Antioxidant peptides from the sturgeon gill protein: purification, identification, molecular docking analysis and antioxidative mechanisms evaluation

Yan Zhu,Lingling Zhu,Zitao Guo,Wei Zhang,Wengang Jin,Ruichang Gao

Sturgeon gills, a by-product of sturgeon processing, constitute approximately 2% of the fish's total body weight but are largely underutilized, resulting in significant protein resource waste. This study aimed to isolate antioxidant peptides from sturgeon gill protein hydrolysates and evaluate their protective effects against oxidative damage in HepG2 cells. Using ultrafiltration, gel filtration, and RP-HPLC, 1,396 peptides were identified, among which FPGLP (FP5), GIPGF (GF5), and APDPF (AF5) showed the lowest binding energies to MPO (-8.6, -8.3, and -8.1 kcal/mol, respectively). These peptides significantly reduced intracellular ROS levels, and decreased cell apoptotic rates and inhibited ROS-mediated MAPK pathway phosphorylation. At 50 μM, FP5, GF5, and AF5 decreased ROS levels by 36.14%, 38.56%, and 33.75%, respectively, and suppressed phosphorylation of ERK, JNK, and p38. In conclusion, this study provides a theoretical basis for the high-value utilization of sturgeon by-products and their potential applications.

Mechanism of purslane extract inhibiting oxidation and modifying physicochemical properties of pork myofibrillar proteins under oxidation system

Haijun Chang,Yu Huang,Yuanwei Shi,Yu Hu,Wenbin Zhou

The research aimed to elucidate how purslane extract (PE) inhibited pork myofibrillar proteins (MPs) oxidation and modulated the physicochemical properties in the Fenton oxidation system. This study employed UHPLC-QE-Orbitrap-MS/MS to comprehensively analyze the PE constituents and evaluate their antioxidant properties. Furthermore, the effects of various PE concentrations (0.05, 0.10, 0.15, 0.20, and 0.40 mg/mL) on the structural characteristics of pork MPs in a hydroxyl radical oxidation system were evaluated. The findings indicated that PE was rich in phenolics, with luteolin, kaempferol, genistein, rutin, diosmetin, morin, kaempferol-3-O-rutinoside, isoquercitrin, and isorhamnetin-3-O-neohesperidoside being the nine most prevalent compounds. These compounds contributed to the extract's strong free radical scavenging activity. Experimental results demonstrated that adding 0.20 mg/mL PE significantly reduced the carbonyl content compared with oxidized MPs while increasing total thiol, free thiol, and free amino group levels ( P  < 0.05). The protective effect of PE on MP amino acid side chains was superior to that of Butylated hydroxyanisole (BHA). Moreover, PE addition protected the secondary structure of MPs and minimized the α-helix loss caused by oxidation. In addition, PE decreased the intrinsic fluorescence intensity of MPs, enhanced their surface hydrophobicity, and reduced their solubility. SDS-PAGE results indicated that oxidation and PE addition induced MP aggregation, with most of it being reversible. These findings support the potential of PE as a natural antioxidant in meat products and offer a theoretical basis for its functional applications in meat processing.

Quantitative Analysis of Ginger Maturity and Pulsed Electric Field Thresholds: Effects on Microstructure and Juice’s Nutritional Profile

Zhong Han,Pan He,Yu-Huan Geng,Muhammad Faisal Manzoor,Xin-An Zeng,Suqlain Hassan,Muhammad Talha Afraz

This study used fresh (young) and old (mature) ginger tissues as model systems to investigate how plant maturity modulates the response to pulsed electric field (PEF), a non-thermal processing technology. Specifically, the influence of tissue maturity on dielectric behavior and its downstream effect on juice yield and bioactive compound extraction was systematically evaluated. At 2.5 kV/cm, old ginger exhibited a pronounced dielectric breakdown effect due to enhanced electrolyte content and cell wall lignification, resulting in a higher degree of cell disintegration (0.65) compared with fresh ginger (0.44). This translated into a significantly improved juice yield of 90.85% for old ginger, surpassing the 84.16% limit observed in fresh ginger. HPLC analysis revealed that the extraction efficiency of 6-gingerol and 6-shogaol increased from 1739.16 to 2233.60 µg/g and 310.31 to 339.63 µg/g, respectively, in old ginger after PEF treatment, while fresh ginger showed increases from 1257.88 to 1824.05 µg/g and 166.43 to 213.52 µg/g, respectively. Total phenolic content (TPC) and total flavonoid content (TFC) also increased in both tissues, with OG-2.5 reaching 789.57 µg GAE/mL and 336.49 µg RE/mL, compared with 738.19 µg GAE/mL and 329.62 µg RE/mL in FG-2.5. Antioxidant capacity, as measured by ABTS•+and DPPH•inhibition, improved more markedly in OG-2.5 (37.8% and 18.7%, respectively) than in FG-2.5. Moreover, volatile compound concentrations increased by 177.9% in OG-2.5 and 137.0% in FG-2.5 compared with their respective controls, indicating differential aroma intensification and compound transformation. Structural characterization by SEM and FT-IR further corroborated enhanced cellular disruption and biochemical release in mature tissue. Collectively, these results reveal a maturity-dependent mechanism of electro-permeabilization in plant tissues, offering new insights into optimizing non-thermal processing for functional food production.

