in combination with copper ions induces cuproptosis in HCC cells. Compared to the cuproptosis observed in HCC as a control, miR-133a-5p further suppresses the proliferation and migration of HCC cells, enhances their death, and increases the expression of cuproptosis-related proteins more prominently. Bioinformatics analysis suggested that ATP7B might be a downstream target gene of miR-133a-5p. This was confirmed by dual luciferase assays, which identified a binding site between miR-133a-5p and ATP7B. Additionally, the expression levels of ATP7B were found to decrease or increase in response to the regulation by miR-133a-5p.Conclusion MiR-133a-5p facilitates cuproptosis in hepatocellular carcinoma through targeting of ATP7B.

Thermal interface material with graphene enhanced sintered copper for high temperature power electronics

Shaojia Deng,Xin Zhang,Guowei David Xiao,Kai Zhang,Xiaowu He,Shihan Xin,Xinlu Liu,Anhui Zhong,Yang Chai

Sintered nano-copper is becoming a promising candidate as thermal interface material (TIM) for die attaching in high power electronics. It exhibits much higher thermal conductivity and operating temperature than conventional TIMs based on polymer and solder joints, and higher electromigration resistance and lower cost than sintered nano-silver TIM. However, the performance of existing sintered nano-copper is lower than expected because of high porosity resulted from poor sintering of copper particles with oxide shell. Here we demonstrate a method of improving the thermal conductivity of sintered copper by addition of graphene/Cu–CuxO with controllable diameter of ∼163 nm. The measured thermal conductivity of the sintered composite TIM is enhanced by up to 123.5% compared with that of sintered pure copper. It can be understood as a result of the formation of graphene heat transfer network in sintered TIM. In addition, the C–O–Cu bonds formed at the interface between graphene and copper nanoparticles are critical for improving thermal performance as well as electrical and mechanical performance of the TIM. The developed TIM can be widely used in high power electronic packaging especially for high temperature applications, including IGBT, SiC and GaN power devices.
