Bioinspired Hydrogel Remodels the Niche after Spinal Cord Injury for Neural Reconnection by Promoting Angiogenesis and Reducing Inflammation

Haifei Cao,Yicheng Ding,Xingzhi Liu,Junchao Luo,Junjie Niu,Huan Zhao,Xichao Zhou,Xiaozhong Zhou,Yong Xu,Huilin Yang,Qin Shi

Spinal cord injury (SCI) disrupts the local microenvironmental niche, resulting in persistent neuroinflammation, vascular dysfunction, and impaired neuronal regeneration, presenting a significant challenge for tissue repair. In this context, hydrogels offer a versatile strategy for targeted and sustained therapeutic delivery and localized microenvironmental modulation. To facilitate niche restoration following SCI, we developed a bioinspired platform by integrating KTPSLEQRTVYAKGGIKRGK-brain-derived neurotrophic factor mimetic peptides (TPS-BDNF) into a dopamine-modified gelatin methacryloyl hydrogel. In vitro and in vivo studies demonstrated that the proposed hydrogel exhibits optimal injectability, shear-thinning behavior, and intrinsic antioxidant and anti-inflammatory properties, thereby facilitating minimally invasive delivery and localized microenvironment modulation. Furthermore, the TPS-BDNF endows the multifunctional hydrogel with sustained neurotrophic and pro-angiogenic cues that promote the restoration of the neurovascular niche and subsequent functional recovery after SCI. In summary, the bioinspired hydrogel offers a promising, clinically translatable approach to synergistic niche remodeling for the restoration of neuronal function following SCI. Statement of significance The gelatin methacryloyl (GelMA) /dopamine/Polyethylene glycol-conjugated KTPSLEQRTVYAKGGIKRGK-brain-derived neurotrophic factor mimetic peptides (TPS-BDNF) (GDP) hydrogel represents a significant advance for spinal cord injury (SCI) therapy by simultaneously promoting neurogenesis and angiogenesis while delivering strong anti-inflammatory and antioxidant effects. By integrating TPS-BDNF with a GelMA-based matrix, GDP remodels the lesion microenvironment, enhances endogenous neural stem cell mobilization and neuronal differentiation, and improves functional recovery in SCI animals, all with injectable, controllable release and minimal systemic toxicity. This platform holds strong potential for translation to human SCI treatment. Download: Download high-res image (372KB) Download: Download full-size image

Suture-Free Piezoelectric Band-Aid Membrane for Complex Peripheral Nerve Defects

Tong Qi,Xiao Wang,Yijing Han,Chengqi He

The regeneration and functional rehabilitation of peripheral nerve defects remain major clinical challenges. Piezoelectric materials have the potential to restore both nerve morphology and function. Conventional grafts struggle to repair nerves with complex structures. To address this issue, band-aid–inspired tissue-adhesive membranes were designed to accommodate diverse nerve diameters and branching structures. Electrospinning was used to prepare a Janus membrane that served as a suture-free piezoelectric nerve repair membrane. The inner layer comprised oriented adhesive hydrogel fibers fabricated from gelatin methacryloyl and tannic acid, whereas the outer layer comprised piezoelectric silk fibroin fibers that provided barrier function and structural support. In vivo experiments demonstrated that this membrane streamlined surgery, enabled rapid and facile bridging of complex peripheral nerve defects, and significantly facilitated morphological and functional nerve recovery. This finding holds significant promise for advancing personalized, simplified, and more effective clinical management of peripheral nerve defects. Additionally, it is anticipated to treat defects in other tissues and organs, specifically those with irregular morphologies.

