Konjac glucomannan/Pullulan films incorporated with cellulose nanofibrils-stabilized tea tree essential oil Pickering emulsions

Nitong Bu,Liying Huang,Guoyu Cao,Huanglong Lin,Jie Pang,Lin Wang,Ruojun Mu

Biopolymers , such as polysaccharides are increasingly concern in preparation of food packaging. However, those materials have weakness in properties of mechanics, hydrophobicity , and antibacterial, which limit their applications. In this work, we prepared emulsion (K-PL-CEOs) films with double network (DN) matrix by using konjac glucomannan (KGM) and Pullulan (PL), and incorporated with cellulose nanofibrils (CNF)-stabilized tea tree essential oil (TTEO) Pickering emulsions (CEOs). The effect of CEOs content on the structural and physicochemical properties of K-PL films was investigated. The results showed that CEOs were well dispersed in K-PL film forming matrix. The addition of CEOs could improve the thermal stability, UV-shielding, water resistance and mechanical properties of K-PL-CEOs films. Moreover, K-PL-CEOs films exhibited good antibacterial activity against E. coli and S. aureus . This paper demonstrated that K-PL-CEOs films can be potentially applied as food packaging.

Covalently phenolated-β-lactoglobulin-pullulan as a green halochromic biosensor efficiency monitored Barramundi fish's spoilage

Ibrahim Khalifa,Krisana Nilsuwan,Thummanoon Prodpran,Soottawat Benjakul

The effect of the covalent binding between anthocyanins extracted from purple potato peels and beta-lactoglobulin (β-Lg) on its ability to fabricate a green/smart halochromic biosensor combined with pullulan (Pul) was studied. The physical, mechanical, colorimetry, optical, morphological, stability, functionality, biodegradability , and applicability of β-Lg/Pul/Anthocyanin biosensors to monitor the Barramundi fish's freshness during storage were entirely evaluated. The docking and multispectral results proved that β-Lg could be successfully phenolated with anthocyanins and subsequently interacted with Pul via H-bonding and other forces which mainly subsequently form the smart biosensors. Phenolation with anthocyanins significantly heightened the mechanical, moisture resistance, and thermal steadiness of β-Lg/Pul biosensors. Anthocyanins also nearly duplicated the bacteriostatic and antioxidant activities of β-Lg/Pul biosensors. The biosensors changed the color associated with the loss in freshness of the Barramundi fish, mostly due to the ammonia production and pH-alteration throughout fish deterioration. Most importantly, β-Lg/Pul/Anthocyanin biosensors are biodegradable and decomposed within ∼30 d of simulated environmental circumstances. Overall, β-Lg/Pul/Anthocyanin smart biosensors could minimize the usage of plastic packaging materials and employ to monitor the freshness of stored fish and fish-stuffs.

Ting Su,Hongxia Shang,Xinru Su,Zhixian Sun,Yuehan Li,Linwen Li,Zhen Zhang,Renyong Geng,Shuisheng Chen

In this investigation, a hydrogel adsorbent featuring remarkable efficiency in dye adsorption was successfully synthesized by the integration of natural polysaccharide (pullulan) and nanoparticles (ZIF-8@PDA). The prepared natural polysaccharide nanocomposite hydrogels not only exhibit superior mechanical strength and biocompatibility, but also demonstrate adeptness in the removal of dye pollutants. The dye removal capacities were 615.4 mg/g for malachite green (MG) and 525.8 mg/g for Congo red (CR), respectively. Notably, the adsorption process exhibits minimal susceptibility to variations in water quality and the presence of co-existing ions. The pH-responsive surface charge conversion capability of the adsorbent renders it recyclable, maintaining a dye adsorption performance exceeding 88 % even after 5 cycles of repeated usage. Overall, these environmentally friendly natural polysaccharide nanocomposite hydrogels hold potential for addressing complex wastewater treatment challenges and long-term use.

High temperature degradable pullulan microsphere/polymer composite for filtration loss control of water-based drilling fluids

