Cuproptosis is a newly recognized copper-dependent nonapoptotic form of cell death, which stimulate studies exploring copper-based nanomaterials to treat cancer through distinct mechanistic action. However, it remains a challenge to completely eradicate tumors via monotherapy. Herein, a copper-doped BiSe x (CBS) nanozyme was developed to boost αPD-L1-mediated immune checkpoint blocking (ICB) via synergetic apoptosis/cuproptosis-induced immunogenic cell death (ICD). The defect-engineered CBS nanozyme exhibits strong peroxidase-mimicking activities and generates abundant reactive oxygen species (ROS) production causing cell apoptosis, which could be further augmented by NIR photoirradiation. Meanwhile, the CBS could cause mitochondrial lipoylated protein aggregation, leading to cell cuproptosis. The photothermal/catalytic/cuproprosis synergistic therapy triggered by CBS nanozyme combined with αPD-L1 antibody effectively inhibits the growth of primary tumors and distant tumors in prostate cancer model. Furthermore, CBS nanozyme exhibited significant germicidal effect in wound infection. Collectively, this work provides a new insight into cancer treatment of copper-based nanozyme based on cuproptosis/apoptosis-related combined therapy.

Methicillin-resistant Staphylococcus aureus (MRSA) has been associated with significant mortality and longer hospital stays. Artificial nanozymes not requiring antibiotics can generate enough reactive oxygen species (ROS) to kill bacteria. In this study, we synthesized hexagonal copper sulfide nanosheets (CuS NSs) using a modified one-pot synthesis. The CuS NSs demonstrated photothermal stability and superior photothermal conversion efficiency due to large surface area and existence in the second near-infrared (NIR-II) window (1270 nm), promoting the production of ROS. Upon NIR-II irradiation, CuS NSs displayed an excellent biocatalytic antibacterial role in vitro and accelerated abscess resolution, and enhanced the recovery of MRSA-infected wounds in vivo . Moreover, CuS NSs demonstrated negligible toxicity following an amplifying peroxidase-like activity through hyperthermia. Overall, CuS NSs provide a feasible strategy for using the synergistic therapeutic platform for effective therapy of deep-seated MRSA-infected wounds.

Quality evaluation and consistency evaluation of drugs are the keys to ensure the therapeutic effect and safety of drugs. In this study, attenuated total refraction infrared (ATR-IR) spectroscopy and near-infrared (NIR) spectroscopy combined with chemometrics were used for rapid detection and quality evaluation of active components of Shuang-Huang-Lian injection (SHLI), a traditional Chinese medicine preparation commonly used in China. Taking the chromatographic detection results as a reference, the partial least squares (PLS) model based on ATR-IR and NIR data was constructed by removing the bands with serious noise interference and low signal frequency band. The results showed that the PLS model achieved satisfactory results for the prediction of the three active components (chlorogenic acid, baicalin and phillyrin) in SHLI, indicating that the two spectral techniques combined with the PLS regression method could be successfully used for rapid quantitative analysis of the three active ingredients in SHLI. Relatively, the PLS model based on the ATR-IR spectrum has higher R 2 and smaller RMSE than it based on the NIR spectrum. Furthermore, based on the rapid quantitative analysis of the three active ingredients in SHLI, the quality of 140 SHLI samples from seven manufacturers was evaluated by TOPSIS (technique for order preference by similarity to the ideal solution) analysis, and the consistency of different batches of SHLI products from the same manufacturer was evaluated. The results showed that there were differences in the quality of SHLI produced by different manufacturers, and the quality of different batches of SHLI produced by the same manufacturer was not completely consistent. In conclusion, ATR-IR and NIR spectroscopy combined with chemometrics can be used for accurate and rapid quantitative analysis and quality evaluation of SHLI. This study provides a good idea for the rapid quantitative analysis and quality evaluation of drugs or food based on spectroscopic technology and chemometrics.

Bacterial diseases pose a serious threat to human health. Continued development of precise diagnostic methods and synergistic therapy techniques for combating bacteria are needed. Herein a hybrid nanosystem (Ag2S@ZIF-Van NS) was constructed by one-step self-assembly of Zn2+, vancomycin (Van) and Ag2S quantum dots (QDs). The nanosystem possesses excellent second near-infrared transparency window (NIR-II) fluorescence properties (∼1200 nm emission wavelength), good photothermal conversion properties, and biocompatibility. The material system enables precise, targeted NIR-II fluorescent imaging of bacterial inflammation in vivo as well as promoting anti-bacterial and wound healing effects.

