BACKGROUND Giant salamander protein peptide is a peptide with rich functional properties. Giant salamander protein peptide KGEYNK (KK-6) is a peptide with both antioxidant and anti-inflammatory properties. The antioxidant and anti-inflammatory mechanisms of KK-6 are still unclear. When we studied the functional mechanism of KK-6, we found that the antioxidant property of KK-6 has a synergistic and promoting effect on anti-inflammatory properties. RESULTS KK-6 enhances cellular resistance to LPS via the MAPK/NF-κB signaling pathway, leading to increased levels of inflammatory factors: interleukin-1β (764.81 ng mL −1 ), interleukin-6 (1.06 ng mL −1 ) and tumor necrosis factor-α (4440.45 ng mL −1 ). KK-6 demonstrates potent antioxidant properties by activating the Nrf2 signaling pathway, resulting in elevated levels of antioxidant enzymes (glutathione peroxidase: 0.03 μg mL −1 ; superoxide dismutase: 0.589 μg mL −1 ) and a reduction in the concentration of the oxidative product malondialdehyde (967.05 μg mL −1 ). CONCLUSION Our findings highlight the great potential of KK-6, a peptide extracted from giant salamander protein, as a remedy for intestinal inflammation. Through its dual role as an antioxidant and anti-inflammatory agent, KK-6 offers a promising avenue for alleviating inflammation-related damage and oxidative stress. This study lays the foundation for further exploration of giant salamander products and highlights their importance in health and novel food development. © 2024 Society of Chemical Industry.

The present work provides a novel route for forming one-dimensional (1D) gallium (Ga) nanoribbon materials with a host molecule calix[6]arene (CA-6) as a template by a facile, one-step and low temperature chemical bath method. Field-emission scanning electron microscopy and transmission electron microscopy showed that the uniform 1D Ga nanoribbon material (Ga-a) can be constructed only in the presence of CA-6, and the formation of ribbon structures is highly dependent on doping ratios and deposition times. Our data indicate that the unusual effect of CA-6 is due to a combination of two factors: a high density of OH groups in the outer surface of its cavity and an appropriate cavity diameter. Especially, the uniform 1D Ga nanoribbon material exhibits a distinct electronic structure and very rare magnetic behaviour when compared to those 3D Ga materials obtained by means of other host molecules: calix[4]arene, γ-cyclodextrin and 18-crown-6. For example, of all the Ga materials, the uniform 1D nanoribbon material has the lowest electron density of Ga core levels in light of X-ray photoelectron spectroscopy analysis. This result suggests that there is a stronger molecule–atom interaction between Ga atoms and CA-6 molecules compared with those in other host–guest systems. More importantly, the uniform 1D Ga nanoribbon material exhibits a magnetic transformation from a diamagnetic to a paramagnetic state under the influence of an applied field, which is completely different from those of all the 3D Ga materials and all the irregular Ga nanoribbon materials. Such a transformation is novel in metals and particularly useful in the chemistry of materials since it allows dramatic modifications of magnetic properties of metal nanocrystals. Finally, a strong surface-enhanced Raman scattering of the uniform 1D Ga nanoribbon material has been observed for organic molecules adsorbed on their surface. Taken together, we believe this work opens a new channel for development of 1D metal-based nanomaterials.

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) has become a robust tool for analyzing a variety of biomacromolecules. However, the strong background interference produced by conventional organic matrices hinders the detection of small molecule analytes, which restricts the widespread application of MALDI-MS in metabolomics studies. Consequently, developing new organic matrices is urgently needed to overcome these issues. In this study, 1,4-Dioxo-1,2,3,4-tetrahydrophthalazine-6-carboxylic acid (DTCA) was firstly employed as a new matrix for MALDI-MS to enhance the detection of low molecular weight compounds because of its strong UV absorption, less matrix background interference, high ionization efficiency for metabolites, and good reproducibility. Considering these advantages, DTCA was used to analyze endogenous metabolites in the serum samples of imidacloprid (IMI)-exposed mice via MALDI-MS in positive ion mode. By combining with machine learning, the differentiation between imidacloprid-exposed mice and control mice was successfully achieved, and 39 metabolites were estimated as potential biomarkers. Additionally, potentially disrupted metabolic pathways were revealed. These results indicate that DTCA, as a new and powerful matrix in positive ion mode, has great potential for applications in the detection of small molecules.

