PTIO as a redox mediator to enhance organic contaminants oxidation by permanganate

Lin Zhao,Jing Zhang,Jun Ma

Although permanganate (Mn(VII)) is extensively utilized as a strong oxidizer for the purification of water, the direct reaction rates between some refractory pollutants and Mn(VII) are moderate or relatively low. In this study, we found that 2-phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl (PTIO), could act as a redox mediator to enhance bisphenol A (BPA) degradation by Mn(VII) at pH 5.0 − 9.0, with a removal higher than 80% over 5 min. Moreover, the Mn(VII)/PTIO system is highly efficient toward a broad spectrum of contaminants. Mechanism was elucidated as following: PTIO was oxidized by Mn(VII) to PTIO + , an oxoammonium cation. As a newly generated reactive species, PTIO + could oxidize organics and be reduced to PTIOH (PTIO hydroxylamine) or PTIO simultaneously. The redox cycle of PTIO in consecutive runs as an electron shuttle proved its stability and reusability in Mn(VII) oxidation. In addition to being an electron shuttle, PTIO also acts as an activator of Mn(VII) to promote the production of MnO 2 , which plays a vital role in enhancing BPA abatement at the acidic condition. For the purpose of further understanding the interaction between PTIO and target contaminants, three corresponding degradation pathways for BPA were proposed. Notably, the transformation products of BPA coupling with PTIO were detected, indicating PTIO inhibited the self-coupling of BPA and facilitated the ring-opening pathway. In addition, the ubiquitous humic acid has a positive effect on the Mn(VII)/PTIO system, suggesting a high promise of this system for practical application.

Liping Yu,Wang Cheng,Meifen Tian,Zhigang Wu,Xiaoli Wei,Xing Cheng,Mingwei Yang,Xuan Ma

The medicinal value ofP. zhennanhas been documented in traditional Chinese medicine books. The aim of this paper was to study the antioxidant activity of alcoholic extracts ofP. zhennanleavesusing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2-phenyl-4,4,5,5-tetramethylimidazolineoxyl-1-oxyl-3-oxide (PTIO) radical scavenging and ferric ion reducing antioxidant power (FRAP) assays. The active components of the leaves were identified via headspace solid-phase microextraction and gas chromatography–mass spectrometry (HS-SPME-GC-MS). The results showed that the scavenging rate of DPPH was 94.67%with an EC50 value of 0.674 mg/mL at a concentration of 2 mg/mL. The maximum scavenging rate was 47.40% at a Trolox equivalent of 0.33 mg TE/mL for PTIO radicals. The FRAP reached 84.80% at 0.20 mg/mL concentration. The results confirmed the strong antioxidant activity of the extracts. Furthermore, 44 compounds, mostly terpenoids, obtained from the alcoholic extracts ofP. zhennanleaves were analyzed using HS-SPME-GC-MS and 15 of these compounds had a relative content exceeding 1%. The strong antioxidant activity of the alcoholic extracts ofP. zhennanleaves could be attributed to the presence of copaene (33.97%), β-caryophyllene (4.42%), δ-cadinene (11.04%), γ-muurolene (4.78%), cis-calamenene (2.02%), linalool (1.04%), α-pinene (1.46%), borneol acetate (1.5%), and γ-terpinene (0.66%). This study demonstrates the potential medicinal value of alcoholic extracts ofP. zhennanleaves.

2-Phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-Oxide (PTIO•) Radical Scavenging: A New and Simple Antioxidant Assay In Vitro

Xican Li

Current in vitro antioxidant assays have several limitations, which frequently cause inconsistent results. The study develops a new antioxidant assay using the 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-oxide radical (PTIO•). After the investigation of various factors, the experimental protocol was briefly recommended as follows: PTIO• and the sample solution were added to phosphate buffer (pH 7.4, 50 mM), incubated at 37 °C for 2 h, and then spectrophotometrically measured at 557 nm. The validation test based on 20 pure compounds and 30 lyophilized aqueous extracts suggested that PTIO• scavenging had a good linear relationship, stability, and reproducibility. In the ultra-performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry analysis, PTIO• was observed to give m/z 234 when encountering l-ascorbic acid. As an antioxidant assay, PTIO• scavenging possesses four advantages, i.e., oxygen-centered radical, physiological aqueous solution, simple and direct measurement, and less interference from the tested sample. It can also satisfactorily analyze the antioxidant structure–activity relationship. PTIO• scavenging has no stereospecificity and is at least involved in H+ transfer.