A ROS-responsive degradable hydrogel integrating MnO2 nanoparticles and gemcitabine for synergistic chemodynamic-chemotherapy of breast cancer

Pingan Li,Xiaodan Chen,Jingyang Su,Qin Zhang,Lin Lin

A ROS-responsive degradable hydrogel is developed for localized breast cancer therapy. • The hydrogel integrates MnO 2 -mediated chemodynamic therapy with gemcitabine chemotherapy. • Tumor microenvironment-triggered ROS generation accelerates hydrogel degradation and drug release. • The self-amplifying therapeutic cascade enhances antitumour efficacy with favourable biosafety. Breast cancer therapy remains limited by systemic side effects, intratumoral drug accumulation and tumor recurrence, highlighting the need for effective localized treatment strategies. Herein, we develop a reactive oxygen species (ROS)-responsive degradable hydrogel for localized chemodynamic-chemotherapeutic breast cancer therapy. Bovine serum albumin-templated MnO 2 (BM) nanoparticles and gemcitabine (Gem) are incorporated into a thiol-norbornene photocrosslinked polyethylene glycol (PEG) network containing ROS-cleavable thioketal (TK) linkers. Such a hydrogel serves not only as a local drug depot, but also a tumor microenvironment-activated therapeutic system. Within this microenvironment, MnO 2 consumes endogenous glutathione (GSH) and reacts with hydrogen peroxide (H 2 O 2 ) to generate Mn 2+ and hydroxyl radicals (·OH), inducing oxidative damage and perturbing tumour redox homeostasis. The resulting ROS further cleave TK linkers, accelerating hydrogel degradation and Gem release. This self-amplifying feedback couples chemodynamic therapy with chemotherapy, enabling localized, stimuli-responsive drug delivery and enhanced antitumour efficacy. In vitro and in vivo evaluations confirm significant tumor suppression and favourable biosafety. These findings highlight a microenvironment-responsive hydrogel platform for regional combination therapy of breast cancer. After implantation at the tumor site, MnO 2 reacts with tumor-overexpressed glutathione and hydrogen peroxide to generate Mn 2+ and hydroxyl radicals, thereby depleting intracellular antioxidants and enhancing chemodynamic therapy. The elevated ROS simultaneously cleave thioketal linkers, triggering hydrogel degradation and accelerating gemcitabine release. This tumor microenvironment-activated cascade integrates ROS-amplified chemodynamic therapy with localized chemotherapy, achieving potent tumor cell killing and improved therapeutic safety.

Capillary-Wave-Induced ‘Endocytosis’ Enables Programmable Core-Shell Capsules Formation for Controlled Release

Yi Huang,Sihang Liu,Shuai Yin,Lian Xiao,Yugang Zhao,Zhi Tao,Haiwang Li

Understanding droplet-liquid pool interactions in immiscible systems is critical for advancing fluidic encapsulation strategies. Here, we uncover a previously unreported ‘endocytosis’ phenomenon in which an aqueous droplet impacts a deep immiscible pool and spontaneously engulfs the pool liquid, forming a stable double-layered liquid structure. This process origins from the focusing of capillary waves at the droplet-pool interface and followed by a pinch-off of a central oil column via Rayleigh-Plateau instability. We propose a dimensionless criterion for central column formation, determined by whether viscous dissipation of the droplet's kinetic energy. By introducing a UV-crosslinkable precursor, gelatin methacryloyl (GelMA) into the droplet, we succeed in encapsulating pool liquid into hydrogel capsules with tunable core-shell ratios by simply adjusting the impact velocity. This enables precise control of release mechanism: thick-shell capsules exhibit non-Fickian sustained release, while thin-shell capsules display delayed burst Case II release upon swelling. The proposed ‘endocytosis’ phenomena offer a surfactant-free, single-parameter controlled strategy compared to conventional liquid encapsulation methods, which offers a new opportunity for on-demand release in drug delivery and responsive materials.

