Multifunctional CeO2/PCL Nanofibrous Scaffold with Integrated pH Sensing and Enhanced Stem Cell Adhesion

Chi Chunyu,Gao Yuantao,Chen Jiazhu,Ge Xuejun,Gao Zexing,Li Yinggang,Yan Shuang

The delayed healing of skin wounds poses significant health risks and increases medical burdens, necessitating the development of multifunctional dressings that can actively monitor and promote the healing process. In this study, a multifunctional CeO 2 /PCL nanofibrous scaffold integrating real-time pH sensing, enhanced stem cell adhesion, and antioxidant functions was developed. A series of composite scaffolds with tunable CeO 2 nanoparticle loadings (7.1–31.6 wt %) were fabricated via a facile one-step electrospinning technique. Notably, the CeO 2 /PCL-13.3 wt % scaffold achieved an optimal balance of tensile strength (1.927 MPa) and elongation at break (70%), fully satisfying clinical wound dressing requirements. Phenol red loading enabled naked-eye pH monitoring (4.0–10.0) via metal−phenolic coordination. Comprehensive evaluations demonstrated that the composite scaffolds exhibited significant antibacterial activity, potent reactive oxygen species scavenging capability, and excellent biocompatibility. The scaffolds maintained over 80% human umbilical cord mesenchymal stem cells viability within 72 h and significantly promoted stem cell migration in wound healing assays. This integrated platform unifies visual diagnostics and therapeutic functions for advanced wound management.

Multifunctional emulsion-electrospun PCL-PVA/SA@nano CB:Que nanofiber membrane for infected wound healing

Shuyue Long,Ying Huang,Zhihao Lin,Sujuan Yan,Xiaoyi Luo,Xia Ren,Xianjun Fu,Zhengao Wang,Wuyi Zhou

Emulsion electrospinning co-delivered nano cuttlebone and quercetin in fibers. • Nano cuttlebone enabled rapid hemostasis and > 96% antibacterial activity. • Nanofiber membrane afforded antioxidant activity and promoted hemostasis. • The membrane suppressed biofilms and coordinated staged infected-wound repair. Persistent bleeding, bacterial colonization, oxidative stress, and prolonged inflammation are major barriers to the repair of infected wounds. An ideal dressing should rapidly control bleeding and bacterial contamination, while regulating oxidative and inflammatory microenvironments to support tissue regeneration. Herein, a multifunctional PCL-PVA/SA@nano CB composite nanofiber membrane was developed by emulsion electrospinning for infected-wound therapy. In this system, polycaprolactone (PCL) provided structural stability, while the hydrophilic polyvinyl alcohol/sodium alginate (PVA/SA) phase improves wettability, water absorption, breathability, and bioactive-agent delivery. Nano Cuttlebone acted as a calcium-rich component for rapid hemostasis and antibacterial activity, whereas quercetin contributes antioxidant and anti-inflammatory functions. In vitro , the optimized membrane achieved antibacterial efficiencies of 96.72% against S. aureus and 96.58% against E. coli , inhibited S. aureus biofilm formation, scavenged DPPH radicals, and promoted L929 fibroblast proliferation and migration. In vivo , the membrane accelerated hemostasis in a mouse liver bleeding model and promoted S. aureus -infected full-thickness wound healing, reaching 98.9% closure on day 14. The nanofiber membrane also reduced bacterial burden, regulated macrophage polarization, suppressed CD86/TNF-α, enhanced CD206/IL-10, and promoted collagen deposition and re-epithelialization. This study provides a promising multifunctional electrospun dressing for complex infected wounds.