An analytical method for the determination of flavonoids in Houttuynia cordata Thunb. by UPLC-TQ-MS and analysis of its antioxidant activity

Zhang Zhiqi,Li Lin,Wang Xinyang,Liu Liyan

Houttuynia cordata Thunb. (H.cordata) , as a traditional Chinese medicinal herb and food, has been used to treat various diseases because of rich content in flavonoids. Here, an ultra-high performance liquid chromatography with triple quadrupole tandem mass spectrometry (UPLC-TQ-MS/MS) method was proposed for the determination of flavonoids profiling in H.cordata . Single factor experiment and response surface methodology were used to optimize the pretreatment process of H.cordata samples. The optimal conditions were as follows: ultrasonic temperature: room temperature (25 °C); ultrasonic time: 5 min; solid-liquid ratio: 1:40 and concentration of ethanol: 60%. The method showed the good linearity ( r  ≥ 0.9855), the satisfactory intra-day (RSD%≤10.94%) and inter-day precision (RSD% ≤ 11.2%) and low limit of quantification (< 0.01 µg/mL). The content of flavonoids in flowers and leaves was much higher than in roots and stems. The flavonoids in the leaves of H.cordata from Chongqing was the highest (9.22 mg/g), and that in the flowers of H.cordata from Hubei province was the highest (6.69 mg/g). In addition, the antioxidant activity of the extracts of H.cordata was evaluated using the scavenging effect of the extracts on five free radicals and the iron ion reducing ability. The best antioxidant activity was observed in scavenging DPPH free radicals with EC 50 value of 295.16 µg/mL. The scavenging of ABTS•+ free radicals and PTIO• free radicals was also effective with EC 50 values of 818.82 µg/mL and 685.43 685.43 µg/mL, respectively. Thus, this study will be helpful to the development and utilization of flavonoids from H.cordata in food and pharmaceutical industries.

A natural polyphenolic nanoparticle--knotted hydrogel scavenger for osteoarthritis therapy

Qinfeng Ding,Yitong Wang,Tianyou Wang,Chengyao Zhang,Shengbing Yang,Lu Mao,Yiyun Cheng,Yiwen Li,Kaili Lin

Exploring highly efficient and cost-effective biomaterials for osteoarthritis (OA) treatment remains challenging, as current therapeutic strategies are difficult to eradicate the excessive reactive oxygen species (ROS) and nitric oxide (NO) at damaged sites. Tea polyphenol (TP) nanoparticles (NPs), a nature-inspired antioxidant in combination with 2-(4-Carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (carboxy-PTIO), a NO scavenger, could provide maximized positive therapeutic effects on OA by eradicating both ROS and NO. Notably, this combination not only improves the half-life of the TP monomer and the drug loading efficiency of carboxy-PTIO but also prevents nitrite from being harmful to tissue. Moreover, the protonation ability of carboxy-PTIO allows smart acid-responsive release in response to environmental pH, which provides conditioned treatment strategies for OA. In in vitro experiments, TP/PTIO NPs downregulated proinflammatory cytokine release via synergistic removal of ROS and NO and suppression of ROS/NF-κB and iNOS/NO/Caspase-3 signaling. For in vivo experiments, NPs were cross-linked with 4-arm-PEG-SH to form an injectable hydrogel system. The release of TP and carboxy-PTIO from the system efficiently prevents cartilage inflammation and damage via similar signaling pathways. Overall, the proposed system provides an efficient approach for OA therapy.