Autotrophic photosynthetic 3D-bioprinting scaffolds for infected wound healing

Jinxuan Jia,Lu Fan,Zhuhao Wu,Xiaocheng Wang,Yuanjin Zhao

An autotrophic, photosynthetic 3D-bioprinted core-shell scaffold (MA-SHC-PDA) has been developed for treating infected wounds. • The microalgae core enables sustained oxygen production under light, whereas the NIR-responsive PDA shell simultaneously triggers SHC carbon release and eradicates bacteria. • The MA-SHC-PDA scaffold alleviates hypoxia, suppresses infection, and accelerates infected wound healing. Bioengineered scaffolds have demonstrated significant potential in enhancing wound healing; while there remains ample room for improvement in mitigating localized hypoxia and preventing infections at wound sites. Here, we introduce a microfluidic in situ 3D bioprinted autotrophic photosynthetic scaffold for promoting the healing of infected wounds. The scaffold incorporates oxygen-producing microalgae (MA) along with its essential nutrients (agarose and sodium bicarbonate). Its exterior is coated with a photothermal material, forming a core-shell structured scaffold capable of autonomously cultivating microalgae while exhibiting excellent oxygen-producing activity and photothermal performance. By employing 3D bioprinting to apply this living autotrophic scaffold to infected wounds, we have achieved effective alleviation of local hypoxia, reduction of oxidative stress, and concurrent inhibition of bacterial infections. These combined effects significantly promoted neoangiogenesis, collagen accumulation, and tissue regeneration in infected wounds. Thus, our autotrophic photosynthetic 3D-bioprinting scaffolds hold substantial potential and value for clinical applications in wound healing.

Versatile molecular slide-rings for the construction of composite hydrogel sensors

Lai Xiangting,Sun Qian,Zhang Meng,Jiang Zhongtian,Xiong Xueru,Wang Kaojin,Wu Gang,Sun Guoxing,Jia Yong-Guang,Zhu Xiaoxia

Hydrogel matrices for flexible electronic devices need to meet multiple criteria, including robust mechanical properties, appropriate skin adhesion, and high conductivity. Conventional hydrogels, however, often suffer from limitations such as hysteresis fatigue, poor adaptability, and structural fragility. In this work, we designed a series of polymerizable, water-soluble molecular slide-ring (MSR) units with precise host-guest molar ratios. These MSRs are based on bile acid guests functionalized with hydrophilic poly(ethylene glycol) chains of varying lengths, which specifically recognize host acrylate β-cyclodextrins. Subsequent photopolymerization of the MSRs with acrylamide yielded a range of SR hydrogels exhibiting wide tensile fracture strains and low hysteresis. Furthermore, the incorporation of nanoclay particles of laponite-RDS led to a composite slide-ring hydrogel system with improved toughness, high strength, and stable sensing performance. The composite hydrogel-based sensors not only demonstrate excellent skin adhesion and minimal skin irritation, but also monitor human motion and electrocardiogram signals with high sensitivity in both humans and mice, highlighting their potential for flexible wearable electronics. This study addresses key synthetic challenges associated with traditional slide-ring hydrogels and establishes a new paradigm for developing high-performance slide-ring materials using versatile MSRs.

Negative-Pressure-Actuated Microfluidics: A Dual-Mode Point-of-Care Sensor for Allergen-Specific IgE in Interstitial Fluid

Jun Zhang,Huiting Lian,Guangming Liu,Qingmei Liu,Bin Liu,Xiaofeng Wei

This study reports an integrated dual-signal microfluidic immunosensor for point-of-care detection of ovalbumin-specific IgE (OVA-sIgE) in interstitial fluid. A pH-responsive composite material, ZIF-8 encapsulating phenolphthalein and surface-conjugated with antimouse OVA-sIgE (ZIF-8@PP∼Abs), was synthesized and characterized. Structural and elemental analyses confirmed the successful synthesis and antibody functionalization, while the composite retained strong alkaline-triggered colorimetric response due to ZIF-8 decomposition and subsequent phenolphthalein release. Carbon dots (CDs) show concentration-dependent fluorescence, which is effectively quenched by ZIF-8@PP∼Abs. A paper-based chip coimmobilized with CDs and OVA antigen enabled simultaneous colorimetric and fluorescent detection. The colorimetric pathway operates as a “turn-on” system through alkaline-induced color change, while the fluorescent pathway functions as a “turn-off” system via CDs quenching by ZIF-8@PP∼Abs. Quantitative analysis using the green channel─identified as the most sensitive through RGB decomposition─showed linear responses across 0.06–8.00 ng/mL OVA-sIgE, with detection limits of 0.026 ng/mL (colorimetric) and 0.032 ng/mL (fluorescence). The dual-signal design, featuring built-in cross-verification, demonstrates high reliability through favorable spike-and-recovery results. ROC analysis shows AUC values of 0.9999 for both signals, with a significant reduction in false positives. To achieve operational integration, a custom-designed negative-pressure-driven microfluidic chip was developed, incorporating all necessary steps into a negative-pressure valve-controlled microfluidic chip. The chip demonstrated robust fluid handling, repeatable operation, and user-friendly functionality. The integrated sensor exhibited excellent selectivity, strong anti-interference capability against common interstitial fluid constituents. This work presents a practical, accurate, and reliable platform for decentralized allergy testing, merging advanced nanomaterials, dual-mode transduction, and microfluidic engineering.