Perovskite hydroxystannate (MSn(OH) 6 ) has garnered considerable interest in recent years as a novel flame retardant characterized by its low toxicity and environmentally benign properties. In order to improve the flame retardancy and smoke suppression properties of epoxy resin (EP) matrices, transition metal hydroxystannate (MSn(OH) 6 , M = Fe, Co, Mn) was prepared by a ultrasonic-coprecipitation method and used as flame retardants. The synthesized composites were assessed for their flame retardant characteristics and mechanical properties through the measurement of the limiting oxygen index (LOI), the cone calorimetry test (CCT), and universal tensile testing. Upon the incorporation of 10 wt% of the MSn(OH) 6 flame retardants, the flame retardant of EP composites exhibited marked enhancement, especially manganese hydroxystannate (MHS). Compared with pure EP, the EP/MHS-10 composite demonstrated the best performance, reducing the peak of heat release rate (pHRR), total heat release (THR), total smoke production (TSP) by 60.0%, 14.4%, and 32.6%, respectively. During the process of combustion, the decomposition of MSn(OH) 6 results in the generation of non-flammable gases and water vapor. In the condensed phase, tin (Sn) and transition metals contribute to the formation of a more protective char residue. The residue serves as a physical barrier, effectively isolating the underlying epoxy (EP) matrix material from heat and oxygen.

Herein, we report a ratio-based electrochemical biosensor for the detection of interleukin-6 (IL-6). We electropolymerized methylene blue (MB) on the surface of screen-printed carbon electrodes; introduced an internal reference signal probe; modified the carboxylate multi-walled carbon nanotubes on the electrode surface to increase the electrochemically active area; and finally linked the amino-modified IL-6 aptamer to the electrode surface through the Schiff base reaction, with bovine serum albumin (BSA) added to mask non-specific adsorption. After adding IL-6 to the samples, the signal of I MB remained almost unchanged, while the signal of I[Fe(CN) 6 ] 3−/4− decreased with increasing IL-6 concentration. Thus, a novel ratiometric electrochemical sensor with a linear range of 0.001~1000.0 ng/mL and a low detection limit of 0.54 pg/mL was successfully developed. The sensor had high repeatability, stability, sensitivity, and practicability. It provides a new method for constructing proportional electrochemical sensors and detecting IL-6.

Graphical In this paper, We developed a novel ECL sensor of the 6-MC/S 2 O 8 2− system by using 2-NSA as co-reaction accelerator. 2-NSA was first proposed as co-reaction accelerator, promoting the generation of additional oxidant SO 4 − from S 2 O 8 2− , thus amplifying the ECL signal. Moreover, this research provides a highly sensitive platform for the detection of MV. In this paper, the novel ECL sensor of the 6-methoxychromone (6-MC)/S 2 O 8 2− system was designed by using 2-naphthalene sulfonic acid (2-NSA) as a co-reaction accelerator. Fluorescence, ECL spectrum, and cyclic voltammetry (CV) were used to explore extensively the ECL mechanism of the system. It is worth pointing out that 2-naphthalene sulfonic acid (2-NSA) was first proposed as a co-reaction accelerator, reacting with the coreactant K 2 S 2 O 8 rather than the luminophore, promoting the generation of additional oxidant SO 4 − from S 2 O 8 2− , thus amplifying the ECL signal. Furthermore, under the optimal experimental conditions, the high sensitivity of the 2-naphthalene sulfonic acid (2-NSA)+6-methoxychromone (6-MC)/S 2 O 8 2− system was realized for the detection of methyl violet (MV) in the linear range of 10 −8 ~10 −2 M, with a detection limit of 5.14×10 −10 M. In addition, this find offers some guidance for further developing co-reaction accelerators with excellent performance in the future.

In this study, the Janus nanosheets (JNSs) as compatibilizers have been applied in the immiscible polyamide 6/acrylonitrile-butadiene-styrene copolymer (PA6/ABS) blends. In the JNSs, polystyrene (PS) chains and epoxy groups were grafted on the opposite sides successfully where the epoxy functional groups can react with terminal carboxyl/amino groups of PA6 with ring-opening condensation reactions during melt blending. The obtained JNSs with grafting PS and PA6 chains on each side of silica nanosheets can gradually migrate toward and be anchored at the phase interface with amphiphilicity and the pickering effect. The JNSs exhibited higher interfacial selectivity and compatibilization efficiency. At 2.0 wt% JNSs loading, the ABS dispersed phase transformed from spherical to irregular ellipsoidal shapes, likely due to the rigid characteristics of JNSs altering phase interface curvature and consequently modifying the blend's phase morphology. The gas barrier properties of the blending films can be significantly enhanced from 3.10 × 10 −15 cm 3 cm·cm −2 s −1 Pa −1 to 1.70 × 10 −15 cm 3 cm·cm −2 s −1 Pa −1 with 2.0 wt% JNSs due to the irregular dispersed phase structure and the presence of silica nanosheets at the interface. This study investigates interfacial structure and mechanical properties with JNSs compatibilizers, providing new insights for interface compatibilization strategies and construction of functional composites.