Electrospinning PCL-CA packaging membranes with transmittance response to ethanol for monitoring grape freshness

Li Xia,Zhina Chen,Hui Liu,Ying Xue,Hualin Wang

In recent years, there has been an increasing focus on the development of active films for monitoring and maintaining food freshness. However, its further functional diversification remains highly desirable. In this study, the active polycaprolactone (PCL) based membranes loaded with carvacrol (CA) are fabricated via electrospinning technique for grape preservation. Upon touching with ethanol exudate, the resulting membrane is capable of changing from opaque to a reversible transparent packaging (RTP) through interfibrillar fusion during food storage. The RTP exhibits good reusability with light transparency above 80 % after 10 cycles. CA is well-dispersed in the fiber matrix by hydrogen bonding and enriching the membrane with strong antibacterial activity. Furthermore, the release of CA from film matrix is well controlled and predominantly governed by Fickian diffusion. In addition to freshness monitoring, PCL-CA15 can effectively extend the shelf life of grape by about 4 days at 25 °C.

Synergistic enzyme/acid-degradable PCL-based polyurethanes with enhanced biocompatibility and shape memory properties

Meng Wu,Yingying Han,Hui You,Zhiwei Yang,Weihua Xu,Shijing Yan,Fang Wu,Taotao Fan,Jun Du

Addressing conventional polyurethane (PU) challenges including non-renewable resource reliance, recycling difficulties, and demands for functionalization, this study designed an enzymatic and acidic-synergistically degradable PU material with biocompatibility and shape-memory behavior. By incorporating acid-hydrolyzable hexahydrotriazine (HT) units and bio-based isosorbide (ISO) into polycaprolactone (PCL)-based PU chains, crosslinked PCL-PU films with dual degradability were synthesized. By regulating the ratio of ISO units and HT chain extender, the PCL-PU films exhibited superior mechanical properties, with a tensile strength of ∼31.1 MPa and elongation at break of ∼963 %, while maintaining high thermal stability. Thanks to their unique chemical structure, PCL-PU films exhibited superior synergistic degradation properties in both acidic and enzymatic environments. Specifically, they achieved rapid and full breakdown in a phosphoric acid/ethanol solution at room temperature, while exhibiting partial enzymatic degradation (∼28.7 % weight loss after 6 weeks in lipase solution). Furthermore, the PCL-PU films also demonstrated good biocompatibility, and possessed shape-memory functionality, enabling spontaneous recovery to their original shape above 37 °C. This study not only provides a new direction for the sustainable development of PU materials but also lays the foundation for medical materials, flexible sensors, smart wearable devices, adaptive structures, and environmentally friendly biomedical applications.

Mechanical, thermal properties and shape memory behaviors of PLA/PCL/PLA-g-GMA blends

Gaoyuan Ye,Tongfei Gu,Boyuan Chen,Hongjie Bi,Yingcheng Hu

To improve the toughness of polylactic acid (PLA), in this research, a tough polymer polycaprolactone (PCL) was combined with PLA to prepare blends, and a compatibilizer of glycidyl methacrylate grafted PLA (PLA-g-GMA) was produced to enhance the compatibility of PLA/PCL blend. Compared with PLA/PCL blend, the compatibilized blend exhibited a lower melting temperature and greater thermal stability. The tensile test indicates that the addition of compatibilizer greatly improved the toughness of the PLA/PCL blend. Particularly, the elongation at break of PLA/PCL blend with 6 wt% PLA-g-GMA increased to 370% and reached about 66 times of neat PLA. In addition, compatibilized blends have good shape memory performance. The research provides a simple and feasible method to overcome the brittleness of PLA, supplying a new strategy to modify PLA.

Enzymatic degradation of fluorinated Poly(ε-caprolactone) (PCL) block copolymer films with improved hydrophobicity