Copper Nanocluster-Decorated Magnesium Silicate-Based Microneedle Enhances Antimicrobial Effects and Tissue Remodeling for Diabetic Wounds

Shuo Tan,Hua Zeng,Wenshuya Li,Haibo Liu,Xuefeng Gu,Xiong Luo,Xinyu Zhao

Diabetic skin lesions, as one of the most common complications of diabetes, present chronic nonhealing wounds that face dual challenges of antibiotic-resistant bacteria threat and insufficient microenvironment regulation due to hyperglycemic conditions, bacterial infections, and multiple pathological factors (e.g., hypoxia and reactive oxygen species (ROS) accumulation and growth factor deficiency). This study develops a microneedle (MN) system integrated with copper nanocluster-decorated magnesium silicate nanoparticles (denoted as MS@Cu MNs), which enables efficient diabetic wound healing via a synergistic multimechanism strategy. Leveraging the unique enzyme-mimetic activity of copper nanoclusters (CuNCs) and the angiogenic properties of magnesium silicate nanoparticles (MS NPs), the engineered MS@Cu nanocomposites demonstrate: 1) broad-spectrum antibacterial efficacy (sterilization rate >99.9%), 2) microenvironment regulation via simultaneous hypoxia mitigation, ROS scavenging, and angiogenesis promotion, and 3) enhanced fibroblast proliferation and migration through PI3K-AKT signaling pathway activation. The MN system using γ-polyglutamic acid (γPGA) as a matrix exhibits both superior mechanical strength and excellent biodegradability. In vivo studies demonstrated accelerated closure of infected diabetic wounds in animal models, with histological analysis revealing robust mature collagen deposition and tissue regeneration. This study develops an integrated strategy for chronic diabetic wound management, combining potentiated antibacterial activity with targeted microenvironment remodeling.

Chitosan-sodium tripolyphosphate-zinc nanogel for synergistic hydrogen and ion release to eradicate Helicobacter pylori and promote gastric mucosal healing

Xiaoyi Zheng,Yijie Xie,Linlin Zhao,Xiaojuan Huang,Shige Wang,Yongkang Lai,Peng Fan,Yanbo Zeng,Jiulong Zhao,Zhaoshen Li

This study developed a chitosan (CS)‑sodium tripolyphosphate (TPP)‑zinc (Zn) nanogel for treating Helicobacter pylori ( H. pylori ) through synergistic antibacterial, anti-inflammatory, and mucosal repair mechanisms. The nanogel was synthesized via an emulsion method, leveraging electrostatic interactions between CS amino groups and TPP phosphate ions, followed by Zn incorporation. Characterization revealed uniform spherical nanoparticles (~200 nm) with high Zn content (20.18 %) and controlled hydrogen (H 2 ) and Zn 2+ release (15 h) in acidic environments. In vitro assays demonstrated potent antibacterial activity against H. pylori (95.83 % inhibition at pH = 2; 28.54 % inhibition at pH = 7) attributed to Zn 2+ -mediated enzyme disruption. The nanogel exhibited significant antioxidant capacity, scavenging 82.3 % of PTIO and 65.48 % of DPPH radicals. Biocompatibility testing confirmed minimal haemolysis (<5 %) and high cell viability (>95 %). In vivo studies in H. pylori -infected mice demonstrated a bacterial inhibition rate of 97.54 % through upregulation of β-catenin and tight junction proteins. Concurrently, mucosal repair was enhanced, achieving migration rates of 44.2 ± 5.6 % and 72.34 ± 2.1 % at 12 and 24 h respectively. The nanogel also preserved gut microbiota diversity post-treatment, unlike conventional antibiotics. This multifunctional nanogel platform combines biocompatibility, ease of manufacture and dual therapeutic effects. In summary, this study proposes a multifunctional therapeutic strategy capable of simultaneously addressing H. pylori infection, gastric inflammation and mucosal damage. It offers a paradigm shift from traditional antibiotic-centred therapies, providing a promising strategy for eradicating Helicobacter pylori and restoring gastric injury.

Cu–Fe3O4 Nanozyme Reverses Age-Impaired Alveolar Healing via Reactive Oxygen–Nitrogen Species Scavenging and Mitophagy Reactivation

Zheng Cheng,Jikang Cao,Haobo Lin,Yuchen Wang,Ran Qin,Yuhao Ruan,Xinyu Hu,Jingxuan Cang,Chang Sun,Dong Qiu,Yifeng Bian,Yifei Du,Yongchu Pan

Age-related decline in bone healing capacity poses significant challenges for tooth extraction recovery in the elderly. Elevated reactive oxygen–nitrogen species (RONS) and impaired mitophagy drive this pathology. We engineered bimetallic Cu–Fe3O4 nanoclusters with oxygen vacancies (OVs) via hydrothermal synthesis. The abundant oxygen vacancies and bimetallic synergy endow Cu–Fe3O4 with exceptional enzyme-mimicking activity, enabling efficient scavenging of excess RONS and activation of mitophagy in senescent cells in vitro, thereby reversing functional decline. Owing to its excellent biocompatibility and negligible in vivo toxicity, Cu–Fe3O4 accelerates tooth extraction wound healing in aged animal models.

