3. 完全培养基:血清配制完全培养液后,2~8℃一周内使用完毕;
4. 禁止:室温、37℃长时间放置,易造成生长因子失活、蛋白析出。
标准解冻与使用方法
1. 标准解冻流程
① 从 - 20℃取出血清,置于 2~8℃冷藏 12~24 h 预解冻;
② 剩余少量冰块转移至室温缓慢完全融化;
③ 融化过程每隔 30min 轻柔颠倒混匀,禁止剧烈摇晃;
④ 完全解冻后,可按需分装为小份冷冻,规避反复冻融损伤品质。
2. 常规使用添加比例
① 常规传代细胞培养:培养基添加10%(V/V)血清;
② 原代细胞、克隆培养、复苏细胞:添加15%~20%(V/V);
③ 细胞维持培养:可降至 5%;
④ 更换血清批号前,建议先小量细胞适应性测试,避免细胞状态波动。
注意事项
1. 严禁反复冻融:多次冻融会破坏生长因子,产生大量蛋白絮状沉淀,降低细胞促生长能力;
2. 沉淀说明:解冻后少量细微蛋白沉淀为天然蛋白复合物,不影响使用;若大量褐色沉淀、重度溶血,禁止投入细胞培养;
3. 无菌操作:分装、配制培养基全程在 A 级无菌操作台操作,避免引入外源微生物;使用后剩余血清不可倒回原瓶,防止整瓶污染;
4. 热灭活说明:本品出厂病毒去除工艺完善,常规细胞培养无需 56℃热灭活;若实验特殊需求灭活补体,56℃水浴 30min,每 5min 轻柔混匀一次,灭活后快速冷却;
5. 安全提示:本品为动物源性生物原料,仅用于体外细胞培养,禁止人、动物体内注射/治疗使用;操作时佩戴一次性手套,皮肤、黏膜接触后清水冲洗;
6. 批次管理:不同批号血清蛋白、生长因子含量存在天然差异,规模化生产尽量固定同一批号;更换批号需同步开展细胞适应性验证。

Is it sterile: Yes
This product is Premium‑Grade Newborn Calf Serum. The blood source is collected from healthy newborn Holstein cattle (Bos taurus) within 14 hours after birth, which have not ingested colostrum. Blood collection is performed under fully aseptic closed conditions. Each batch can be fully traced back to ranch, quarantine and blood‑collection records to guarantee clear and reliable source information.
The manufacturing process adopts aseptic closed blood collection, constant‑temperature coagulation, low‑temperature centrifugation, and five‑stage serial terminal sterile filtration with 0.1 μm filters. Aseptic filling is carried out in a Class‑A laminar‑flow environment, followed by immediate quick‑freezing and warehousing, so as to maximize product quality and batch‑to‑batch consistency.
As Premium‑Grade Newborn Calf Serum, this product features low endotoxin levels and exhibits superior proliferation‑promoting performance compared with conventional‑grade newborn calf serum, with performance falling between standard newborn calf serum and fetal bovine serum. It is especially suitable for cell‑culture applications with high serum‑quality requirements, such as primary cells, vaccine cell lines and hybridoma cells.
Scope of Application
1. In vitro culture of various mammalian subcultured cells and delicate primary cells
2. Large‑scale cell culture for veterinary vaccines and human biological products
3. Monoclonal antibody preparation and hybridoma cell culture
4. Preparation of blocking solutions and protein diluents for immunological assays
5. Cost‑effective alternative raw material for fetal bovine serum in mid‑to‑high‑end cell‑culture applications
Storage Conditions and Shelf‑life
1. Long‑term storage: Store sealed in the dark at −20 °C under frozen conditions.
2. Short‑term storage after thawing: Store at 2‑8 °C and use within 1 month.
3. Complete culture medium: After serum is formulated into complete medium, use within one week at 2‑8 °C.
4. Prohibition: Prolonged placement at room temperature or 37 °C shall be avoided, as it may cause growth‑factor inactivation and protein precipitation.
Standard Thawing and Usage Protocols
1. Standard thawing procedure
① Remove serum from -20 °C and pre‑thaw at 2‑8 °C for 12‑24 h.
② Transfer with residual small ice cubes to room temperature for slow full thawing.
③ Gently invert and mix every 30 min during thawing; violent shaking is prohibited.
④ Upon complete thawing, aliquot into small portions for frozen storage as required to prevent quality impairment caused by repeated freeze‑thaw cycles.
2. Routine addition ratios
① Routine subcultured cell culture: Add serum at 10% (V/V) to medium.
② Primary cell culture, cloning culture and cell resuscitation: Add 15%‑20% (V/V).
③ Cell maintenance culture: May be reduced to 5%.
④ Prior to switching serum batches, perform small‑scale cell adaptability testing to prevent cell‑status fluctuations.
Precautions
1. Strictly avoid repeated freeze‑thaw cycles: Multiple freeze‑thaw cycles degrade growth factors, generate substantial flocculent protein precipitates and reduce cell‑growth‑promoting activity.
2. Precipitate explanation: Minor fine protein precipitates after thawing represent natural protein complexes and do not affect usage. Do not use for cell culture in case of massive brown precipitates or severe hemolysis.
3. Aseptic manipulation: Perform aliquoting and medium preparation entirely within an A‑grade biosafety cabinet to avert exogenous microbial contamination. Do not pour leftover serum back into the original bottle after use to prevent whole‑bottle contamination.
4. Heat‑inactivation note: This product undergoes comprehensive viral‑removal processing at manufacture; heat‑inactivation at 56 °C is unnecessary for general cell culture. If complement inactivation is required for specific experiments, perform water‑bath incubation at 56 °C for 30 min with gentle mixing every 5 min, then cool rapidly after inactivation.
5. Safety reminder: This product is an animal‑derived biological raw material intended solely for in‑vitro cell culture. Injection or therapeutic administration in humans or animals is forbidden. Wear disposable gloves during handling; rinse skin and mucous membranes thoroughly with clean water upon contact.
6. Batch management: Natural variations in protein and growth‑factor contents exist among different serum batches. For large‑scale production, use a fixed batch whenever possible. Perform parallel cell‑adaptability validation upon batch change.