Zhengwen Lin,Fang Li,Xinyu Liu,Junlin Su

The effective control of water-based drilling fluid filtration rates in deep and ultra-deep wells poses a significant challenge. This study introduced a novel approach utilizing pullulan microsphere (PM) grafted with N,N-dimethylacrylamide, 2-acrylamido-2-methylpropanesulfonic acid, and dimethyl diallyl ammonium chloride as a filtrate reducer (PM-DAD). The polymer's structure was analyzed through Fourier transform infrared spectroscopy, nuclear magnetic resonance, thermogravimetric analysis, and scanning electron microscopy. Structural characterization confirmed the successful polymerization and good thermal stability of PM-DAD. PM-DAD showed the pH response characteristics of particle size decreasing with the increase of alkalinity and the temperature response characteristics of degradation into nanoparticles after high temperature aging. The filtration loss and rheological properties of PM-DAD were assessed in accordance with the American Petroleum Institute standards. The rheological evaluation results indicated that PM-DAD could effectively maintain optimal rheological properties of the base mud after aging. PM-DAD showed remarkable filtration loss reduction ability. After aging at 200 °C, the filtration loss of the base mud was 4.7 mL, and that of the base mud containing 15 wt% NaCl was 8.2 mL. The filtration reduction mechanism of PM-DAD was investigated through mud cake analysis, particle size distribution, and X-ray diffraction. The filtration loss reduction mechanism showed that degraded nano PM-DAD intercalated between smectite clay minerals, creating electrostatic attraction and hydrogen bonds with the smectite surface, thereby facilitating the formation of a dense filter cake This research not only presents a novel approach for the utilization of pullulan but also offers a new path for controlling high-temperature filtration in water-based drilling fluids.

Tiantian Min,Xiaoli Sun,Liping Zhou,Haiyu Du,Zhu Zhu,Yongqiang Wen

Pathogenic microorganisms posed perniciousness for postharvest fruits and vegetables, as well as brought potential risks for human health. In this work, pullulan/polyvinyl alcohol (PUL/PVA) nanofibers incorporated with thymol-loaded porphyrin metal-organic framework nanoparticles ( [email protected] NPs) were developed for antibacterial food packaging. PCN-224 MOFs not only act as thymol loading carriers but also highly produce singlet oxygen ( 1 O 2 ) with bactericidal activity . PUL/PVA nanofiber was a promising sustainable substrate because of its good flexibility, biocompatibility and biodegradability . The loading capacity of PCN-224 for thymol was about 20%. The [email protected] /PUL/PVA nanofibers exhibited synergistic antibacterial activities against E. coli (~99%) and S. aureus (~98%) under light irradiation . The cell viability assays and fruit preservation study demonstrated good biosafety of the polymeric film. The results suggested that this novel nanofiber has potential application prospects for food packaging.

Preparation of enteric capsules with pulsatile drug delivery potential using pullulan and polyacrylic acid resin III

Shengan Hu,Chongmei Xu,Yinghui Zhang,Yue Du,Jinbao Tang,Lu Chen

In this study, the enteric swelling layer of capsules was prepared by optimizing the ratio of pullulan to gellan gum. The enteric-coating solution was prepared by dissolving polyacrylic acid resin III in 85 % ethanol, and the two were combined to form pullulan enteric-coated capsules (PECs). To elaborate further, Pullulan enteric-coated thin films (PEFs) were fabricated using the casting method, and their oxygen permeability (OP) and water vapor permeability (WVP) were assessed. Enteric capsules were prepared employing a secondary impregnation method. The cap gap and surface roughness of the capsules were characterized using Scanning Electron Microscopy (SEM) and Atomic Force Microscopy (AFM). The water content and hygroscopicity of the capsules were examined by employing the drying weight loss method. The pullulan-based enteric capsules (PECs) exhibited attributes such as low water content, hygroscopicity, brittleness, leakage resistance, and high impermeability. Finally, the in vitro release of aspirin from the enteric capsules was investigated. Results demonstrated the excellent combination of the enteric-coating layer and the swelling layer in the PECs. Compared with Gelatin enteric-coated capsules (GECs) on the market, it had a smaller cap gap, lower brittleness, weaker moisture absorption, and a good dissolution effect. Thus, it met the requirements of Chinese pharmacopeia, i.e., the ability to maintain integrity for 2 h in simulated gastric fluid and a drug-release rate reaching 80 % within 30 min in simulated intestinal fluid. This capsule also exhibited certain characteristics of a pulsatile drug-delivery system. This work proposed a potential method of preparing enteric hard capsules by using pullulan combined with polyacrylic acid resin III.

