To prepare and characterize low-bitter angiotensin-converting enzyme (ACE)-inhibitory peptides from sesame protein, a triple-enzyme hydrolysis system was optimized using mixture design and response surface methodology. The resulting hydrolysate was separated by ultrafiltration and medium-pressure chromatography, followed by identification through nano-liquid chromatography–electrospray ionization-tandem mass spectrometry. Finally, the mechanism of typical low-bitter ACE-inhibitory peptides was elucidated by molecular docking and molecular dynamics simulation. Results showed that the optimal enzyme activity ratio of 1:0.94:1.07 for Alcalase, trypsin, and Flavourzyme, combined with optimized hydrolysis conditions (E/S ratio of 126,793.03 nkat/g, pH 8.40, 4.82 h hydrolysis time, and 45 °C), resulted in a peptide yield of 93.19 ± 0.14%, ACE-inhibitory rate of 95.92 ± 0.23%, and bitter value of 3.15 ± 0.09. APQLGR and APWLR exhibited high ACE-inhibitory activity and minimal bitterness among the seventeen identified peptides. Although both peptides bound to the S1 pocket and Zn2+catalytic site of ACE, APWLR exhibited an additional interaction with the S2 pocket. Both peptides were predicted to antagonize the bitter taste receptor T2R14 by forming stable complexes with key residues, but two complexes exhibited distinct mechanisms of stabilization. This work demonstrates a method for producing dual-functional peptides from sesame protein, paving the way for their application in functional foods.

Lithium (Li) reportedly has anti-bacterial properties. Thus, it is an ideal option to modify barrier membranes used for guided bone regeneration to inhibit the bacterial adhesion. The aims of this study were to fabricate and characterize nanofibrous poly(L-lactic acid) (PLLA) membranes containing Li, and investigate their antibacterial effects on Porphyromonas gingivalis and Actinobacillus actinomycetemcomitans in vitro. Li (5%Li, 10%Li, and 15%Li)-loaded nanofibrous PLLA membranes were fabricated using an electrospinning technique, and characterized via scanning electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, a contact angle measuring device, and a universal testing machine. Sustained release of Li ions was measured over a 14-day period and biocompatibility of the Li-PLLA membranes was investigated. Evaluation of bacterial adhesion and antibacterial activity were conducted by bacterial colony counting, LIVE/DEAD staining and inhibition zone method using P.gingivalis and A.actinomycetemcomitans. Of the three Li-loaded membranes assessed, the 10%Li-PLLA membrane had the best mechanical properties and biocompatibility. Adhesion of both P.gingivalis and A.actinomycetemcomitans on Li-PLLA membranes was significantly lower than adhesion on pure PLLA membranes, particularly with regard to the 10%Li and 15%Li membranes. Significant antibacterial activity of Li-PLLA were also observed against according to the inhibition zone test. Given their better mechanical properties, biocompatibility, and antibacterial activity, PLLAs with 10%Li are a better choice for future clinical utilization. The pronounced antibacterial effects of Li-loaded PLLA membranes sets the stage for further application in guided bone regeneration.

A random terpolymer of poly(L-lactide-co-glycolide-co-ε-caprolactone) (PLLGC) was synthesized from L-lactide, glycolide and ε-caprolactone. The in-vitro hydrolytic degradation behavior of the PLLGC terpolymer was investigated as a function of the degradation time; the results showed that the degradation rate of PLLGC was lower than that of PLGA due to a reduction in the acidic degradation products. Therefore, we suggest the degradation rate of the PLGA could be controllable by introduction of ε-caprolactone into the PLGA martrix, while the physical properties (mechanical, etc.) were superior to PLGA.

N -(phosphonomethyl)- l -proline is an analogue of glyphosate. The protonation for N -(phosphonomethyl)- l -proline was studied by potentiometry, calorimetry, 31 P NMR spectroscopy and quantum chemical calculations to further understand the protonation process of glyphosate. The results confirmed that the order of successive protonation sites of totally deprotonated N -(phosphonomethyl)- l -proline are a phosphonate oxygen, amino nitrogen, and finally the carboxylate oxygen. The results can improve the understanding of the biological activity of these types of molecules in solution.

In this research, extraction optimization, preliminary characterization and immunological activities in vitro of polysaccharides from Elaeagnus angustifolia L. pulp were investigated. A response surface methodology (RSM) with a Box-Behnken design (BBD) was used to optimize the extraction process. The maximum EAP yield was 9.82 ± 0.38%, which is in good agreement with the predicted value (9.93 ± 0.24%). Two homogeneous polysaccharides, EAP-1a and EAP-1b with molecular weights of 8.70 kDa and 4.39 kDa respectively, were prepared by DEAE-52 cellulose and Sephadex G-100 columns and characterized by HPLC, HPGPC, and FT-IR. Three polysaccharides (EAP, EAP-1a and EAP-1b) could stimulate macrophages to release NO and enhance phagocytic activities of RAW 264.7 cells in dose-dependent manner. Moreover, there was no significant difference between crude EAP group (400 μg/mL) and positive control group (LPS) in effects on macrophages. The results implied that EAP had the potential to be developed as natural medicines or health foods .