Muscle-Targeted Nanocomposite Therapy Alleviates Age-Related Sarcopenia via Antioxidant and Metabolic Reprogramming

Zhang Liu,Zile Shen,Hengli Lu,Bingqiang Lu,Peng Zhang,Wenhao Chen,Guowei Huang,Xinyu Qu,Zhen Yu,Feng Chen,Wangfu Zang

Sarcopenia, a progressive skeletal muscle disorder marked by loss of mass and function, presents growing societal challenges due to limited therapeutic options. Here, we identify mitochondrial dysfunction and oxidative stress as central drivers of sarcopenia through integrated bioinformatics and clinical validation. To address this pathophysiology, we engineer a muscle-targeted nanocomposite (BP-PEG-MOTS-c, BM) combining mitochondrial-derived peptide MOTS-c with antioxidant black phosphorus nanosheets (BP). BM exhibits dual functionality: MOTS-c restores mitochondrial function, while BP synergistically amplifies ROS scavenging capacity. In cellular and murine models with age-related sarcopenia, BM treatment alleviates muscle dysfunction and muscle loss, concurrently normalizing mitochondrial function and reducing lipid peroxidation. Mechanistic profiling via RNA-seq reveals BM’s activation of PI3K/AKT/Nrf2 and suppression of ROS/p38 MAPK signaling pathway, mediating antioxidant responses and maintenance of mitochondrial homeostasis. The nanocomposite demonstrats superior biocompatibility in toxicity assays, outperforming conventional delivery systems. Our findings establish that BM has been established as a promising mitochondrial redox modulator with translational potential for sarcopenia and related age-associated pathologies.

Engineered Targeted Ce-Based MOF Nanozymes for ROS Scavenging and Inflammatory Reprogramming in Chronic Pancreatitis

Yongkang Lai,Yongliang Ouyang,Xiaojing Yin,Tao Yu,Jianhua Wan,Xueyang Li,Yi Hu,Xu Shu,Huan Wang

Chronic pancreatitis (CP) is a lifelong progressive fibrotic inflammatory disorder for which no effective cure is currently available. Persistent and recurrent inflammatory stimulation induced by reactive oxygen species (ROS) is a key driver of pancreatic fibrogenesis, making oxidative stress a promising therapeutic target to halt disease progression. In this study, we developed a nanosystem, HC@CeMOF, consisting of a small-sized cerium-based metal–organic framework (CeMOF) core loaded with curcumin and coated with hyaluronic acid (HA), enabling precise targeting of inflamed pancreatic tissue. HC@CeMOF exhibits a small-sized particle size along with favorable cellular and biological safety profiles. Once administered in vivo , the nanosystem exploits the specific binding affinity of HA to CD44 receptors on macrophages to selectively accumulate at inflamed pancreatic sites. Subsequently, the cerium-based nanozyme efficiently scavenges ROS through the reversible redox cycling between Ce 3+ and Ce 4+ , while the slow release of curcumin further suppresses the NF-κB signaling pathway and modulates inflammatory cytokine levels, thereby achieving synergistic anti-inflammatory and antioxidant effects. Collectively, these mechanisms substantially attenuate CP progression. This targeted ROS-scavenging and anti-inflammatory strategy holds promise as an alternative therapeutic approach for chronic pancreatitis.

An asymmetric Janus PCL/chitosan bilayer membrane harmonizing mechanical integrity and Osteo-immunomodulatory bioactivity for periodontal regeneration

Lin Huang,Fengxiao He,Tao Wu,Jinghao Fan,Lei Liu,Xiang Zhang,Jiangchen Liu,Shiqian Huang,Decheng Wu,Waruna Lakmal Dissanayaka