LC-MS/MS Reveals Variation in Bioactive Flavonoid Profiles and MYB Regulatory Networks in Wild vs. Cultivated Strawberries (White, Pink, Red)

Mingzheng Duan,Ting Jiang,Xi Wang,Kangjian Song,Xu Xiao,Xiaoting Fu,Sijiu He,Jieming Feng,Muhammad Junaid Rao,Hengcui Guo

Strawberries ( Fragaria × ananassa ) are prized for bioactive flavonoids and anthocyanins, contributing to antioxidant capacity and health benefits. However, decades of breeding for yield and shelf-life have eroded their phytochemical diversity. To explore this trade-off, we compared wild strawberries ( Fragaria nilgerrensis , HM) with three cultivated varieties (pink-FY, red-RF, white-DX) using integrated LC-MS/MS metabolomics and transcriptomics. Wild HM strawberries contained twice the vitamin C (134.2 mg/100g), 2.5× more total flavonoids (138.7 mg/100g), and higher antioxidant activity (367 μg Trolox/g) than commercial varieties. In contrast, red-fruited RF showed abundant anthocyanins (pelargonidin-3-glucoside 6.6% abundance), while the wild HM genotype accumulated higher levels of flavonoids, with neohesperidin, lonicerin, hyperin and ternatumoside II collectively representing 20.9% of total flavonoids - concentrations 3-169 times greater than those observed in the three commercial varieties (FY, RF, DX). Transcriptomic analysis (RNA-seq, Illumina HiSeq 2500) revealed distinct MYB transcription factor associations: FxaYL_611g0111200 (MYB66) showed strong positive correlation with anthocyanin biosynthesis in RF (r=0.92, **p<0.005 ), while FxaYL_242g0472670 (MYB44-like) was significantly associated with HM's unique antioxidant flavonoid profile (r=0.96, ***p<0.0001 ). These regulatory relationships were corroborated by hierarchical clustering demonstrating varietal-specific expression patterns and principal component analysis (82.6% variance explained), collectively providing molecular targets for enhancing nutritional quality in strawberry breeding programs.

ROS-responsive hydrogel loaded with berberine liposome protects against intervertebral disc degeneration by inhibiting mitochondrial fission and apoptosis

Cunliang Guo,Lilong Du,Xiaoxun Du,Rui Gao,Xinyi Jiao,Zhenhua Li,Tongxing Zhang,Pingsheng Huang,Chuangnian Zhang,Zujian Feng,Weiwei Wang,Baoshan Xu

Intervertebral disc degeneration (IVDD) is closely associated with mitochondrial dysfunction caused by the excessive accumulation of reactive oxygen species (ROS). Current therapeutic strategies are limited in their ability to precisely intervene in mitochondrial dynamics imbalance. This study develops a ROS-responsive hydrogel crosslinked via dynamic borate ester bonds between boric acid-modified hyaluronic acid and polyvinyl alcohol (PVA), which was further loaded with berberine liposomes (HP-Lipo@BBR), aiming to scavenge excessive ROS, and restore mitochondrial function. The hydrogel exhibited excellent degradability, injectability, self-healing properties, antioxidative effects, and biocompatibility. It triggers intelligent drug release in the high-ROS microenvironment of degenerated discs. The liposomes efficiently encapsulate berberine, facilitating intracellular drug delivery via the endocytosis of nucleus pulposus cells. Mechanistically, berberine scavenges ROS, inhibits abnormal phosphorylation of dynamin-related protein 1 (p-Drp1), and reduces excessive mitochondrial fission, thereby blocking the caspase-3-mediated apoptosis pathway. In a rat disc puncture degeneration model, intradiscal injection of HP-Lipo@BBR effectively maintains intervertebral disc height and the hydration of nucleus pulposus tissue, inhibits mitochondrial fission-apoptosis axis, and improves the regeneration of extracellular matrix, with therapeutic effects significantly superior over free berberine or hydrogel. This study provides a new strategy for minimally invasive treatment of IVDD and other related degenerative diseases.