Glass fiber-reinforced polyamide 6 (PA6/GF) composites are widely used due to their excellent mechanical and thermal properties, but the glass fibers exacerbate flammability via the “candlewick effect”. Conventional flame retardants require high loadings (>15 wt%), which degrade mechanical performance and processing, limiting practical use. Herein, we developed a dual-functional interfacial flame-retardant strategy through incorporating a DOPO-functionalized compatibilizer, poly(DOPO-hydroxyethyl acrylate- co -maleic anhydride) [P(DH-co-MA)]. P(DH-co-MA) covalently bonds to the glass fiber surface via maleic anhydride groups, significantly improving fiber-matrix interfacial adhesion and composite integrity. With a minimal addition of 0.6 wt% P(DH-co-MA) and 10 wt% base flame-retardant system, the modified composites achieve a limiting oxygen index (LOI) of 33.8 %, UL-94 V -0 rating, and a 30.2 % reduction in peak heat release rate (pk-HRR). Moreover, tensile strength, flexural modulus, and impact strength increase by 4.2 %, 5.5 %, and 17.4 %, respectively. Thermogravimetric analysis and char residue characterization indicate that P(DH-co-MA) promotes interfacial char formation and gas-phase radical quenching, exhibiting a dominant gas-phase flame-retardant mechanism. This study offers a viable and efficient route for developing high-performance, interface-engineered flame-retardant polymer composites, achieving simultaneous improvements in mechanical robustness and fire safety at low additive levels. Notably, the DOPO-functionalized compatibilizer can be readily applied for surface-tailored sizing of glass fibers, enabling in situ modification without altering existing processing protocols.

1,3-propanediol (1,3-PDO) is a high-value chemical that can be produced via glycerol hydrogenolysis or biological fermentation. The main products of glycerol hydrogenolysis include 1,3-PDO and 1,2-propanediol (1,2-PDO). The properties of propanediol isomers are extremely similar, which are difficult to effectively separate them. Based on the differences in the shape of ellipsoidal 1,3-PDO and spherical 1,2-PDO, ZIF-300 with a 6-member ring (6-MR) trap between cha and d6r cages that can match spherical 1,2-PDO have been selected for the separation of propanediol isomers. ZIF-300 series materials (named ZIF-300-S1, ZIF-300-S2 and ZIF-300-S3) with different 6-MR trap orientations have been constructed by variable the ratio between the small-sized 2-methylimidazole (mIm) and large-sized 5-bromobenzimidazole (bbIm) links. Theoretical and experimental results show that ZIF-300-S3 (Zn(mIm) 1.04 (bbIm) 0.96 ) preferentially adsorbed 1,2-PDO over 1,3-PDO because the 6-MR trap with uniform large bbIm/small mIm links distribution faces essentially the small d6r cage, which can better match spherical 1,2-PDO. In contrast, ZIF-300-S1 (Zn(mIm) 0.82 (bbIm) 1.18 ) show low uptake of 1,2-PDO and 1,3-PDO because the 6-MR trap with main large bbIm links distribution faces essentially the larger cha cage, which show weak confinement of 1,2-PDO and 1,3-PDO. The dynamic column adsorption (1,2-PDO/1,3-PDO: 50/50 g L −1 ) results exhibited that the adsorption capacity of ZIF-300-S3 for 1,2-PDO can be as high as 64.7 mg g −1 and selectivity for 1,2-PDO/1,3-PDO was 2.0. This work provides valuable insights into the construction of ZIFs materials with tunable 6-MR trap for effective separation of isomers.