Extraction optimization, characterization and antioxidant activity in vitro of polysaccharides from mulberry leaves (MLP) were investigated in the present study. The optimal extraction conditions with an extraction yield of 10.0 ± 0.5% for MLP were determined as follows: extraction temperature 92 °C, extraction time 3.5 h and ratio (v/w, mL/g) of extraction solvent (water) to raw material 34. Two purified fractions, MLP-3a and MLP-3b with molecular weights of 80.99 and 3.64 kDa, respectively, were obtained from crude MLP by chromatography of DEAE-Cellulose 52 and Sephadex G-100. Fourier transform-infrared spectroscopy revealed that crude MLP, MLP-3a and MLP-3b were acidic polysaccharides. Furthermore, crude MLP and MLP-3a had more complicated monosaccharide compositions, while MLP-3b had a relatively higher content of uronic acid. Crude MLP, MLP-3a and MLP-3b exhibited potent Fe 2+ chelating power and scavenging activities on 1,1-diphenyl-2-picrylhydrazyl, hydroxyl, superoxide and 2,2′-azinobis-(3-ethyl-benzothiazolin-6-sulfonic acid) radicals. The results suggested that MLP could be explored as natural antioxidant.

Background Artemisia dracunculus L. (A. dracunculus) is a distinctive plant resource in Xinjiang, possesses both medicinal and culinary value, with distinct regional characteristics. Methods In this study, total flavonoids extracted from A. dracunculus (TFAD) were obtained using a reflux method, with extraction parameters optimized through single-factor experimental design. Ultraviolet-visible spectrophotometry was used for both qualitative and quantitative assessment of TFAD. Samples collected from Jimusaer, Hami, and Tacheng in Xinjiang were analyzed for total flavonoid content and antioxidant activity. Results The optimized extraction conditions included 50% ethanol as the solvent, a solid-to-liquid ratio of 1:30, an extraction temperature of 80 °C, and a duration of 8 h. Under these conditions, the TFAD content in samples from Jimusaer, Hami, and Tacheng was 107.0146 ± 1.4986, 111.9548 ± 1.6136, and 124.8996 ± 1.3801 mg g⁻¹, respectively. Statistically significant regional differences were observed ( p < 0.01), with the highest flavonoid concentration found in samples from Tacheng. Antioxidant activities of TFAD from the three regions were assessed using in vitro free radical scavenging assays, with ascorbic acid (AA) serving as the positive control. TFAD extracted from Jimusaer exhibited the strongest antioxidant capacity, with IC₅₀ values of 31.31 ± 1.19 and 114.04 ± 5.56 µg mL⁻¹ against DPPH and ABTS⁺· radicals, respectively, compared with 7.25 ± 0.56 and 154.18 ± 9.54 µg mL⁻¹ for AA. Conclusion These results suggest that TFAD from the Jimusaer region demonstrates superior antioxidant activity, providing a basis for further exploration and potential utilization of A. dracunculus as a functional resource in Xinjiang.

The development of highly sensitive and selective platforms for trace Zn 2+ detection is crucial for environmental monitoring and biomedical diagnostics. Herein, a photoelectrochemical (PEC) sensor was constructed using an L-cysteine (L-Cys)-functionalized BiOBr composite, in which surface-interface engineering was adopted to enrich available recognition sites and modulate the interfacial PEC response. Compared with pristine BiOBr, the incorporation of L-Cys introduces abundant Zn 2+ binding functional groups and modifies the local surface environment of BiOBr, thereby enhancing the photocurrent response and enabling the selective recognition of Zn 2+ . The sensor operates through a signal-off mechanism, in which the coordination of Zn 2+ with surface-bound L-Cys forms an interfacial coordination layer that partially hinders mass transport and interfacial charge transfer, resulting in a decrease in photocurrent. Under optimized conditions, the platform exhibits a wide linear response ranging from 0.5 to 40 nmol/L, with a low detection limit of 2.69 nmol/L ( S / N = 3). Furthermore, the applicability of the sensor is validated in real-world scenarios, demonstrating excellent recoveries in tap water, lake water, and complex biological matrices such as HepG2 cell lysates (recoveries, 96.3–108.5%). This work provides a robust strategy for engineering bio-semiconductor interfaces for the detection of essential metal ions in complicated samples.