This study introduces a new bioactive Janus membrane for guided bone regeneration (GBR) to treat periodontal bone defects. • Unlike conventional barrier membranes that lack long-term stability or biological function, our bilayer membrane, composed of poly(ε-caprolactone) (PCL), chitosan (CS), and the natural polyphenol proanthocyanidin (PA), combines mechanical strength with antioxidant, antibacterial, and anti-inflammatory properties. • Its asymmetric design, with in situ biomineralized hydroxyapatite particles (HAp) on a chitosan layer, supports bone growth on one side while preventing bacterial invasion on the PA-incorporated side. • This innovative approach addresses major challenges in periodontal tissue engineering and offers a promising platform for more effective and biologically intelligent therapies in clinical bone regeneration. The achievement of periodontal bone regeneration using a barrier membrane in guided bone regeneration (GBR) technology is often limited by either its inferior mechanical strength or bioactivity. To address this limitation, we developed a novel Janus bi-layer membrane by sequentially electrospinning poly(ε-caprolactone) (PCL) mixed with proanthocyanidin (PCL-PA) and chitosan (CS) in acetic acid, then biomineralized it in simulated body fluid (BSF) to form hydroxyapatite particles (HAp) in situ on a CS nanofiber layer (CS-HAp): PCL-PA/CS-HAp. This bi-layer membrane exhibited superior mechanical performance compared to single-layer membranes, with a peeling strength of 58 J/m 2 and tensile strength exceeding 5.0 MPa under hydrated conditions. In vitro antioxidant assays demonstrated the strong free-radical-scavenging ability of PCL-PA/CS-HAp. In addition, the PA-incorporated membranes exhibited greater than 95% bacterial inhibition against both Gram-negative E. coli and Gram-positive S. aureus . The bilayer membrane also alleviated oxidative stress induced by hydrogen peroxide (H 2 O 2 ), thereby promoting the proliferation of MC3T3-E1 cells, as monitored by cytoskeletal staining. Flow cytometry analysis showed that the PA-incorporated membrane can stabilize immunomodulatory effects by about 10% in lipopolysaccharides (LPS)-stimulated RAW264.7 cells. When used in a rat periodontal bone defect model, significant bone regeneration was observed in the PCL-PA/CS-HAp group, with a bone volume to total volume ratio of 36.2% and a significant reduction in bone defect size after 8 weeks, outperforming other treatment groups ( p  < 0.001). The PCL-PA/CS-HAp bi-layer membrane represents a promising strategy for guided bone regeneration, effectively combining mechanical stability and bioactive properties. A bi-layer barrier membrane, PCL-PA/CS-HAp, prepared via electrospinning, was effective for periodontal bone regeneration in a rat model. Stage-Adaptive Janus Microneedle System for Redox-Immune Regulation and Mitochondrial Protection in Infected Diabetic Wound Healing

Mengting Yin,Yu Zhang,Xinyu Qu,Jiayi Liu,Zhongyi Sun,Haibo Liu,Ziyan Chen,Jing Ru,Jingwen Han,Bingqiang Lu,Yan Lu,Yan Wang,Xinyu Zhao,Feng Chen

Infected diabetic wounds are sustained by a vicious cycle of hyperglycemia-driven bacterial infection, persistent oxidative stress, and excessive inflammation, which collectively disrupt the ordered progression of tissue repair. Here, we engineered a stage-adaptive Janus microneedle patch (MN-FeSAC-PPE) to enable a staged therapeutic process from early antibacterial intervention to subsequent redox–immune microenvironment remodeling and regenerative tissue repair. This stage-adaptive design integrates Fe single-atom nanozymes (Fe-SACs) into the microneedle base to rapidly kill bacteria using near-infrared light, which activates reactive oxygen species (ROS) production, enabling rapid antibacterial activity against wound pathogens. Meanwhile, propolis extract-loaded (PPE) tips deliver antioxidant bioactive compounds into the wound bed to mitigate oxidative stress, modulate the redox-immune microenvironment, and support the inflammatory-to-regenerative transition. In vitro, MN-FeSAC-PPE enhanced antioxidant defense, suppressed pro-inflammatory factors, and protected fibroblasts from oxidative stress-induced mitochondrial dysfunction. Transcriptomic analysis further supported reduced inflammatory signaling and enhanced metabolism-related programs. In S. aureus -infected diabetic wounds, NIR-activated MN-FeSAC-PPE accelerated wound closure, promoted angiogenesis and collagen remodeling, and alleviated inflammation. These findings establish a stageadaptive redox-immune and bioenergetic regulatory microneedle platform for infected diabetic wound repair.