Eco-friendly Chitosan-Pullulan hydrogels with polyethylene glycol diglycidyl ether for superior adsorption of congo red

Zhu Xiaoji,Hou Haohao,Wang Jie,Ma Hui,Zhou Hongfu,Zang Xiaoling,Xiang Aimin

The urgent necessity to create affordable adsorbents for the effective elimination of leftover contaminants from wastewater arises from the significant water pollution resulting from the excessive use of dyes. A novel chitosan (CS) /pullulan (PL) hydrogel, chemically cross-linked using polyethylene glycol diglycidyl ether (PEGDE), exhibited high swelling capacity, excellent mechanical stability, and superior adsorption performance. Adsorption tests indicated that CS/PL-2 hydrogel attained a 93.23% removal efficiency for congo red (CR) dye at the concentration of 1000 mg/L. The adsorption capacity of CS/PL-2 hydrogel was investigated in relation to various factors, such as temperature, initial concentration, pH, contact time, and the quantity of adsorbent used. Adsorption kinetics for CS/PL-2 hydrogel were examined utilizing both pseudo-first-order and pseudo-second-order models, with the findings demonstrating that physical adsorption primarily governed the process. The maximum adsorption capacity identified for CS/PL-2 hydrogel, as the Langmuir model, was observed to be 953.60 mg/g, and the thermodynamic analysis of adsorption suggested that the process occurs spontaneously. After five cycles of adsorption, the removal rate of CS/PL-2 hydrogel remained at 70%. In conclusion, this hydrogel adsorbent offers a practical solution for treating CR contaminated wastewater.

Gallic acid-grafted chitosan and pullulan as a hydrogen bonding assembled, multifunctional coating for fruit preservation

Haihua Wang,Jianxia Yao,Ying Hou,Zhuoya Liu,Dong Yang

The development of eco-friendly and multifunctional edible coatings is crucial for mitigating postharvest losses of perishable fruits. Hence, a novel composite coating was engineered based on chitosan (CS) and pullulan (PUL) matrix, enhanced by a gallic acid-grafted chitosan (GS) conjugate synthesized through free radical grafting. The GS conjugate served as a multifunctional crosslinker, establishing a dense intermolecular network within the CS-PUL matrix primarily (CP) through hydrogen bonding, as confirmed by structural characterization. This molecular-level interaction significantly improved the mechanical strength (Tensile stress = 4.95 MPa, Elongation at break =121.21%) and barrier properties of the resulting coating. The chitosan/pullulan-gallic acid grafted chitosan (CP-GS) coating, applicable by spraying or dipping, exhibited remarkable antioxidant activity, achieving 97.8% DPPH radical scavenging (25 °C, 50% RH), the CP-GS coating significantly reduced weight loss, delayed the decline in titratable acidity, and effectively suppressed microbial growth, thereby extending the shelf life by 7 to 9 days compared to the uncoated samples. This study highlights the effectiveness of molecular grafting in designing advanced bio-based coatings and presents a promising strategy for fruit preservation using biological macromolecules.

A QbD-informed study of permeation enhancer–octreotide nanoscale interactions influencing performance in pullulan buccal films

Sahil Malhotra,Sandeep Karki,Holly Linford,Muhammad Ijaz,Jimmy Muldoon,Ehsan Rezvani,Antonio Benedetto,Arun HS Kumar,Eoin O’ Cearbhaill,David J. Brayden

The peptide, octreotide, is used to treat acromegaly and neuroendocrine tumours. It is administered by either a long-acting injection or by an oral capsule. Although the oral route is preferrable, food interference necessitates dosing octreotide on an empty stomach and bioavailability is only ∼0.7%. Buccal film administration avoids the fasting constraints of oral octreotide and offers the potential for improved systemic bioavailability. This study presents a Quality by Design (QbD)- informed strategy to create a mucoadhesive pullulan-based buccal film co-entrapping octreotide with a bile salt permeation enhancer, sodium glycodeoxycholate (GDC). Pullulan was selected from a panel of candidate polymers based on film-forming performance and compatibility with octreotide and GDC, enabling reproducible formulation at relatively high entrapment levels despite the tendency of them to associate. Atomic force microscopy of films revealed concentration-dependent nanoscale roughness and microdomain formation at high GDC and octreotide levels. However, these structural features did not compromise key critical quality attributes (CQAs) including mechanical robustness, rapid hydration/disintegration, and acceptable content uniformity. Critical performance attributes (CPAs) using isolated porcine buccal mucosae demonstrated that films containing 10% w/w GDC (relative to pullulan) enhanced tissue uptake and octreotide permeation and via reversible non-destructive epithelial modulation. Stability testing confirmed >90% chemical recovery and conformational integrity of octreotide in films. UHPLC–HR-MS/MS confirmed identical fragmentation patterns for film-released and native octreotide, confirming the peptide’s intact cyclic backbone and the absence of chemical modification after formulation and after >60 days storage at ambient environment. This is the first report to integrate nanoscale structural analysis with systematic QbD- informed evaluation of formulation quality and performance, establishing pullulan–GDC buccal films as a robust adaptable platform for future multilayer oromucosal peptide film development.