Ethnopharmacological relevance Trametes robiniophila Murr (Huaier), a traditional Chinese medicine (TCM), has shown promising effects as a complementary therapy in the treatment of multiple kinds of cancers. Aim of the study This study aimed to systematically evaluate whether Huaier could enhance the antitumor effects of palbociclib, a highly selective CDK4/6 inhibitor. Materials and methods A breast cancer xenograft mouse model was constructed to assess the anticancer effects of Huaier and palbociclib. Transcriptomic analysis was performed to identify the differentially expressed genes and associated pathways. Flow cytometry, immunofluorescence and immunohistochemical staining were then conducted to observe the infiltration of immune cells in the tumor microenvironment (TME). The composition of the gut microbiota was explored through 16S rDNA sequencing, and a pseudosterile mouse model was established to verify the contribution of the gut microbiota to the antitumor effects of Huaier and palbociclib. Results Oral administration of Huaier enhanced the antitumor effects of palbociclib in breast cancer. Transcriptomic analysis of tumor tissues revealed that the differentially expressed genes were related to immune functions. Indeed, the addition of Huaier to palbociclib therapy further increased the ratio of CD8 + /CD3 + T cells and the abundance of GzmB + CD8 + T cells in the TME. Mechanistically, sequencing of 16S rDNA revealed that Huaier reshaped the gut microbiota composition in favor of Akkermansia , which positively modulated antitumor immune responses. The pseudosterile mouse model verified the necessity of the gut microbiota for the synergistic effect of Huaier on palbociclib. Furthermore, butyrate, a metabolite of Akkermansia , synergized with palbociclib to suppress tumor progression and promote the infiltration of CD8 + T cells in the TME. Conclusions Our data suggested that Huaier augmented the antitumor effects of palbociclib by increasing the abundance of Akkermansia in gut microbiota, which contributed to the enhancement of anti-tumor immunity. Consequently, combining palbociclib with Huaier may serve as a promising strategy for the treatment of breast cancer.

Previous studies have indicated that 6:2 chlorinated polyfluoroalkyl ether sulfonate (6:2 Cl-PFAES), a substitute for perfluorooctane sulfonate (PFOS), caused disruptions in glucose metabolism, but its toxicological mechanisms remain unclear. Pancreatic β cells are responsible for regulating glucose metabolism. This study used mouse insulinoma β cells (β-TC-6) to study the toxic effects of 6:2 Cl-PFAES and elucidate its potential mechanisms. Through cell counting kit-8, trypan blue staining, and apoptosis assays, we found that 6:2 Cl-PFAES showed toxicity to β-TC-6 cells, even surpassing that of PFOS. Mechanistic studies demonstrated that 6:2 Cl-PFAES induced oxidative stress by ascendant levels of reactive oxygen species (ROS) and malondialdehyde (MDA), alongside increased superoxide dismutase (SOD) activity. The oxidative stress-inducing capability of 6:2 Cl-PFAES was stronger than PFOS. Co-exposure with N-acetyl-L-cysteine (NAC), as the ROS scavenger, which obviously reduced the degree of cell apoptosis and the production of ROS, highlighting oxidative stress as a crucial mechanism of 6:2 Cl-PFAES-induced cell death. Transcriptomic analysis further revealed that 6:2 Cl-PFAES affected genes association with the oxidative phosphorylation pathway, confirming the oxidative stress effect, and altered diabetes-related genes expression, suggesting a heightened risk of diabetes. This study provides insights into the toxicity of 6:2 Cl-PFAES on β cells and its potential risks on glucose metabolism disruption.

BackgroundGiven an epidemiological association between hypertension and osteoarthritis, we aim to elucidate how vascular pathology triggers avascular articular cartilage loss by comparing vascular and joint phenotypes between spontaneously hypertensive rats and normotensive Wistar Kyoto rats at tissue, cellular, and molecular levels.MethodsSystemic and local endothelial function were assessed via blood pressure monitoring and photoacoustic imaging of knee joints, respectively (n=5–6). Tissue and cellular damages in articular cartilage, subchondral bone, and synovium were systemically evaluated by radiological and histological examination (n=6). Endothelial transcriptional changes were delineated via bulk RNA‐seq (n=3). The secretome of human endothelial cells under oxidative stress was analyzed via proteomics to identify key factors in chondrocyte senescence (n=4). Captopril, an antihypertensive medication, was adopted as a tool to rescue chondrocyte senescence and osteoarthritis both in vitro (n=3) and in vivo (n=6).ResultsCompared with Wistar Kyoto rats, spontaneously hypertensive rats exhibited higher blood pressure and local joint hypoxia with increased endothelial oxidative stress as early as 3 months old. Notably, these endothelial dysfunctions preceded the onset of structural joint damage. By 9 months of age, spontaneously hypertensive rats developed osteoarthritis‐like pathology characterized by senescent chondrocyte accumulation and cartilage degradation, which could be mitigated by captopril. Transcriptomic and proteomic analyses of rat and human endothelial cells revealed upregulated IGFBPs (insulin‐like growth factor‐binding proteins). Among them, IGFBP6 was identified to induce chondrocyte senescence in vitro. This effect can be modified by captopril treatment.ConclusionsSystemic vascular dysfunction may induce local joint damage with upregulation of endothelial IGFBPs secretion such as IGFBP6. This study highlights a potential therapeutic target for preventing joint structural damage by addressing endothelial dysfunction.