IR820, an analog of FDA-approved indocyanine green, is a promising second near-infrared window (NIR-II) fluorescence probe with better NIR-II fluorescence stability and great clinical transformation potential. Moreover, its fluorescence can be further remarkably enhanced by the interaction with albumin. Therefore, it is significant to flexibly design IR820–albumin complex using endogenous or exogenetic albumin to meet the requirements of different biological applications. Herein, we show the rational synthesis of IR820–albumin complex for NIR-II fluorescence imaging-guided surgical treatment of tumors and gastrointestinal obstruction. We compared the NIR-II fluorescence imaging ability of IR820 pre-incubated with albumin or not to visualize tumors and the gastrointestinal tract in vivo and found that the formation of IR820–albumin was essential for the intense NIR-II fluorescence. For imaging-guided tumor treatment, after intravenous injection of free IR820, IR820–albumin complex can be formed in vivo due to the presence of plenty of albumin in the blood. For imaging-guided gastrointestinal obstruction removal, IR820–albumin complex should be synthesized in vitro before oral administration. NIR-II fluorescence imaging-guided surgeries were successfully realized in both tumor resection and gastrointestinal obstruction removal. Besides, toxicity assessments in vitro and in vivo confirmed the good biocompatibility of IR820. Our study provides a flexible paradigm for IR820-based NIR-II fluorescence imaging and surgical navigation towards different diseases.

The simultaneous detection of multiple serum biomarkers of cancer with high sensitivity and point-of-care manner is of great significance for facile and early screening of cancers. In this work, the sensitivity of lateral-flow immunoassay (LFIA) platform was improved by two strategies. The NIR-Ⅱ emitting lead sulfide quantum dots (PbS QDs) were employed to replace common visible region fluorophores to reduce background interference from sample matrix. Meanwhile, numerous hydrophobic PbS QDs were concentrated and assembled within single dendritic scaffold via three-dimensional distribution manner to enhance the fluorescence intensity of individual colloidal label. The LFIA strips with multi-testing lines were performed to realize simultaneous diagnosis of carcino-embryonic antigen (CEA), cytokeratin 19 fragment (Cyfra21-1) and neuron-specific enolase (NSE). The multi-target LFIA could be completed within 12 min with precise quantitative results by a portable NIR-Ⅱ strip scanner. The analytical data illustrated wide linear detection ranges of 0.11–100 ng mL −1 , 0.18–100 ng mL −1 and 0.28–100 ng mL −1 and limit of detection of 0.11, 0.18 and 0.28 ng mL −1 for CEA, Cyfra21-1 and NSE, respectively. The multi-target LFIA revealed minor cross-reactivity of three specific antibodies conjugated labels with non-specific cancer biomarkers and interfering proteins in human serum. For real serums of lung cancer patients and healthy persons, the combination detection of the three biomarkers exhibited an improved sensitivity up to 92.7%, which was significantly higher than any individual biomarker detection, with a satisfactory specificity of 92.0%. The multi-target LFIA platform with high sensitivity and specificity holds great significance for point-of-care and simultaneous diagnosis of multiple cancer biomarkers.

The enriched surface functional group such as OH termination and unique 2D structure of Ti 3 C 2 T x MXene provide the feasibility to form hydrogen bonds with organic photosensitive materials as van der Waals heterostructures, thus can realize the improved photoresponsivity of near infrared (NIR) photodetectors (PDs). In this work, an organic–inorganic van der Waals heterostructure-based NIR PD array is designed using Ti 3 C 2 T x MXene as conductive electrodes and RAN film as active materials (Ti 3 C 2 T x -RAN PDs). The on-off ratio of the fabricated devices shows 6.25 times higher than that of PDs using thermal evaporated Au electrode (Au-RAN PDs) under 1064 nm laser excitations. The fabricated Ti 3 C 2 T x -RAN NIR PDs also possess outstanding mechanical stability with no obvious photoresponse degradation under different bending states and high transparency with a transmittance above 70% in the visible region. Owing to the high performance of the Ti 3 C 2 T x -RAN PDs, a 1024-pixel (32 × 32) image sensor array is fabricated to achieve delicate, vivid, and higher resolution imaging of a “deer,” opening up a new avenue for the biomimetic vision and flexible wearable accurate image sensing.

It is essential to develop novel multifunctional and easily synthesized stable NIR-II fluorescent probes to guide photothermal therapy for tumors. Here, we propose a new strategy to construct boron dipyrromethene (BODIPY) J-aggregates by intermolecular hydrogen bonding (H-bond) and π-π stacking interactions to achieve fluorescence emission in the second near-infrared window (NIR-II, 1000–1700 nm). A novel meso -benzamide galactose hexanoate-BODIPY (Gal-OH-BDP) amphiphilic small molecular dye was synthesized and it formed nanoparticles spontaneously in aqueous solution with a maximum emission wavelength near 1060 nm, which works as a smart nanomedicine for targeting NIR-II imaging-guided photothermal therapy (PTT) of hepatocellular carcinoma. Galactose not only provided hydrogen bonds to regulate the aggregation pattern of the molecules but also effectively targeted hepatocellular carcinoma cells and promoted the formation of well-dispersed nanoparticles of dye molecules due to their hydrophilicity . Moreover, due to high photothermal conversion efficiency (PCE = 55%), Gal-OH-BDP NPs achieve galactose-targeted NIR-II imaging and PTT, which is important for the precise diagnosis and treatment of tumors ( Scheme 1 ). In the present research work, H-bond was introduced for the first time into BODIPY for building J-aggregates to achieve the NIR-II fluorescence.