Efficient recovery and high-value utilization of keratin from waste human hair are of significant interest for sustainable materials development. However, the densely cross-linked disulfide bonds and extensive hydrogen-bond network of keratin make its mild and efficient extraction challenging. In this work, a redox-active deep eutectic solvent (DES) composed of L-cysteine (L-Cys) and monoethanolamine (MEA) was developed for keratin recovery from waste human hair. Under optimized conditions of 80 °C, 3.5 h, and 10 wt% solid loading, the keratin recovery yield reached 96.5%, and the solvent maintained stable performance over five recycling cycles. Experimental results, together with DFT and MD simulations, indicate that the dissolution process is driven by synergistic disulfide bond cleavage and disruption of the hydrogen-bond network, while the protein backbone remains largely intact. The recovered keratin was further phosphorylated and co-regenerated with cellulose to fabricate a phosphorylated keratin/cellulose composite membrane (P-KCM). The membrane exhibited efficient and selective Fe 3+ adsorption, with a maximum adsorption capacity of 297.8 mg·g −1 . FT-IR and XPS analyses reveal that phosphate groups serve as the primary coordination sites for Fe 3+ . In tests with real LiFePO₄ leachate, P-KCM removed 97.3% of Fe 3+ while leaving 80.1% of Li + in solution, and it retained 84.7% of its initial adsorption capacity after four adsorption-desorption cycles. This work provides a mild, efficient, and recyclable strategy for keratin valorization and offers a feasible approach for selective metal ion separation and pre-treatment in battery recycling.

Plant invasions and microplastic (MP) pollution are widespread environmental challenges with increasing impacts on soil ecosystems. Although their individual effects on soil faunal diversity are well-documented, the interactive effects of these co-occurring stressors remain poorly understood. We investigated the short-term (two months) responses of soil faunal communities to conventional (low-density polyethylene, LDPE) and biodegradable (polylactic acid, PLA) MPs in habitats with and without invasion by Solidago canadensis L. We found that invasion by S. canadensis significantly increased overall abundance and richness of soil fauna, particularly microbivores. However, MP addition—either alone or in combination with plant invasion—did not significantly affect the abundance or richness of the total soil fauna or most trophic groups. Structural equation modeling revealed that MPs not only attenuated the positive effect of invasion on overall abundance by 48.4%, but also reversed its effect on richness from positive to a net decrease of 120.6%. Moreover, while S. canadensis invasion significantly altered soil faunal community composition in plots without MPs, this effect disappeared in most MP-amended treatments. These findings suggest that short-term MP pollution may alter the impact of S. canadensis invasion on soil fauna. However, the magnitude of MP effects varied across trophic groups, with microbivores exhibiting the most notable response. This study underscores the necessity of considering interactions among co-occurring environmental stressors in soil biodiversity assessments, as their combined effects can reshape outcomes for key functional groups within ecosystems.

LPA treatment alleviates eggplant pericarp shrinkage. • The biosynthesis of functional amino acids depresses pericarp shrinkage. • The maintenance of TCA cycle decreases pericarp shrinkage. • The activation of linoleic acid metabolism lowers pericarp shrinkage under enhanced antioxidant capacity. • The enhancement of cell wall rigidity and hydrophobicity reduces pericarp shrinkage. Eggplant is prone to pericarp shrinkage during room-temperature storage, reducing its commercial value. While L -pyroglutamic acid (LPA), a glutathione cycle derivative, shows potential in post-harvest preservation, its exact mechanisms in eggplant post-harvest storage remain unclear. This study found that LPA treatment alleviated pericarp shrinkage and darkening, reduced water loss, and maintained the contents of nutrients (soluble sugar, soluble protein, and amino acid) in eggplant. Through integrated transcriptomic, metabolomic, physiological, and biochemical analyses, we elucidated the regulatory effects of LPA. LPA promoted the biosynthesis of osmoregulatory and resistance-related amino acids, such as proline, glutamate, arginine, and glutamine. LPA enhanced the accumulation of citrate, stimulated the TCA cycle, and regulated energy supply. Additionally, LPA activated linoleic acid metabolism, leading to the oxidation of linoleic acid by lipoxygenase to produce oxylipins, including 9(S)-HPODE, 9-OxoODE, and 13(S)-HODE. Under the buffering effect of the antioxidant system, these oxylipins may act as defense signal activators, enhancing fruit defense capacity. Furthermore, LPA enhanced the mechanical strength and hydrophobicity of the cell wall by promoting the accumulation of polysaccharides, including cellulose, hemicellulose, pectin, and lignin, thereby reinforcing the epidermal barrier function. This study elucidates the molecular mechanisms by which LPA maintains post-harvest eggplant quality through the regulation of primary metabolism, defense signaling, and cell wall barrier, providing a theoretical basis for its application in fruit and vegetable preservation.