Cunshi Zhao,Lili Song,Yuanxun You,Yi Yang,Dong Wang,Zefang Xiao,Yanjun Xie,Qingbo Wang,Yonggui Wang

Polylactic acid (PLA), as a biodegradable material, is considered an ideal alternative to petroleum-based plastics. The intrinsic mechanical fragility and lack of multifunctional characteristics significantly constrain its practical utilization in advanced applications. This study designed and prepared a ternary PLA composite material with uniform surface and enhanced functionality by uniformly dispersing esterified CNC (E n CNC) Curcumin (Cur) hybrid nanostructures in the polymer matrix. The synergistic interaction between the E n CNC and Cur significantly enhanced their interfacial compatibility with the PLA matrix, leading to uniform dispersion and significantly improved flexibility. The flexibility of resulting E n CNC_Cur_PLA composite films was significantly improved, with elongation at break increased to 64.42 %, representing a 628.59 % increase compared to pure PLA. The addition of EnCNC increased the bioavailability of Cur and demonstrated multifunctional effects, including controlled release of Cur in ethanol, enhanced antioxidant activity (80.6 % DPPH clearance) and antibacterial activity (over 99 % against Escherichia coli, 87.7 % against Staphylococcus aureus ). Overall, these results highlight a simple and effective strategy to fabricate multifunctional, sustainable bio-based films with improved mechanical performance and biological activity, suitable for advanced food packaging applications.

Lei Yang,Xuanxuan Yuan,Dian Dai,Shuqiang Wang,Ying Wu,Haihua Wang,Zhongguo Shan

The growing concern over food safety and environmental pollution has stimulated the development of multifunctional biodegradable films. Herein, β-Cyclodextrin metal-organic frameworks (β-CD-MOFs) were prepared as loading carriers for chlorogenic acid (CGA) and silver nanoparticles (Ag NPs). These materials (CGA-Ag@CD-MOFs) were then added into a zein matrix to construct bioactive packaging films (MOFs-Zein). SEM showed that appropriate content of CGA-Ag@CD-MOFs (2 wt%) promoted the formation of a more uniform and denser film structure. FT-IR verified that hydrogen bonding between CGA-Ag@CD-MOFs and zein matrix endowed MOFs-Zein-2 % with superior tensile strength (24.61 ± 2.58 MPa), elongation at break (5.01 ± 0.71 %), barrier properties of water vapor (1.84 × 10 −7  g·m −1 ·h −1 ·Pa −1 ) and oxygen (0.63 × 10 −10  g·cm/cm 2 ·s). Moreover, it can control the release of both CGA and Ag + , which displayed high-efficiency antioxidant and antimicrobial activities against Escherichia coli O157:H7 and Staphylococcus aureus . Comprehensive biosafety evaluation through cytotoxicity (Caco-2 viability >98 %), hemolysis (< 5 %), and acute toxicity studies confirmed material safety. When applied for grape storage, MOFs-Zein-2 % effectively preserved nutritional quality and texture, and prolonged the shelf-life of grapes by 16 days. Edible Films Based on Fish Gelatin and Soluble Soybean Polysaccharide Enriched with Tea Polyphenol for Active Food Packaging

Jie Liu,Zhongfeng Song,Yiwei Wang,Ying Pei,Keyong Tang

The increasing demand for environmentally friendly alternatives to conventional plastic packaging has driven interest in the development of biodegradable edible films with functional properties. In this work, edible blend films were developed based on fish gelatin (FG), soluble soybean polysaccharide (SSPS), and tea polyphenol (TP) for active food packaging applications. The FG/SSPS/TP films were prepared by solvent casting and characterized in terms of their structural, mechanical, optical, thermal, and barrier properties. FTIR, SEM, and XRD analyses revealed TP-induced morphological and structure changes in the biopolymer matrix. The incorporation of TP significantly enhanced the antioxidant activity and UV-shielding properties of the films, while also modifying their flexibility and surface hydrophilicity. The packaging performance of FG/SSPS/TP films was evaluated using beef tallow as a model food product. Compared to neat FG/SSPS and polyethylene films, the FG/SSPS/TP films effectively suppressed lipid oxidation of the samples during storage. The results demonstrated that the prepared FG/SSPS/TP films possess strong potential for use as edible and active packaging materials for food products.