Composite Patch with Heterogeneous Structural Design for Improved Rotator Cuff Repair

Sun Yikun,Li Mingjun,Hao GuiPeng,Yang Zhen,Yang Lei

Postoperative re-tear remains a prevalent challenge in rotator cuff repair. To address this, we developed a bioactive glass composite rotator cuff patch (BCP) with heterogeneous structural design to promote tendon−bone interface regeneration. The BCP features an outer barrier of hydrophobic poly (lactic acid) (PLA) electrospun membrane, which prevents infiltration of inflammatory fibrotic tissue. The inner structure, fabricated via a dual-nozzle 3D printing strategy, comprises gelatin methacryloyl (GelMA) hydrogel layers differentially loaded with zinc-doped (ZnBG) or strontium-doped (SrBG) bioactive glass, targeting the tendon and bone regions, respectively. Bioactive glass enables sustained ion release within the physiological environment, establishing distinct ionic microenvironments across the tissue interface. In vitro , ZnBG-released ions significantly promoted the proliferation and migration of tendon-derived stem cells, upregulated mature tendon matrix expression, and inhibited fibrotic scarring. Conversely, SrBG-released ions enhanced the osteogenic differentiation and mineralization potential of bone marrow mesenchymal stem cells. In vivo evaluation further demonstrated that the BCP effectively facilitated organized collagen regeneration and improved the quality of its integration with bone. Additionally, the BCP favorably modulated the local immune microenvironment toward a reparative phenotype. In summary, this study presents an innovative biomaterial strategy for achieving comprehensive structural and functional regeneration of the tendon−bone interface following rotator cuff injury.

Multidimensional Oriented Piezoelectric Conduits for Peripheral Nerve Defect Regeneration

Tong Qi,Xiao Wang,Lina Wu,Min Han,Changchun Zhou,Bo Wu,Xuan Pei,Chengqi He

Peripheral nerve regeneration and functional recovery hold immense clinical significance, as they are critical for reinnervating the motor and sensory functions of organs. In this study, piezoelectric nerve guidance conduits (NGCs) with multidimensional oriented structures and zinc oxide (ZnO) nanoparticles were developed for this purpose, fabricated via a combination of digital light processing (DLP) 3D printing and directional freezing technologies. The therapeutic efficacy of the NGCs was evaluated in a rat model of sciatic nerve defect, with continuous monitoring of neuromorphological and functional recovery metrics. Results indicated that the oriented structure of the NGCs and the piezoelectrically generated electric field significantly promoted both morphological regeneration and functional restoration of peripheral nerves, achieving repair effects comparable to the gold-standard autologous nerve transplantation. This work presents a promising strategy for the clinical treatment of peripheral nerve defects.

Skin-Adaptive and Visually Switchable Adhesive Hydrogels for 3D Printing with On-Demand Detachability

Xincong Dong,Xiaochen Wang,Song Hu,Xinqiang Xu,Zhengwei Lin,Zhihan Hong,Yang Lyu,Zhongying Ji,Xiaolong Wang