Characterisation of pullulan polysaccharide membranes cross-linked with epigallocatechin gallate and Fe3+ for passion fruit preservation

Jiale Zhao,Guanglong Yao,Jian Chen,Lin Zhu

Traditional packaging materials can cause serious environmental pollution, prompting increased interest in the development of green bioplastic packaging alternatives. Herein, epigallocatechin gallate (EGCG) and Fe 3+ cross-linked pullulan polysaccharide (PUL) composite films were prepared using the solution casting method. Their structures and properties were measured. The application of PUL composite coatings in passion fruit preservation was also investigated. Fourier-transform infrared spectroscopy and X-ray diffraction analyses confirmed strong biocompatibility and interaction among PUL, EGCG and Fe 3+ . Scanning electron microscopy results further demonstrated good compatibility between the polymer and both EGCG and Fe 3+ . The metal-phenol-aldehyde network structure formed by EGCG and Fe 3+ crosslinks with the PUL film, enhancing its structural density and significantly reducing water vapor permeability by 15.24 %. Additionally, the mechanical properties of the composite film were enhanced, with tensile strength increasing from 14.36 MPa to 27.68 MPa, 92.76 % improvement over pure PUL film. Concurrently, the incorporation of EGCG and Fe 3+ substantially boosted the PUL film's UV barrier properties and antioxidant capacity. In passion fruit preservation experiments, PUL/EGCG/Fe 3+ -coated passion fruit exhibited the lowest weight loss rate (19.60 %) and shrinking index (3.3) after 10 days of storage compared to other treatment groups. This effectively delayed fruit quality deterioration and maintained appearance.

3D nanofiber sponge based on natural insect quaternized chitosan/pullulan/citric acid for accelerating wound healing

Qing Li,Xiaomin Lai,Yun Duan,Fuchen Jiang,Yingxi Li,Zhen Huang,Shuang Liu,Ying Wang,Can Jiang,Chen Zhang,Xiaoli Pan

Extensive traumatic injuries and difficult-to-heal wounds, induced by many circumstances, impose a significant social and economic burden on an annual basis. Thus, innovative wound dressings that encourage wound healing are greatly needed. In this work, we prepared a novel insect chitosan (MCS) using waste pupal shells from housefly ( Musca domestica L.) culture. After conducting comparative investigations with commercially available chitosan, it was shown that MCS exhibited comparable qualities and may be used as a substitute source of commercial chitosan. A quaternized chitosan/pullulan/citric acid three-dimensional nanofiber sponge (3D-NS) of natural origin was prepared by electrostatic spinning and gas foaming techniques after MCS was quaternized. In vitro , tests showed that the 3D-NS had a higher liquid absorption capacity than the two-dimensional nanofibrous membrane (2D-NM). Additionally, the 3D-NS showed improved hemostatic, pro-cell proliferation, antibacterial, and anti-inflammatory qualities. In vitro , tests demonstrated that 3D-NS could inhibit the release of inflammatory factors, promote angiogenesis, accelerate collagen deposition, and promote wound contraction. These effects considerably facilitated the healing process of wounds in rats with full-thickness skin damage. In conclusion, the great bioactivity and physicochemical properties of 3D-NS render it an optimal candidate for developing novel wound dressings.

Structural Characterization and Properties of Pullulan/Curdlan Blend Films and the Coating for Cherry Tomato Preservation

Kai Chen,Yanlin Tian,Hong Qian,Shibo Qiu,Gang Wei,Kao Wu,Junping Yin,Fatang Jiang

Fruits and vegetables (F&V) are prone to moisture loss and deterioration at postharvest. The strategy of a composite coating based on a single chain-triple helix network was applied to inhibit water escape and respiration of F&V. In this study, pullulan (PUL) with a high oxygen barrier was incorporated with the hydrophobic curdlan (CUD) to prepare the PC solutions and films. The results indicated that the linear PUL molecules were entangled with the CUD triple-helix micelle network, forming a continuous, dense three-dimensional network based on hydrogen bonds, and showing good biocompatibility. The addition of CUD enhanced the surface roughness of PC films, and significantly improved the elongation at break by 89%, hydrophobicity, water vapor and oxygen barrier, and thermal stability. Overall, the P 7 C 3 coating was selected for the cherry tomato preservation, significantly delaying weight loss by 2.5% and ascorbic acid decline by 16.8%, maintaining firmness, total solids, and acid content. The coating significantly inhibited respiratory intensity and sustained the sensory attributes (appearance, texture, taste, and smell) of cherry tomatoes at a high level, which suggested that the P 7 C 3 coating can effectively extend the shelf life of cherry tomatoes. This work would provide a reference for the application of pullulan and curdlan complex in F&V preservation and show a good prospect.