Novel galloyl acylglycerols amphiphilic antioxidants for multilayer and multivesicular liposomes protection

Humera Rani,Songbai Liu

In this study, novel amphiphilic antioxidants, galloyl acylglycerols (GAGs), were successfully synthesized by integrating gallic acid (GA) with acylglycerols of varying acyl chain length (C 12 -C 18 ) using mild Steglich esterification. The structural identities of these derivatives were confirmed through NMR and MS analysis. Further evaluation revealed that GAG-C 18 exhibited excellent radical scavenging activity and superior antioxidant performance in both simple and complex liposomal dispersions compared to other gallic acid derivatives. Additionally, GAG-C 18 effectively minimized the degradation and leakage of bioactives including curcumin and anthocyanins in multilayer and multivesicular liposomes. Microscopic investigation by confocal microscopy further demonstrated the visual results of the GAG-C 18 to protect the liposome under oxidation. As an amphiphilic antioxidant, GAG-C 18 demonstrated increased efficiency in lipid dispersions by preferentially accumulating in the oil-water interface, where oxidation predominantly occurs. The remarkable antioxidant performance of naturally derived amphiphilic GAG antioxidants makes them ideal candidates for ensuring oxidative stability in delivery systems.

An active packaging film prepared from aqueous two-phase Pickering emulsion and chitosan for fruit preservation

Jiayan Zhang,Changyong Cai,Yuxiao Hu,Zhijian Tan

Aqueous two-phase Pickering emulsions (ATPPEs) are advantageous because they are biocompatible and environmentally friendly. Developing active packaging films using ATPPE with antimicrobial, antioxidant, and sustained-release properties is an effective strategy to extend food shelf life. In this study, an ATPPE stabilized by cellulose nanocrystals (CNC) was prepared using a polyethylene glycol (PEG)/dextran (Dex) aqueous two-phase system (ATPS), encapsulating epigallocatechin gallate (EGCG) as a bioactive component. An active packaging films based on chitosan and EGCG-loaded ATPPE were prepared and characterized, which revealed that the ATPPE is highly biocompatible with the chitosan matrix. And the prepared composite film provides protection and sustained release for EGCG, as well as excellent mechanical, barrier, antibacterial and antioxidant properties. This composite film significantly inhibited the spoilage of bananas and tomatoes, extending their shelf life to 8 and 12 d, respectively. Hence, the developed active packaging film is a promising material for fruit preservation.

Selenized neural stem cell-derived exosomes: A neotype therapeutic agent for traumatic injuries of the central nervous system

Wenjing Wang,Guihong Lu,Peilin Guo,Haochong Zhang,Yan Wang,Diwei Zheng,Chengliang Lyu,Dongfang Wang,Shang Li,Feng Li,Jiawei Zhao,Meng Qin,Weiping Li,Hui Tan,Guanghui Ma,Wei Wei

Oxidative damage and neuroinflammation are the key features of central nervous system (CNS) injury. Inspired by the neuroprotective properties of neural stem cell-derived exosomes (NExo) and the reactive oxygen species (ROS) scavenging ability of selenium, we develop an advanced NExo bearing ultrasmall nano-selenium (∼3.5 nm) via lipid-mediated nucleation (SeNExo). In addition to maintaining the biological components of NExo, the resulting SeNExo exhibits a Se–O bond that dramatically enhances its ROS-scavenging performance. SeNExo penetrates the blood-brain barrier (BBB) via the apolipoprotein E and prolow-density lipoprotein receptor-related protein 1 (APOE_LRP-1) interaction. Through proteomics, microRNA (miRNA) omics, and single-nucleus RNA sequencing, we find that SeNExo can alleviate neuronal apoptosis, restore glia homeostasis, and remodel glia-neuron networks. Therefore, SeNExo confers potent therapeutic benefits, significantly reducing cerebral lesions in a murine traumatic brain injury model. Even extending to a murine spinal cord injury model, SeNExo promotes locomotory recovery, further supporting SeNExo as a neotype and a promising therapeutic agent for treating traumatic CNS injury.