Functional hydrogels with on-demand detachability and a visually intelligible adhesion switch hold significant promise for wound healing, biomedical diagnostics, and wearable sensors. Conventional hydrogels offer desirable properties such as flexibility, biocompatibility, and tissue-like mechanical properties. However, developing multifunctional hydrogels that integrate skin-conformable adhesion, controllable detachment, visual adhesion switching via tunable transparency, body-temperature responsiveness, and excellent biocompatibility remains challenging. Inspired by octopus suckers and leveraging hydrogen-bond-driven adhesion between monomers, we fabricated a hydrogel with suction-cup-like structures using vat photopolymerization-based 3D printing. The resulting hydrogel exhibits strong, reversible adhesion, with adhesive strength varying with temperature. Simultaneously, its tunable optical properties enable visual monitoring of adhesion through transparency changes. Specifically, the hydrogel adheres firmly to human skin at body temperature to aid wound healing. Upon recovery, it detaches cleanly by heating to 50 °C, leaving no residue and minimizing skin damage. Moreover, the hydrogel demonstrates excellent biocompatibility, with cell survival exceeding 98%, and antibacterial activity against Gram-negative E. coli and Gram-positive S. aureus, with inhibition zones of 14 mm and 24 mm. In summary, this bioinspired 3D-printed hydrogel unites skin-adaptive adhesion, visually switchable adhesion, high biocompatibility, and on-demand detachability, offering great potential for advanced wound care, wearable electronics, and intelligent medical patches.

Janus Microgel Robots for Actively Boosting Catalytic Efficiency and Recovery of Living Materials

Changming Lan,Jing Liang,Jie Xu,Xinxin Wang,Xiao Liu,Yiran Ni,Yingnan Zhou,Ming-Bang Wu,Chao Zhang,Junqiu Liu,Baiheng Wu

Leveraging living materials such as algae as sustainable photocatalytic platform is highly promising for mitigating antibiotic water pollution; however, they are confronted with low catalytic efficiency and difficulty in recovery, as imposed by their passive, static working mode. Herein, we report a Janus microgel robot (JMR) that features the integration of magnetic-controlled mobility and living photocatalytic function, allowing for substantially boosting antibiotic degradation efficiency and efficient recovery in an actively magnetic-controlled manner. The key to the JMR lies in harnessing gas shear microfluidic technique to manipulate the spatial distribution of TiO2–Chlorella pyrenoidosa and Fe3O4 phases into a Janus architecture, followed by gel encapsulation to prevent cell leakage. Under simulated sunlight, the JMR system achieves 77% antibiotic degradation within 10 h, which is 10 times that of the free C. pyrenoidosa (7.6%). Moreover, we demonstrate that the JMR can be imparted with enhanced degradation efficiency by 10.6% and over 95% effectiveness through 3 consecutive operational cycles in actively magnetic-controlled mode. This work establishes a prototype for sustainable environmental biorobots and provides a novel strategy for photocatalytic-biological hybrid system design, advancing the next-generation living materials for water treatment.

An Injectable, Osteoconductive Gelatin-Enabled GelMA/HAp Hydrogel Scaffold for Minimally Invasive Bone Tissue Engineering

Juhan Li,Nan Xiang,Lingbin Che,Jianfeng Wu,Dianwen Song

Despite extensive exploration of gelatin methacryloyl (GelMA)-based hydrogels for bone tissue engineering, their clinical translation is hindered by a critical trade-off: poor precursor stability leads to rapid sedimentation of bioactive fillers like hydroxyapatite (HAp), while formulations optimized for injectability often sacrifice mechanical integrity or handling precision. To overcome this challenge, we report a rheologically engineered, injectable composite hydrogel scaffold that integrates unmodified gelatin as a thermoresponsive viscosity modulator into a GelMA/HAp matrix. The incorporation of gelatin yields a stable, paste-like precursor at physiological temperature, which effectively prevents HAp sedimentation and enables precise, filamentous extrusion. Subsequent UV crosslinking locks the homogeneous structure in place, resulting in a mechanically robust scaffold with significantly enhanced compressive modulus. In vitro studies demonstrate that this biomimetic microenvironment not only supports high viability and proliferation of bone marrow stromal cells (BMSCs) but also potently enhances their osteogenic differentiation, as evidenced by upregulated alkaline phosphatase activity, Runx2 expression, and matrix mineralization. This simple, one-step strategy successfully reconciles injectability, structural fidelity, and bioactivity, offering a highly promising and clinically translatable platform for minimally invasive bone regeneration.