Tumor recurrence and drug-resistant bacterial infection are the main reasons that wounds heal with difficulty after skin tumor treatment. The near infrared- (NIR-) and pH-responsive, bionic , cellulose nanofiber-based (CNF-based) nanocage wound dressing with biocompatibility, bioviscosity, and shape adaptability is designed for dual NIR-triggered photothermal therapy of tumor and infection-induced wound healing. The wound dressing with the intertwining three dimensional (3D) nanocage network structure is skillfully constructed using NIR-responsive cellulose nanofibers and pH-responsive cellulose nanofibers as the skeleton, which endows the dressing with a high drug-loading capacity of doxorubicin (400 mg·g -1 ), and indocyanine green (25 mg·g - 1 ). Moreover, the NIR- and pH-responsive bionic “On/Off” switches of the dressing enable a controllable and efficient drug release onto the wound area. The dual NIR-triggered wound dressing with excellent photothermal conversion performance possesses good antibacterial properties against Escherichia coli , Staphylococcus aureus , and drug-resistant Staphylococcus aureus . It could effectively eliminate bacterial biofilms and kill A375 tumor cells. Interestingly, the bionic wound dressing with shape adaptability could adapt and treat irregular postoperative skin tumor wounds and drug-resistant bacterial infection via the synergistic therapy of photothermal, photodynamic , and chemotherapy, which provides an ideal strategy for clinical intervention.

UV-charged optical stimulated single-band phosphors are being used for single-level optical data storage because of their ability to easily write/read information by light. In contrast, UV–visible–NIR rechargeable optical stimulated double-band phosphor is urgently necessary for multiplexed high-throughput optical data storage. Here, the authors design and synthesize Mn 2+ ,Cr 3+ co-doped zinc gallate storage persistent phosphor that exhibits strong red-green bicolor persistent luminescence (PersL). Besides the traditional ultraviolet charging, the phosphor can be effectively and repeatedly charged by using visible and NIR light by one- or two-photon process. Interestingly, red and green PersL exhibit the opposite temperature dependence, which is extremely important for multilevel information storage. The mechanism of temperature-dependent PersL intensity based on different trap types and filling approaches for the double luminescence centers is proposed. The phosphor promises a sophisticated application in optical data storage of wavelength multiplexing.

Sentinel lymph node imaging is important for breast tumor staging and prediction of postoperative metastasis . However, clinical sentinel lymph node imaging has limitations such as low specificity, low contrast, and short retention time. The combination of bio-conjugates chemistry and luminescence technology may achieve the specific targeting effect. In this research, we designed a dual-targeting composite nanoprobe (∼50 nm) using a metal-organic framework (MOF) as carrier, loaded with lanthanide and ICG , and combined with hyaluronic acid and folic acid to detect metastatic lymph nodes. The coupled hyaluronic acid and folic acid can target to the tumor cells and dentritic cells with a dual-targeting effect. The FA-HA/ [email protected] nanoprobes can accumulate rapidly in sentinel lymph node with a stronger luminescence intensity (1.6 times) than that of normal popliteal lymph nodes in vivo , thus distinguish metastatic sentinel lymph node from normal effectively. Furthermore, due to the MOF carrier, the integrated lanthanide and near-infrared dye by transferring the absorbed excitation energy from ICG to Nd 3+ can enhance the signal-to-background ratio of NIR II imaging and have long retention time in vivo imaging. Finally, the FA-HA/ [email protected] @ZIF-8 nanoplatform increased the penetration depth and contrast of imaging, prolonged the retention time, and achieved the sentinel lymph nodes surgical resection. This study has important implications for lymph node imaging and surgical navigation.

Orthotopic advanced hepatic tumor resection without precise location and preoperative downstaging may cause clinical postoperative recurrence and metastasis. Early accurate monitoring and tumor size reduction based on the multifunctional diagnostic-therapeutic integration platform could improve real-time imaging-guided resection efficacy. Here, a Near-Infrared II/Photoacoustic Imaging/Magnetic Resonance Imaging (NIR-II/PAI/MRI) organic nanoplatform IRFEP-FA-DOTA-Gd (IFDG) is developed for integrated diagnosis and treatment of orthotopic hepatic tumor. The IFDG is designed rationally based on the core “S-D-A-D-S” NIR-II probe IRFEP modified with folic acid (FA) for active tumor targeting and Gd-DOTA agent for MR imaging. The IFDG exhibits several advantages, including efficient tumor tissue accumulation, good tumor margin imaging effect, and excellent photothermal conversion effect. Therefore, the IFDG could realize accurate long-term monitoring and photothermal therapy non-invasively of the hepatic tumor to reduce its size. Next, the complete resection of the hepatic tumor in situ lesions could be realized by the intraoperative real-time NIR-II imaging guidance. Notably, the preoperative downstaging strategy is confirmed to lower the postoperative recurrence rate of the liver cancer patients under middle and advanced stage effectively with fewer side effects. Overall, the designed nanoplatform demonstrates great potential as a diagnostic-therapeutic integration platform for precise imaging-guided surgical navigation of orthotopic hepatic tumors with a low recurrence rate after surgery, providing a paradigm for diagnosing and treating the advanced tumors in the future clinical translation application.