Qun Liu,Haipeng Wang,Lei Chen,Wulong Li,Yakun Zong,Yi Sun,Zhanxiong Li

Fluorinated Poly(ε-caprolactone) (PCL) block copolymers PCL-PTFOA(2 h), PCL-PTFOA(4 h), PCL-PTFOA(6 h), PCL-PTFOA(8 h) were prepared via esterification by using poly(1H,1H,2H,2H-perfluorooctylacrylate) (PTFOA) with different molecular weights as reactants. It was found that PCL-PTFOA(6 h) and PCL-PTFOA(8 h) possessed the similar molecular weight via GPC analysis. PCL and fluorinated PCL block copolymers films were prepared by solution casting method. The structure and properties of film surface of the fluorinated PCL block copolymers were analyzed by energy dispersive X-ray spectrometer (EDS), scanning electron microscope (SEM), Differential scanning calorimetry (DSC), water contact angles (WCAs) and the surface free energy. The results showed that the fluorinated PCL films possessed improved hydrophobicity because of the microphase separation between fluoropolymer chain and PCL chain. The enzymatic degradation of PCL, PCL-PTFOA(2 h), PCL-PTFOA(4 h) and PCL-PTFOA(6 h) films were performed at 37 °C in a phosphate buffer solution (PBS, 0.1 M, with a pH of 7.2–7.4) with lipase and analyzed by weight loss, SEM and EDS . It was found that the introduction of fluorine-containing segment reduced the rate of enzymatic degradation of the copolymer film in the early period, while increased the finally weight loss rate with a prolonged time. In addition, the enzymatic degradation rate of PCL-PTFOA(2 h) film and PCL-PTFOA(4 h) film was faster than that of PCL-PTFOA(6 h) film.

Synthesis of PCL-branched P(MMA-co-HEMA) to toughen electrospun PLLA fiber membrane

Suchen Jiang,Huanchao Liu,Xue Zhang,Yajun Ren,Xinxiang Cui,Xiaofeng Song

Electrospun biodegradable fiber mesh is a promising alternative scaffold for delivering progenitor cells for repairing damaged or diseased tissue, but its cripple mechanical stability has not met the requirement of tissue engineering yet. In this work, the well-defined poly(ε-caprolactone)-branched poly(methyl methacrylate- co -hydroxyethylmethacrylate) (PCL-PMH) has been successfully synthesized to toughen electrospun poly( l -lactide) (PLLA) fiber membrane. Characterization of the obtained nanofibrous meshes indicates that PCL-PMH and PLLA can be well blended to make smooth fibers, and fibrous diameter vary little with blending PCL-PMH. The aggregation state of two macromolecules is closely correlated with blend ratio, molecular structure, and molecular weight of PCL-PMH, and only when PCL-PMH and PLLA form good interfacial adhesion can PMH give full play to its potential for toughening the fiber membrane. The tensile strength and elongation at break of the blend are 6.20 MPa and 63.40% under the optimal conditions, respectively, and it also exhibits the representative feature of toughness materials. The blending fiber membrane is as no cytotoxic as original PLLA. This work will provide a new way for toughness of electrospun fiber membrane in practice.

Preparation and Characterization of Simvastatin-Loaded PCL/PEG Nanofiber Membranes for Drug Sustained Release

Lu Pan,Jiahao Yang,Lan Xu

Simvastatin (SIM) particles are liposoluble drugs with large particle sizes, resulting in poor compatibility with electrospun polycaprolactone (PCL)/polyethylene glycol (PEG) nanofibers, so that part of them will be exposed to the electrospun nanofiber surface, which is easy to cause the burst release of drugs. Therefore, in this paper, stearic acid (SA) with good biocompatibility was innovatively added to increase the dispersion uniformity of SIM in the spinning solution, thus improving the performances of SIM-loaded PCL/PEG nanofiber membranes (NFMs). Accordingly, the effects of SA addition on the morphologies, mechanical properties, wettability, and drug release properties of the SIM-loaded NFMs were studied. The results showed that after SIM was dissolved in SA solution, the particle size of SIM was significantly reduced and could be evenly dispersed in the polymer spinning solution, thus obtaining the SIM-loaded composite NFMs with the best morphology and performance. Keywords:electrospinning;simvastatin;composite nanofiber membranes;drug release

A mussel-bioinspired coating strategy to licence PCL osteoinductive activity for repairing bone defects

Wentao Shi,Zhijia Shen,Lu Bian,Yan Gao,Yiqing Wu,Feifei Zhao,Hong Tang,Xiaojie Lu

3,4-Dihydroxyphenylalanine (DOPA) is believed to play an essential role in the solidification and adhesion of liquid mussel adhesives; this thin polymer layer very tightly adheres to the surface of scaffolds, making it stable enough to prevent it from being peeled off. Fascinatingly, the deposition of DOPA on various types of surfaces via the self-polymerization of DOPA under alkaline conditions is a simple approach for creating biomimetic coatings. In this study, DOPA derivatives were used as monomers to form primary coatings on polycaprolactone (PCL) surfaces. After that, a second chemical crosslinking method was utilized to graft alendronate (AL) on the DOPA coatings via genipin (GP) crosslinking ( [email protected] ). X-ray photoelectron spectroscopy, Fourier transform infrared reflection and scanning electron microscopy analyses indicated that AL was distributed homogeneously on the surface of PCL. This new organic–inorganic hybridized coating could support EMSC growth and osteogenic differentiation. Additionally, animal studies were also performed to verify the function of [email protected] in promoting osteogenesis. The method is simple, cheap and highly efficient, which makes it very convenient to apply on a large scale.

3D PCL/collagen nanofibrous medical dressing for one-time treatment of diabetic foot ulcers

Ting Chang,Huiyi Yin,Xi Yu,Lihuan Wang,Longfei Fan,John H. Xin,Hui Yu

Nanofibrous dressings exhibit high specific surface areas, good histocompatibility , enhanced wound healing, and reduced inflammation, which have broad technological implications for treating diabetic foot ulcers (DFUs). However, current nanofibrous dressings still suffer from high resistance to cell infiltration and multiple dressing changes. In this study, polycaprolactone (PCL) and collagen were adopted as electrospinning materials to prepare a 3D PCL/Collagen (PC) nanofibrous dressing (3D-PC) using aqueous phase fibre reassembly technology. The matrix metalloproteinases (MMPs) inhibitor doxycycline hyclate (DCH)-loaded halloysite nanotubes (HNTs) ( [email protected] ) and antibacterial agent cephalexin (CEX) were loaded onto the dressing to prepare a multifunctional 3D drug-loaded PCL/Collagen nanofibrous dressing to promote DFU wound healing. The obtained 3D nanofibrous dressing exhibited high water absorption capacity and swelling capacity. It showed good antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) in vitro antibacterial test. In addition, the 3D nanofibrous dressing demonstrated good biocompatibility. It could significantly reduce the frequency of dressing changes and improve the healing of DFU wounds compared with the conventional multiple dressing changes method, suggesting a potential candidate for healing diabetic wounds.

An asymmetric wettable PCL/chitosan composite scaffold loaded with IGF-2 for wound dressing

Shuang Yang,Linhao Lan,Mingda Gong,Ke Yang,Xiaoming Li

An effective dressing is essential for wound healing. In fact, the wettability performance is one of the most important factors of a wound dressing. The fundamental functions of a wound dressing involve the absorption of excess exudates and maintenance of optimal moisture at the wound by controlling water evaporation. Here, we designed a type of chitosan (CS) sponge and PCL nanofibrous membrane composite dressing with asymmetric wettability surfaces as wound healing materials for biomedical applications. The hydrophobic surfaces of the composite dressing were waterproof and could efficiently control the water vapor transmission rate, whereas the hydrophilic surface of the CS sponge had good cytocompatibility and water-absorbing capability. Insulin-like growth factor-2 (IGF-2) was added to the CS sponge, and exhibited a stimulatory effect on fibroblasts migration and proliferation. Therefore, the fabricated CS sponge and PCL membrane composite dressing had excellent cytocompatibility, vapor transmission rate, and liquid absorption and asymmetric wettability, suggesting its potential as a promising alternative to traditional wound dressing.
